Functional metabolomics analysis of the protective mechanism of total flavonoids of Scutellaria baicalensis on acute myocardial ischemia rats

Background:The study aimed to investigate the protective effect and mechanism of total flavonoids of Scutellaria baicalensis(TFSB)on acute myocardial ischemia(AMI)rats by using functional ***:Rats were divided into the Control,Model,AMI positive control(Propranolol hydrochloride,30 mg/kg),low dose TFSB(50 mg/kg),and high dose TFSB(100 mg/kg)*** received the corresponding treatment by intragastric administration once daily for 10 consecutive ***,myocardial enzyme,triphenyltetrazolium chloride staining,hematoxylin-eosin,and enzyme-linked immunosorbent assay were performed to evaluate the protective effect of TFSB on AMI ***,the UHPLC-Q-Orbitrap MS method based on serum metabolomics was utilised to search for metabolic biomarkers and metabolic ***,Western blot and RT-PCR techniques were employed to identify the respective genes and ***:Pharmacodynamics revealed that TFSB could ameliorate AMI in *** results of the metabolomics analysis indicated that the alterations in metabolic profile observed in rats with AMI were partially improved by treatment with ***,the mRNA expression levels of 5-lipoxygenase(5-LOX)and 15-lipoxygenase(15-LOX)and the protein expression levels of 5-LOX,15-LOX,interleukin-1β(IL-1β),and NF-κB p65 were reduced following treatment with ***:The potential treatment of TFSB in AMI may be ascribed to its ability to regulate arachidonic acid metabolism.

Cereal-legume intercropping stimulates straw decomposition and promotes soil organic carbon stability

Increasing carbon（C） sequestration and stability in agricultural soils is a key strategy to mitigate climate change towards C neutrality. Crop diversification is an initiative to increase C sequestration in fields, but it is unclear how legume-based crop diversification impacts the functional components of soil organic carbon（SOC） in dryland, including the formation and transformation of particulate organic carbon（POC） and mineral-associated organic carbon（MAOC）. We investigated the decomposition of straw residues, the fate of photosynthesized C,as well as the formation of MAOC and POC fractions using an in situ13C labeling technique in the soybean-wheat intercropping, soybeanmaize intercropping and their respective monocropping systems, with and without cover crops. After 4-year treatments, the total SOC content in bulk soil remained unchanged, while MAOC content increased significantly by 5.6% with intercropping. Moreover, the in situ13C labeling results confirmed that more photosynthesized C was transferred to MAOC, and less was retained in the POC fraction. Intercropping significantly increased total soil N and mineral N content by 15.3% and 13.4%, respectively, and decreased soil and microbial C/N ratio by11.3% and 17.4%, respectively. This outcome, therefore, relieved microbial N limitation and accelerated straw residue ***, the potential of MAOC formation was strengthened for better SOC persistence. Our study suggests that legume-based crop diversification can effectively enrich N and support POC transformation to MAOC, accordingly contributing to the persistent SOC pool and thus potentially achieving C neutrality under climate change in dryland agroecosystems.

Background:Insomnia is a prevalent clinical condition and Shangxia Liangji formula(SXLJF)is a well-established method of ***,the specific mechanism of action of SXLJF remains ***:The mouse model of insomnia was established by intraperitoneal injection of ***-two mice were randomly divided into a negative control group,model group,SXLJF group(18.72 g/kg/day),and positive control group(diazepam,2 mg/kg)and treated with the corresponding drugs for 7 consecutive *** open field test and pentobarbital-induced sleeping test were ***-MS-based untargeted metabolomics and network pharmacology were applied to explore the potential targets of SXLJF for treating ***,key targets were validated using ***:Behavioral tests demonstrated that SXLJF reduced the total distance,average velocity,central distance,and sleep latency,and prolonged sleep *** and network pharmacology revealed potential targets,signaling pathways,metabolic pathways,and metabolites associated with the anti-insomnia effects of ***,tyrosine hydroxylase(TH)and tyrosine metabolism emerged as crucial metabolic pathways and targets,***-qPCR results supported the role of TH in the mechanism of SXLJF in treating ***:In conclusion,TH and tyrosine metabolism may represent significant targets and pathways for SXLJF in treating insomnia.

In the era of time-domain,multi-messenger astronomy,the detection of transient events on the high-energy electromagnetic sky has become more important than *** gravitational wave high-energy electromagnetic counterpart all-sky monitor(GECAM)is a dedicated mission to monitor gamma-ray transients,launched in December,2020.A real-time on-board trigger and location software,using the traditional signal-to-noise ratio(SNR)method for blind search,is constrained to relatively bright signals due to the limitations in on-board computing resources and the need for real-time *** this work,we developed a ground-based pipeline for GECAM to search for various transients,especially for weak bursts missed by on-board *** pipeline includes both automatic and manual mode,offering options for blind search and targeted *** targeted search is specifically designed to search for interesting weak bursts,such as gravitational wave-associated gamma-ray bursts(GRBs).From the ground search of the data in the first year,GECAM has been triggered by 54 GRBs and other transients,including soft gamma-ray repeaters,X-ray binaries,solar flares,terrestrial gamma-ray *** report the properties of each type of triggers,such as trigger time and light *** this search pipeline and assuming a soft Band spectrum,the GRB detection sensitivity of GECAM is increased to about 1.1×10^(-8)erg cm^(-2)s^(-1)(10-1000 ke V,burst duration of 20 s).These results demonstrate that the GECAM ground search system(both blind search and targeted search)is a versatile pipeline to recover true astrophysical signals which were too weak to be found in the on-board search.

To the Editor:Irreversible electroporation(IRE)is a promising ablative technique for hepatobiliary *** has unique advantages,including no detrimental heat sink effect[1]and no injury of the surrounding tissues[2].For lesions in complex areas,bile duct[3],and large blood vessels[4],IRE has been developed as an alternative to thermal ablation such as radiofrequency and *** IRE tumor ablative technique combines cell biology and pulsed high-voltage *** high-voltage,high-frequency and transient pulses can create irreversible electroporation in the phospholipid bilayer of the cell membrane and nuclear envelope,resulting in cell *** with radiofrequency ablation and microwave ablation[5],IRE does not cause damage to important vessels and *** has some unique shortcomings in delivering high currents,which has limited its clinical applications in the *** animal and clinical studies,side effects including muscle contraction,procedural hypertension,and arrhythmia are frequently reported[6].However,during the ablation process,if the pulse is synchronized with the heart rhythm,the risk of arrhythmia is *** muscle relaxant eliminates muscle ***,arrhythmia[7],hypertension[8],and myocardial infarction[9]are potential risks requiring proper *** adverse events are mostly moderate and self-limiting,but they may have serious consequences for patients with coronary artery *** to the potential risks,patients with a history of cardiovascular disease are not recommended for IRE treatment.

目的:探讨人胚胎滋养层细胞中染色体数目异常与着丝粒特异性蛋白质CENP-I表达水平的相关性。方法:T-A克隆制备CENP-I、GAPDH定量PCR标准品;用Ni2+-NTA亲和层析柱纯化CENP-I重组蛋白,免疫兔制备抗人CENP-I多克隆抗体;采用FQ-PCR和Western-blot检测23例染色体数目异常的滋养层细胞(实验组)和30例具有正常核型的滋养层细胞(对照组)中CENP-ImRNA和蛋白表达水平。结果:成功制备CENP-I、GAPDHFQ-PCR标准品,表达和纯化CENP-I重组蛋白,获得兔抗人CENP-I多克隆抗体;FQ-PCR和Western-blot结果显示CENP-I在实验组和对照组中均有表达,但实验组中CENP-I的表达水平明显降低(P<0.05)。结论:CENP-I表达的降低可能是导致人胚胎滋养层细胞中染色体数目异常的原因之一。

Impact of Alpine Wetland Succession to Soil Bacterial Diversity

This study aims to explore the characteristics of change and the rules of response of the soil bacterial community structure and diversity during the evolution of alpine wetlands. It also seeks to identify the leading factors affecting changes in the soil bacterial community，with the goal of providing a scientific basis for the restoration and management of alpine wetland ecosystems. Three different types of wetlands （original alpine wetland，alpine wet meadow，and alpine meadow） were selected to represent the succession process of alpine wetlands. The composition and diversity of the soil bacterial communities were analyzed using high-throughput sequencing technology. The findings revealed the following：① Proteobacteria and Acidobacteria were the predominant bacteria，with their relative abundance exceeding 20.08%. In the alpine wetland，Nitrosomonadaceae and RB41 were the dominant bacteria. Meanwhile，Chthoniobacteraceae and Candidatus_Udaeobacter were the dominant genera in the alpine wet meadow and alpine meadow，respectively. ② Although no significant change was observed in community diversity，significant differences were present in the structure of the soil bacterial community at different succession stages （P © 2025 Science Press.
