Ovarian endometrioma（OE）, also known as “chocolate cysts,” is a cystic mass that develops in the ovaries due to endometriosis and is a common gynecological condition characterized by the growth of endometrial tissue outside the uterus, leading to symptoms such as dysmenorrhea, pelvic pain, and infertility. However, the precise molecular and cellular mechanisms driving this pathophysiology remain largely unknown, posing challenges for diagnosis and treatment. Here, we employed integrated single-cell transcriptomic profiling of over 52,000 individual cells from endometrial tissues of OE patients and healthy donors and identified twelve major cell populations. We identified notable alterations in cell type-specific proportions and molecular signatures associated with OE. Notably, the activation of IGFBP5+macrophages with pro-inflammatory properties, NK cell exhaustion, and aberrant proliferation of IQCG+and KLF2+epithelium are key features and may be the potential mechanisms underlying the pathogenesis of OE. Collectively, our data contribute to a better understanding of OE at the single cell level and may pave the way for the development of novel therapeutic strategies.

AIM: The human cytochrome P-450 2C18(CYP2C18) hasbeen characterized. However, the protein has not beenpurified from liver and very little is known regarding thespecific substrate of CYP2C18. In order to study its enzymaticactivity for drug metabolism, the CYP2C18cDNA was clonedand a stable CHL cell line expressing recombinant CYP 2C18was ***: The human CYP2C18cDNA was amplified withreverse transcription-polymerase chain reaction (RT-PCR)from total RNAs extracted from human liver and cloned intopgeM-T vector. The cDNA segment was identified by DNAsequencing and subcloned into a mammalian expressionvector pREP9. A transgenic cell line was established bytransfecting the recombinant plasmid of pREPg-CYP2C18toChinese hamster lung (CHL) cell. The enzyme activity ofCYP2C18 catalyzing oxidation of tolbutamide tohydroxytolbutamide in postmitochondrial supernant(Sg)fraction of the cell was determined by high performanceliquid chromatography(HPLC).RESULTS: The amino acid sequence predicted from thecloned cDNA segment was identical to that of reported byRomkes et al(genBank accession number: M61856,J05326).The S9 fraction of the established cell line metabolizestolbutamide to hydroxytolbutamide. Tolbutamide hydroxylaseactivity was found to be 0.509±0.052 μ***-1.g-1 S9protein or 8.82±0.90 ***-1 CYP, but wasundetectable in parental CHL cell. In addition, we haveidentified a CYP2C18cDNA clone with exon 5 ***: The cDNA of human CYP2C18 wassuccessfully cloned and a cell line, CHL-CYP2C18, efficientlyexpressing the protein of CYP2C18, was established. Aspliced variant of CYP2C18 with exon 5 missing was identifiedin the cloning process.

Reverse modulation of defect density of Ce-MOF-808 by solventassisted ligand exchange for dicyclopentadiene hydrogenation

Defect engineering is a pivotal strategy for tailoring the electronic structure and physicochemical properties of catalysts, yet the straightforward manipulation of defect density in porous materials presents a current challenge. In this work, we achieved precise control over the coordination unsaturated site（CUS） content and pore architecture of Ce-MOF-808 by employing 2-fluorobenzoic acid（2-FBA） as a modifier. Leveraging the p Kadisparities among three ligands（1,3,5-tricarboxylic benzoic acid,formic acid, and 2-FBA）, we preserved the spn topology of Ce-MOF within an aqueous system, while realizing partial substitution of the auxiliary ligands and saturation of the CUS. The amount of 2-FBA was negatively correlated with the number of ligand defects, and the consequent decrease in Ce3+content and the increase in the number of ligand linkages also induced crystal structure transformation. The modulation of the defect structure was correlated with the catalytic performance of dicyclopentadiene（DCPD） hydrogenation reaction, in which Ce-MMF0.15, constructed by the modulation of 0.15 mol amount of 2-fluorobenzoic acid, exhibited an optimal ligand defect density and micro-mesoporous structure, enabling almost complete conversion of DCPD at 100 ℃ and 2 MPa. This article also elucidates the correlation between the regulation of defect density and the accessibility of frustrated Lewis pair（FLP） active sites, guiding the application of novel porous intrinsic FLP materials in hydrogenation catalysis.

The natural element method （NEM） is a newly- developed numerical method based on Voronoi diagram and Delaunay triangulation of scattered points, which adopts natural neighbour interpolation to construct trial functions in the framework of Galerkin method. Owing to its distinctive advantages, the NEM is used widely in many problems of computational mechanics. Utilizing the NEM, this paper deals with numerical limit analysis of structures made up of perfectly rigid-plastic material. According to kinematic the- orem of plastic limit analysis, a mathematical programming natural element formulation is established for determining the upper bound multiplier of plane problems, and a direct iteration algorithm is proposed accordingly to solve it. In this algorithm, the plastic incompressibility condition is handled by two different treatments, and the nonlinearity and nons- moothness of the goal function are overcome by distinguishing the rigid zones from the plastic zones at each iteration. The procedure implementation of iterative process is quite simple and effective because each iteration is equivalent to solving an associated elastic problem. The obtained limit load multiplier is proved to monotonically converge to the upper bound of true solution. Several benchmark examples are investigated to validate the significant performance of the NEM in the application field of limit analysis.

The dynamics of polymer chains in layered silicate/polystyrene nanocomposites was studied by anelastic spectroscopy. Two thermal activated peaks （or and α＇ peaks） appeared when the specimens were heated to a high temperature and they were related to glass transition and liquid-liquid transition, respectively. The activation energy was calculated based on Arrhenius equation and it showed that the activation energy of glass transition （Eg） is much higher than that of liquid-liquid transition （EH）. Furthermore, the most interesting result for the activation energy was that there were two contrary trends for Eg and Ell, Eg decreased and Ell increased with the addition of clay platelets. The fragile parameter was analyzed and the variation of fragile parameters for the two transitions was also contrary to each other with the addition of clay platelets. All the results indicated that the confinement effect of clay platelets on the dynamics of polymer chain was scale dependent, and perhaps, the two transitions were produced by different mechanisms.
