Protein arginine methyltransferase-6 regulates heterogeneous nuclear ribonucleoprotein-F expression and is a potential target for the treatment of neuropathic pain

Protein arginine methyltransferase-6 participates in a range of biological functions,particularly RNA processing,transcription,chromatin remodeling,and endosomal ***,it remains unclear whether protein arginine methyl transferase-6 modifies neuropathic pain and,if so,what the mechanisms of this *** this study,protein arginine methyltransferase-6 expression levels and its effect on neuropathic pain were investigated in the spared nerve injury model,chronic constriction injury model and bone cancer pain model,using immunohistochemistry,western blotting,immunoprecipitation,and label-free proteomic *** results showed that protein arginine methyltransferase-6 mostly co-localized withβ-tubulinⅢin the dorsal root ganglion,and that its expression decreased following spared nerve injury,chronic constriction injury and bone cancer *** addition,PRMT6 knockout(Prmt6~(-/-))mice exhibited pain ***,the development of spared nerve injury-induced hypersensitivity to mechanical pain was attenuated by blocking the decrease in protein arginine methyltransferase-6 ***,when protein arginine methyltransferase-6 expression was downregulated in the dorsal root ganglion in mice without spared nerve injury,increased levels of phosphorylated extracellular signal-regulated kinases were observed in the ipsilateral dorsal horn,and the response to mechanical stimuli was ***,protein arginine methyltransferase-6 appeared to contribute to spared nerve injury-induced neuropathic pain by regulating the expression of heterogeneous nuclear ***,protein arginine methyltransfe rase-6-mediated modulation of hete rogeneous nuclear ribonucleoprotein-F expression required amino atids 319 to 388,but not classical H3R2 *** findings indicated that protein arginine methyltransferase-6 is a potential therapeutic target fo r the treatment of peripheral neuro pathic pain.

Fifteen 1-（4-substituted phenyl）-4-（4-bromophenyl）-5-（halo-o-hydroxyphenyl）imino-1,2,3-triazoles were designed and synthe- sized based on rational combination of 1,2,3-triazoles and （halo）o-hydroxyphenyl group according to the superposition principle of reinforcement of biological activities. All the compounds were tested to an in vitro antimicrobial screening against M.a. and E.c.. Compounds IIe-IIo exhibited more potent antimicrobial activities against M.a. and E.c. than triclosan and fluconazole, which provided valuable information to further study of novel antimicrobial research.

MODELING PHOTONIC CRYSTALS WITH COMPLEX UNIT CELLS BY DIRICHLET-TO-NEUMANN MAPS

For a photonic crystal （PhC） of finite size, it is important to calculate its transmission and reflection spectra. For two-dimensional （2-D） PhCs composed of a square lattice of circular cylinders, the problem can be solved by an efficient method based on the Dirichlet-to-Neumann （DtN） map of the unit cell and a marching scheme using a pair of operators. In this paper, the DtN operator marching method is extended to handle 2-D PhCs with complex unit cells and arbitrary lattice structures.

Study of damage to a high concrete dam subjected to underwater shock waves

To study the effect of a strong underwater shock wave on a concrete dam, this research aims to improve hammer impact methods in model tests. Six 1:200 scale models were designed and tested under distributed impact loads. A device was deployed for a direct measurement of the impact force at the upstream face of the dams. The model dam bases were anchored to prevent displacement. The experimental results indicate that the top part of the concrete dam is a weak zone, and the impact failure initiates with a fracture on the top of the dam. The peak value of impact stress increases when the second crack appears in the concrete dam from the upstream face to the downstream face. And, the level of the second crack in the dam body is lower as the peak value of impact stress increases. In this study, dynamic analysis was conducted by calculating the results to verify the effectiveness of a device to directly measure the impact force. This method may be used to approximately forecast the damage of concrete dam and may also be useful in other engineering applications.

Analysis of the axial fitting clearance between the fuel rod and the end seat in CIADS

The fuel assembly is key structure in China Initiative Accelerator Driven System,and the axial fitting clearance(AFC) for the fuel assembly design is an essential subject of *** this paper,different methods are used to calculate critical stress in cylindrical *** the thermal expansion of fuel assembly outer tube is larger than that of the cladding of fuel rod,enough space should be reserved between the upper end plug and upper seat *** collapse critical compressive stress of the cladding is obtained numerically through ANSYS simulation *** AFC range between the fuel rod cladding and the end seat due to the displacement of thermal expansion is given by the theoretical formulas and ANSYS buckling *** provide a reference for the AFC design of the reactor fuel assembly.

益心口服液对大鼠离体心肌缺血的作用研究

目的观察益心口服液预处理对心肌缺血大鼠的影响并初步探讨其作用机制。方法利用Langendorff离体心脏灌注装置,结扎Wistar大鼠冠状动脉左前降支建立心肌缺血模型。将大鼠随机分为假手术组、模型组、地尔硫组、复方丹参滴丸组,以及益心口服液大、中、小剂量组,观察各组结扎前后心脏血流动力学的改变,测定各组心肌超氧化物歧化酶(SOD)的活性和丙二醛(MDA)的含量,检测各组心肌组织中腺苷酸(ATP、ADP、AMP)含量以及能荷(EC)的改变。结果①不同浓度的益心口服液预处理均能改善结扎导致的左室收缩压(LVSP)的降低(P0.05)。③益心口服液大、中剂量和复方丹参滴丸均能缓解缺血心肌组织中ATP含量的衰减(P<0.05)。结论益心口服液预处理可对抗离体大鼠心肌缺血损伤,且有剂量依从性,其保护作用可能涉及到抗氧化损伤以及对能量的有效保存。

Net energy yield and carbon footprint of summer corn under different N fertilizer rates in the North China Plain

Excessive use of N fertilizer in intensive agriculture can increase crop yield and at the same time cause high carbon（C） *** study was conducted to determine optimized N fertilizer application for high grain yield and lower C emissions in summer corn（Zea mays L.）.A field experiment, including 0（N0）, 75（N75）, 150（N150）, 225（N225）, and 300（N300） kg N ha–1 treatments, was carried out during 2010–2012 in the North China Plain（NCP）.The results showed that grain yield, input energy, greenhouse gas（GHG） emissions, and carbon footprint（CF） were all increased with the increase of N rate, except net energy yield（NEY）.The treatment of N225 had the highest grain yield（10 364.7 kg ha–1） and NEY（6.8%）, but the CF（0.25） was lower than that of N300, which indicates that a rate of 225 kg N ha–1 can be optimal for summer corn in *** GHG emision compontents, N fertilizer（0–51.1%） was the highest and followed by electricity for irrigation（19.73–49.35%）.We conclude that optimazing N fertilizer application rate and reducing electricity for irrigation are the two key measures to increase crop yield, improve energy efficiency and decrease GHG emissions in corn production.

Pterygial body epithelium domination of pterygial proliferation with TCF4 as a potential key factor

AIM： To characterize the proliferative capacity of pterygial epithelium in different regions（head, neck and body） of pterygium and explore the function of transcription factor 4（TCF4） in pterygium ***： Thirty pterygium tissues and 10 normal conjunctival tissues were obtained from Zhongshan Ophthalmic Center（ZOC） and Guangdong Eye Bank, respectively. Proliferative capacity of head, neck and body in pterygial epithelium was measured using clonal analysis, fold growth analysis and expression profile of proliferative markers revealed by immunofluorescent staining and real-time PCR. The expression of TCF4 was highlighted by double immunofluorescent staining with other proliferation related markers such as proliferating cell nuclear antigen（PCNA） and ATP-binding cassette subfamily G member 2（ABCG2）.RESULTS： The proliferative potential of pterygial epithelium was higher than that of normal conjunctival epithelium. High expression levels of proliferative markers（P63α, PCNA and ABCG2） in pterygial body epithelium were observed in immunofluorescent staining and real-time PCR（P〈0.05）. Also, epithelial cells isolated from pterygial body demonstrated higher proliferative capacity in clonal analysis and fold growth analysis, than those isolated from the head and neck regions. The TCF4 expression in pterygial epithelium was similar to other proliferative markers（P63α, PCNA and ABCG2）, as higher in pterygial body than head and neck. Moreover, TCF4 showed coexpression with other proliferation-related markers（PCNA and ABCG2） in the double immunofluorescent staining ***： The proliferative capacity in pterygial body epithelium is prominent than the head and neck regions, and upregulated TCF4 may be associated with enhanced proliferation in the pterygium.

Diffuse-type tenosynovial giant cell tumor(dTGCT) is a destructive but rare benign proliferative synovial neoplasm. Although surgery is currently the main treatment modality for dTGCT, the recurrence risk is up to 50%. Therefore, there is a great need for effective drugs against dTGCT with minor side effects. The Janus kinase(JAK)/signal transducer and activator of transcription(STAT) signaling plays a central role in rheumatoid arthritis(RA), a disease with similar characteristics as dTGCT, but its function in dTGCT remains *** fibroblast-like synoviocytes(FLS) and macrophages were isolated from 10 synovial tissue samples from dTGCT patients for the screening and validation of the five clinically approved JAK inhibitors to treat RA against dTGCT. Cell viability, cell death, inflammation and the activity of the JAK family members of cultured dTGCT FLS(both 2-D and 3-D) and macrophages were investigated for the efficacy of the JAK inhibitors. Here, we found that similar to RA, JAK/STAT signaling was markedly activated in the dTGCT synovium. Of the 5 JAK inhibitors, peficitinib was shown to have the most potency in addressing some of the pathological responses of dTGCT FLS and *** potency of peficitinib was much higher than pexidartinib, which is the only FDA-approved drug for dTGCT. Mechanistically, peficitinib inhibited tyrosine kinase 2(TYK2), a JAK family member necessary for the pathological progression of dTGCT FLS and macrophages. In summary, we not only revealed JAK/STAT(especially TYK2) signaling as the major mechanism underlying dTGCT, but also identified peficitinib as a promising drug against dTGCT.

Influences of preparation techniques on glass-forming ability of Fe-P-B-Si-C amorphous alloys

It has been widely accepted that the ultrafast cooling rate is required for the glass formation of amorphous alloys. Here, the larger glass-forming ability （GFA） of Fe76P5（B0.5Si0.3C0.2）19 amorphous alloy was achieved by water quenching at lower cooling rate under argon atmosphere. Cylindrical rods with diameters of 1-2 mm were prepared by water quenching without flux treatment, Cu-mold injection casting, and Cu-mold suction casting, respectively. The influences of the preparation techniques with different cooling rates on GFA, thermal property, and nucleation/growth behavior were examined. The critical diameter of the Fe76P5（B0.5Si0.3C0.2）19 amorphous alloys is 1.7 mm for water quenching while smaller than 1.0 mm for injection casting. Microstructure analysis indicates that the crystallization and solidification processes are quite different between the water-quenched and the injection-cast rods. These findings could deepen fun-damental understanding on the relationship between the cooling rate, techniques, and GFA of Fe-based amorphous alloys.
