Effects of Mycobacterium vaccae vaccine in a mouse model of tuberculosis: protective action and differentially expressed genes

Background:Tuberculosis is a leading cause of death *** is an effective vaccine,but not widely used in many parts of the world due to a variety of *** vaccae(***)is another vaccine used in human subjects to prevent *** the current study,we investigated the potential mechanisms of *** vaccination by determining differentially expressed genes in mice infected with *** before and after *** ***:Three days after exposure to *** H37 Rv strain(5×10~5 CFU),adult BALB/c mice randomly received either *** vaccine(22.5μg)or vehicle via intramuscular injection(n=8).Booster immunization was conducted 14 and 28 days after the primary *** expressed genes were identified by microarray followed by standard bioinformatics ***:*** vaccination provided protection against *** infection(most prominent in the lungs).We identified 2,326 upregulated and 2,221 downregulated genes in vaccinated *** changes could be mapped to a total of 123 signaling pathways(68 upregulated and 55 downregulated).Further analysis pinpointed to the MyD88-dependent TLR signaling pathway and PI3 K-Akt signaling pathway as most likely to be ***:*** vaccine provided good protection in mice against *** infection,via a highly complex set of molecular *** findings may provide clue to guide development of more effective vaccine against tuberculosis.

Sirtuin 1(SIRT1)is a protein deacetylase,which regulates various physiological activities by deacetylating different protein *** increasing number of studies have revealed critical roles of SIRT1 in different aspects of cancers including metabolism,proliferation,genomic instability,and chemotherapy *** on the protein targets in a certain oncogenic context,SIRT1 may play a unique role in each individual blood cancer *** previous work showed that activation of SIRT1 in primitive leukemia cells of acute myeloid leukemia(AML)and chronic myelogenous leukemia(CML)promotes disease *** the other hand,an SIRT1 agonist was shown to disrupt maintenance of myelodysplastic syndrome(MDS)stem cells and holds promise as a potential therapeutic ***,we present a concise summary of the different functions of SIRT1 in hematologic malignancies.

Synergy of first-and second-sphere interactions in a covalent organic framework boosts highly selective platinum uptake

Platinum recovery from waste electrical and electronic equipment(WEEE) in highly acidic solutions is significant to the electronics industry and environmental remediation. However, the lack of ingenious design and synergetic coordination gives rise to unsatisfied PtCl_(4)^(2-)extraction capacities and selectivities in most previously reported adsorbents(e.g., polymeric and inorganic materials). Herein, we proposed a synergistic strategy that realizes highly selective PtCl_(4)^(2-)uptake through first-and second-sphere coordinations. The proof-of-concept imine-linked covalent organic framework(SCU-COF-3) was found to chelate Pt Cl42-via the direct N…Pt coordination and the synergistically interlaminar N–H…Cl hydrogen bonds, which was disclosed by the comprehensive analysis of extended X-ray adsorption fine structure(EXAFS) characterizations and density functional theory(DFT) calculations. The unique adsorption mechanism imparts a superior adsorption capacity(168.4 mg g-1)and extraordinary Pt(II) selectivity to SCU-COF-3 under static conditions. In addition, SCU-COF-3 exhibits an upgraded distribution coefficient of 1.62 × 10^(5)mL· g^(-1), one order of magnitude higher than those of reported natural adsorbents. Specifically, SCU-COF-3 can extract PtCl_(4)^(2- )quantitatively from a simulated acidic waste solution coexisting with other 12 competitive ions, suggesting its promising application in practical scenarios.

Application of defin让ive screening design to quantify the effects of process parameters on key granule characteristics and optimize operating parameters in pulsed-spray fluid-bed granulation

The pulsed-spray fluid-bed granulation (PSFBG) process was investigated and optimized using definitive scree ning design, a recently proposed new class of three-level desig n of experiment method. Such a design enabled quadratic models to be established that described the effect of six in put process parameters - inlet air temperature, inlet air humidity, binder spray rate, atomization pressure, pulse period, and pulse width - on the granule quality in a PSFBG process. Mathematical models of the mean particle size, relative size distribution width, production yield, and porosity were developed to quantify the relationships between the in flue ncing factors and critical quality attributes. On the basis of con strai nts on the desired granule properties, a design space for PSFBG was determined and ranges of the operating parameters were defined. An acceptable degree of prediction was confirmed by validation experiments, demonstrating the reliability and effectiveness of using definitive screening design to study the PSFBG process. This method can accelerate screening and optimization of this process within a large multidimensional design space.

Defect of SLC38A3 promotes epithelial-mesenchymal transition and predicts poor prognosis in esophageal squamous cell carcinoma

Objective: Solute carrier family 38(SLC38 s) transporters play important roles in amino acid transportation and signaling transduction. However, their genetic alterations and biological roles in tumors are still largely *** study aimed to elucidate the genetic signatures of SLC38 s transporters and their implications in esophageal squamous cell carcinoma(ESCC).Methods: Analyses on somatic mutation and copy number alterations(CNAs) of SLC38 A3 were performed as described. Immunohistochemistry(IHC) assay and Western blot assay were used to detect the protein expression level. MTS assay, colony formation assay, transwell assay and wound healing assay were used to explore the malignant phenotypes of ESCC cells. Immunofluorescence assay was used to verify the colocalization of two indicated proteins and immunopreciptation assay was performed to confirm the interaction of ***: Our findings revealed that SLC38 s family was significantly disrupted in ESCC, with high frequent CNAs and few somatic mutations. SLC38 A3 was the most frequent loss gene among them and was linked to poor survival and lymph node metastasis. The expression of SLC38 A3 was lower in tumor tissues compared to that in normal tissues, which was also significantly associated with worse clinical outcome. Further experiments revealed that depletion of SLC38 A3 could promote EMT in ESCC cell lines, and the interaction of SLC38 A3 and SETDB1 might lead to the reduced transcription of Snail. Pharmacogenomic analyses demonstrated that fifteen inhibitors were showed significantly correlated with SLC38 A3 ***: Our investigations have provided insights that SLC38 A3 could act as a suppressor in EMT pathway and serve as a prognostic factor and predictor of differential drug sensitivities in ESCC.

BACKGROUND Immunoinflammatory markers such as the peripheral blood neutrophil-tolymphocyte ratio(NLR)and the platelet-to-lymphocyte ratio(PLR)have gained considerable attention as prognostic markers in gastrointestinal stromal tumors(GISTs).AIM To assess the prognostic value of Onodera’s Prognostic Nutritional Index(OPNI)for *** All patients who had undergone surgical resection for a primary,localized GIST from 2009 to 2016 at our cancer center were initially and retrospectively ***-free survival(RFS)was calculated by the Kaplan-Meier method and compared by the log-rank *** used multivariate Cox proportional hazard regression models to identify associations with outcome *** A total of 235 GISTs were identified and included for analysis under our inclusion *** and multivariate analyses both identified the OPNI as an independent prognostic marker,and the OPNI was associated with the primary site,tumor size,mitotic index,tumor rupture,necrosis,and modified NIH risk *** OPNI(<51.30;hazard ratio=5.852;95% confidence interval:1.072–31.964;P=0.0414)was associated with worse *** 2-and 5-year RFS rates of the patients with a low OPNI were 92.83% and 76.22%,respectively,whereas 100% and 98.41% were achieved by the patients with a high *** The preoperative OPNI is a novel and useful prognostic marker for GISTs.

Curbing Charging Currents in Pulsed Field Emission by Prolonging Pulse Edges

In field emission under a non-dc voltage, a displacement current is inevitable due to charging the cathode–anode condenser. Under an often-used square voltage pulse, in which the voltage rises from zero to a certain value abruptly, the charging current in the circuit is very large at the rising and falling edges. This large charging current makes measurement of the actual emissive current from the cathode difficult, constitutes a threat to the components in the circuit and causes attenuation of the emissive current within the pulse. To alleviate these drawbacks, trapezoid voltage pulses, whose rising edges are extended dramatically in comparison with square voltage pulses, are employed to extract the field emission. Under a trapezoid voltage pulse, the charging current is clearly lowered as expected. Furthermore, the heat generated by the charging current under the trapezoid voltage pulse is much smaller than that under the square voltage pulse. Hence the emissive current does not show any attenuation within the pulse. Finally, the average emissive currents are found to decrease with the repetition frequency of the pulses.
