KISS-1 inhibits the proliferation and invasion of gastric carcinoma cells

AIM:To investigate the function of the KISS-1 gene in gastric carcinoma cells and to explore its potential mechanism.METHODS:A KISS-1 eukaryotic expression vector was constructed and transfected into BGC-823 cells.Resistant clones were obtained through G418 selection.reverse transcription-polymerase chain reaction and western blotting were used to detect KISS-1 and matrix metalloproteinase-9(MMP-9)expression in transfected cells.The growth of transfected cells was investigated by 3-(4,5-dimethylthiazolyl-2)-2,5-diphenyltetrazolium bromide(MTT)proliferation assays,and the cells'invasive potential was analyzed by basement membrane(Matrigel)invasion assays.The anti-tumor effects of KISS-1 were tested in vivo using allografts in nude mice.RESULTS:The expression level of KISS-1 mRNA andprotein in BGC-823/KISS-1 transfected cells were significantly higher than in BGC-823/pcDNA3.1 transfected cells(P<0.05)or the parental BGC-823 cell line(P< 0.05).The expression level of MMP-9 mRNA and protein in BGC-823/KISS-1 were significantly less than in BGC-823/pcDNA3.1(P<0.05)or BGC-823 cells(P< 0.05).MTT growth assays show the proliferation of BGC-823/KISS-1 cells at 48 h(0.642±0.130)and 72 h(0.530±0.164)were significantly reduced compared to BGC-823/pcDNA3.1(0.750±0.163,0.645±0.140)(P<0.05)and BGC-823 cells(0.782±0.137,0.685± 0.111)(P<0.05).Invasion assays indicate the invasive potential of BGC-823/KISS-1 cells(16.50±14.88)is significantly reduced compared to BGC-823/pcDNA3.1(20.22±14.87)(P<0.05)and BGC-823 cells after 24 h(22.12±16.12)(P<0.05).In vivo studies demonstrate the rate of pcDNA3.1-KISS-1 tumor growth is significantly slower than pcDNA3.1 and control cell tumor growth in nude mice.Furthermore,tumor volume of pcDNA3.1-KISS-1 tumors(939.38±82.08 mm3)was significantly less than pcDNA3.1(1250.46±44.36 mm3) and control tumors(1284.36±55.26 mm3)(P<0.05).Moreover,the tumor mass of pcDNA3.1-KISS-1 tumors(0.494±0.84 g)was significantly less than pcDNA3.1(0.668±0.55 g)and control tumors(0.682±0.38 g)(P <0.05).CONCLUSION:KISS-1

A statistical distribution texton feature for synthetic aperture radar image classification

We propose a novel statistical distribution texton(s-texton) feature for synthetic aperture radar(SAR) image classification. Motivated by the traditional texton feature, the framework of texture analysis, and the importance of statistical distribution in SAR images, the s-texton feature is developed based on the idea that parameter estimation of the statistical distribution can replace the filtering operation in the traditional texture analysis of SAR images. In the process of extracting the s-texton feature, several strategies are adopted, including pre-processing, spatial gridding, parameter estimation, texton clustering, and histogram statistics. Experimental results on Terra SAR data demonstrate the effectiveness of the proposed s-texton feature.

Alzheimer’s disease is the most prevalent neurodegenerative disease affecting older adults.Primary features of Alzheimer’s disease include extra cellular aggregation of amyloid-βplaques and the accumulation of neurofibrillary tangles,fo rmed by tau protein,in the cells.While there are amyloid-β-ta rgeting therapies for the treatment of Alzheimer’s disease,these therapies are costly and exhibit potential negative side effects.Mounting evidence suggests significant involvement of tau protein in Alzheimer’s disease-related neurodegeneration.As an important microtubule-associated protein,tau plays an important role in maintaining the stability of neuronal microtubules and promoting axonal growth.In fact,clinical studies have shown that abnormal phosphorylation of tau protein occurs before accumulation of amyloid-βin the brain.Various therapeutic strategies targeting tau protein have begun to emerge,and are considered possible methods to prevent and treat Alzheimer’s disease.Specifically,abnormalities in post-translational modifications of the tau protein,including aberrant phosphorylation,ubiquitination,small ubiquitin-like modifier(SUMO)ylation,acetylation,and truncation,contribute to its microtubule dissociation,misfolding,and subcellular missorting.This causes mitochondrial damage,synaptic impairments,gliosis,and neuroinflammation,eventually leading to neurodegeneration and cognitive deficits.This review summarizes the recent findings on the underlying mechanisms of tau protein in the onset and progression of Alzheimer’s disease and discusses tau-targeted treatment of Alzheimer’s disease.

Encouraging Solution to the Problem of Synthesizing Protein α-Thioester

Expressed protein ligation(EPL)provides a powerful tool to access large-size proteins with precise structures.Existing methods for constructing the critical protein thioester for EPL have predominantly relied on the recombinant intein fusion expressed in Escherichia coli(E.coli).Despite its powerful applications,the expression of thioester derived from eukaryotic protein in E.coli inherently suffers from its limited solubility,the inactivity of intein,premature hydrolysis and low yields.To overcome these obstacles,we present herein the facile one-flask synthesis of inaccessible proteinα-thioester via a SUMO-protein-intein(SPI)sandwich model.The utility of SUMO enhances the protein fusion yield and solubility,prevents premature hydrolysis and simplifies the purification process.The inaccessible protein thioester with internal Cys residues can be readily produced and is compatible with the EPL-desulfurization protocol used to prepare complex proteins,which is otherwise difficult to obtain using traditional methods.Its utility has been highlighted through the synthesis of human granulocyte colony-stimulating factor(G-CSF).

Xiao Tan San Jie Fang Hampers the Growth of Colon Cancer Stem Cells through the Wnt/β-catenin Signaling Pathway

Objective: The purpose of this research was to examine the potential anticancer properties of Xiao Tan San Jie Fang(XTSJF) and its potential mechanism of action against colorectal cancer. Materials and Methods: HCT116 cells were induced into HCT116 spheres in DMEM/F12 medium by treatment with epidermal growth factor + fibroblast growth factor + leukemia inhibitory factor + B27. The proliferation ability and stemness of HCT116 spheres was examined. Various concentrations of XTSJF were used to treat HCT116 spheres to observe the impact on proliferation, apoptosis, and expression of stem cell markers. Next, Wnt/β-catenin pathway-related factor proteins were detected. Results: The findings revealed that XTSJF suppressed cell growth and induced cell death in HCT116 cells in a dosage-dependent manner. Similarly, XTSJF promotes apoptosis, inhibits cell proliferation, prolongs survival, and maintains the expression of stem cells through the Wnt/-catenin/TCF4 axis. XTSJF also inhibits AKT activity and subsequently activates glycogen synthesis kinase-3β expression, inhibiting Wnt/beta-catenin pathway activity and downstream target gene transcript expression. The Wnt/β-catenin signaling pathway is inhibited by the XTSJF, leading to the suppression of colon cancer stem cell proliferation. Conclusion: Xiaotan Sanjie Prescription inhibited colon cancer stem cell growth through Wnt/β-catenin signaling pathway.

Spiking neural network(SNN),widely known as the third-generation neural network,has been frequently investigated due to its excellent spatiotemporal information processing capability,high biological plausibility,and low energy consumption characteristics.Analogous to the working mechanism of human brain,the SNN system transmits information through the spiking action of neurons.Therefore,artificial neurons are critical building blocks for constructing SNN in hardware.Memristors are drawing growing attention due to low consumption,high speed,and nonlinearity characteristics,which are recently introduced to mimic the functions of biological neurons.Researchers have proposed multifarious memristive materials including organic materials,inorganic materials,or even two-dimensional materials.Taking advantage of the unique electrical behavior of these materials,several neuron models are successfully implemented,such as Hodgkin–Huxley model,leaky integrate-and-fire model and integrate-and-fire model.In this review,the recent reports of artificial neurons based on memristive devices are discussed.In addition,we highlight the models and applications through combining artificial neuronal devices with sensors or other electronic devices.Finally,the future challenges and outlooks of memristor-based artificial neurons are discussed,and the development of hardware implementation of brain-like intelligence system based on SNN is also prospected.

Clinical study of different prediction models in predicting diabetic nephropathy in patients with type 2 diabetes mellitus

BACKGROUND Among older adults,type 2 diabetes mellitus(T2DM)is widely recognized as one of the most prevalent diseases.Diabetic nephropathy(DN)is a frequent com-plication of DM,mainly characterized by renal microvascular damage.Early detection,aggressive prevention,and cure of DN are key to improving prognosis.Establishing a diagnostic and predictive model for DN is crucial in auxiliary diagnosis.AIM To investigate the factors that impact T2DM complicated with DN and utilize this information to develop a predictive model.METHODS The clinical data of 210 patients diagnosed with T2DM and admitted to the First People’s Hospital of Wenling between August 2019 and August 2022 were retrospectively analyzed.According to whether the patients had DN,they were divided into the DN group(complicated with DN)and the non-DN group(without DN).Multivariate logistic regression analysis was used to explore factors affecting DN in patients with T2DM.The data were randomly split into a training set(n=147)and a test set(n=63)in a 7:3 ratio using a random function.The training set was used to construct the nomogram,decision tree,and random forest models,and the test set was used to evaluate the prediction performance of the model by comparing the sensitivity,specificity,accuracy,recall,precision,and area under the receiver operating characteristic curve.RESULTS Among the 210 patients with T2DM,74(35.34%)had DN.The validation dataset showed that the accuracies of the nomogram,decision tree,and random forest models in predicting DN in patients with T2DM were 0.746,0.714,and 0.730,respectively.The sensitivities were 0.710,0.710,and 0.806,respectively;the specificities were 0.844,0.875,and 0.844,respectively;the area under the receiver operating characteristic curve(AUC)of the patients were 0.811,0.735,and 0.850,respectively.The Delong test results revealed that the AUC values of the decision tree model were lower than those of the random forest and nomogram models(P<0.05),whereas the difference in AUC values of the random forest and colu

Optical Transient Source AT2021lfa:A Possible“Dirty Fireball”

AT2021lfa,also known as ZTF21aayokph,was detected by the Zwicky Transient Facility on 2021 May 4,a 05:34:48 UTC.Follow-up observations were conducted using a range of ground-based optical telescopes,as wel as Swift/XRT and VLA instruments.AT2021lfa is classified as an“orphan afterglow”candidate due to its rapid flux decline and its reddened color(g-r=0.17±0.14 mag).For an optical transient source without promp gamma-ray detection,one key point is to determine its burst time.Here we measure the burst time through fitting the initial bump feature of AT2021lfa and obtain its burst time as 2021 May 3,at 22:09:50 UTC.Using afterglowpy,we model the multi-band afterglow of AT2021lfa and find that the standard model canno reproduce the late radio observations well.Considering that the microphysical parameters ε_(e),ε_(B)(the energy fraction given to electrons and magnetic field),andξN(the fraction of accelerated electrons)may vary with time we then model the afterglow of AT2021lfa taking into account the temporal evolution of the physical parameters ε_(e),ε^(B),and ξ_(N) and find in this case the multi-wavelength observations can be reproduced well.The initial Lorentz factor of AT2021lfa can be estimated from the peak time of the early afterglow,which yields a value of about 18 suggesting that AT2021lfa should be classified as a“dirty fireball.”From the upper limit for the prompt emission energy of AT2021lfa,we obtain that the radiation efficiency is less than 0.02%,which is much smaller than that of ordinary gamma-ray bursts(GRBs).It is also interesting that the fitted values of jet angle and viewing angle are very large,θ_(c)~0.66 rad,θ_(v)~0.53 rad,which may lead to the low Lorentz factor and radiation efficiency.When compared with GRB afterglow samples,it is evident that the onset bump timescale of AT2021lfa satisfies the empirical relationships observed in GRB samples.Additionally,the luminosity of AT2021lfa falls within the range of observations for GRB samples;however,approximately 1 day after the burst,its lumino

Cultivating the uncultured:Harnessing the“sandwich agar plate”approach to isolate heme-dependent bacteria from marine sediment

In the classical microbial isolation technique,the isolation process inevitably destroys all microbial interactions and thus makes it difficult to culture the many microorganisms that rely on these interactions for survival.In this study,we designed a simple coculture technique named the“sandwich agar plate method,”which maintains microbial interactions throughout the isolation and pure culture processes.The total yield of uncultured species in sandwich agar plates based on eight helper strains was almost 10-fold that of the control group.Many uncultured species displayed commensal lifestyles.Further study found that heme was the growth-promoting factor of some marine commensal bacteria.Subsequent genomic analysis revealed that heme auxotrophies were common in various biotopes and prevalent in many uncultured microbial taxa.Moreover,our study supported that the survival strategies of heme auxotrophy in different habitats varied considerably.These findings highlight that cocultivation based on the“sandwich agar plate method”could be developed and used to isolate more uncultured bacteria.

Influence of sea-land breeze on the formation and dissipation of severe dense fog and its burst reinforcement in the yellow Sea coastal area,China

Based on the global reanalysis data of the National Centers for Environmental Prediction(NCEP)/National Center for Atmospheric Research,the surface meteorological observation data,sounding data and satellite observation data,this paper comprehensively analyzes the evolution process and formation mechanism of a persistent severe dense fog process occurred on February 15–17,2015 in yancheng,eastern China.Through the numerical simulation experiment of Weather Research and Forecast(WRF)model,we further analyze the impact of sea-land breeze on the formation and burst reinforcement of fog.Results show that the precipitation caused by the southwesterly airflow in front of the upper-level trough and the low-pressure inverted trough are conducive to the formation of early rain fog,while the nighttime clear radiance under the control of surface cold high and the infiltration of weak cold advection are conducive to the formation and development of later radiation-advection fog.The WRF model simulates the fog evolution process,which is basically consistent with the actual fog area,and the simulation results are credible to a certain extent.The simulation results show that the establishment of sea breeze has an advection cooling effect on the near surface layer,which is conducive to the formation and development of the inversion layer on the near surface,providing stable stratification conditions for the formation and burst reinforcement of fog.On one hand,the strengthening of sea breeze circulation can continuously transport water vapor to the study area.On the other,the occurrence of ultra-low level jet is favorable for the accumulation of low-level water vapor.At the same time,the inversion intensity further strengthens,which is in favor of the burst reinforcement and long-term maintenance of fog.
