Chromosome-level assembly of triploid genome of Sichuan pepper(Zanthoxylum armatum)

As an important spice species in Rutaceae, the Sichuan pepper (Zanthoxylum armatum) can provide pungent and numbing taste, as well as aroma in its mature fruit. Here we assembled a chromosome-level genome of green prickly ash which was widely cultivated in a major production area including Chongqing and Sichuan province, china. We generated 712 Gb (~112×) PacBio long reads and 511 Gb (~82×) Hi-C data, and yielded an assembly of 99 pseudochromosomes with total size of 5.32 Gb and contig N50 of 796 kb. The genomic analyses and cytogenetic experiments both indicated that the cultivarZhuye Huajiao’ was a triploid. We identified a Zanthoxylum-specific whole genome duplication event emerging about 24.8 million years ago (Mya). We also detected a transposition burst event (0.3-0.4 Mya) responsible for the large genome size of Z. armatum. Metabolomic analysis of the Zanthoxylum fruits during development stages revealed profiles of39 volatile aroma compounds and 528 secondary metabolites, from which six types of sanshools were identified. Based on metabolomic and transcriptomic network analysis, we screened candidate genes encoding long chain acyl-CoA synthetase, fatty acid desaturase,branched-chain amino acid aminotransferase involved in sanshool biosynthesis and three genes encoding terpene synthase during fruit development. The multi-omics data provide insights into the evolution of Zanthoxylum and molecular basis of numbing and aroma flavor of Sichuan pepper.

Deep sowing is a traditional method for drought resistance in maize production,and mesocotyl elongation is strongly associated with the ability of maize to germinate from deep ***,little is known about the functional genes and mechanisms regulating maize mesocotyl *** the present study,we identified a plant-specific SIMILAR TO RCD-ONE(SRO) protein family member,ZmSRO1e,involved in maize mesocotyl *** expression of ZmSRO1e is strongly inhibited upon transfer from dark to white *** loss-of-function zmsro1e mutant exhibited a dramatically shorter mesocotyl than the wild-type in both constant light and darkness,while overexpression of ZmSRO1e significantly promoted mesocotyl elongation,indicating that ZmSRO1e positively regulates mesocotyl *** showed that ZmSRO1e physically interacted with Zmb ZIP61,an ortholog of Arabidopsis ELONGATED HYPOCOTYL 5(HY5) and showed a function similar to that of HY5 in regulating *** found that ZmSRO1e repressed the transcriptional activity of Zmb ZIP61 toward target genes involved in the regulation of cell expansion,such as ZmEXPB4 and ZmEXPB6,by interfering with the binding of ZmbZIP61 to the promoters of target *** results provide a new understanding of the mechanism by which SRO regulates photomorphogenesis and highlight its potential application in deep sowing-resistant breeding.

It is essential to enhance the thickness of the absorber layer for perovskite solar cells(PSCs)to improve device performance and reduce industry ***,thick perovskite films(>1μm)are difficult to be fabricated by employing traditional solvents,such as N,N-dimethylformamide(DMF),dimethyl sulfoxide(DMSO).Besides,it is a challenge to fabricate thick-film perovskite owing to the deteriorated surface morphology and serious defect ***,a simple method was reported to deposit uniform pinhole-free perovskite films with a thickness of more than 2μm utilizing the methylamine acetate(MAAc)ionic liquid as the *** with methylammonium chloride(MACl)as an additive,thick-film perovskite with~2μm in grain size and few grain boundaries(GBs)was prepared,which dramatically improved the perovskite crystal quality and enhanced carrier transport *** final PSCs exhibited a power conversion efficiency(PCE)of 20.16%.The device showed improved stability with 95%of its initial efficiency in a nitrogen environment over 5000 *** work provides an alternative strategy to produce extremely efficient and stable thick-film *** can be believed that this device has great potential in the application of large areas and laminated PSCs.

Background Deep convolutional neural networks have garnered considerable attention in numerous machine learning applications,particularly in visual recognition tasks such as image and video *** is a growing interest in applying this technology to diverse applications in medical image *** three dimensional Breast Ultrasound is a vital tool for detecting breast cancer,and computer-assisted diagnosis software,developed based on deep learning,can effectively assist radiologists in ***,the network model is prone to overfitting during training,owing to challenges such as insufficient training *** study attempts to solve the problem caused by small datasets and improve model detection *** We propose a breast cancer detection framework based on deep learning(a transfer learning method based on cross-organ cancer detection)and a contrastive learning method based on breast imaging reporting and data systems(BI-RADS).Results When using cross organ transfer learning and BIRADS based contrastive learning,the average sensitivity of the model increased by a maximum of 16.05%.Conclusion Our experiments have demonstrated that the parameters and experiences of cross-organ cancer detection can be mutually referenced,and contrastive learning method based on BI-RADS can improve the detection performance of the model.

BACKGROUND Cardioneuroablation(CNA)has shown encouraging results in patients with vasovagal syncope(VVS).However,data on different subtypes was *** This observational study retrospectively enrolled 141 patients[mean age:40±18 years,51 males(36.2%)]with the diagnosis of *** characteristics among different types of VVS and the outcomes after CNA were *** After a mean follow-up of 4.3±1.5 years,41 patients(29.1%)experienced syncope/pre-syncope events after ***/pre-syncope recurrence significantly differed in each subtype(P=0.04).The cardioinhibitory type of VVS had the lowest recurrence rate after the procedure(n=6,16.7%),followed by mixed(n=26,30.6%)and vasodepressive(n=9,45.0%).Additionally,a significant difference was observed in the analyses of the Kaplan-Meier survival curve(P=0.02).Syncope/pre-syncope burden was significantly reduced after CNA in the vasodepressive type(Pline deceleration capacity(DC)level than those without(7.4±1.0 ms vs.9.0±1.6 ms,P=0.01).Patients with DC<8.4 ms had an 8.1(HR=8.1,95%CI:2.2-30.0,P=0.02)times risk of syncope/pre-syncope recurrence after CNA compared to patients with DC≥8.4 ms,and this association still existed after adjusting for age and sex(HR=8.1,95%CI:2.2-30.1,P=0.02).CONCLUSIONS Different subtypes exhibit different event-free *** vasodepressive type exhibited the lowest event-free rate,but those patients with DC≥8.4 ms might benefit from CNA.
