To understand better the pathogenesis of duck virus enteritis (DVE),the levels of viral DNA in various tissues of duckling during acute stage of virulent duck enteritis virus (DEV) infection were investigated by using quantitative real-time *** results show that the viral levels of DEV in systemic organs have a close correlation with the progression of *** rapid dissemination of virulent DEV to systemic organs and active replication in lymphoid tissues at the early phase of acute infection accelerate the progression of *** levels of viral DNA increased sharply soon after develop clinical signs of disease,and the extent of increase and the final magnitude of DEV DNA load in various tissues of ducklings immediately following the exhibition of clinical signs may be a critical determinant of the outcome of DEV *** relatively high level of DEV infection and replication in bursa and intestine tissues of dead ducklings most likely reflects the abundance of target epithelial and lymphoid cells in these tissues,and therefore play a key role in the pathogenesis of acute DVE manifested as severe lesions on the lymphoid tissues and small intestine.

Duck virus enteritis (DVE) is an acute and contagious herpesvirus infection of duck,geese and swans with high morbidity and *** kinetics of viral levels and immunohistochemical localization of virulent duck enteritis virus (DEV),as well as histopathological examination in various tissues of ducks following oral infection were *** time course for the appearance of viral antigen and tissue lesions in various tissues was coincident with the viral levels of DEV in sites,suggesting that the viral levels of DEV in systemic organs have a close correlation with the progression of *** abundance of target epithelial and lymphoid cells may contribute to the high levels of virus infection and replication in lymphoid and intestinal tissues.

To understand better attenuated DEV vaccine induced immune mechanisms,the levels of viral DNA in various duckling’s tissues during different immune stage of attenuated DEV vaccine by the mucosal or systemic route were investigated by using quantitative TaqMan MGB real-time *** the immune organs examined were positive for DEV DNA at 60 min post-immunization (p.i.) and contained the viral DNA levels varied from 7.60±0.51 to 3.88±*** kinetics of DEV DNA loads in immune organs were markedly different among various immune groups,and subcutaneously immunized DVE vaccine distributed and replicated relatively *** samples examined in orally,nasally and subcutaneously immunized ducklings were positive for DEV DNA at 60 min,90 min and 12 h p.i., *** immunized DVE vaccine distributed and replicated relatively rapidly in digestive and respiratory tract tissues and *** all the immune ducklings,The DVE vaccine loads in the heart, brain,duodenum and cecum were significantly higher than other samples examined (P<0.05).However, the DVE vaccine concentrations in most of samples examined decreased at various rates after peaked successively during 60 min to 6 days *** the end of the study,the most of samples examined atill contained a moderate attenuated DEV loads ranging from 5.59±0.24 to 2.64±*** together,these results suggest that the rapid dissemination and replication of DVE vaccine during the early stage of vaccination in multiple organs and tissues,especially in systemic immune organs and mucosal-associated tissues,plays a key role in immediate induction of DEV specific systemic and mucosal immunity in immunized *** initial sites of DVE vaccine distribution and replication have a good correlation with the immune routes and anatomic structure on duck.A continuous low-level of DVE vaccine replication persisted in multiple samples examined will contribute to the continual induction of host immune system to produce DEV sp

Duck immunoglobulin A (IgA) at electrophoresis scale was achieved by polyethylene glycol (PEG) extraction followed by Micro-Prep S cellulose ion-exchange chromatography,and monospecific antiserum was prepared in rabbits and purified anti-duck IgA rabbit IgG was labeled with Horseradish Peroxidase (HRP) for measurement of IgA antibodies in intestinal secretions and serum of *** kinetics of viral level,pathology,humoral immune responses,and localization of IgA antibody-secreting cell in intestine and lymphoid organs of ducklings were evaluated after orally infected ducklings with virulent duck enteritis virus (DEV).The levels of DEV-specific IgA in intestinal secretions appeared to be reciprocal to the kinetics of DEV load and the severity of pathological *** results suggest that oral infection of virulent DEV in ducklings induces both mucosal and system humoral immune responses, and specific IgA antibodies may have a role in controlling DEV infection in the *** severe lesions of villus epithelial cells may inhibit humoral immune responses on the intestinal mucosal surface and cause a increased level of DEV in ***,IgA-producing B cells were observed in the intestine,but also in the bursa as well as the *** results may imply that the exposure of the intestinal mucosa to DEV can lead to IgA production in the local as well as distant mucosal surface.
