Mkrn2 deficiency induces teratozoospermia and male infertility through p53/PERP-mediated apoptosis in testis

The apoptosis that occurs in the immature testis under physiological conditions is necessary for male germ cell development,whereas improper activation of apoptosis can impair spermatogenesis and cause defects in *** previously demonstrated that in mice,the makorin-2(Mkrn 2)gene is expressed exclusively in the testis and its deletion leads to male *** understand the potential molecular mechanism,in this study,we found that levels of apoptosis in the testis were abnormally high in the absence of Mkrn *** identify specific gene(s)involved,we performed digital gene expression profiling(DGE)and pathway analysis via gene set enrichment analysis(GSEA)and the Kyoto Encyclopedia of Genes and Genomes(KEGG)database,and we found that MKRN2 inhibits p53 apoptosis effector related to PMP22(PERP)expression and that levels of the protein in sperm samples have an inverse correlation with infertility *** additionally indicated that PERP is a negative regulator of spermatogenesis and that its ectopic expression induces male ***,Gene Expression Omnibus(GEO)dataset analysis showed that p53,upstream of PERP,was upregulated in oligoasthenoteratozoospermia(OAT).These observations suggest that Mkrn 2 is crucial for protecting germ cells from excessive apoptosis and implicate Mkrn 2-based suppression of the p53/PERP signaling pathway in spermatogenesis and male fertility.

Nonlinear dynamics of electroencephalography study in schizophrenic patients

Background Few characteristic changes of linear electroencephalograph （EEG） have been reported in schizophrenia. The aim of the present study was to investigate the changes in temporal-spatial dimensional properties of EEG under different cognitive tasks in patients with schizophrenia. Methods EEG was recorded by using EEG-1518K system and mapping system （Nihon Kohden Tomioka Corporation, Japan） in 45 schizophrenic patients and 47 healthy adults （normal control, NC） under five states： eyes c^osed, eyes open, mental arithmetic test with eyes closed, memory test with eyes open, and number cancellation test. Correlation dimension （D2） and point-wise correlation dimension （PD2） were calculated for all EEG analyses. Results （1） There were no significant differences of D2 and PD2 between NC and schizophrenic patients under states of eyes open and closed. （2） Compared with NC, schizophrenic patients showed decreased performance of D2 in mental arithmetic test with eyes closed and number cancellation test （mental arithmetic test with eyes closed： Nc 5.9±0.6, Sch 3.0＋0.8; number cancellation test： Nc 6.0±0.6, Sch 4.4±0.7; P 〈0.05 or P 〈0.01）. （3） Schizophrenic patients also showed decrease performance of PD2 in mental arithmetic test with eyes closed, memory test with eyes open, and number cancellation test （mental arithmetic test with eyes closed： Nc 6.9±0.7, Sch 4.0±0.8; memory test with eyes open： Nc 6.6±0.8, Sch 5.0±0.9; number cancellation test： Nc 7.1±0.7, Sch 4.8±0.9; P 〈0.05 or P 〈0.01）. Conclusions Nonlinear dynamic analysis provided a new approach in clinical investigation of EEG signals. It was helpful to further understand the cerebral mechanism in schizophrenic cognitive process.

Optimization of the co-closing law of guide vanes and blades for bulb turbines based on CFD

The load rejection transient process of bulb turbine units is critical to safety of hydropower stations,and determining appropriate closing laws of guide vanes(GVs)and runner blades(RBs)for this process is of *** this study,we proposed a procedure to optimize the co-closing law of GVs and RBs by using computational fluid dynamics(CFD),combined with the design of experiment(DOE)method,approximation model,and genetic optimization *** sensitivity of closing law parameters on the histories of head,speed,and thrust was analyzed,and a two-stage GVs’closing law associating with a linear RBs’closing law was *** results show that GVs dominate the transient characteristics by controlling the change of *** GVs’first-stage closing speed while shortening first-stage closing time can not only significantly reduce the maximum rotational speed but also suppress the water hammer pressure;slowing GVs’second-stage closing speed is conducive to controlling the maximum reverse axial *** directly affect the runner *** RBs’closing speed can further reduce the rotational speed and the maximum reverse axial *** safety margin of each control parameter,flow patterns,and pressure pulsations of a practical hydropower station were all improved after optimization,demonstrating the effectiveness of this method.

Improved multi-scale inverse bottleneck residual network based on triplet parallel attention for apple leaf disease identification

Accurate diagnosis of apple leaf diseases is crucial for improving the quality of apple production and promoting the development of the apple industry. However, apple leaf diseases do not differ significantly from image texture and structural information. The difficulties in disease feature extraction in complex backgrounds slow the related research progress. To address the problems, this paper proposes an improved multi-scale inverse bottleneck residual network model based on a triplet parallel attention mechanism, which is built upon ResNet-50, while improving and combining the inception module and ResNext inverse bottleneck blocks, to recognize seven types of apple leaf(including six diseases of alternaria leaf spot, brown spot, grey spot, mosaic, rust, scab, and one healthy). First, the 3×3 convolutions in some of the residual modules are replaced by multi-scale residual convolutions, the convolution kernels of different sizes contained in each branch of the multi-scale convolution are applied to extract feature maps of different sizes, and the outputs of these branches are multi-scale fused by summing to enrich the output features of the images. Second, the global layer-wise dynamic coordinated inverse bottleneck structure is used to reduce the network feature loss. The inverse bottleneck structure makes the image information less lossy when transforming from different dimensional feature spaces. The fusion of multi-scale and layer-wise dynamic coordinated inverse bottlenecks makes the model effectively balances computational efficiency and feature representation capability, and more robust with a combination of horizontal and vertical features in the fine identification of apple leaf diseases. Performance of iron-air battery with iron nanoparticle-encapsulated C-N composite electrode

Highly efficient and stable iron electrodes are of great significant to the development of iron-air battery(IAB).In this paper,iron nanoparticle-encapsulated C–N composite(NanoFe@CN)was synthesized by pyrolysis using polyaniline as the C–N *** performance of the NanoFe@CN in different electrolytes(alkaline,neutral,and quasi-neutral)was investigated via cyclic voltammetry(CV).The IAB was assembled with NanoFe@CN as the anode and IrO_(2)+Pt/C as the *** effects of different discharging/charging current densities and electrolytes on the battery performance were also *** K_(2)SO_(4)electrolyte can effectively suppress the passivation of iron electrode,and the battery showed a good cycling stability during 180 charging/discharging *** to the pure nano-iron(NanoFe)battery,the NanoFe@CN battery has a more stable cycling stability either in KOH or NH_(4)Cl+KCl electrolyte.

AIM:To investigate systemic immune-inflammation index(SII),neutrophil-to-lymphocyte ratio(NLR),and plateletto-lymphocyte ratio(PLR)levels in patients with type 2 diabetes at different stages of diabetic retinopathy(DR).METHODS:This retrospective study included 141 patients with type 2 diabetes mellitus(DM):45 without diabetic retinopathy(NDR),47 with non-proliferative diabetic retinopathy(NPDR),and 49 with proliferative diabetic retinopathy(PDR).Complete blood counts were obtained,and NLR,PLR,and SII were *** study analysed the ability of inflammatory markers to predict DR using receiver operating characteristic(ROC)*** relationships between DR stages and SII,PLR,and NLP were assessed using multivariate logistic ***:The average NLR,PLR,and SII were higher in the PDR group than in the NPDR group(P=0.011,0.043,0.009,respectively);higher in the NPDR group than in the NDR group(P<0.001 for all);and higher in the PDR group than in the NDR group(P<0.001 for all).In the ROC curve analysis,the NLR,PLR,and SII were significant predictors of DR(P<0.001 for all).The highest area under the curve(AUC)was for the PLR(0.929 for PLR,0.925 for SII,and 0.821 for NLR).Multivariate regression analysis indicated that NLR,PLR,and SII were statistically significantly positive and independent predictors for the DR stages in patients with DM[odds ratio(OR)=1.122,95%confidence interval(CI):0.200–2.043,P<0.05;OR=0.038,95%CI:0.018–0.058,P<0.05;OR=0.007,95%CI:0.001–0.01,P<0.05,respectively).CONCLUSION:The NLR,PLR,and SII may be used as predictors of DR.

Effects of formal credit on pastoral household expense: Evidence from the Qinghai-Xizang Plateau of China

Formal credit is critical in agricultural production,allowing more expenditure and productive input,thereby improving farmers'*** pastoral China,formal financial institutions are gradually ***,a limited understanding remains of how formal credit affects herders'household *** on a survey of 544 herders from the Qinghai-Xizang Plateau of China,this study adopted the propensity score matching approach to identify the effect of formal credit on herders'total household expenses,daily expenses,and productive *** results found that average age,grassland mortgage,and other variables significantly affected herders'participation in formal *** credit could significantly improve household expenses,especially productive expenses.A heterogeneity analysis showed that formal credit had a greater impact on the household total expense for those at higher levels of wealth;however,it significantly affected the productive expense of herders at lower wealth ***,the mediating effect indicated that formal credit could affect herders'household income,thus influencing their household ***,this study suggests that policies should improve herders'accessibility to formal credit.
