Dose-individualization Efficiently Maintains Sufficient Exposure to Methotrexate without Additional Toxicity in High-dose Methotrexate Regimens for Pediatric Acute Lymphoblastic Leukemia

Objective:Methotrexate(MTX)can be safely administered to most patients but may cause severe toxicity in *** study aimed to summarize the characteristics of high-dose methotrexate(HD-MTX)chemotherapy and to evaluate whether the modified dose-adjustment program was able to improve the maintenance of sufficient MTX exposure levels while minimizing ***:We evaluated 1172 cycles of high-dose MTX chemotherapy from 294 patients who were treated according to the CCCG-ALL-2015 protocol(clinical trial number:ChiCTR-IPR-14005706)and analyzed the data of actual MTX dosage,MTX concentration,toxicity,and *** compared data between the dose-adjustment Program 1(fixed 20%reduction in dose)and the dose-adjustment Program 2(dose-individualization based on reassessment of the creatine clearance rate and the MTX concentration-monitoring point at 16 h),which were applied if the MTX clearance was delayed in the previous ***:The patients who used Program 2 had higher actual MTX infusion doses and infusion rates and were able to better maintain the MTX concentration at 44 h at the established target value than those on Program 1(P<0.001).No significant differences in toxicities were found between these two programs except that abnormal serum potassium levels and prolonged myelosuppression in intermediate-risk/high-risk patients were more frequently observed in patients using Program 2(P<0.001).No significant correlations were observed between the MTX dose,dose-adjustment programs,or MTX concentrations and relapse-free ***:Adjusting the MTX dose using Program 2 is more efficient for maintaining sufficient MTX exposure without significantly increasing the toxicity.

A New Immunosuppressive Therapy for Very Severe Aplastic Anemia in Children with Autoantibodies

Huai Qi Huang-induced Apoptosis via Down-regulating PRKCH and Inhibiting RAF/MEK/ERK Pathway in Ph+ Leukemia Cells

Imatinib mesylate(IM)is the first-line treatment for Philadelphia(Ph)chromosomal positive leukemia by inhibiting phosphorylation of substrates via binding to the ABL kinase *** of the drug resistance,side effects and the high cost of IM,it is necessary to find anti-cancer drugs with relatively low toxicity and cost,and enhanced efficacy,such as traditional Chinese medicines(TCMs).As one of TCMs,Huai Qi Huang(HQH)was chosen to treat BV173 and K562 *** concentrations of HQH were added to cells for 24-72 ***-treatment of HQH and trametinib,an MEK inhibitor,was used to verify the synergistic effects on cell viability and *** and overexpression of mitogen-activated protein kinase kinase 4(MEK4)were implemented to demonstrate the role of MEK in cell *** viability and apoptosis were measured by cell counting kit-8 assay(CCK8)and flow cytometry,*** blotting and real-time quantitative PCR(RT-qPCR)were used to assess protein and mRNA expression levels,*** results showed that HQH inhibited survival and promoted apoptosis of BV173 and K562 cells in a dose-dependent manner,accompanied with down-regulation of PRKCH mRNA as well as CRAF,MEK4,phospho-ERK(pERK)and BCL2 proteins,and up-regulation of cleaved caspase3 ***-treatment of HQH and trametinib had.a synergistic effect on inhibiting survival and promoting ***4 knockdown increased apoptosis,and had a synergistic effeet with *** contrast,MEK4 overexpression ***,and had the opposite efect with ***,the results of this study may identify a therapeutic mechanism of HQH on promoting apoptosis,and provide a potential option for treatment of Pht leukemia.

Mixed-charge glycopolypeptides as antibacterial coatings with long-term activity

Planktonic bacteria adhere and subsequently form biofilms on implantable medical devices can cause severe infections that have become the major types of hospital-acquired infections. Traditional coatings for the implants are frequently lack of long-term antifouling and bactericidal activities. It is still a big challenge to simultaneously improve the antifouling and bactericidal activities of the coatings. Herein,we report that mixed-charge glycopolypeptide coatings are of long-term antibacterial activities to efficiently inhibit the biofilm growth. The glycosylation of mixed-charge polypeptides has led to a significant improvement of both antifouling and bactericidal activities. The cooperative effect of the saccharide residues and mixed-charge residues improved the resistance of the polypeptide coatings against protein adsorption. The saccharide and L-glutamic acid(E) residues collectively enhanced the bacterial membrane-disruption of cationic L-lysine(K) residues, leading to potent bactericidal activity. Meanwhile,the glycopolypeptide coatings showed superior biocompatibility, long-term antibiofilm and anti-infection properties in two types of mouse subcutaneous infection models and one type of mouse urinary tract infection model. This work provides a new strategy to achieve antibacterial coatings with long-term activities for preventing implantable medical device associated infections.

The textured roll and polished roll were applied instead of the ground roll in a 20-high mill to conduct two-pass rolling of 316L stainless steel strip with thickness of 0.027 *** the two-pass rolling with the textured roll and polished roll(TPR),the surface roughness of the strip is dramatically reduced,and the surface topographical anisotropy index is diminished to 30.9%of the initial *** with the strip rolled using the ground roll in both passes(GGR),the elongation of TPR rolled strip is obviously improved,and the mechanical property anisotropy is greatly *** anisotropy index of tensile strength and elongation are 42.58%and 52.59%of that of GGR rolled strip,which is mainly attributed to the significant decrease of the texture intensity of the strip by TPR *** results indicate that TPR process can obtain the stainless steel ultra-thin strip with smooth and uniform surface topography and good mechanical properties.

This article summarizes the epidemiological characteristics and clinical manifest-ations of nonalcoholic fatty liver disease(NAFLD).The incidence of NAFLD has been increased dramatically and become the leading cause of chronic liver disease *** addition to its adverse outcomes of liver fibrosis,cirrhosis,and hepatocellular carcinoma,and related complications,NAFLD has recently been found to be associated with the high-risk extrahepatic carcinomas,such as various types of lung cancer(i.e.,lung adenocarcinoma,squamous cell carcinoma,and small cell lung cancer).The presence of hepatic steatosis also predisposes lung cancer to liver metastasis,but has better response to immune checkpoint *** other factors(i.e.,gender,smoking,etc.)are associated with NAFLD and lung cancer remains *** also comment on the reciprocal rela-tionships between NAFLD and components of metabolic *** meta-bolic syndrome components are suggested to facilitate lung cancer development via activating insulin/insulin-like growth factor *** addition,suppressed anti-tumor immunity and accelerated tumor progression could be attributed to the cell-specific metabolic reprogramming in condition of high-fat diet and related *** findings may reveal the role of NAFLD in pulmonary carcinoma and help develop new treatment strategies for this disease.

As a highly promising conductive polymer material,the synthesis method,structure regulation,and performance improvement of polyaniline(PANI)are hot research *** this work,the radiation-induced polymerization of aniline in HNO_(3)solution was successfully achieved at room temperature without the use of chemical *** the analysis of the radiation chemical reactions of inorganic acids and nitrate salt solutions,the characterization of the intermediate free radicals in the irradiated systems,and the influence of the pH of the solutions on the polymerization activity and product morphologies,the radiation-induced polymerization mechanism of aniline is discussed in detail and *** at a condition of[HNO_(3)]>[aniline],i.e.,pH<2.5,PANI can be successfully obtained underγ-ray *** polymerization begins with the oxidation of aniline cations to aniline cation radicals by·NO_(3)generated by radiolysis reactions,and undergoes repeated three steps of monomer free radical recombination,deprotonation,and oxidation reaction of·NO_(3),thus forming a PANI *** addition to the polymerization reaction,the aniline units are protonated and oxidized because of the strongly acidity and oxidation of the reaction system under γ-ray irradiation,which means that the molecular chain structure of the radiation-synthesized PANI can be regulated by pH,nitrate concentration,and irradiation ***-synthesized PANI has a moderate protonation and oxidation state,which can be used for the preparation of PANI supercapacitors with better electrochemical properties than those prepared by chemical oxidation under the same *** work presents a new radiation-synthesis method and polymerization mechanism of PANI,which not only expands the application of radiation technique in the field of polymer synthesis,but also provides a new idea for the structural regulation and electrochemical property optimization of PANI.
