Quantitative proteomic analysis for high-throughput screening of differential glycoproteins in hepatocellular carcinoma serum

Objective： Hepatocellular carcinoma （HCC） is a leading cause of cancer-related deaths. Novel serum biomarkers are required to increase the sensitivity and specificity of serum screening for early HCC diagnosis. This study employed a quantitative proteomic strategy to analyze the differential expression of serum glycoproteins between HCC and normal control serum samples. Methods： Lectin affinity chromatography （LAC） was used to enrich glycoproteins from the serum samples. Quantitative mass spectrometric analysis combined with stable isotope dimethyl labeling and 2D liquid chromatography （LC） separations were performed to examine the differential levels of the detected proteins between HCC and control serum samples. Western blot was used to analyze the differential expression levels of the three serum proteins. Results： A total of 2,280 protein groups were identified in the serum samples from HCC patients by using the 2D LC-MS/MS method. Up to 36 proteins were up-regulated in the HCC serum, whereas 19 proteins were down-regulated. Three differential glycoproteins, namely, fibrinogen gamma chain （FGG）, FOS-like antigen 2 （FOSL2）, and a-l, 6-mannosylglycoprotein 6-^-N-acetylglucosaminyltransferase B （MGATSB） were validated by Western blot. All these three proteins were up-regulated in the HCC serum samples. Conclusion： A quantitative glycoproteomic method was established and proven useful to determine potential novel biomarkers for HCC.

Atmospheric carbon dioxide （CO2） is assimilated by the most abundant but sluggish enzyme, ribulose- 1,5-bisphosphate carboxylase/oxygenase （Rubisco）. Here we show that acetylation of lysine residues of the Rubisco large subunit （RbcL）, including Lys201 and Lys334 in the active sites, may be an important mechanism in the regulation of Rubisco activities. It is well known that Lys201 reacts with CO2 for carba- mylation, a prerequisite for both carboxylase and oxygenase activities of Rubisco, and Lys334 contacts with ribulose-1,5-bisphosphate （RuBP）. The acetylation level of RbcL in plants is lower during the day and higher at night, inversely correlating with the Rubisco carboxylation activity. A search of the chloroplast proteome database did not reveal a canonical acetyltransferase; instead, we found that a plant-derived metabolite, 7-acetoxy-4-methylcoumarin （AMC）, can non-enzymatically acetylate both native Rubisco and synthesized RbcL peptides spanning Lys334 or Lys201. Furthermore, lysine residues were modified by synthesized 4-methylumbelliferone esters with different electro- and stereo-substitutes, resulting in varied Rubisco activities. 1-Chloroethyl 4-methylcoumarin-7-yl carbonate （CIMC） could transfer the chloroethyl carbamate group to lysine residues of RbcL and completely inactivate Rubisco, whereas bis（4-methylcoumarin-7-yl） carbonate （BMC） improved Rubisco activity through increasing the level of Lys201 carbamylation. Our findings indicate that RbcL acetylation negatively regulates Rubisco activity, and metabolic derivatives can be designed to dissect and improve CO2 fixation efficiency of plants through lysine modification.

Human brain development is a complex process,and animal models often have significant *** address this,researchers have developed pluripotent stem cell-derived three-dimensional structures,known as brain-like organoids,to more accurately model early human brain development and *** enable more consistent and intuitive reproduction of early brain development,in this study,we incorporated forebrain organoid culture technology into the traditional unguided method of brain organoid *** involved embedding organoids in matrigel for only 7 days during the rapid expansion phase of the neural epithelium and then removing them from the matrigel for further cultivation,resulting in a new type of human brain organoid *** cerebral organoid system replicated the temporospatial characteristics of early human brain development,including neuroepithelium derivation,neural progenitor cell production and maintenance,neuron differentiation and migration,and cortical layer patterning and formation,providing more consistent and reproducible organoids for developmental modeling and toxicology *** a proof of concept,we applied the heavy metal cadmium to this newly improved organoid system to test whether it could be used to evaluate the neurotoxicity of environmental *** organoids exposed to cadmium for 7 or 14 days manifested severe damage and abnormalities in their neurodevelopmental patterns,including bursts of cortical cell death and premature *** exposure caused progressive depletion of neural progenitor cells and loss of organoid integrity,accompanied by compensatory cell proliferation at ectopic *** convenience,flexibility,and controllability of this newly developed organoid platform make it a powerful and affordable alternative to animal models for use in neurodevelopmental,neurological,and neurotoxicological studies.

Photocatalytic Cr(Ⅵ)reduction over MIL-88A(Fe)on polyurethane sponge: From batch to continuous-flow operation

MIL-88A(Fe)@sponge(MS) was synthesized by a dip-coating method, which displayed efficient photocatalytic Cr(Ⅵ) reduction efficiency under both low power LED UV light and real solar light irradiation. It was observed that MS(0.2 g/L) could remove 100% Cr(Ⅵ)(10 mg/L) by adding 0.4 mmol/L tartaric acid(TA) without adjusting pH(pH 5.05) within 6.0 min and 3.0 min under UV light and real solar light irradiation, respectively. Besides, the photo-induced e-and radicals(O_(2)^(·-) and CO_(2)^(·-)) were found to play the momentous roles in the MS/TA/UVL/Cr(Ⅵ) system by the scavenger experiments and electron spin resonance(ESR) tests. MS was also filled into a fixed-bed reactor to test the possibility of long-term Cr(Ⅵ)reduction operation in TA/UVL system. As expected, the results revealed that MS could still maintain 100% activity up to 60 h. These results demonstrated that MIL-88A(Fe) might be the potentially efficient catalyst for large-scale wastewater treatment in the near future.

Gradient magnetic heterointerfaces have injected infinite vitality in optimizing impedance matching,adjusting dielectric/magnetic resonance and promoting electromagnetic(EM)wave absorption,but still exist a significant challenging in regulating local phase ***,accordion-shaped Co/Co_(3)O_(4)@N-doped carbon nanosheets(Co/Co_(3)O_(4)@NC)with gradient magnetic heterointerfaces have been fabricated via the cooperative high-temperature carbonization and lowtemperature oxidation *** results indicate that the surface epitaxial growth of crystal Co_(3)O_(4) domains on local Co nanoparticles realizes the adjustment of magnetic-heteroatomic components,which are beneficial for optimizing impedance matching and interfacial ***,gradient magnetic heterointerfaces simultaneously realize magnetic coupling,and long-range magnetic ***,the synthesized Co/Co_(3)O_(4)@NC absorbents display the strong electromagnetic wave attenuation capability of−53.5 dB at a thickness of 3.0 mm with an effective absorption bandwidth of 5.36 GHz,both are superior to those of single magnetic domains embedded in carbon *** design concept provides us an inspiration in optimizing interfacial polarization,regulating magnetic coupling and promoting electromagnetic wave absorption.

Non-monotonous Sequential Subgradient Projection Algorithm for Convex Feasibility Problem

The existing methods of projection for solving convex feasibility problem may lead to slow conver- gence when the sequences enter some narrow＂corridor＂ between two or more convex sets. In this paper, we apply a technique that may interrupt the monotonity of the constructed sequence to the sequential subgradient pro- jection algorithm to construct a nommonotonous sequential subgradient projection algorithm for solving convex feasibility problem, which can leave such corridor by taking a big step at different steps during the iteration. Under some suitable conditions, the convergence is *** also compare the numerical performance of the proposed algorithm with that of the monotonous algorithm by numerical experiments.

On geometric interpretation of extended state observer: a preliminary study

Borrowing the framework of the geometric approach, this paper tries to analyze and explain why it is possible for the extended state observer (ESO) to estimate the state vector and total disturbance accurately. The geometric approach has provided an elegant and rigorous framework to redefine some key concepts in modern control theory, such as controllability and observability. Moreover, those concepts can be extended to deal with systems in the presence of inaccessible disturbances, such as controlled invariants and conditioned invariants. It is shown in this paper that the augmented system of the ESO is unknown-state unknown-input completely reconstructable in finite time interval. A numerical simulation is given to verify the state vector and total disturbance can be estimated accurately by the ESO if the augmented system is unknown-state unknown-input completely reconstructable.

The effect of drug position on the properties of paclitaxel-conjugated gold nanoparticles for liver tumor treatment

Structure-efficacy effect of small molecular drug attracts wide attentions,but it has always been ignored in nanomedicine *** reveal the efficacy modulation of nanomedicine,we developed a new type of paclitaxel(PTX)-conjugated gold nanoparticles(PTX-co njugated GNPs)to investigate the influence of drug position in controlling their in vitro properties and in vivo *** therapeutic ligands(TA-PEG-NH-N=PTX and TA-PTX=N-NH-PEG)were synthesized to conjugate PTX on the surface of GNPs at different positions,locating on the surface of gold conjugate and inserting between GNPs and polyethylene glycol(PEG,molecular weight 1000 Da),*** was found that PEG-PTX@GNPs with PTX located between GNP and PEG exhibited higher aqueous solubility,biocompatibility,and *** addition,an acid sensitive hydrazone bond has been inserted between PTX and PEG in both ligands for drug release of PTX and PTX-PEG segment,respectively,at the tumor *** release of PTX from PTX-PEG segment is based on the esterase hydrolysis of an ester bond between PTX and *** two-step drug release mechanism offers PEG-PTX@GNPs effective and sustained release behavior for desirable anticancer activity,enhanced therapeutic efficacy,and lower systematic toxicity in Hepsbearing animal models.

Air-sea carbon fluxes and their controlling factors in the Prydz Bay in the Antarctic

The Prydz Bay in the Antarctic is an important area in the Southern Ocean due to its unique geographic feature. It plays an important role in the carbon cycle in the Southern Ocean. To investigate the distributions of carbon dioxide in the atmosphere and surface seawater and its air-sea exchange rates in this region, the Chinese National Antarctic Research Expedition （CHINARE） had set up several sections in the Prydz Bay. Here we present the results from the CHINARE-XVI cruises were presented onboard R/V Xue/ong from November 1999 to April 2000 and the main driving forces were discussed controlling the distributions of partial pressure of carbon dioxide. According to the partial pressure of carbon dioxide distributions, the Prydz Bay can be divided into the inside and outside regions. The partial pressure of carbon dioxide was low in the inside region but higher in the outside region during the measurement period. This distribution had a good negative correlation with the concentrations of ehlorophyll-a in general, suggesting that the partial pressure of carbon dioxide was substantially affected by biological production. The results also indicate that the biological produetion is most likely the main driving force in the marginal ice zone in the Southern Ocean in summer. However, in the Antarctic divergence sector of the Prydz Bay （about 64°S）, the hydrological processes become the controlling factor as the sea surface partial pressure of carbon dioxide is much higher than the atmospheric one due to the upwelling of the high DIC CDW, and this made the outside of Prydz Bay a source of carbon dioxide. On the basis of the calculations, the CO2 flux in January （austral summer） was -3.23 mmol/（m^2 · d） in the inner part of Prydz Bay, i.e. , a sink of atmospheric CO2, and was 0.62 mmol/（m^2 · d） in the outside part of the bay, a weak source of atmospheric CO2. The average air-sea flux of CO2 in the Prydz Bay was 2.50 mmol/（m^2 · d）.
