AIM： To study the effect of nicotine on the migration and invasion of human esophageal squamous carcinoma cells and to investigate whether nimesulide can inhibit the effect of ***： The esophageal squamous carcinoma cell line （TE-13） was treated with different concentrations of nicotine （100 μg/mL and 200 μg/mL） or 200 μg/mL nicotine plus 100 μmol/L nimesulide. Cell migration and invasion were measured using migration and invasion chamber systems. COX-2 expression was determined by Western blotting. Matrix metalloproteinase-2 （MMP-2） was analyzed by zymography and ***： Nicotine （100 μg/mL, 200 μg/mL） enhanced TE-13 cells migration and invasion, and increased the protein expression of COX-2 and the activity of MMP-2. Nicotine （200 μ/mL） stimulated TE-13 cells migration and invasion which were partly blocked by nimesulide. This was associated with decreased protein expression of COX-2 and decreased activity and protein expression of MMP-2. CONCLUSION： Nicotine enhances the migration and invasion of the esophageal squamous carcinoma cell line, and nimesulide partly blocks the effect ofnicotine-enhanced esophageal squamous carcinoma cell migration and invasion.

On July 27, 2022, a high-precision system for measuring the magnetic field based on a coherent population trapping magnetometer（referred to as the CPT system） was launched onboard the Space Advanced Technology demonstration satellite（SATech-01） by a ZK-1A rocket. The payload comprises a scalar coherent population trapping magnetometer（CPTM）, triaxial anisotropic magnetoresistance magnetometer（AMRM）, and nano star tracker mounted on a non-magnetic telescopic tubular mast. This configuration enables synchronized measurement of scalar and vector geomagnetic fields, as well as the attitude,making the payload with its compact sensors suitable for applications on miniature satellite platforms. On November 7, 2022,the novel telescopic tubular mast was deployed in orbit, extending to a length of 5.28 m. The CPTM, an absolute scalar magnetometer with an omnidirectional sensor, is the first China-developed quantum/atomic magnetometer successfully operating in space. In orbit, the CPTM has sensitivity of ～10 pT/Hz1/2and can automatically operate under complex magnetic field variations. The difference between the scalar fields obtained using the CPTM and CHAOS model has a mean deviation of–3.73 n T and a standard deviation of 26.11 nT（1σ）, without on-orbit calibration or correction. The miniaturized AMRM has a sensitivity of ～0.3 n T/Hz1/2and maintains stable and reliable operation in orbit. The low-power and miniaturized nano star tracker has a measurement uncertainty of 14.23″（1σ） in orbit. The mission has realized the on-orbit technical verification of the integrated and synchronized detection of the vector magnetic field and attitude and fully validated the system's performance,automation level, and reliability.

AIM:To identify the novel methylation-silenced gene pentraxin 3(PTX3) in esophageal squamous cell carcinoma(ESCC).METHODS:PTX3 mRNA expression was examined in six human ESCC cell lines,one human immortalized normal esophageal epithelial cell line,primary ESCC tumor tissue,and paired adjacent nontumor tissue using reverse transcription polymerase chain reaction(RTPCR).Semi-quantitative immunohistochemistry was used to examine cellular localisation and protein *** specific PCR and bisulphite genomic sequencing were employed to investigate the methylation of the candidate ***:In the majority of ESCC cell lines,we found that PTX3 expression was down-regulated due to gene promoter hypermethylation,which was further confirmed by bisulphite genomic *** treatment with 5-aza-2'-deoxycytidine restored PTX3 mRNA expression in ESCC cell *** was more common in tumor tissues(85%) than in adjacent nontumor tissues(25%)(P < 0.01).CONCLUSION:PTX3 is down-regulated through promoter hypermethylation in ESCC,and could potentially serve as a biomarker of ESCC.

Background:Esophageal cancer is the sixth leading cause of cancer-related death ***-3(PTX3) is a member of the PTX ***,we investigated the role of PTX3 in esophageal squamous cell carcinoma(ESCC).Methods:The effect of PTX3 on ESCC cell proliferation,colony formation,apoptosis,migration,and invasion was investigated using cell viability assays,colony formation assays,flow cytometry,and migration and invasion *** effect of PTX3 on the tumorigenicity of ESCC in vivo was investigated with xenograft studies in nude ***:PTX3 overexpression in ESCC cells reduced cellular proliferation and colony formation(P < 0.05) and increased the rate of apoptosis(P < 0.05).PTX3 expression had no significant effect on the migratory or invasive potential of ESCC *** our mouse model of human ESCC,we achieved 100%successful tumor *** with the control and empty vector-expressing groups,the PTX3-expressing group formed significantly smaller tumors(P < 0.05).Conclusions:This study indicates that PTX3 might play an inhibitory role in ESCC.

AIM： To investigate the expression and methylation status of the secreted frizzled-related protein 2 （SFRP2） in esophageal squamous cell carcinoma （ESCC） and ex- plore its role in ESCC ***： Seven ESCC cell lines （KYSE 30, KYSE150, KYSE410, KYSE510, EC109, EC9706 and TE-1） and one immortalized human esophageal epithelial cell line （Het- 1A）, 20 ESCC tissue samples and 20 paired adjacent non-tumor esophageal epithelial tissues were analyzed in this study. Reverse-transcription polymerase chain reaction （RT-PCR） was employed to investigate the expression of SFRP2 in cell lines, primary ESCC tumor tissue, and paired adjacent normal tissue. Methylation status was evaluated by methylation-specific PCR and bisulfite sequencing. The correlation between expres- sion and promoter methylation of the SFRP2 gene was confirmed with treatment of 5-aza-2＇-deoxycytidine. To assess the potential role of SFRP2 in ESCC, we es-tablished stable SFRP2-transfected cells and examined them with regard to cell proliferation, colony formation, apoptosis and cell cycle in vivo and in ***： SFRP2 mRNA was expressed in the im- mortalized normal esophageal epithelial cell line but not in seven ESCC cell lines. By methylation-specific PCR, complete methylation was detected in three cell lines with silenced SFRP2 expression, and extensive methylation was observed in the other four ESCC cell lines. 5-aza-2＇-deoxycytidine could restore the expres- sion of SFRP2 mRNA in the three ESCC cell lines lack- ing SFRP2 expression. SFRP2 mRNA expression was obviously lower in primary ESCC tissue than in adjacent normal tissue （0.939 ± 0.398 vs 1.51 ± 0.399, P 〈 0.01）. SFRP2 methylation was higher in tumor tissue than in paired normal tissue （95% vs 65%, P 〈 0.05）. The DNA methylation status of the SFRP2 correlated inversely with the SFRP2 expression. Kindlin-2 belongs to a subfamily of FERM domain con-taining proteins,which plays key roles in activating integrin transmembrane receptors and mediating cell *** to conventional FERM domains,kindlin-2 FERM contains an inserted pleckstrin homology(PH)domain that specifically binds to phosphatidy-linositol(3,4,5)trisphosphate(PIP3)and regulates the kindlin-2 *** have determined the crystal struc-ture of kindlin-2 PH domain at 1.9Åresolution,which reveals a conserved PH domain fold with a highly charged and open binding pocket for PIP3 head *** comparison with a previously reported solution structure of kindlin-2 PH domain bound to PIP3 head group reveals that upon PIP3 insertion,there is a significant conformational change of both the highly positively charged loop at the entry of the PIP3 binding pocket and the entireβbarrel of the PH *** propose that such“induced-fit”type change is crucial for the tight binding of PIP3 to anchor kindlin-2 onto the membrane surface,thereby promoting its binding to *** results provide important structural insight into kindlin-2-mediated membrane anchoring and integrin activation.

Objective： Pancreatic fistula （PF） is a common complication after pancreaticoduodenectomy （PD） and there is no consensus regarding the criteria to define PF. The study was undertaken to determine the risk factors for PF according to the definition of the International Study Group on Pancreatic Fistula （ISGPF） and to delineate its impact on patient outcome. Methods： Between March 1994 and May 2009, data from 153 consecutive patients with malignant tumors underwent a PD with pancreaticojejunostomy in the Peking University People＇s Hospital were recorded prospectively A total of 24 factors were examined with univariate analysis and multivariate logistic regression analysis was used to estimate relative risks, and their 95% confidence intervals （95% CI） and odds ratio （OR）. Results： Our institution belonged to medium-volume center and PF occurred in 30 patients （19.6%）. Pancreatic texture, early postoperative hemorrhage and pancreatic pathologies correlated with PF rates significantly in univariate analyses. But in multivariate regression, soft gland （OR, 4.934; 95% CI, 1.132-7.312） and early postoperative hemorrhage with conservative therapy （OR, 4.130; 95% CI, 1.057-21.112） were predictive. The mean postoperative length of stay in patients with PF was longer （32.7±23.9 versus 60.5±56.2 days） than patients without PF （P=0.001）. Overall 30-day mortality was not affected by the development of PF （P=0.657）. There was no difference in reoperation rates between patients with and without PF （10.0% versus 6.5%, P=0.787）. Concerning the sum of postoperative complications, there were 36 complications for 30 patients with PF, while 64 for 123 patients without PF. When patients with distal cholangiocarcinoma, ampullary and duodenal cancer were considered as a whole for survival analysis, the median survival for patients with PF was 20 months, whereas the median survival for patients without PF was 26 months.
