Progesterone receptor membrane component 1 as a potential prognostic biomarker for hepatocellular carcinoma

AIM To investigate the clinicopathological significance of progesterone receptor membrane component 1(PGRMC1) and PGRMC2 in hepatocellular carcinoma(HCC). METHODS We performed immunohistochemical staining to evaluate the estrogen receptor(ER), progesterone receptor(PR), PGRMC1, and PGRMC2 in a clinical cohort consisting of 89 paired HCC and non-tumor liver samples. We also analyzed HCC data(n = 373) from The Cancer Genome Atlas(TCGA). We correlated the expression status of PGRMC1 and PGRMC2 with clinicopathological indicators and the clinical outcomes of the HCC patients. We knocked down or overexpressed PGRMC1 in HCC cell lines to evaluate its biological significance in HCC cell proliferation, differentiation, migration, and invasion. RESULTS We found that few HCC cases expressed ER(5.6%) and PR(4.5%). In contrast, most HCC cases expressed PGRMC1(89.9%) and PGRMC2(100%). PGRMC1 and PGRMC2 exhibited significantly lower expression in tumor tissue than in non-tumor tissue(P < 0.001). Lower PGRMC1 expression in HCC was significantly associated with higher serum alpha-fetoprotein expression(P = 0.004), poorer tumor differentiation(P = 0.045) and liver capsule penetration(P = 0.038). Low PGRMC1 expression was an independent predictor for worse disease-free survival(P = 0.002, HR = 2.384,CI: 1.377-4.128) in our cases, as well as in the TCGA cohort(P < 0.001, HR = 2.857, CI: 1.781-4.584). The expression of PGRMC2 did not relate to patient outcome. PGRMC1 knockdown promoted a poorly differentiated phenotype and proliferation of HCC cells in vitro, while PGRMC1 overexpression caused the opposite effects.CONCLUSION PGRMC1 is a non-classical hormonal receptor that negatively regulates hepatocarcinogenesis. PGRMC1 down-regulation is associated with progression of HCC and is a poor prognostic indicator.

AIM: To demonstrate that administering heparanase inhibitor PI-88 at 160 mg/d is safe and promising in reducing hepatocellular carcinoma(HCC) recurrence for up to 3 year following curative resection. METHODS: A total of 143 patients(83.1% of the 172 participants in the phase Ⅱ study) participated in the follow-up study. Of these patients, 50 had received no treatment, 48 had received 160 mg/d PI-88, and 45 had received 250 mg/d PI-88 during the phase Ⅱ trial. Safety parameters and the following efficacy endpoints were investigated:(1) time to recurrence;(2) diseasefree survival; and(3) overall survival. RESULTS: PI-88 at 160 mg/d delayed the onset and frequency of HCC recurrence, and provided a clinically significant survival advantage for up to 3 years after treatment compared with those of the control group:(1) the recurrence-free rate increased from 50% to 63%, and(2) time to recurrence at the 36 th percentile was postponed by 78%. The efficacy of administering PI-88 at 250 mg/d was confounded by a high dropout rate(11 out of 54 patients). Additionally, subgroup analyses of patients with(1) multiple tumors or a single tumor ≥ 2 cm; and(2) hepatitis B or C revealed that administering PI-88 at 160 mg/d conferred the most significant survival advantage(56.8% improvement in disease-free survival, P = 0.045) for patients with both risk factors for recurrence. CONCLUSION: Administering PI-88 at 160 mg/d is a safe and well-tolerated dosage that may confer significant clinical benefits for patients with HCC.

Theoretical Analysis of A Buoyant Jet Interacting with Small Amplitude Waves

A new theoretical solution is presented here for the dynamic characteristics of a buoyant jet due to opposing small amplitude waves. The conservation equations of mass, tangential moment^n and vertical momentum are solved by the integral method which encompasses the Gaussian profiles of velocity and density. The action of waves is incorporated into the equations of motion as an external force and a new exact solution is obtained to predict the trajectory, velocity distribution and boundary thickness of the buoyant jet over an arbitrary lateral cross section. It is found that the velocity along the centerline is inversely proportional to the ratio of the momentum of the wave to the buoyant jet. The averaged bound- ary width varies with the fluctuation of the boundary width, the distance from the orifice and the velocity correction function. Owing to the motion of waves, the fluctuation of the boundary width is proportional to the wave steepness.

Hydro-mechanical response with respect to the air ventilation for water filtration in homogeneous soil

When water penetrates into soil,interstitial air can become trapped by the infiltrating water.Neglecting the effect of air ventilation could cause deviations in the predicted pore water pressure and the associated effective stress.This study aims at the effect of air ventilation on the coupled hydro-mechanical responses in homogeneous soil during infiltration.A schematic concept of infiltration conditions(open-and closed-valve)in homogeneous soil is proposed for investigating their impacts on the pore water pressure and effective stress.Experiments of vertical soil column filled with Ottawa sand(ASTM C77820/30)were designed for two types of air ventilation(namely,open and closed infiltration).The evolution of pore water pressure at the cylinder bottom was recorded,and served as a benchmark problem for evaluating the coupled hydro-mechanical response.Coding with the commercial software,GeoStudio,was employed for the dynamic behaviors of pore-water and-air pressures as well as the evolving effective stress.It was found in both the experiments and numerical investigations that the infiltration condition plays a crucial role for the ascending rate of pore water pressure as well as the associated effective stress.These results illustrate the inevitable impacts of the air ventilation conditions on the mechanical properties of the soil during infiltration.

Embryonic Development and Eclosion Season of New Species Berastagia (Lepidoptera: Pyralidae) from Taiwan

This paper describes a new species of the snout moth Berastagia tainanica sp. nov. (Lepidoptera: Pyralidae) from Taiwan. From 2009 to 2016, a biology study was conducted on population dynamics and embryonic development. Spring season is the peak of the eclosion of overwintering larvae or pupae. The average longevity of adult was 14.8 ± 6.2 days (N = 174), the average number of eggs laid was 259 ± 3 eggs/moth (N = 2), the hatching rate of eggs was 95.4% (N = 262), and the average hatching time of eggs was 99.6 ± 18.6 hours (N = 68). The average body length of males was 5.64 mm ± 0.91 mm (N = 30), and the average body length of females was 6.28 mm ± 0.84 mm (N = 30). This finding indicates that female snout moths are larger than males (Global R = 0.058, P = 0.012). The snout moth eclosion rate was 16.9 moths/100 pods in the first year (2010/2011, N = 2,224 pods) and 10.9 moths/100 pods in the second year (2014/2015, N = 6,382 pods). The pod borer rate was 31.8% (N = 707) and the seed borer rate was 41.2% (N = 3,628) in the first year, whereas the pod borer rate was 76.2% (N = 6,382) in the second year.

Sexual Dimorphism, Length-Weight Relationships, Fecundity, and Diet of the Striped Eel Catfish Plotosus lineatus (Plotosidae) on Taiwan’s Southwest Coast

This study of feeding guild fish from the same coast of subtropics helps to suggest conservation of fishery resources. A total of 2,537 specimens of Plotosus lineatus (Plotosidae) were collected from July 2013 to March 2019 on the coast of Taiwan. The average total body length (TL) was 23.7 cm ± 4.7 cm and 20.3 cm ± 2.9 cm, and the average body weight (W) was 94.1 g ± 59.6 g and 56.6 g ± 30.8 g among 174 males and 630 females, respectively. The LWRs of male and female were ln(W)M = 3.2914 × ln (TL) −6.0395, and ln(W)F = 3.0917 × ln(TL) −5.3629. The male specimens were large than female in TL and W. The average OSW/TL ratios were 0.722 ± 0.044 and 0.649 ± 0.029 among 41 male and 49 female specimens, respectively. The OSW of the male head was wider than that of the female head (F = 91.5;p lineatus exhibits significant sexual dimorphism. The average eggs number of each female was 2375.6 eggs/individual ± 1140.9 eggs/individual (N = 161). The major prey of P. lineatus were shrimps, crabs, and fishes;the total relative frequencies of occurrence of the prey were 57.1%, 32.6%, and 37.2%, respectively (N = 282), and their total relative abundance levels were 40.2%, 26.3%, and 25.8%, respectively (N = 445). Proper scientific knowledge management will improve the development of fisheries.

BACKGROUND Although much information is available regarding hepatitis C virus infection and diabetes,less is known about the relationship between hepatitis A virus(HAV)infection and diabetes.AIM To examine the roles of HAV in diabetes risk.METHODS This cross-sectional study population included data from the National Health and Nutrition Examination Survey collected between 2005-2012.Adult subjects(≥20 years old)with available body mass index measurements,defined diabetes status,history of HAV vaccination,and HAV serology were included.HAV vaccination was based on self-reported history.Successful HAV immunization was defined as the presence of both vaccination and anti-HAV antibody.HAV infection was defined by the absence of vaccination but presence of anti-hepatitis A antibody.The odds ratio(OR)for diabetes with 95%confidence intervals(95%CI)was calculated for each HAV status and then adjusted for covariates.Sensitivity tests,based on different definitions of diabetes,were performed to verify the results.RESULTS Among 19942 subjects,4229 subjects(21.21%)received HAV vaccination and HAV antibody was present in 9224 subjects(46.25%).Although HAV infection was associated with an increased risk of diabetes(OR:1.13;95%CI:1.08-1.18),HAV vaccination was not associated with diabetes(OR:1.06;95%CI:0.95-1.18),and successful HAV immunization had no impact on the risk of diabetes(OR:1.11;95%CI:0.97-1.27).Thus,HAV infection was an unlikely cause of diabetes.Alternatively,in non-vaccinated subjects,diabetes increased the risk of HAV infection by 40%(OR:1.40,95%CI:1.27-1.54).CONCLUSION An association between HAV infection and diabetes is observed which is best explained by an increased risk of HAV infection in diabetic patients.Diabetic subjects are more susceptible to HAV.Thus,HAV vaccination is highly recommended in diabetic patients.

Monitoring of PON System Using Compound Surveillance Technique

A passive optical network（PON） monitoring system combined light pulse and frequency sweep techniques is proposed and verified in a field test. The light pulse surveys over the all whole network and the frequency sweep are used to investigate any fault in the link. The field test is performed with 4 PONs. Each PON is monitored at 4 ports, one is the splitter port and the other three are arbitrary chosen multiple optical units（ONUs）. All the tested PONs are monitored in turns once per hour. Faults at the feeder and branch fiber have been observed in this field test and have been analyzed with the monitoring system.

Loss of heterozygosity: An independent prognostic factor of colorectal cancer

AIM: Colorectal cancers result from the accumulation of several distinct genetic alterations. This study was to investigate the frequency and prognostic value of loss of heterozygosity (LOH) and microsatellite instability (MSI) at 14 genetic loci located near or within regions containing important genes implicated in colorectal tumorigenesis.METHODS: We studied colorectal cancers with corresponding normal mucosae in 207 patients (139 males and 68 females,mean age at the time of tumor resection 66.2±12.4 years,range 22-88 years). There were 37 right-sided colonic tumors, 85 left-sided colonic tumors and 85 rectal tumors.The distribution of tumor staging was stage Ⅰ in 25, stage Ⅱ in 73, stage Ⅲ in 68, and stage Ⅳ in 41. We analyzed the LOH and MSI of HPC1, hMSH2, hMLH1, APC, MET,P53, NH23-H1, DCC, BAT25, BAT26, D17S250, MYCL1 and D8S254 with fluorescent polymerase chain reaction and denatured gel electrophoresis. High-frequency LOH was determined to be greater than three, or more than 50%of the informative marker with LOH. High-frequency MSI (MSI-H) was determined as more than four markers with instability (＞30%). Correlations of LOH and MSI with clinical outcomes and pathological features were analyzed and compared.RESULTS: The occurrence of MSI-H was 7.25%, located predominantly in the right colons (7/15) and had a higher frequency of poor differentiation (6/15) and mucin production (7/15). LOH in at least one genetic locus occurred in 78.7% of the tumors and was significantly associated with disease progression. Of the 166 potentially cured patients, 45 developed tumor recurrence within 36 mo of follow-up. Clinicopathological factors affecting 3-year disease-free survival (DFS) were TNM staging, grade of differentiation, preoperative CEA level, and high LOH status. Patients with high LOH tumors had a significantly lower DFS (50%) compared with patients with low LOH tumors (84%).
