Characteristic dysbiosis in gout and the impact of a uric acid-lowering treatment,febuxostat on the gut microbiota

Gut dysbiosis is suggested to play a critical role in the pathogenesis of *** aim of our study was to identify the characteristic dysbiosis of the gut microbiota in gout patients and the impact of a commonly used uric acid-lowering treatment,febuxostat on gut microbiota in gout.16S ribosomal RNA sequencing and metagenomic shotgun sequencing was performed on fecal DNA isolated from 38 untreated gout patients,38 gout patients treated with febuxostat,and 26 healthy controls.A restriction of gut microbiota biodiversity was detected in the untreated gout patients,and the alteration was partly restored by *** metabolic indexes involved in liver and kidney metabolism were significantly associated with the gut microbiota composition in gout *** analysis revealed that the gut microbiome of gout patients had an enriched function on carbohydrate metabolism but a lower potential for purine metabolism,which was comparatively enhanced in the febuxostat treated gout patients.A classification microbial model obtained a high mean area under the curve up to ***,gut dysbiosis characterizings gout could potentially serve as a noninvasive diagnostic tool for gout and may be a promising target of future preventive interventions.

In order to study the migration and transformation mechanism of Hg content and occurrence form in subsurface flow zone of gold mining area in Loess Plateau and its influence on water environment,the field in-situ infiltration test and laboratory test were carried out in three typical sections of river-side loess,alluvial and proluvial strata in Tongguan gold mining area of Shaanxi Province,and the following results were obtained:(1)The source of Hg in subsurface flow zone is mainly caused by mineral processing activities;(2)the subsurface flow zone in the study area is in alkaline environment,and the residual state,iron and manganese oxidation state,strong organic state and humic acid state of mercury in loess are equally divided in dry and oxidizing environment;mercury in river alluvial or diluvial strata is mainly concentrated in silt,tailings and clayey silt soil layer,and mercury has certain stability,and the form of mercury in loess is easier to transform than the other two media;(3)under the flooding condition,most of mercury is trapped in the silt layer in the undercurrent zone where the sand and silt layers alternate with each other and the river water and groundwater are disjointed,and the migration capacity of mercury is far less than that of loess layer and alluvial layer with close hydraulic connection;(4)infiltration at the flood level accelerates the migration of pollutants to the ground;(5)the soil in the undercurrent zone is overloaded and has seriously exceeded the *** the groundwater monitoring results are safe this time,relevant enterprises or departments should continue to pay attention to improving the gold extraction process,especially vigorously rectify the small workshops for illegal gold extraction and the substandard discharge of the three wastes,and intensify efforts to solve the geological environmental problems of mines left over from *** present,the occurrence form of mercury in the undercurrent zone is relatively s

Pathological diagnosis is maybe non-essential for special gastric cancer: Case reports and review

Histopathological results are critical for the diagnosis and surgical decision regarding gastric cancer. How-ever, opposite opinions from radiology and pathology can sometimes affect clinical decisions. The two cases reported in this article were both highly suspected as gastric cancer by clinical manifestations and radiologic findings, although both showed negative results in the first biopsy examination. One was confirmed as gastric cancer by the time of the 6 th biopsy, while the other was still negative even after 8 biopsies. With a definite pathologic result and the agreement of the patient for the latter case, both of them finally received surgery. Postoperative pathological examination revealed find-ings that were the same as Borrmann type Ⅳ gastric cancer. We believed that duplicate biopsies under ra-diologic guidance were necessary for highly suspected gastric cancer cases in the absence of a definite pathol-ogy result, and patients should be under close follow-up. We propose that, if gastric cancer is highly sus-pected when typical radiology changes of widely diffuse gastric parietal lesions suffice to exclude lymphoma and other similar situations, and even in absence of a posi-tive biopsy result, a diagnostic laparotomy under lapa-roscopy and even radical gastrectomy may be reason-ably performed by an experienced gastric cancer center with the agreement of the patient after being decided by a multidisciplinary discussion team.

目的对单克隆抗体制品中残余SP2/0细胞蛋白含量ELISA检测试剂盒进行验证。方法对Cygnus Technologies公司制备的SP2/0Host Cell Protein(sHCPs)ELISA试剂盒进行灵敏度、精密性验证及基质干扰试验和样品稀释回收试验,并采用该试剂盒对本公司制备的10批单抗制品原液及冻干品进行检测。结果标准蛋白含量的对数与相应的光吸收值的对数,在2～200ng/ml范围内呈线性关系;该试剂盒检测限可达0.6ng/ml,定量限可达2ng/ml,灵敏度符合要求;检测低、中、高3个浓度的SP2/0HCPs样品试验内及试验间变异系数分别为15.4%、3.0%、2.1%及12.0%、3.3%、1.5%;样品基质对检测无明显干扰;单抗样品检测不存在Hook效应;检测本公司10批单抗制品原液和冻干品中SP2/0细胞蛋白残留率均小于0.01%。结论该试剂盒灵敏度高,精密性好,可用于单抗制品中残余SP2/0细胞蛋白含量的测定。

Baddeleyite and zircon U-Pb ages of the ultramafic rocks in Chigu Tso area,Southeastern Tibet and their constraints on the timing of Comei Large Igneous Province

A suite of ultramafic and mafic rocks developed in the Chigu Tso area,eastern Tethyan *** and zircon U-Pb ages acquired by SIMS and LA-ICP-MS from olivine pyroxenite rocks in the Chigu Tso area are 138.9±3.0 Ma and 139.0±1.9 Ma,*** two Early Cretaceous ages are similar with the ages of the more abundant mafic rocks in the eastern Tethyan Himalaya,indicating that this suite of ultramafic and mafic rocks in the Chigu Tso area should be included in the outcrop area of the Comei Large Igneous Province(LIP).These ultramafic rocks provide significant evidence that the involvement of mantle plume/hot spot activities in the formation of the Comei *** U-Pb dating by SIMS is one reliable and convenient method to constrain the formation time of ultramafic *** dating results of baddeleyite and zircon from the olivine pyroxenite samples in this paper are consistent with each other within analytical uncertainties,suggesting that baddeleyite and zircon were both formed during the same magmatic *** consistency of baddeleyite U-Pb ages in the Chigu Tso area with zircon U-Pb ages for a large number of Early Cretaceous mafic rocks in the eastern Tethyan Himalaya further support that zircon grains from such mafic rocks yielding Early Cretaceous ages are also magmatic in origin.

Characteristics of the TCR Vβ repertoire in imatinib-resistant chronic myeloid leukemia patients with ABL mutations

Diversity in the T cell receptor(TCR) repertoire provides a miniature defense ability for the T cell immune system that may be related to tumor initiation and progression. Understanding the T cell immune status of leukemia patients is critical for establishing specific immunotherapies. Previous studies have reported abnormal TCR repertoires and clonally expanded TCR V? T cells in chronic myeloid leukemia in chronic phase(CP-CML). In this study, we investigated the distribution and clonality of the TCR V? repertoire in 4 cases with imatinib-resistant CML in blast crisis(BC-CML) with abelson murine leukemia viral oncogene homolog 1(ABL1) kinase domain mutations(KDMs). Examination of TCR V? expression and clonality was performed by reverse transcription-polymerase chain reaction(RT-PCR) and Gene Scan analysis. Significantly skewed TCR V? repertoires were observed in BC-CML patients with different KDMs, and 4 to 8 oligoclonally expanded TCR V? subfamilies could be identified in each sample. Intriguingly, a relatively highly expanded V?9 clone with the same length as complementarity-determining region 3(CDR3)(139 bp) was found in all three CML patients in lymphoid blast crisis(LBC-CML) who had different KDMs, but the clone was not detected in the only CML patient in myeloid blast crisis(MBC-CML). In conclusion, restricted TCR V? repertoire expression and decreased clone complexity was a general phenomenon observed in the BC-CML patients with different KDMs, indicating the T-cell immunodeficiency of these patients. In addition, clonally expanded V?9 T cell clones may indicate a specific immune response to leukemia-associated antigens in LBC-CML patients.

Visceral hypersensitive rats share common dysbiosis features with irritable bowel syndrome patients

AIM: To evaluate gut microbial dysbiosis in two visceral hypersensitive models in comparison with irritable bowel syndrome(IBS) patients and to explore the extent to which these models capture the dysbiosis of IBS patients. METHODS: Visceral hypersensitivity was developed using the maternal separation(MS) rat model and post-inflammatory rat model. The visceral sensitivity of the model groups and control group was evaluated using the abdominal withdraw reflex score and electromyography in response to graded colorectal distention. The 16 S ribosomal RNA gene from fecal samples was pyrosequenced and analyzed. The correlation between dysbiosis in the microbiota and visceral hypersensitivity was calculated. Positive findings were compared to sequencing data from a published human IBS ***: Dysbiosis triggered by neonatal maternal separation was lasting but not static. Both MS and postinflammatory rat fecal microbiota deviated from that of the control rats to an extent that was larger than the co-housing effect. Two short chain fatty acid producing genera, Fusobacterium and Clostridium XI, were shared by the human IBS cohort and by the maternal separation rats and post-inflammatory rats, respectively, to different extents. Fusobacterium was significantly increased in the MS group, and its abundance positively correlated with the degree of visceral hypersensitivity. Porphyromonadaceae was a protective biomarker for both the rat control group and healthy human controls. CONCLUSION: The dysbiosis MS rat model and the post-inflammatory rat model captured some of the dysbiosis features of IBS patients. Fusobacterium, Clostridium XI and Porphyromonadaceae were identified as targets for future mechanistic research.

Multi-stage dual replica bit-line delay technique for process-variation-robust timing of low voltage SRAM sense amplifier

A multi-stage dual replica bit-line delay （MDRBD） technique is proposed for reducing access time by suppressing the sense-amplifier enable （SAE） timing variation of low voltage static randomaccess memory （SRAM） applications. Compared with the traditional technique, this strategy, using statistical theory, reduces the timing variation by using multi-stage ideas, meanwhile doubling the replica bit-fine （RBL） capacitance and discharge path simultaneously in each stage. At a supply voltage of 0.6 V, the simulation results show that the standard deviations of the SAE timing and cycle time with the proposed technique are 69.2% and 47.2%, respectively, smaller than that with a conventional RBL delay technique in TSMC 65 nm CMOS technology （Taiwan Semiconductor Manufacturing Company, Taiwan）.
