PHR1(PHOSPHATE STARVATION RESPONSE1)plays key roles in the inorganic phosphate(Pi)starvation response and in Pi deficiency-induced anthocyanin biosynthesis in plants. However, the post-translational regulation of PHR1 is unclear,and the molecular basis of PHR1-mediated anthocyanin biosynthesis remains elusive. In this study, we determined that MdPHR1 was essential for Pi deficiency-induced anthocyanin accumulation in apple(Malus × domestica). MdPHR1 interacted with MdWRKY75, a positive regulator of anthocyanin biosynthesis, to enhance the MdWRKY75-activated transcription of MdMYB1,leading to anthocyanin accumulation. In addition,the E3 ubiquitin ligase SEVEN IN ABSENTIA1(MdSINA1) negatively regulated MdPHR1-promoted anthocyanin biosynthesis via the ubiquitination-mediated degradation of ***, the protein kinase apple BRASSINOSTEROID INSENSITIVE2(MdBIN2) phosphorylated MdPHR1 and positively regulated MdPHR1-mediated anthocyanin accumulation by attenuating the MdSINA1-mediated ubiquitination degradation of MdPHR1. Taken together,these findings not only demonstrate the regulatory role of MdPHR1 in Pi starvation induced anthocyanin accumulation, but also provide an insight into the post-translational regulation of PHR1.

Monogenic disorders refer to a group of human diseases caused by mutations in single genes. While disease-modifying therapies have offered some relief from symptoms and delayed progression for some monogenic diseases, most of these diseases still lack effective treatments. In recent decades, gene therapy has emerged as a promising therapeutic strategy for genetic disorders. Researchers have developed various gene manipulation tools and gene delivery systems to treat monogenic diseases. Despite this progress, concerns about inefficient delivery, persistent expression, immunogenicity, toxicity, capacity limitation, genomic integration, and limited tissue specificity still need to be addressed. This review gives an overview of commonly used gene therapy and delivery tools, along with the challenges they face and potential strategies to counter them.

Purpose:Minimal data exist on brain injury in patients with exertional heatstroke(EHS)in developing *** this study,we explored the risk factors for brain injury induced by EHS 90-day after ***:A retrospective cohort study of patients with EHS was conducted in the intensive care unit of the General Hospital of Southern Theater Command of PLA in China from April 2014 to June *** were divided into non-brain injury(fully recovered)and brain injury groups(comprising deceased patients or those with neurological sequelae).The brain injury group was further subdivided into a death group and a sequela group for detailed *** information,neurological performance and information on important organ injuries in the acute stage were recorded and *** logistic regression was used to identify risk factors for brain injury after EHS and mortality risk factors for brain injury,and Kaplan-Meier survival curve was used to evaluate the effect of the neurological dysfunction on ***:Out of the 147 EHS patients,117 were enrolled,of which 96(82.1%)recovered,13(11.1%)died,and 8(6.8%)experienced neurological *** significant differences were found between non-brain injury and brain injury groups in age,hypotension,duration of consciousness disorders,time to drop core body temperature below 38.5℃,lymphocyte counts,platelet counts,procalcitonin,alanine aminotransferase,aspartate aminotransferase,creatinine,cystatin C,coagulation parameters,international normalized ratio,acute physiology and chronic health evaluation II scores,sequential organ failure assessment(SOFA)scores,and Glasgow coma scale scores(all p<0.05).Multivariate logistic regression showed that age(OR=1.090,95%CI:1.02-1.17,p=0.008),time to drop core temperature(OR=8.223,95%CI:2.30-29.40,p=0.001),and SOFA scores(OR=1.676,95%CI:1.29-2.18,p<0.001)are independent risk factors for brain injury induced by *** Kaplan-Meier curves suggest significa

At present, the main controlling factors of helium accumulation is one of the key scientific problems restricting the exploration and development of helium reservoir. In this paper, based on the calculation results of He generation rate and the geochemical characteristics of the produced gas, both the similarities and differences between natural gas and He resources in the Bohai Bay, Ordos and the surrounding Songliao Basin are compared and analyzed, discussing the main controlling factors of helium resources in the three main petroliferous basins of the North China Craton. It is found that the three basins of Bohai Bay, Ordos and Songliao have similar characteristics of source rocks, reservoirs and cap rocks, that's why their methane resource characteristics are essentially the same. The calculated ~4He generation per cubic metamorphic crystalline basement in the three basins is roughly equivalent, which is consistent with the measured He resources, and it is believed that the ~4He of radiogenic from the crust is the main factor controlling the overall He accumulation in the three basins;there is almost no contribution of the mantle-derived CH_4, which suggests that the transport and uplift of mantle-derived ~3He carried by the present-day magmatic activities along the deep-large faults is not the main reason for the mantle-derived ~3He mixing in the basins. Combined with the results of regional volcanic and geophysical studies,it is concluded that under the background of the destruction of North China Craton, magma intrusion carried a large amount of mantle-derived material and formed basic volcanic rocks in the Bohai Bay Basin and Songliao Basin, which replenished mantle-derived ~3He for the interior of the basins, and that strong seismic activities in and around the basins also promoted the upward migration of mantle source ~3He. Neighborhood predictors of short sleep duration and bedtime irregularity among children in the United States: results from the 2019-2020 National Surveyof Children's Health

Background Environmental factors may contribute to short sleep duration and irregular bedtime in *** factors and children's sleep duration and bedtime regularity remain a less investigated *** aim of this study was to investigate the national and state-level proportions of children with short sleep duration and irregular bedtime and their neighborhood *** A total of 67,598 children whose parents completed the National Survey of Children's Health in 2019-2020 were included in the ***-weighted Poisson regression was used to explore the neighborhood predictors of children's short sleep duration and irregular *** The prevalence of short sleep duration and irregular bedtime among children in the United States(US)was 34.6%[95%confidence interval(CI)=33.8%-35.4%]and 16.4%(95%CI=15.6%-17.2%)in 2019-2020,*** neighborhoods,supportive neighborhoods,and neighborhoods with amenities were found to be protective factors against children's short sleep duration,with risk ratios ranging between 0.92 and 0.94,P<*** with detracting elements were associated with an increased risk of short sleep duration[risk ratio(RR)=1.06,95%CI=1.00-1.12]and irregular bedtime(RR=1.15,95%CI=1.03-1.28).Child race/ethnicity moderated the relationship between neighborhood with amenities and short *** Insufficient sleep duration and irregular bedtime were highly prevalent among US children.A favorable neighborhood environment can decrease children's risk of short sleep duration and irregular *** the neighborhood environment has implications for children's sleep health,especially for children from minority racial/ethnic groups.
