Validation of Bacteroidales-based microbial source tracking markers for pig fecal pollution and their application in two rivers of North China

In China,pig feces is the predominant source of excrement produced b y animal husbandry.Improper use or direct discharge of pig feces can result in contamination of natural water systems.Microbial source tracking(MST)technology can identify the sources of fecal pollution in environmental water,and contribute to the management of pig fecal pollution by local environmental protection agencies.However,the accuracy of such assays can be context-dependent,and they have not been comprehensively evaluated under Chinese conditions.We aimed to compare the performance of five previously reported nig-specific MST assays(PF,Pig-Bac 1 STBR,Pig-Bac2 SYBR,Pig-1-Bac TaqMan,and Pig-2-Bac TaqMan,which are based on Bacteroidales 16S rRNA gene markers)and apply them in two rivers of North China.We collected a total of 173 fecal samples from pigs,cows,goats,chickens,humans,and horses across China.The PF assay optimized in this study showed outstanding qualitative performance and achieved 100%specificity and sensitivity.However,the two SYBR green qPCR assays(Pig-Bac1 SYBR and Pig-Bac2 SYBR)cross-reacted with most non-pig fecal samples.In contrast,both the Pig-l-Bac TaqMan and Pig-2-Bacr TaqMan assays gave 100%specificity and sensitivity.Of these,the Pig-2-Bac TaqMan assay showed higher reproducibility.Our results regarding the specificity of these pig-specific MST assays differ from those reported in Thailand,Japan,and America.Using the PF and Pig-2-Bac TaqMan assays,a field test comparing the levels of pig fecal pollution in rivers near a pig farm before and after comprehensive environmental pollution governance indicated that pig fecal pollution was effectively controlled at this location.

Previous studies showed that acetyl-11-keto-beta-boswellic acid(AKBA),the active ingredient in the natural Chinese medicine Boswellia,can stimulate sciatic nerve injury repair via promoting Schwann cell proliferation.However,the underlying molecular mechanism remains poorly understood.In this study,we performed genomic sequencing in a rat model of sciatic nerve crush injury after gastric AKBA administration for 30 days.We found that the phagosome pathway was related to AKBA treatment,and brain-derived neurotrophic factor expression in the neurotrophic factor signaling pathway was also highly up-regulated.We further investigated gene and protein expression changes in the phagosome pathway and neurotrophic factor signaling pathway.Myeloperoxidase expression in the phagosome pathway was markedly decreased,and brain-derived neurotrophic factor,nerve growth factor,and nerve growth factor receptor expression levels in the neurotrophic factor signaling pathway were greatly increased.Additionally,expression levels of the inflammatory factors CD68,interleukin-1β,pro-interleukin-1β,and tumor necrosis factor-αwere also decreased.Myelin basic protein-andβ3-tubulin-positive expression as well as the axon diameter-to-total nerve diameter ratio in the injured sciatic nerve were also increased.These findings suggest that,at the molecular level,AKBA can increase neurotrophic factor expression through inhibiting myeloperoxidase expression and reducing inflammatory reactions,which could promote myelin sheath and axon regeneration in the injured sciatic nerve.

Experimental and numerical investigations on wave motions over platform reef-flat

Previous studies have shown that high-frequency(HF)waves,low-frequency(LF)waves and wave set-up coexist on shallow coral reef-flat and jointly contribute to potential floods and subsequent damages of infrastructures and islands on it.To better understand the reef-flat wave dynamics with incident waves and still water level,a wave-flume experiment was performed based on an idealized platform reef composed of a steep reef-face(1:4),a relatively mild reef-rim(1:14)and a horizontal reef-flat.Also,the non-hydrostatic phase-resolving model SWASH was validated against the experiment and then applied to further numerically investigate the effects of reef-rim topographic features on the reef-flat wave motions.The results show that incident waves of a larger wave height and a longer wave period can generate larger LF waves and wave set-up,thereby inducing greater HF waves on the reef-flat.Higher still water level can lead to larger HF waves but result in smaller wave set-up.In contrast to HF waves and wave set-up,LF waves are minimally affected by the still water level.A rim of milder slope and larger edge depth will induce smaller HF and LF waves and set-up on the reef-flat,and thus provide better protection for the reef-flat region.Furthermore,on the reef-flat,the ratio of HF significant wave height to water depth H_(s_h)/(h_(r)+η^(-))is approximately constant;the dimensionless LF significant wave height Hs_l/H_(s0) and the dimensionless wave set-upη^(-)/[T_(p)(gH_(s0))1/2]can be related to the inverse wave steepness parameter gT_(p)^(2)/H_(s0) and the relative reef-flat submergence(h_(r)+η^(-))/H_(s0) respectively.

BACKGROUND Pancreatic adenocarcinoma is one of the most common malignant tumors of the digestive system.More than 80%of patients with pancreatic adenocarcinoma are not diagnosed until late stage and have distant or local metastases.AIM To investigate the value of computed tomography(CT)perfusion imaging in the evaluation of angiogenesis in pancreatic adenocarcinoma patients.METHODS This is a retrospective cohort study.Patients with pancreatic adenocarcinoma and volunteers without pancreatic diseases underwent CT perfusion imaging from December 2014 to August 2017 in Huashan Hospital,Fudan University Shanghai,China.RESULTS A total number of 35 pancreatic adenocarcinoma patients and 33 volunteers were enrolled.The relative blood flow(r BF),and relative blood volume(r BV)were significantly lower in patients with pancreatic adenocarcinoma than in the control group(P<0.05).Conversely,the relative permeability in patients with pancreatic adenocarcinoma was significantly higher than that in controls(P<0.05).In addition,r BF,r BV,and the vascular maturity index(VMI)were significantly lower in gradeⅢ-Ⅳpancreatic adenocarcinoma than in gradeⅠ-Ⅱpancreatic adenocarcinoma(P<0.05).Vascular endothelial growth factor(VEGF),CD105-MVD,CD34-MVD,and angiogenesis rate(AR)were significantly higher in gradeⅢ-Ⅳpancreatic adenocarcinoma than in gradeⅠ-Ⅱpancreatic adenocarcinoma(P<0.05).Significant correlations between r BF and VEGF,CD105-MVD,AR,and VMI(P<0.01)were observed.Moreover,the levels of r BV were statistically significantly correlated with those of VEGF,CD105-MVD,CD34-MVD,and VMI(P<0.01).CONCLUSION Perfusion CT imaging may be an appropriate approach for quantitative assessment of tumor angiogenesis in pancreatic adenocarcinoma.

Objective:To explore the influence of artificial luminous environment and preventive function of traditional Chinese medicine(TCM) intervention based on "Theory of yin-yang clock" on myopia.Methods:A total of 45 New Zealand young rabbits were randomly divided into 5groups,9 for each group.Control group was exposed in natural light.Fluorescent group and full spectrum group were exposed in fluorescent light and full spectrum light,on which basis fluorescent TCM group and full spectrum TCM group were added with "Rizhong Yinyang Formulas",respectively.Optical parameters were measured and the influence of different lights on the serum and retinal dopamine(DA) levels as well as the retinal histopathological tissues was observed.Results:The spectrum of fluorescent light mainly focused at 420-490 nm with the peak value of wavelength near 450 nm,whereas that of full spectrum was wider(400-800 nm) with the peak value near 600 nm.After 4 and 12 weeks,fluorescent group was evidently lower in serum and retinal DA levels(P0.05).Histopathological observation showed that there was significant difference in pigment epithelium layer,photoreceptor and nerve fiber layer between fluorescent group and control group,but the difference among the test groups was not significant.Conclusions:Fluorescent light has certain influence on retinal histological construction and visual performance.However,TCM intervention may have some degree of protective function on retina.

Sealing Performance and Optimization of a Subsea Pipeline Mechanical Connector

Researchers seldom study the optimum design of a mechanical connector for subsea oil-gas pipeline based upon the sealing performance. An optimal design method of a novel subsea pipeline mechanical connector is presented. By analyzing the static metal sealing mechanism, the critical condition of the sealing performance is established for this connector and the formulation method of the contact pressure on the sealing surface is created. By the method the minimum mean contact pressure of the 8.625 inch connector is calculated as 361 MPa, which is the constraint condition in the optimum design of connector.The finite element model is created in ANSYS Parametric Design Language(APDL) and the structure is optimized by the zero-order method, with variance of contact pressure as the objective function, and mean contact pressures and plastic strains as constraint variables. The optimization shows that variances of contact pressure on two sealing surfaces decrease by 72.41% and 89.33%, respectively, and mean contact pressures increase by 31.18% and 52.84%, respectively. The comparison of the optimal connectors and non-optimal connectors in the water pressure experiments and bending experiments shows that the sealing ability of optimized connectors is much higher than the rated pressure of 4.5 MPa, and the optimal connectors don’t leak under the bending moment of 52.2 kN·m.This research provides the formulation to solve contact pressure on the sealing surface and a structure optimization method to design the connectors with various dimensions.

China is the global leader in rare earth （RE） production using hydrometallurgical processes. Advantageous extraction techniques of rare earths from Baotou mixed rare earth minerals, bastnaesite, and ion-adsorption clays of rare earth deposits have been developed in China. The separation and purification technologies have also achieved rapid development. The industrial application processes for rare earth hydrometallurgy were summarized in the present paper. With the large demands and rapid development of rare earths, the issues of resources and environment are more prominent. This review gives an overview of the main processes that were developed in the past toward greener hydrometallurgy of rare earths in China. Based on the development of the rare earth industry, comprehensive utilization and cleaner production should still be focused on in the future, to support the sustainable development.

The pathogenesis of diabetic foot disease is complex and its clinical manifestations are diverse.Western medicine mainly focuses on hypoglycemic,improving circulation,controlling infection,interventional and surgical treatment,and the clinical effect is not good.Chinese medicine has obvious advantages in the treatment of diabetic foot,including syndrome differentiation,staging treatment,prescription plus or minus treatment,Chinese patent medicine,external foot bath,fumigation and washing,massage,acupuncture and moxibustion,ointment powder application and comprehensive treatment of traditional Chinese and western medicine.This paper discusses the treatment of diabetic foot with traditional Chinese medicine.

Influence of hydrogen concentration on Fe_2O_3 particle reduction in fluidized beds under constant drag force

The fixed-gas drag force from a model calculation method that stabilizes the agitation capabilities of different gas ratios was used to explore the influence of temperature and hydrogen concentration on fluidizing duration, metallization ratio, utilization rate of reduction gas, and sticking behavior. Different hydrogen concentrations from 5vol%to 100vol%at 1073 and 1273 K were used while the drag force with the flow of N2 and H2 （N2：2 L·min^-1;H2：2 L·min^-1） at 1073 K was chosen as the standard drag force. The metallization ratio, mean reduc-tion rate, and utilization rate of reduction gas were observed to generally increase with increasing hydrogen concentration. Faster reduction rates and higher metallization ratios were obtained when the reduction temperature decreased from 1273 to 1073 K. A numerical relation among particle diameter, particle drag force, and fluidization state was plotted in a diagram by this model.
