BACKGROUND Although highly effective as a component of Helicobacter pylori(***)treatment regimen,tetracycline is associated with a high incidence of medicationrelated adverse *** dosing of tetracycline as part of quadruple therapy may improve safety while providing comparable eradication *** To evaluate the efficacy and safety of modified dosing of tetracycline in patients receiving tetracycline and furazolidone-containing quadruple therapy in patients with *** *** Consecutive patients(10/2020-12/2021)who received tetracycline and furazolidone quadruple therapy for *** infection at Sir run run Shaw Hospital were *** patients received tetracycline,furazolidone,proton pump inhibitor,and bismuth for 14 d as primary or rescue *** tetracycline dose group received tetracycline 500 mg twice daily while standard group received 750 mg twice daily or 500 mg three times *** Three hundred and ninety-four patients[mean age=46.3±13.9,male=137(34.8%),and 309(78.4%)primary therapy]completed tetracycline and furazolidone quadruple therapy for *** infection including those who received modified tetracycline dose in 157 and standard doses in 118(750 mg twice daily)and 119(500 mg three times daily).Eradication rates in the modified tetracycline dose group were 92.40%and in the standard groups,eradication rates were 93.20%for 750 mg twice daily group and 92.43%for 500 mg three times daily group,respectively,without statistical difference(P=0.959).The incidence of adverse events was lower in the modified tetracycline dose(15.3%vs 32.3%and 29.4%;P=0.002)compared to the standard dose *** In a real-world experience,modified tetracycline dosing as part of tetracycline and furazolidone quadruple therapy for 14 d demonstrated high efficacy,comparable to standard tetracycline dose regimens,with a favorable safety profile.

β-ray-induced X-ray spectroscopy(BIXS)is a promising method for tritium detection in solid materials because of its unique advantages,such as large detection depth,nondestructive testing capabilities,and low requirements for sample ***,high-accuracy reconstruction of the tritium depth profile remains a significant challenge for this *** this study,a novel reconstruction method based on a backpropagation(BP)neural network algorithm that demonstrates high accuracy,broad applicability,and robust noise resistance is *** average reconstruction error calculated using the BP network(8.0%)was much lower than that obtained using traditional numerical methods(26.5%).In addition,the BP method can accurately reconstruct BIX spectra of samples with an unknown range of tritium and exhibits wide applicability to spectra with various tritium ***,the BP network demonstrates superior accuracy and stability compared to numerical methods when reconstructing the spectra,with a relative uncertainty ranging from 0 to 10%.This study highlights the advantages of BP networks in accurately reconstructing the tritium depth profile from BIXS and promotes their further application in tritium detection.

Alzheimer’s disease not only affects the brain,but also induces metabolic dysfunction in peripheral organs and alters the gut *** aim of this study was to investigate systemic changes that occur in Alzheimer’s disease,in particular the association between changes in peripheral organ metabolism,changes in gut microbial composition,and Alzheimer’s disease *** do this,we analyzed peripheral organ metabolism and the gut microbiota in amyloid precursor protein-presenilin 1(APP/PS1)transgenic and control mice at 3,6,9,and 12 months of ***-month-old APP/PS1 mice exhibited cognitive impairment,Alzheimer’s disease-related brain changes,distinctive metabolic disturbances in peripheral organs and fecal samples(as detected by untargeted metabolomics sequencing),and substantial changes in gut microbial composition compared with younger APP/PS1 ***,a strong correlation emerged between the gut microbiota and kidney metabolism in APP/PS1 *** findings suggest that alterations in peripheral organ metabolism and the gut microbiota are closely related to Alzheimer’s disease development,indicating potential new directions for therapeutic strategies.

Leber先天性黑矇(Leber's congenital amaurosis,LCA)是一种遗传性致盲性眼病,在婴儿早期出现严重的视力低下或丧失丧失。该疾病的LCA2型与RPE65的突变相关。既往对于LCA2在内的遗传性视网膜疾病无有效治疗方法。近年来,随着基因治疗技术的进步,遗传性视网膜疾病的治疗进展取得了巨大进步,其中最成功的便是LCA2的基因治疗。本文简要介绍了LCA2基因治疗的发展,并对既往LCA2临床试验中的注射剂型、剂量、注射方式、测量方法、治疗效果与年龄的相关性和治疗效果的稳定性进行综述,为LCA2基因治疗进入我国临床工作提供参考及临床治疗经验。

A dual-protease-triggered chemiluminescent probe for precise tumor imaging

Imaging detection of interlinked dual proteases is imperative for precise tumor imaging, which remains challenging due to limited modification position of specific substrate and possible steric ***, we have developed a unimolecular chemiluminescent probe(LGP-CL) tandemly activated by two proteases interlinked with liver cancer to achieve precise tumor imaging. Probe LGP-CL consists of a phenoxy-dioxetane scaffold caged by a tripeptide substrate(LGP, leucine-glycine-proline) as the sensing layer, which can be cleaved sequentially by aminopeptidase N(APN) and dipeptidyl peptidase IV(DPPIV)to turn on a strong chemiluminescent signal, and silenced by specific inhibitor of each enzyme, which accounts for an integrated logic gate(AND, OR and INHIBIT). The successful cleavage of dual proteases on the metabolic site depends on the proper structure of the tripeptide substrate, as confirmed by two probes design. Probe LGP-CL(LGP as the substrate) enables the excellent “dual-lock-dual-key” fit with a382-fold enhancement of chemiluminescent emission while no obvious signal is observed by using GPLCL(GPL as the substrate). By virtue of its rapid response(several minutes), high sensitivity and good cell viability, probe LGP-CL has been utilized to evaluate upregulated levels of proteases in vitro and in living systems, especially to distinguish liver tumor cells(Hep G2) from others(LO2, MCF-7, MCF-10a and RAW264.7). Overall, the newly developed CL probe may facilitate rapid investigation into the role played by proteases in liver diseases, enabling timely selection appropriate treatment. Therefore, our work not only sheds light on the rational design of optical probes for dual protease imaging, but provides a promising tool for clinical diagnosis and even drug discovery.
