Background: Thyroid cancer(TC) is the most common endocrine cancer around the world, of which papillary thyroid cancer(PTC) accounts for about 85%. Fine-needle aspiration biopsy(FNAB) is the preferred method for the diagnosis of thyroid nodules in the clinical application. However, about 20-30% of cases are indeterminate nodules according to FNAB, of which approximately 69% are classified as benign by postoperative pathology. Besides, the sensitivities of FNAB are 65-99% across studies. It is thus of great importance to develop novel biomarkers to distinguish the malignant tumors from the benign ones. DNA methylation is commonly dysregulated in a variety of diseases including cancers. Our previous Infinium Methylation EPIC 850 K beadchip assay indicated a differentially methylated CpG site(cg02183083) within hyaluronidase 1(HYAL1) gene. Here, we conducted a validation study to explore the association between HYAL1 and thyroid *** and methods: We measured the HYAL1 methylation level in formalin-fixed paraffin-embedded(FFPE) tissues of 195 early-stage PTC(91.05% PTC at stage Ⅰ) and 195 age-and gender-matched benign tumors collected during 2019-2020. The median(interquartile range, IQR) ages of malignant and benign cases were 51.00(44.00-57.00) and 53.00(46.00-60.25) years old, respectively. Females accounted for 78.95%(150/190) in both PTC and benign cases. Protein expression of HYAL1 was evaluated in another 55 pairs of early-stage PTC(89.09% PTC at stage Ⅰ) and benign tumors collected during 2013-2016, of which 34 pairs of cases were females. The median age was 43.00(31.00-51.00)and 47.00(36.00-52.00) years in PTC and benign *** methylation was quantified by matrixassisted laser desorption ionization time-of-flight mass spectrometry and protein expression was detected by immunohistochemical(IHC) staining. Aims: Mahogunin ring finger 1(MGRN1), an E3 ubiquitin ligase of the Really Interesting New Gene finger family, plays a pivotal role in many biological and cellular mechanisms. The DNA hypomethylation in MGRN1 has been reported to be associated with breast cancer(BC) in the Caucasian ***, the association has not been studied in other population. In this case-control study, we attempted to study the relationship between MGRN1 DNA methylation and BC in the Chinese *** and Methods: 4 MGRN1 CpG sites were quantitatively measured through mass spectrometry in the case-control study with 234 sporadic BC cases and 222 controls. The interquartile analysis was performed by calculating the quartiles from the methylation data from the total participants. The logistic regression adjusted for covariants and non-parametric tests were used to investigate the association between MGRN1 methylation level and the status of ***: We observed four MGRN1 CpG sites(MGRN1pG, MGRN1pG, MGRN1pG,4)were associated with BC. A significant tendency was also found for two MGRN1 CpG sites(P≤ 0.016,Table 1). In addition, we found significant associations between BC and altered MGRN1 methylation at different age groups, mostly in women at postmenopausal(age ≥ 50 years old, odds ratios(ORs)per-10% methylation: 1.58-1.85, P ≤ 0.031 for three of four CpG loci;age < 50 years old, ORs per-10% methylation = 0.77, P=0.033 for one CpG site, Table 2). This age-related MGRN1 methylation was also modified by the status of human epidermal growth factor receptor 2(HER2)(HER2 negative,ORs per-10% methylation: 0.86-1.28, P ≤ 0.045 for all four CpG loci;HER2 positive, ORs per-10%methylation = 0.79, P = 0.022 for one CpG site, Table 2).Conclusion: In the Chinese population, we reported a significant association between BC and the bloodbased MGRN1 methylation modified by age and the status of HER2. Using the observation data in Yongxing Island,South China Sea(SCS)from December 2013 to November 2018,the multiple time scales variation of atmospheric CO_(2)and CH_(4)were analyzed to understand their temporal variation characteristics and controlling *** regional-averaged background mole fractions of CO_(2)and CH_(4)both show a single-period sinusoidal variation with a lower value at noon and a higher value in the wee *** the seasonal scale,they exhibited a significant seasonal difference with higher values in winter and lower values in *** the annual scale,CO_(2)and CH_(4)both show an increasing trend,with an annual growth rate of approximately 3.2 ppm and 12 ppb,*** annual growth rate at this site was higher than the global *** change in atmospheric CO_(2)and CH_(4)in Yongxing Island was probably caused by the higher emission of the surrounding areas and the airflows driven by ***,the long-term and high-resolution greenhouse gases(GHGs)dataset will aid relevent researchers and decision-makers in performing more in-depth studies for GHG sources in order to derive effective strategies.

Real-Time 3D Ultrasound Imaging System Based on a Hybrid Reconstruction Algorithm

As a safe and convenient imaging technology in clinical routine diagnosis, ultrasound imaging can provide real-time 2D images of internal tissues and organs. To realize real-time 3D image reconstruction, pixel nearest neighbor interpolation(PNN) reconstruction algorithm and Bezier interpolation algorithm are combined into a hybrid reconstruction algorithm. On this basis, a real-time interactive 3D ultrasound imaging system is *** temporal calibration and spatial calibration, the six degrees of freedom poses of 2D ultrasound images can be accurately collected. The 3D volume reconstructed by the proposed 3D reconstruction algorithm is visualized by volume rendering. A multi-thread software system allows parallel operation of data acquisition, 3D reconstruction,volume visualization and other functions. 3D imaging experiments on a 3D printing femur model, a neck phantom and the neck of human volunteers were performed for systematic evaluation. When the reconstruction voxel size was set to be(0.53 mm3, 1.03 mm3, 1.53 mm3), the reconstruction errors of the femur and trachea model were respectively(0.23 mm, 0.31 mm, 0.56 mm) and(0.62 mm, 0.88 mm, 1.41 mm). Clinical feasibility was demonstrated by application of the 3D ultrasound imaging on the neck of human volunteers.

Causal relationship between circulating vitamin C and 25-hydroxyvitamin D concentrations and common mental disorders-a Mendelian randomization study

Mental disorders seriously affect people’s health and social *** Mendelian randomization(MR)study was designed to investigate the causal relationship between circulating vitamin C(VC)or 25-hydroxyvitamin D(25(OH)D)levels and mental *** data used for the MR analysis were derived from the summary genome-wide association studies(GWAS)database for VC and 25(OH)D and from the Finn Gen consortium for fourteen mental *** on the inverse variance weighted(IVW)method,we found a potential causal association between circulating VC and anxiety disorders(IVW:OR=1.139,95%CI:1.023-1.269,P=0.018).However,no causal association was found between VC or 25(OH)D and other mental disorders(P>0.05).In the reverse MR analysis,individuals with Alzheimer’s disease was causally associated with higher concentrations of circulating VC(P=0.012),while individuals with anxiety disorders had a negative association between the concentrations of 25(OH)D(P=0.012).However,the current evidence does not support a causal relationship between VC or 25(OH)D and other mental *** addition,there was no causal association between circulating VC and 25(OH)D(P>0.05).Future studies are needed to confirm these findings and to elucidate the mechanisms of potential causality.

Mesenchymal stem cells(MSCs)transplantation is a promising strategy for osteoporosis ***,limited sources and poor tissue-homing efficiency limit their clinical *** this study,we isolated a kind of MSCs from women’s menstrual blood(MenSCs)noninvasively and established a novel MSCs bone marrow-targeted delivery system by utilizing waterin-oil-in-water droplet *** were encapsulated withinβ-cyclodextrin-functionalized alginate microcapsules loaded with zoledronates,which has a high affinity for *** this delivery system,MenSCs could be preferentially delivered to the bone marrow tissues via intravenous infusion,and restored bone mass by remodeling the bone marrow niche in situ in ovariectomized mouse ***,scRNA-seq analysis demonstrated that those MenSCs homed to the bone marrow recruited CD4^(+)FOXP3^(+)natural regulatory T(nT_(reg))cells by secreting *** recruited nTreg promoted CD8^(+)T cells to secret Wnt family member 10B(WNT10B),activating the Wnt signaling in osteoblasts and thus promoting bone formation in situ in the bone *** study reveals a promising application of MenSCs in postmenopausal osteoporosis treatment and highlights the clinical value of MenSCs by encouraging women to reserve autologous MenSCs before menopause to prevent and alleviate postmenopausal osteoporosis.

Re-entry rocket basic flow characteristics and thermal environment of different retro-propulsion modes

During the supersonic re-entry of multi-nozzle heavy rockets into the atmosphere,the basic flow state becomes increasingly complex due to the coupling effect between the retropropulsion plumes and the freestream.A numerical method using the hybrid Reynolds-Averaged Navier-Stokes and Large Eddy Simulation(RES)method and discrete coordinate method is developed to accurately estimate the thermal *** addition,finite rate chemical kinetics is used to calculate the afterburning *** numerical results agree well with wind tunnel data,which confirms the validity and accuracy of the numerical *** are conducted for the heavy carrier rocket re-entry from 53.1 km to 39.5 km altitude with 180°angle of attack by using three different Supersonic Retro-Propulsion(SRP)*** numerical results reveal that these three SRP flow fields are all Short Penetration Models(SPM).As the re-entry altitudes decrease,both the plume-plume interaction and the plume-freestream interaction become *** highest temperatures in the plume shear layers of the three SRP modes increase by 8.36%,7.33%and 6.92%respectively after considering afterburning reactions,and all occur at a reentry altitude of 39.5 *** the rocket re-enters the atmosphere,the maximum heat flux on the rocket base plate of three SRP modes stabilizes at 290,170 and 200 kW/m^(2) respectively,but the maximum heat flux on the side wall increases *** the altitude declines to 39.5 km,the extreme heat flux of the three modes increase by 84.16%,49.45%and 62.97%respectively compared to that at 53.1 km.

Effects of retained dry material on the impact,overflow and landing dynamics

During long-term operation,the performance of obstacles would be changed due to the material accumulating upslope the ***,the effects of retained material on impact,overflow and landing dynamics of granular flow have not yet been *** address this gap,physical flume tests and discrete element simulations are conducted considering a range of normalized deposition height h0/H from 0 to 1,where h0 and H represent the deposition height and obstacle height,*** analytical model is modified to evaluate the flow velocity and flow depth after interacting with the retained materials,which further serve to calculate the peak impact force on the ***,the computed impact forces successfully predict the experimental results when a≥25°.In addition,the results indicate that a higher h0/H leads to a lower dynamic impact force,a greater landing distance L,and a larger landing coefficient Cr,where Cr is the ratio of slope-parallel component of landing velocity to flow velocity just before *** to the existing overflow model,the measured landing distance L is underestimated by up to 30%,and therefore it is insufficient for obstacle design when there is retained ***,the recommended Cr in current design practice is found to be nonconservative for estimating the landing velocity of geophysical *** study provides insightful scientific basis for designing obstacles with deposition.

Optimization of Random Feature Method in the High-Precision Regime

Machine learning has been widely used for solving partial differential equations(PDEs)in recent years,among which the random feature method(RFM)exhibits spectral accuracy and can compete with traditional solvers in terms of both accuracy and ***,the optimization problem in the RFM is more difficult to solve than those that arise in traditional *** the broader machine-learning research,which frequently targets tasks within the low-precision regime,our study focuses on the high-precision regime crucial for solving *** this work,we study this problem from the following aspects:(i)we analyze the coeffcient matrix that arises in the RFM by studying the distribution of singular values;(ii)we investigate whether the continuous training causes the overfitting issue;(ii)we test direct and iterative methods as well as randomized methods for solving the optimization *** on these results,we find that direct methods are superior to other methods if memory is not an issue,while iterative methods typically have low accuracy and can be improved by preconditioning to some extent.
