Background:In this study,we used network pharmacology and molecular docking combined with vitro experiments to explore the potential mechanism of action of Gualou Qumai pill(GLQMP)against ***:We screened effective compounds and drug targets using Chinese medicine systemic pharmacology database and analysis platform and Chinese medicine molecular mechanism bioinformatics analysis tools;and searched for DKD targets using human online Mendelian genetics and gene *** potential targets of GLQMP for DKD were obtained through the intersection of drug targets and disease *** software was applied to build herbal medicine-active compound-target-disease networks and analyze them;protein-protein interaction networks were analyzed using the STRING database platform;gene ontology and Kyoto Encyclopedia of Genes and Genomes were used for gene ontology and gene and genome encyclopedia to enrich potential targets using the DAVID database;and the AutoDock Vina 1.1.2 software for molecular docking of key targets with corresponding key *** vitro experiments were validated by CCK8,oil red O staining,TC,TG,RT-qPCR,and Western ***:Through network pharmacology analysis,a total of 99 potential therapeutic targets of GLQMP for DKD and the corresponding 38 active compounds were obtained,and 5 core compounds were *** constructing the protein-protein interaction network and performing network topology analysis,we found that PPARA and PPARG were the key targets,and then we molecularly docked these two key targets with the 38 active compounds,especially the 5 core compounds,and found that PPARA and PPARG had good binding ability with a variety of *** vitro experiments showed that GLQMP was able to ameliorate HK-2 cell injury under high glucose stress,improve cell viability,reduce TC and TG levels as well as decrease the accumulation of lipid droplets,and RT-qPCR and Western blot confirmed that GLQMP was able to promote the expressi

Effect of emodin on mobility signal transduction system of gallbladder smooth muscle in Guinea pig with cholelithiasis

Objective: To study the effect of emodin on protein and gene expressions of the massagers in mobility signal transduction system of cholecyst smooth muscle cells in guinea pig with cholesterol calculus. Methods: The guinea pigs were randomly divided into 4 groups, such as control group, gall-stone(GS) group, emodin group and ursodesoxycholic acid(UA) group. Cholesterol calculus models were induced in guinea pigs of GS, emodin and UA groups of induced models by lithogenic diet, while emodin or UA were given to the corresponding group for 7 weeks. The histomorphological and ultrastructure change of gallbladder were detected by microscope and electron microscope, the content of plasma cholecystokinin(CCK) and [Ca^(2+)]i were analyzed successively by radioimmunoassay and flow cytometry. The protein and mR NA of Gsα, Giα and Cap in cholecyst cells were determined by western blotting and real time polymerase chain reaction(RT-PCR). Results: Emodin or UA can relieve pathogenic changes in epithelial cells and muscle cells in gallbladder of guinea pig with cholesterol calculus by microscope and transmission electron microscope. In the cholecyst cells of GS group, CCK levels in plasma and [Ca^(2+)]i decreased, the protein and m RNA of GS group were downregulated,the protein and m RNA of Gi and Cap were up-regulated. Emodin significantly decreased the formative rate of gallstone, improved the pathogenic change in epithelial cells and muscle cells, increased CCK levels in plasma and [Ca^(2+)]i in cholecyst cells, enhanced the protein and mR NA of Gs in cholecyst cells, reduced the protein and mR NA of Gi and Cap in cholecyst cells in guinea pig with cholesterol calculus. Conclusion: The dysfunction of gallbladder contraction gives rise to the disorders of mobility signal transduction system in cholecyst smooth muscle cells, including low content of plasma CCK and [Ca^(2+)]i in cholecyst cells, abnormal protein and mRNA of Gs, Gi and Cap. Safe confinement of fission iodine isotopes for long-term radioactive waste disposal remains a formidable challenge,as conventional sorbents provide inherently weak iodine-host *** report here a novel halogen bond(X-bond)directed strategy to sequester volatile iodine in hydrogen-bonded(H-bonded)frameworks with unprecedented ***-assisted Hbonded frameworks bearing open halide sites are developed,showing distinctive iodine encapsulation behaviors without compromising the *** crystallographic evidence indicates the formation of X-bonds,i.e.,I–I···Cl^(−) and I–I···Br^(−),within the confined pore *** polyhalogen anions,i.e.,[I_(2)Cl_(2)]^(2−)and[I_(2)Br_(2)]^(2−),sustained in H-bonded frameworks are identified for the first *** X-bond reinforced host-guest interaction affords robust iodine trapping without leaking out even at elevated temperatures up to 180℃.By integrating the halogen-bond chemistry with H-bonded frameworks,this study offers fresh concepts for developing effective host reservoirs to secure fission iodine isotopes from spent fuel reprocessing off-gases.

We establish an explicit embedding of a quantum affine sl_(n) into a quantum affine sl_(n)+*** embedding serves as a common generalization of two natural,but seemingly unrelated,embed-dings,one on the quantum affine Schur algebra level and the other on the non-quantum *** embedding on the quantum affine Schur algebras is used extensively in the analysis of canonical bases of quantum affine sln and gl_(n).The embedding on the non-quantum level is used crucially in a work of Riche and Williamson on the study of modular representation theory of general linear groups over a finite *** same embedding is also used in a work of Maksimau on the categorical representations of affine general linear *** further provide a more natural compatibility statement of the em-bedding on the idempotent version with that on the quantum affine Schur algebra level.A gl_(n)-variant of the embedding is also established.

Sulfated Cyclocarya paliurus polysaccharides exert immunomodulatory potential on macrophages via Toll-like receptor 4 mediated MAPK/NF-κB signaling pathways

The biological activity of plant polysaccharides can be enhanced by sulfated *** this study,the immunomodulatory effect of sulfated Cyclocarya paliurus polysaccharides(SCP3)on macrophages RAW264.7 and its potential molecular mechanism were *** showed that SCP3 at 25-100μg/m L increased viability and improved phagocytosis of RAW264.7 ***,SCP3 could activate mitogen-activated protein kinase(MAPK)and nuclear factor kappa B(NF-κB)signaling pathways,which increased the phosphorylation of Erk1/2,JNK,p38 and NF-κB p65,promoting secretion of cytokines tumor necrosis factorα(TNF-α),interleukin 6(IL-6)and nitric oxide(NO)as well as the production of reactive oxygen species(ROS).In addition,Toll-like receptor 4(TLR4)receptor inhibitors were able to block the production of NO and TNF-αby SCP3-stimulated *** on Western blot analysis and validation using specific inhibitors against MAPK and NF-κB signaling pathways,the results demonstrated that SCP3 induced macrophages activation and enhanced TNF-αand NO production via TLR4-mediated MAPK and NF-κB *** summary,SCP3 has significant immunomodulatory *** underlying molecular mechanism was that SCP3 activates macrophages via TLR4 receptors to promote ROS production,which in turn activates the downstream MAPK/NF-κB signaling pathway and then increases the secretion levels of cytokines and NO.

TSC prediction and dynamic control of BOF steelmaking with state-of-the-art machine learning and deep learning methods

Mathematical(data-driven)models based on state-of-the-art(SOTA)machine learning and deep learning models and data collected from 12,786 heats were established to predict the values of temperature,sample,and carbon(TSC)test,including temperature of molten steel(TSC-Temp),carbon content(TSC-C)and phosphorus content(TSC-P),which made prepa-ration for eliminating the TSC *** maximize the prediction accuracy of the proposed approach,various models with different inputs were implemented and compared,and the best models were applied to the production process of a Hesteel Group steelmaking plant in China in the *** number of tabular features(hot metal information,scrap,additives,blowing practices,and preset values)was expanded,and time series(off-gas profiles and blowing practice curves)that could reflect the entire steelmaking process were introduced as ***,the latest machine learning models(LightGBM,CatBoost,TabNet,and NODE)were used to make predictions with tabular features,and the best coefficient of determination R^(2)values obtained for TSC-P,TSC-C and TSC-Temp predictions were 0.435(LightGBM),0.857(Cat-Boost)and 0.678(LightGBM),respectively,which were higher than those of classic models(backpropagation and support vector machine).Then,making predictions was performed by using SOTA time series regression models(SCINet,DLinear,Informer,and MLSTM-FCN)with original time series,SOTA image regression models(NesT,CaiT,ResNeXt,and GoogLeNet)with resized time series,and the proposed Concatenate-Model and Parallel-Model with both tabular features and time *** optimization and comparisons,it was finally determined that the Concatenate-Model with MLSTM-FCN,SCINet and Informer as feature extractors performed the best,and its R^(2)values for predicting TSC-P,TSC-C and TSC-Temp reached 0.470,0.858 and 0.710,*** field test accuracies for TSC-P,TSC-C and TSC-Temp were 0.459,0.850 and 0.685,respectively.A related importance analysis was carri
