Currently, there is no effective strategy to promote functional recovery after a spinal cord injury. Collagen scaffolds can not only provide support and guidance for axonal regeneration, but can also serve as a bridge for nerve regeneration at the injury site. They can additionally be used as carriers to retain mesenchymal stem cells at the injury site to enhance their effectiveness. Hence, we hypothesized that transplanting human umbilical cord-mesenchymal stem cells on collagen scaffolds would enhance healing following acute complete spinal cord injury. Here, we test this hypothesis through animal studies and a phase I clinical trial.(1) Animal experiments: Models of completely transected spinal cord injury were established in rats and canines by microsurgery. Mesenchymal stem cells derived from neonatal umbilical cord tissue were adsorbed onto collagen scaffolds and surgically implanted at the injury site in rats and canines;the animals were observed after 1 week–6 months. The transplantation resulted in increased motor scores, enhanced amplitude and shortened latency of the motor evoked potential, and reduced injury area as measured by magnetic resonance imaging.(2) Phase I clinical trial: Forty patients with acute complete cervical injuries were enrolled at the Characteristic Medical Center of Chinese People's Armed Police Force and divided into two groups. The treatment group(n = 20) received collagen scaffolds loaded with mesenchymal stem cells derived from neonatal umbilical cordtissues;the control group(n = 20) did not receive the stem-cell loaded collagen implant. All patients were followed for 12 months. In the treatment group, the American Spinal Injury Association scores and activities of daily life scores were increased, bowel and urinary functions were recovered, and residual urine volume was reduced compared with the pre-treatment baseline. Background Mumps is a common type of respiratory infectious disease caused by mumps virus(MuV),and can be effectively prevented by vaccination.In this study,a reverse genetic system of MuV that can facilitate the rational design of safer,more efficient mumps vaccine candidates is established.Methods MuV-S79 cDNA clone was assembled into a full-length plasmid by means of the GeneArtTM High-Order Genetic Assembly System,and was rescued via reverse genetic technology.RT-PCR,sequencing,and immunofluorescence assays were used for rMuV-S79 authentication.Viral replication kinetics and in vivo experimental models were used to evaluate the replication,safety,and immunogenicity of rMuV-S79.Results A full-length cDNA clone of MuV-S79 in the assembly process was generated by a novel plasmid assemble strategy,and a robust reverse genetic system of MuV-S79 was successfully established.The established rMuV-S79 strain could reach a high virus titer in vitro.The average viral titer of rMuV-S79 in the lung tissues was 2.68±0.14 log10PFU/g lung tissue,and rMuV-S79 group did not induce inflammation in the lung tissues in cotton rats.Neutralizing antibody titers induced by rMuV-S79 were high,long-lasting and could provide complete protection against MuV wild strain challenge.Conclusion We have established a robust reverse genetic system of MuV-S79 which can facilitate the optimization of mumps vaccines.rMuV-S79 rescued could reach a high virus titer and the safety was proven in vivo.It could also provide complete protection against MuV wild strain challenge.

为探究烟草源微生物单菌发酵对雪茄烟叶挥发性致香成分的影响,采用GC-MS检测莫海威芽孢杆菌(Bacillus mojavensis)ZLX10发酵雪茄烟叶中性致香成分,筛选差异致香成分,并与感官评价进行相关性分析,结合ROAV值变化,明确ZLX10提升雪茄烟叶感官品质的关键致香成分。结果表明,ZLX10发酵雪茄烟叶后,香气总量和大多数中性致香成分含量均显著提高,共筛选得到15种差异中性致香成分,共有10种致香成分显著上调(P<0.05),其中巨豆三烯酮A、巨豆三烯酮B、6-甲基-5-庚烯-2-酮、2,6,6-三甲基-2-环己烯-1,4-二酮、茄酮和植物醇与烟叶香气呈显著正相关(P<0.05),可认为是增加烟叶干香和花香的关键差异致香成分;共有5种致香成分显著下调(P<0.05),其中苯乙醛和2,4-二叔丁基-6-硝基苯酚与烟叶刺激性呈显著正相关(P<0.05),对烟叶香气有较大的负面影响。由此可知,ZLX10发酵能促进雪茄烟叶香气质和香气量得到提升,这为人工接菌发酵雪茄烟叶提供了基础。

以锦屏Ⅱ级水电站排水洞为研究对象,采用数值模拟和微震监测方法,分析隧道近断层岩爆形成机理,揭示断层(断层角度、断层与隧道的相对位置)和赋存环境(侧应力系数、岩石强度应力比)对围岩变形破坏的影响。分析结果表明,数值模拟结果与排水洞开挖应力调整过程中的微震特征具有很好的一致性,能够实现深埋隧洞近断层开挖力学响应规律的准确描述。断层会导致隧洞与断层间形成应力异常区,破坏将不同程度地从初始损伤区向断层迁移。断层的相对位置和角度对隧洞围岩的破坏程度和破坏方式影响显著。侧应力系数增大会导致隧洞破坏加剧,尤其在隧洞顶板和底板位置。岩石强度应力比增大会导致近断层隧洞破坏模式由应力控制型转变为复合控制型。研究成果对类似深埋隧洞变形破坏机制具有参考价值。

Objective To comparatively investigate the inorganic composition and crystallographic properties of cortical and cancellous bone via thermal treatment under 700 ℃. Methods Thermogravimetric measurement, infrared spectrometer, X-ray diffraction, chemical analysis and X-ray photo-electron spectrometer were used to test the physical and chemical properties of cortical and cancellous bone at room temperature 250 ℃, 450 ℃, and 650 ℃, respectively. Results The process of heat treatment induced an extension in the a-lattice parameter and changes of the c-lattice parameter, and an increase in the crystallinity reflecting lattice rearrangement after release of lattice carbonate and possible lattice water. The mineral content in cortical and cancellous bone was 73.2wt% and 71.5wt%, respectively. For cortical bone, the weight loss was 6.7% at the temperature from 60 ℃ to 250℃, 17.4% from 250 ℃ to 450 ℃, and 2.7% from 450 ℃ to 700 ℃. While the weight loss for the cancellous bone was 5.8%, 19.9%, and 2.8 % at each temperature range, the Ca/P ratio of cortical bone was 1.69 which is higher than the 1.67 of stoichiometric HA due to the B-type CO32- substitution in apatite lattice. The CaJP ratio of cancellous bone was lower than 1.67, suggesting the presence of more calcium deficient apatite. Conclusion The collagen fibers of cortical bone were arrayed more orderly than those of cancellous bone, while their mineralized fibers ollkded similar. The minerals in both cortical and cancellous bone are composed of poorly crystallized nano-size apatite crystals with lattice carbonate and possible lattice water. The process of heat treatment induces a change of the lattice parameter, resulting in lattice rearrangement after the release of lattice carbonate and lattice water and causing an increase in crystal size and crystallinity. This finding is helpful for future biomaterial design, preparation and application.

DEAR EDITOR Protein arginine methyltransferases (PRMTs)are involved in many cellular processes via the arginine methylation of histone or non-histone proteins.We examined the expression patterns of prmt4,prmt7,and prmt9 during embryogenesis in Xenopus using whole-mount in situ hybridization and quantitative reverse transcription polymerase chain reaction (RT-PCR).Xenopus prmt4 and prmt7 were expressed in the neural crest,brain,and spinal cord,and also detected in the eye,branchial arches,and heart at the tailbud stage.Specific print9 signals were not detected in Xenopus embryos until the late tailbud stage when weak expression was observed in the branchial arches.Quantitative RT-PCR indicated that the expression of prmt4 and prmt7 was up-regulated during the neurula stage,whereas prmt9 maintained its low expression until the late tailbud stage,consistent with the whole-mount in situ hybridization results.Thus,the developmental expression patterns of these three print genes in Xenopus embryos provide a basis for further functional study of such genes.

A spatial color-mixing model based on tricolor angular frequencies is proposed in consideration that the design theory falls behind the application of digital camouflage pattern.The model is based on Fourier transform and Gaussian low-pass filter(LPF).In the model,the tricolor angular frequencies are introduced to the spatial frequency response function of human color vision,and the effects of atmospheric attenuation and air screen brightness on color mixture are considered.The field test shows that the model can simulate the color-mixing process in the aspects of color-mixing order,and shape and position of color-mixing spot.But the color-mixing spot color is not perfect,which can be improved by optimizing the atmospheric parameters and tricolor cut-off angular frequencies.The model provides a tool for the research on digital camouflage pattern.

中药复方治疗疼痛的用药规律

基于国家知识产权局专利数据库收录的镇痛中药复方为数据来源,整理专利中治疗疼痛的中药复方,探析用药规律,为新型镇痛中药研发提供借鉴。该研究采用IBM SPSS Modeler 18.3、SPSS Statistic 26.0联合对专利数据进行关联规则、聚类分析、复杂网络分析。结果显示,纳入的101首口服方中,使用频次排名前5的中药是甘草、当归、白芍、川芎、丹参;纳入的49首外用方中,使用频次排名前5的药物是没药、乳香、白芷、冰片、川芎。口服方和外用方的药性均以温药为主,且药味均以苦、辛、甘为主。根据中药复杂网络分析,口服方以甘草、当归、白芍、川芎为核心药物,外用方以乳香、没药、甘草、川芎、当归为核心药物。总体而言,口服中药复方多以补气养血、行气活血为主,外用中药复方在口服中药的基础上注重活血化瘀、行气止痛。在未来镇痛药的研发中,可加强安神解郁中药的配伍使用。随着中医药现代化发展,坚持在中医辨证论治思想指导下,基于古法、临床经验方开发新的镇痛中药复方专利,满足当下社会对疼痛治疗的新需求,充分发挥中医药治疗疼痛优势。
