Abscisic acid(ABA)is a key regulator of plant responses to abiotic stresses,such as drought.Abscisic acid receptors and coreceptors perceive ABA to activate Snf1-related protein kinase2 s(SnRK2s)that phosphorylate downstream effectors,thereby activating ABA signaling and the stress response.As stress responses come with fitness penalties for plants,it is crucial to tightly control SnRK2 kinase activity to restrict ABA signaling.However,how SnRK2 kinases are inactivated remains elusive.Here,we show that NUCLEAR PORE ANCHOR(NUA),a nuclear pore complex(NPC)component,negatively regulates ABA-mediated inhibition of seed germination and post-germination growth,and drought tolerance in Arabidopsis thaliana.The role of NUA in response to ABA depends on SnRK2.2 and SnRK2.3 for seed germination and on SnRK2.6 for drought.NUA does not directly inhibit the phosphorylation of these SnRK2s or affects their abundance.However,the NUA-interacting protein EARLY IN SHORT DAYS 4(ESD4),a SUMO protease,negatively regulates ABA signaling by directly interacting with and inhibiting SnRK2 phosphorylation and protein levels.More importantly,we demonstrated that SnRK2.6 can be SUMOylated in vitro,and ESD4 inhibits its SUMOylation.Taken together,we identified NUA and ESD4 as SnRK2 kinase inhibitors that block SnRK2 activity,and reveal a mechanism whereby NUA and ESD4 negatively regulate plant responses to ABA and drought stress possibly through SUMOylation-dependent regulation of SnRK2s.

The photo-dimerization characteristics of coumarin pendants within amphiphilic random copolymer micelles in aqueous solution was comprehensively investigated using various selected wavelength light in the UV-Vis-NIR region. The time-dependent photo-dimerization degree （PD） changes in the photo-dimerization experiments showed saturating behaviors with intensity-independent of PDmax values at 28%, 44%, 92%, 85%, 36%, 35%, 32% and 31% for 254, 288, 320, 360, 400, 500, 650 and 900 nm irradiations, respectively. The irradiation experiments at 254 and 288 nm announced the occurring of an asymmetric equilibrium of photo-dimerization and photo-cleavage at the used conditions. Both the alternative irradiation cycles of 360 and 254 nm, 650 and 254 nm showed a partially, but evidently reversible photo-dimerization tendency.

It is of great significance to study the root characteristics of rice to improve water and nitrogen(N) use efficiency and reduce environmental pollution. This study investigated whether root traits and architecture of rice influence grain yield, as well as water and N utilization efficiency. An experiment was conducted using the upland rice cultivar Zhonghan 3(a japonica cultivar) and paddy rice cultivar Huaidao 5(also a japonica cultivar) using three N levels, namely, 2 g urea/pot(low amount, LN), 3 g urea/pot(normal amount, NN), and 4 g urea/pot(high amount, HN), and three soil water potentials(SWPs, namely, well-watered(0 kPa), mildly dried(–20 kPa) and severely dried(–40 kPa). The results showed that with decreasing SWP, the percentage of upland rice roots increased in the 0–5 cm tillage layer, and decreased in the 5–10 and 10–20 cm tillage layers, whereas paddy rice roots showed the opposite trend. With increasing amounts of N, the yield of upland and paddy rice increased, and the percentage of root volume ratios of the two rice cultivars in the 0–5 and 5–10 cm tillage layers increased, whereas that in the 10–20 cm tillage layer decreased. The roots of upland rice are mainly distributed in the 10–20 cm tillage layer, whereas most paddy rice roots are in the 0–5 cm tillage layer. These results indicate that the combination of-20 kPa SWP and NN in upland rice and 0 kPa SWP and LN in paddy rice promotes the growth of the root system during the middle and late stages, which in turn may decrease the requirements for water and N fertilizer and increase rice yield.

This study aims to investigate the potential factors affecting hydraulic fracturing of inter-salt oil shale reservoirs in the Qianjiang Depression,China.Using the inter-salt shale samples,the re-crystallization seepage tests,rock mechanical tests under high temperature and pressure,salt rock creep tests,and direct shear tests were conducted.The testing results suggest several major factors that affect hydraulic fracturing effects in the end.First,the seepage of reservoir and fracturing fluid through hydraulic frac-tures leads to salt dissolution and crystallization,reducing the effective seepage area of fractures.Second,the salt crystal may block the pore throats or micro fractures after brine invades the shale,decreasing the overall permeability.Third,the low strength and obvious plasticity of inter-salt shale and the strong creep characteristics of salt rock raise difficulties for proppant to effectively support fracture walls,thereby sharply narrowing the hydraulic fracture width.Lastly,the weak interfaces(bedding planes and lithology interfaces)in inter-salt oil shale reservoirs restrict the height of hydraulic fractures,resulting in the disconnection of seepage channels between multiple inter-salt shale reservoirs.Thus,several factors together reduce reservoir permeability,weaken the fluid flow capacity in the fracture,narrow the fracture width,and limit the effective stimulation volume,resulting in weaken the effect hydraulic fracturing.

BET proteins,which influence gene expression and contribute to the development of cancer,are epigenetic interpreters.Thus,BET inhibitors represent a novel form of epigenetic anticancer treatment.Although preliminary clinical trials have shown the anticancer potential of BET inhibitors,it appears that these drugs have limited effectiveness when used alone.Therefore,given the limited monotherapeutic activity of BET inhibitors,their use in combination with other drugs warrants attention,including the meaningful variations in pharmacodynamic activity among chosen drug combinations.In this paper,we review the function of BET proteins,the preclinical justification for BET protein targeting in cancer,recent advances in small-molecule BET inhibitors,and preliminary clinical trial findings.We elucidate BET inhibitor resistance mechanisms,shed light on the associated adverse events,investigate the potential of combining these inhibitors with diverse therapeutic agents,present a comprehensive compilation of synergistic treatments involving BET inhibitors,and provide an outlook on their future prospects as potent antitumor agents.We conclude by suggesting that combining BET inhibitors with other anticancer drugs and innovative next-generation agents holds great potential for advancing the effective targeting of BET proteins as a promising anticancer strategy.

A novel Pleurotus nebrodensis polysaccharide(PN-S) was purified andcharacterized,and its immune-stimulating activity was evaluated in RAW264.7 macrophages.PN-S induced the proliferation of RAW264.7 cells in a dose-dependent manner,as determined by the MTT assay.After exposure to PN-S,the phagocytosis of the macrophages was significantly improved,with remarkable changes in morphology being observed.Flow cytometric analysis demonstrated that PN-S promoted RAW264.7 cells to progress through S and G2/M phases.PN-S treatment enhanced the productions of interleukin-6(IL-6),nitric oxide(NO),interferon gamma(INF-γ),and tumor necrosis factor-α(TNF-α) in the macrophages,with up-regulation of m RNA expressions of interleukin-6(IL-6),inducible nitric oxide synthase(i NOS),interferon gamma(INF-γ) and tumor necrosis factor-α(TNF-α) being observed in a dose-dependent manner,as measured by q RT-PCR.In conclusion,these results suggest that the purified PN-S can improve immunity by activating macrophages.

A new X-ray imaging and biomedical application beamline(BL13HB)has been implemented at the Shanghai Radiation Synchrotron Facility(SSRF)as an upgrade to the old X-ray imaging and biomedical application beamline(BL13W1).This is part of the Phase II construction project of the SSRF.The BL13HB is dedicated to 2D and 3D static and dynamic X-ray imaging,with a field of view of up to 48.5 mm×5.2 mm and spatial resolution as high as 0.8μm.A super-bending magnet is used as the X-ray source in BL13HB,which has a maximum magnetic field of 2.293 T.The energy range of monochromatic X-ray photons from a double-multiplayer monochromator was 8–40 keV,and the white beam mode was provided on the beamline for dynamic X-ray imaging and dynamic X-ray micro-CT.While maintaining the previous experimental setup of BL13W1,new equipment was added to the beamline experimental station.The beamline is equipped with different sets of X-ray imaging detectors for several experimental methods such as micro-CT,dynamic micro-CT,and pair distribution function.The experimental station of BL13HB is designed specifically for various in situ dynamic experiments,and BL13HB has been open to users since June 2021.
