A new approach for the separation of antithrombin III with high performance affinity chromatography (HPAC) was described. A novel monodisperse, non-porous, cross-linked poly (glycidyl methacrylate) beads (PGMA) were used as the affinity support. With the water-soluble carbodiimide, heparin was linked covalently to amino-PGMA-beads, which was prepared by amination of PGMA. The adsorbent obtained exhibits high binding activity to antithrombin III (ATIII), good resolution and excellent mechanical properties and can be used under high flow rate.

A simple aqueous two-phase extraction system （ATPS） of PEG/phosphate was proposed for selective separation and enrichment of proteins. The combination of ATPE with HPLC was applied to identify the partition of proteins in two phases. Five proteins （bovine serum albumin, Cytochrome C, lysozyme, myoglobin, and trypsin） were used as model proteins to study the effect of phosphate concentration and pH on proteins partition. The PEG/phosphate system was firstly applied to real human saliva and plasma samples, some proteins showed obviously different partition in two phases. The primary results manifest the selective separation and enrichment of proteins in ATPS provided the potential for high abundance proteins depletion in proteomics. ~ 2009 Feng Qu. The second member of the Lucaogou Formation(P_(2)l_(2))in the Tiaohu and Malang Sags of the Santanghu Basin(study area)underwent periodic volcanic activity and frequent lithological *** study comprehensively analyzed the organic geochemistry,mineral composition,organic matter(OM)types,volcanic cycle,and palaeoenvironment of shale in the study *** such as total organic carbon(TOC),rock pyrolysis(Rock-Eval),organic petrology,scanning electron microscopy(SEM)with energydispersive spectrum(EDS)analysis,X-ray diffraction(XRD),trace elements,and saturated hydrocarbon gas chromatography and mass spectrometry(GC-MS)were *** findings suggest that limited terrigenous input during the sedimentary period of the P_(2)l_(2) led to the deposition of a distinctive mixture of volcanic ash(felsic)and carbonate(dolomite and calcite),with a low average clay mineral content of 6%.The P_(2)l_(2) shale emerged as a high-quality source rock,predominantly composed of type I and II kerogens,with moderate OM *** deposition environment was characterized by hot and arid conditions,high salinity,and intensive reducibility,which was favorable for algae development and conducive to OM ***,two lamalginite types,labeled as lamalginite“A”and lamalginite“B,”were ***“B”-rich shales were deposited in a hotter and drier climate compared to lamalginite“A”-rich ***“B”-rich shale inexhibited high levels of C_(28)regular sterane andβ-carotenes,distinguishing it from lamalginite“A”-rich shale.A comprehensive analysis involving organic petrology,SEM,sedimentary environment,and biomarker characteristics suggests that lamalginite“B”may be a salt-tolerant green alga,while lamalginite“A”may be a ***,an OM enrichment model for the P_(2)l_(2) shale was established.

采用蓝绿温和胶凝胶电泳(blue-nativepolyacrylamidegel-electrophoresis,BN-PAGE),以及改进的第二向SDS-PAGE分离了水稻低叶绿素b突变体ZH249-Y和野生型ZH249-W类囊体膜蛋白复合物,系统比较了突变体和野生型各复合物亚基的表达差异.结果显示,第一向BN-PAGE分离了PSⅠ-LHCⅠ、LHCⅠ缺失的PSⅠ、ATP合成酶、细胞色素b6f、CP43缺失的PSⅡ及LHCⅡ六种复合物.上述各复合物经第二相SDS-Urea-PAGE分离后,利用胶内酶解,高效液相层析分离肽段,电喷雾串联质谱鉴定了复合物的亚基.结合免疫印迹研究,证明和野生型相比,突变体光系统Ⅱ捕光天线复合体的表达量适度下降,但光系统Ⅰ捕光天线破坏严重,同时光系统Ⅱ核心蛋白和ATP合成酶的表达量上升.研究结果对揭示低叶绿素b水稻突变体较高光化学效率和光稳定性的分子基础提供了线索,同时也表明,改进的BN/SDS-PAGE双向电泳不仅可以有效地分离膜蛋白复合物及亚基,也可以进行不同生理条件下,或野生型和突变体之间膜蛋白质组的比较研究.

BACKGROUND The benefits of healthy lifestyles are well recognized. However, the extent to which improving unhealthy lifestyles reduces cardiovascular disease(CVD) risk needs to be discussed. We evaluated the impact of lifestyle improvement on CVD incidence using data from the China-PAR project(Prediction for Atherosclerotic Cardiovascular Disease Risk in China).METHODS A total of 12,588 participants free of CVD were followed up for three visits after the baseline examination. Changes in four lifestyle factors(LFs)(smoking, diet, physical activity, and alcohol consumption) were assessed through questionnaires from the baseline to the first follow-up visit. Cox proportional hazard models were used to estimate hazard ratios(HRs) and corresponding 95% confidence intervals(CIs). The risk advancement periods(RAPs: the age difference between exposed and unexposed participants reaching the same incident CVD risk) and population-attributable risk percentage(PAR%) were also *** A total of 909 incident CVD cases occurred over a median follow-up of 11.14 years. Compared with maintaining 0-1healthy LFs, maintaining 3–4 healthy LFs was associated with a 40% risk reduction of incident CVD(HR = 0.60, 95% CI: 0.45–0.79)and delayed CVD risk by 6.31 years(RAP:-6.31 [-9.92,-2.70] years). The PAR% of maintaining 3–4 unhealthy LFs was 22.0%compared to maintaining 0-1 unhealthy LFs. Besides, compared with maintaining two healthy LFs, improving healthy LFs from 2to 3–4 was associated with a 23% lower risk of CVD(HR = 0.77, 95% CI: 0.60–0.98).CONCLUSIONS Long-term sustenance of healthy lifestyles or improving unhealthy lifestyles can reduce and delay CVD risk.

FMS-like tyrosine kinase 3(FLT3)mutation is strongly associated with poor prognosis in acute myeloid leukemia(AML).Though many FLT3 inhibitors have been developed for clinical application with 34%-56%complete remission rate,patients would develop resistance sooner or later after initial response to tyrosine kinase inhibitors(TKIs),such as *** increasing studies have shown that several resistance related mutations of FLT3 emerged during the AML ***,further investigation is warranted for these FLT3mu,AML patients to achieve a better treatment outcome.4-Hydroxyphenyl retinamide(4-HPR)has been investigated extensively in animal models and clinical trials as an anticancer/chemopreventive agent and is currently used for protection against cancer development/recurrence,with minimal side *** this study,we performed gene-set enrichment analysis and found that down-regulated genes induced by 4-HPR were associated with FLT3-ITD gene ***34+ AML stem/progenitor cells separated from 32 AML samples were treated with *** analysis showed that AML cells with FLT3-ITD genetic alteration were more sensitive to 4-HPR treatment than those without ***,we treated 22 primary AML cells with 4-HPR and found that 4-HPR was more toxic to AML cells with *** results indicated that 4-HPR was preferentially cytotoxic to all FLT3-ITD AML cells irrespective of stem/progenitor cells or blast cells.4-HPR-induced reactive oxygen species(ROS)production and NF-kB inhibition might be the reason of 4-HPR selectivity on FLT3 mutated AML cells.

Background The relationships between dietary intake of soybean products and incident hypertension were still *** study aimed to illustrate the associations between intake of soybean products with risks of incident hypertension and longitudinal changes of blood pressure in a prospective cohort *** We included 67,499 general Chinese adults from the Project of Prediction for Atherosclerosis Cardiovascular Disease Risk in China(China-PAR).Information about soybean products consumption was collected by standardized questionnaires,and study participants were categorized into the ideal(≥125 g/day)or non-ideal(<125 g/day)*** ratios(HRs)and corresponding 95%confidence intervals(95%CIs)for incident hypertension were calculated using Cox proportional hazard *** participants with repeated measures of blood pressure,generalized linear models were used to examine the relationships between soybean products consumption and blood pressure *** During a median follow-up of 7.4 years,compared with participants who consumed<125 g of soybean products per day,multivariable adjusted HR for those in the ideal group was 0.73(0.67-0.80).This inverse association remained robust across most subgroups while significant interactions were tested between soybean products intake and age,sex,urbanization and geographic region(P values for interaction<0.05).The mean systolic and diastolic blood pressure levels were 1.05(0.71-1.39)mmHg and 0.44(0.22-0.66)mmHg lower among participants in the ideal group than those in the non-ideal *** Our study showed that intake of soybean products might reduce the long-term blood pressure levels and hypertension incidence among Chinese population,which has important public health implications for primary prevention of hypertension.

Background Previous studies have demonstrated that micro RNA-204(mi R-204) is involved in atherosclerosis and vascular calcification. However, the value of mi R-204 as the predictive biomarker for cardiovascular disease(CVD) remains unclear. We aimed to evaluate the association between the circulating mi R-204 level and ten-year CVD risk based on the Framingham risk score(FRS). Methods In this retrospective study, we enrolled 194 consecutive patients with type 2 diabetes mellitus(T2DM) without CVD in Beijing Anzhen Hospital between January 2015 and September 2016. We used the FRS to evaluate the risk of CVD for each patient. Circulating mi R-204 levels were measured by quantitative real-time polymerase chain reaction. Results Circulating mi R-204 levels were significantly lower in the group of patients(0.49 ± 0.13) at high risk of CVD(FRS > 20%) than in the low(FRS < 10%) and intermediate(FRS: 10%–20%) risk groups(0.87 ± 0.19 and 0.75 ± 0.25, respectively;P < 0.001). FRS was negatively correlated with mi R-204 levels(r =-0.421, P < 0.001). According to multivariate logistic analyses, reduced mi R-204 level was independently associated with an increased risk of CVD after adjusting for conventional risk factors(OR = 0.876, 95% CI: 0.807–0.950, P = 0.001). Receiver-operating characteristic curve analysis showed that the circulating mi R-204 level can predict the high risk of CVD with higher specificity than the traditional risk factor of high systolic blood pressure or the protective factor of high-density lipoprotein cholesterol. Conclusions Our study demonstrated that patients with lower circulating mi R-204 levels were at high risk for CVD. After adjustment for potential confounders, mi R-204 was independently associated with CVD in patients with T2DM.

Background Transvenous lead placement is the standard approach for left ventricular （LV） pacing in cardiac resynchronization ther- apy （CRT）, while the open chest access epicardial lead placement is currently the most frequently used second choice. Our study aimed to compare the ventricular electromechanical synchronicity in patients with heart failure after CRT with these two different LV pacing tech- niques. Methods We enrolled 33 consecutive patients with refractory heart failure secondly to dilated cardiomyopathy who were eligible for CRT in this study. Nineteen patients received transvenous （TV group） while 14 received open chest （OP group） LV lead pacing. Intraand inter-ventricular electromechanical synchronicity was assessed by tissue Doppler imaging （TDI） before and one year after CRT procedure. Results Before CRT procedure, the mean QRS-duration, maximum time difference to systolic peak velocity among 12 left ventricle segments （LV Ts-12）, standard deviation of time difference to systolic peak velocity of 12 left ventricle segments （LV Ts-SD）, and inter-ventficular mechanical delay （IVMD） in OP and TV group were 166 ± 17 ms and 170 ± 21 ms, 391 ±42 ms and 397 ± 36 ms, 144 ± 30 ms and 148 ± 22 ms, 58 ± 25 ms and 60 ± 36 ms, respectively （all P 〉 0.05）. At one year after the CRT, the mean QRS-duration, LV Ts-12, LV Ts-SD, and IVMD in TV and OP group were 128 ± 14 ms and 141 ± 22 ms （P = 0.031）, 136 ± 37 ms and 294 ± 119 ms （P = 0.023）, 50± 22 ms and 96 ± 34 ms （P = 0.015）, 27 ± 11 ms and 27 ± 26 ms （P = 0.86）, respectively. The LV lead implantation procedure time was 53.4±16.3 rain for OP group and 136 ± 35.1 min for TV group （P = 0.016）. The mean LV pacing threshold increased significantly from 1.7 ± 0.6 V/0.5 ms to 2.3 ± 1.6 V/0.5 ms （P 〈 0.05） in TV group while it remained stable in the OP group.
