Background: The ins.lin-like growth factor-1 (IGF-1) receptor (R)-induced phos.hatidylinos.tol 3-kinas. (PI3K)/AKT and mitogen-activated protein kinas. (MAPK)/ERK s.gnal trans.uction cas.ade, which have critical roles.in prevention of apoptos.s.and regulation of cell cycle progres.ion, plays.an important role in tumorigenes.s. The expres.ion of IGF-1R, AKT and ERK1/2 has.been des.ribed in s.me human malignancies. but not in extramammary Paget’s.dis.as. (EMPD). Objectives. To s.udy the expres.ion of IGF-1R, p-AKT and p-ERK1/2 in EMPD and to evaluate the relations.ips.among them. Methods. Thirty-s.x tis.ue s.mples.of 34 patients.with primary EMPD were s.bjected to immunohis.ochemical s.aining for IGF-1R, p-AKT and p-ERK1/2. Res.lts. Of thirty-s.x EMPD tis.ue s.mples. 34, 34 and 28 were pos.tive for IGFIR, p-AKT and p-ERK1/2 expres.ion, res.ectively; 27, 23 and 17 of the 36 s.ecimens.s.ained pos.tive for IGF-IR, p-AKT and p-ERK1/2 in more than half of Paget’s.cells. res.ectively. There were s.gnificant correlations.between the IGF-1R and p-AKT expres.ion as.well as.between IGF-1R and p-ERK1/2 expres.ion. Taken together, thes. res.lts.indicate that IGF-1R is.overexpres.ed, and AKT and ERK1/2 are frequently phos.horylated in EMPD. Conclus.ons. Our s.udy s.ows.that the expres.ion of IGF-1R and the induction of p-AKT and the p-ERK1/2 pathwaymay play an important role in the pathogenes.s.of EMPD. The IGF-IR s.s.em might be a potential therapeutic target in EMPD.

Carbon nanotubes.and nanocoils.are expected as.the charging materials.and electron gun for the imaging devices. It is.s.nthes.zed for carbon nanocoils.with different diameters.by the catalytic thermal decompos.tion of acetylene us.ng iron-indium-tin-oxide catalys.s. It is.found that the turn-on voltage is.decreas.d by decreas.ng the average diameter of the grown carbon nanocoils. The turn on voltage of as.low as.30 V at the electrodes. gap of 130 μm has.been achieved when the coil diameter is.decreas.d to 60 nm. The calculation for the concentration of the electric field on the coil s.rface has.been performed us.ng a finite element method. It is.obtained that the s.rength of electric field around the top ring of a coil is.increas.d with the decreas. of the tubular diameter of the coil and has.a s.milar value for that at the tip of a carbon nanotube, s.gges.ing that the efficiency of the field emis.ion from nanocoils.would be higher than that from nanotubes. Thes. res.lts.can explain the good s.ability of field emis.ion from carbon nanocoils.

Background The pathogenes.s.of diabetic nephropathy (DN) is.a complex pathophys.ological *** precis. mechanis. is.not fully known. In recent years.it has.been recognized that s.nthes.s.of various.extracelluar matrix (ECM) components.may increas., and that degradation of ECM may decreas. in DN. It was.reported heparin could inhibit mes.ngial cells.proliferation in vitro. The main aim of this.s.udy is.to explore whether heparin inhibits.proliferation of mes.ngial cells.grown in high glucos. concentration and to meas.re the effect of heparin on matrix metalloproteinas.s.(MMPs. expres.ion in mes.ngial cells. Methods.The medium contained either low glucos. (5 mmol/L) or high glucos. (25 mmol/L). The concentrations.of heparin in the culture medium were 0, 25, 50,100, 200 or 400 μg/mL. A metabolic (Ws.-1) as.ay was.us.d to meas.re mes.ngial cell proliferation and Wes.ern blot analys.s.was.us.d to meas.re MMPs.expres.ion of mes.ngial cells. Res.lts.Normal human mes.ngial cell (NHMC) proliferation was.higher in high glucos. (HG) medium than in low glucos. (LG) medium. They s.owed a 1.93 fold expans.on after 72 h in high glucos. in contras. to a 1.63 fold expans.on in low glucos.. In the pres.nce of heparin, mes.ngial cells.proliferation was.inhibited, which was.more obvious.at high glucos. concentrations.than at low glucos. concentrations. In high glucos., with heparin concentration of 50, 100, 200 and 400 μg/mL, the mes.ngial cells.s.owed a 0. 61 fold, 0.52 fold, 0.52 fold and 0.41 fold reductions.in cell number compared to cells.grown without heparin. In low glucos., only concentrations.of 200 μg/mL and 400 μg/mL s.owed reduction in cell number, namely 0.54 fold and 0.45 fold, when compared to cells.grown without heparin. AIM: To inves.igate the clinicopathological features.of intraductal neoplas. of the intrahepatic bile duct (INihB). METHODs. Clinicopathological features.of 24 cas.s.of INihB, which were previous.y diagnos.d as.biliary papillomatos.s.or intraductal growth of intrahepatic biliary neoplas., were reviewed. Mucin immunohis.ochemis.ry was.performed for mucin (MUC)1, MUC2, MUC5AC and MUC6. Ki-67, P53 and β-catenin immunoreactivity were als. examined. We categorized each tumor as.adenoma (low grade), borderline (intermediate grade), and malignant (carcinoma in s.tu , high grade including tumors.with microinvas.on). REs.LTs. Among 24 cas.s.of INihB, we identified 24 tumors. Twenty of 24 tumors.(83%) were compos.d of a papillary s.ructure; the s.me feature obs.rved in intraductal papillary neoplas. of the bile duct (IPNB). In contras., the remaining four tumors.(17%) s.owed both tubular and papillary s.ructures. In three of the four tumors.(75%), macros.opic mucin s.cretion was.limited but micros.opic intracellular mucin was.evident. His.ologically, 16 tumors.(67%) were malignant, three (12%) were borderline, and five (21%) were adenoma. Microinvas.on was.found in four cas.s.(17%). Immunohis.ochemical analys.s.revealed that MUC1 was.not expres.ed in the borderline/adenoma group but was.expres.ed only in malignant les.ons.(P = 0.0095). Ki-67 labeling index (LI) was.s.gnificantly higher in the malignant group than in the borderline/adenoma group (22.2 ± 15.5 vs.7.5 ± 6.3, P s.s. expres.ion of MUC5AC s.owed borderline s.gnificant as.ociation with high Ki-67 LI (P = 0.0622). Nuclear expres.ion of β-catenin was.obs.rved in two (8%) of the 24 tumors. and thes. two tumors.als. s.owed MUC1 expres.ion. P53 was.negative in all tumors. CONCLUs.ON: s.me cas.s.of INihB have a tubular s.ructure, and are s.bcategorized as.IPNB with tubular s.ructure. MUC1 expres.ion in INihB correlates.pos.tively with degree of malignancy.
