Visualizing experimental investigation on gas-liquid replacements in a microcleat model using the reconstruction method

Cleats are the main channels for fluid transport in coal ***,the microscale flow characteristics of both gas and water phases in primary cleats have not been fully studied as ***,the local morphological features of the cleat were determined using image processing technology and a transparent cleat structure model was constructed by microfluidic lithography using the multiphase fluid visualization test ***,the effect of microchannel tortuosity characteristics on two‐phase flow was analyzed in this *** results are as follows:(1)The local width of the original cleat structure of coal was strongly *** cleats showed contraction and expansion in the horizontal direction and undulating characteristics in the vertical direction.(2)The transient flow velocity fluctuated due to the structural characteristics of the primary *** water‐driven gas interface showed concave and convex instability during flow,whereas the gas‐driven water interface presented a relatively stable concave surface.(3)The meniscus advanced in a symmetrical pattern in the flat channel,and the flow stagnated due to the influence of undulation points in a partially curved *** flow would continue only when the meniscus surface bypassed the stagnation point and reached a new equilibrium position.(4)Enhanced shearing at the gas-liquid interface increased the gas‐injection pressure,which in turn increased residual liquids in wall grooves and liquid films on the wall surface.

KLF4 Inhibits the Activation of Human Hepatic Stellate Cell In Vitro

Objective Hepatic stellate cells(HSCs)play a crucial role in liver ***-stage liver fibrosis is reversible and intimately associated with the state of ***-like factor 4(KLF4)plays a pivotal role in a wide array of physiological and pathological *** study aimed to investigate the effect of KLF4 on the proliferation,apoptosis and phenotype of quiescent HSCs Methods We designed a KLF4 lentiviral vector and a KLF4 siRNA lentiviral vector,to upregulate and silence KLF4 expression in human HSC LX-2 cells via *** proliferation was assessed using the CCK-8 *** cytometry was used to detect the cell cycle distribution and apoptosis *** blotting was used to determine the levels of some quiescence and activation markers of HSCs Results Overexpression of KLF4 significantly increased the levels of E-cadherin and ZO-1,which are quiescent HSC markers,while significantly decreased the levels of N-cadherin and a-SMA,known activated HSC *** contrast,cell proliferation and apoptosis rates were elevated in LX-2 cells in which KLF4 expression was silenced Conclusion KLF4 inhibits the proliferation and activation of human LX-2 *** might be a key regulatory protein in the maintenance of HSC quiescence and may serve as a target for the inhibition of hepatic fibrosis.

Effects of daidzein on antioxidant capacity in weaned pigs and IPEC-J2cells

Our previous study found that soybean isoflavones in soybean meal play an important role in improving growth performance and antioxidant capacity in ***,it is still unknown whether long-term supplementation with daidzein,an active molecule deglycosylated from daidzin,in a corn-soybean meal diet can enhance growth performance in ***,in the present study,an animal trial was carried out to investigate the effects of dietary supplementation with daidzein on the growth performance and antioxidant capacity of pigs.A total of 80 weaned piglets(40 barrows and 40 females)were assigned to 4treatments with 5 pens per treatment and 4 piglets per pen and fed a diet supplemented with 0,25,50 and 100 mg/kg daidzein for a 72-day *** addition,porcine intestinal epithelial cells(IPEC-J2)were used as an in vitro model to explore the underlying antioxidant mechanisms of ***-J2 cells were treated with 0.6 mM hydrogen peroxide(H_(2)O_(2))in the presence or absence of 40μM *** results showed that adding 50 mg/kg of daidzein to the diet significantly improved body weight on day72,average daily gain(ADG)during days 0 to 72 and plasma superoxide dismutase(SOD)activity on day42,and decreased malondialdehyde(MDA)content on day 14(P<0.05).Treatment with 0.6 m M H_(2)O_(2)for 1 h significantly decreased cell viability and catalase(CAT)activity and increased intracellular reactive oxygen species(ROS)levels and MDA content(P<0.05),while pretreatment with 40μM daidzein prevented the decrease in cell viability and CAT activity and the increase in intracellular ROS levels and MDA content caused by H_(2)O_(2)(P<0.05).In addition,H_(2)O_(2)stimulation significantly suppressed the expression of nuclear factor erythroid-2-related factor 2(Nrf2),CAT,occludin and zonula occludens-1(ZO-1),while pretreatment with daidzein preserved the expression of Nrf2,CAT and occludin in H_(2)O_(2)-stimulated IPEC-J2 cells(P<0.05).In conclusion,our results suggested that long-term di

Mirror-like tongue is an important predictor of acute heart failure:a cohort study of acute heart failure in Chinese patients

OBJECTIVE:To reveal that mirror-like tongue,observed via a noninvasive inspection,is a powerful indicator of the severity and prognosis of patients with acute heart failure(AHF).METHODS:This was an observational,prospective study.A total of 408 patients who met the inclusion criteria and were diagnosed with AHF for the first time at Taicang Traditional Chinese Medicine Hospital Affiliated to Nanjing University of Chinese Medicine from August 2019 to January 2021 were selected as the research *** to their tongue fur,the patients were divided into group A(mirror-like tongue group)and group B(non-mirror-like tongue group).The clinical characteristics and incidence of major adverse cardiovascular events(MACEs)within 1 year of follow-up were systematically compared between the two ***:Sixty-five patients were included in group A,and 343 patients were included in group *** were 32 males and 33 females in group A and 168 males and 175 females in group *** average age of the overall population was 64 years old,and the average age of group A was significantly higher than that of group B(74 vs 62 years,P<0.001).Compared with that in group B,the left ventricular ejection fraction(LVEF)in group A was significantly lower(35%vs 42%,P<0.001),and the log N-terminal pro B-type natriuretic peptide(NT-proBNP)level was significantly higher(4.0 vs 3.4,P<0.001).The proportion of the combined application of vasoactive drugs in group A was significantly higher than that in group B(64%vs 38%,P<0.001).Group B had a higher proportion of coronary angiography(29.5%vs 16.9%,P=0.038).Group A was more inclined to require mechanical ventilation than group B(33.9%vs 22.5%,P=0.049).The length of hospital stay in group A was significantly longer than that in group B(13.1 vs 7.6,P<0.001).The incidence of MACEs,such as recurrence of AHF,new myocardial infarction and stroke,in group A within one year was higher than that in group B(P=0.007,0.009,<0.001).The incidence of cu

Satellite observations of coastal upwelling in the northern Arafura Sea

Coastal upwelling is significant for marine ecosystems by lifting nutrient-rich deep waters into the euphotic zone,thereby increasing primary and secondary *** satellite observations show that the northern Arafura Sea(NAS),especially in the coastal region,features high chlorophyll-a(chl-a)concentrations,implying a strong coastal ***,coastal upwelling in the NAS has not received much *** on a semi-automatic image processing technology,the seasonal and interannual variability of coastal upwelling in the NAS are investigated in this study using satellite-observed sea surface temperature(SST)and wind *** results suggest that there are seasonal coastal upwelling events in the NAS modulated by upwelling-favorable southeast monsoon(SEM).The annual mean days,mean area,and annual mean intensity of coastal upwelling events during the SEM season are 92 days,6514 km^(2),and-5.31×10^(5),respectively,while the corresponding values during the northwest monsoon(NWM)season are 32 days,5569 km^(2),and-1.41×10^(5).It is also found that the SEM coastal upwelling in the NAS displays prominent interannual *** strong upwelling events are found in 2010,2013,2016,and 2017 when the southeast monsoon winds were *** analysis suggests that at the interannual scale,the upwelling index(UI)averaged in the SEM season is negatively correlated with that of three upwelling *** can be attributed to the limitation of onshore geostrophic flow which is evidenced by the negative correlation between the UI and the alongshore difference in sea surface *** study highlights the important role of the southeast monsoon in the temporal variability of coastal upwelling in the NAS.

Preparation by Wet-spinning and Properties Polyaniline-Polyethersulfone Fibers for Flexible Supercapacitor Electrodes

Flexible PANI-Polyethersulfone (PES) fibers were fabricated using the wet-spinning *** particles were uniformly distributed within the matrix and micropores formed by the phase separation of PES,which prevented PANI particles aggregation and facilitated the formation of continuous ion transport *** experimental results reveals that the electrochemical performance of the fiber electrode material is optimal when the concentration of PES in the spinning solution is 15wt%.The assembled supercapacitor exhibits a commendable specific area capacitance of 162.75 mF·cm^(-2) at a current density of 0.5 mA·cm^(-2) and achieves an energy density of 14.47 mWh·cm^(-2) at a power density of 321.69 mW·cm^(-2).The capacitor retains 98.1% of its capacitance after 1 000 bending ***,the prepared fibers have good electrochemical properties and flexibility,and this simple and efficient preparation method is promising for the scalable production of flexible electrodes.

Chick chorioallantoic membrane model to investigate role of migrasome in angiogenensis

The development of the vascular system is essential for embryonic development, including processes such as angiogenesis. Angiogenesis plays a critical role in many normal physiological and pathological processes. It is driven by a set of angiogenic proteins, including angiogenic growth factors, chemokines,and extracellular matrix proteins. Among various animal model systems, the chorioallantoic membrane(CAM), a specialized and highly vascularized tissue of the avian embryo, has proven to be a valuable tool for analyzing the angiogenic potential of candidate cells or factors. In this protocol, we provide detailed procedures for establishing the CAM model to evaluate the function and mechanism of migrasomes in embryonic angiogenesis. This includes the CAM nylon mesh assay and CAM ex vivo sprouting assay to assess CAM angiogenesis, as well as the observation, purification, and delivery of migrasomes. Additionally, we describe the generation of T4-KO-m Cherry-KI embryos using the CRISPR system within the CAM tissue to investigate the role of migrasomes in angiogenesis.
