Impacts of Urban Expansion on the Urban Thermal Environment:A Case Study of Changchun,China

Urbanization,especially urban land expansion,has a profound influence on the urban thermal *** in Northeast China face remarkably uneven development and environmental issues,and thus it is necessary to strengthen the diagnosis of thermal environmental pressure brought by *** this study,multi remote sensing imageries and statistical approaches,involving piecewise linear regression(PLR),were used to explore urban expansion and its effects on the thermal environment of Changchun City in Jilin Province,*** show that Changchun experienced rapid urban expansion from 2000 to 2020,with urban built-up areas increasing from 171.77 to 525.14 km^(2).The area of the city’s urban heat island(UHI)increased dramatically,during both day and *** PLR,a positive linear correlation of built-up density with land surface temperature(LST)was detected,with critical breakpoints of 70%-80%during the daytime and 40%-50%at *** the thresholds,the magnitude of LST in response to built-up density significantly increased with intensifying urbanization,especially for nighttime *** analysis of the relative frequency distributions(RFDs)of LST reveals that rapid urbanization resulted in a significant increase of mean LST in newly urbanized areas,but had weak effects on daytime LST change in existing urban *** expansion also contributed to a constant decrease of spatial heterogeneity of LST in existing urban area,especially at ***,in newly urbanized areas,the spatial heterogeneity of LST was decreased during the daytime but increased at nighttime due to urbanization.

Distinct evolution process among type I interferon in mammals

Interferon (IFN) is thought to play an important role in the vertebrate immune system, but systemic knowledge of IFN evolution has yet to be elucidated. To evaluate the phylogenic distribution and evolutionary history of type I IFNs, 13genomes were searched using BLASTn program, and a phylogenetic tree of vertebrate type I IFNs was constructed. In the present study, an IFNδ-like gene in the human genome was identified, refuting the concept that humans have no IFNδ genes, and other mammalian IFN genes were also identified. In the phylogenetic tree, the mammalian IFNβ, IFNɛ, and IFNκ formed a clade separate from the other mammalian type I IFNs, while piscine and avian IFNs formed distinct clades. Based on this phylogenetic analysis and the various characteristics of type I IFNs, the evolutionary history of type I IFNs was further evaluated. Our data indicate that an ancestral IFNα-like gene forms a core from which new IFNs divided during vertebrate evolution. In addition, the data suggest how the other type I IFNs evolved from IFNα and shaped the complex type I IFN system. The promoters of type I IFNs were conserved among different mammals, as well as their genic regions. However, the intergenic regions of type I IFN clusters were not conserved among different mammals, demonstrating a high selection pressure upon type I IFNs during their evolution.

Aptamer-induced in-situ growth of acetylcholinesterase-Cu_(3)(PO_(4))_(2)hybrid nanoflowers for electrochemical detection of organophosphorus inhibitors

Enzyme-inorganic hybrid nanoflowers(HNFs)have shown excellent sensing capabilities due to their large specific surface area as well as the simplicity and mildness of the preparation ***,coupling HNFs to electrodes to fabricate a uniform and controllable enzymatic electrochemical sensing interface remains a ***,we proposed an aptamer-induced insitu fabrication strategy for preparing an HNF-based electrochemical sensor with ideal *** to this strategy is the introduction of acetylcholinesterase(AChE)-specific binding aptamer(Apt),which induces the in-situ growth of AChE-copper phosphate(Cu_(3)(PO_(4))_(2))HNFs on the surface of carbon paper(CP).In addition,a dense gold nanoparticle(AuNP)layer was electrodeposited on the CP for anchoring Apt and further extending the electroactive surface *** prepared AChECu_(3)(PO_(4))_(2)HNF/Apt/AuNP/CP biosensor exhibited a wide detection range from 1 to 107 pM for the four organophosphorus inhibitors,including isocarbophos,dichlorvos,methamidophos,and parathion,with detection limits down to 0.016,0.028,0.071,and 0.113 pM,*** the reactivation of pralidoxime chloride,the electrode can still recover 98.1%of the response after five times of repeated *** real sample detection,the biosensor achieved high recoveries from 96.45%to 100.13%.The detection target may be extendable to other AChE inhibitors(e.g.,drugs for Alzheimer’s disease).This study demonstrates for the first time the feasibility of using aptamers as an inducer to fabricate an electrochemical enzyme sensing interface *** strategy can be used to fabricate other enzyme-based biosensors and therefore has broader applications.

Distribution of lanthanum among the chloroplast subcomponents of spinach and its biological effects on photosynthesis:location of the lanthanum binding sites in photosystem II

The effects of lanthanum at different concen-trations on the related photosynthetic activities of Hill reac-tion, Mg2+-ATPase and Ca2+-ATPase in spinach chloroplast were studied. Experimental results showed that lanthanum can increase all the activities at suitable concentration (15-30 mg·L-1), however, it behaves toxically on them when over used (60 mg·L-1). To get an improved understanding of the mechanism of lanthanum effects on the photosynthesis of spinach, the different subcomponents in the chloroplast of the cultured spinach were isolated, and the content of lan-thanum in each subcomponent was determined by ICP-MS. The results obtained indicated that among these different subcomponents, about 90% out of the total chloroplast lan-thanum was located in photosystem II (PS II) while there was little lanthanum in photosystem I (PS I). Moreover, size ex-clusion high performance liquid chromatography (SE-HPLC) coupled with online UV and ICP-MS detections was novelly used for locating lanthanum binding sites in PS II proteins for the first time. It was found that lanthanum has two bind-ing sites in PS II: La associates with chlorophyll together with magnesium in PS II by partly replacing magnesium and also shares the common binding sites of PS II proteins to-gether with the inorganic cofactors of calcium and manga-nese, influencing the process of photosynthesis.

Bioaccumulation and transformation of cadmium by Phaeodactylum tricornutum

In this paper,we report the bioaccumulation and transformation of cadmium (Cd) by Phaeodactylum tricornutum in the presence of ethylenediamine tetra acetic acid (EDTA) and cysteine (Cys). Both EDTA and Cys can alleviate the toxicity of Cd to P. tricornutum. Short term intracellular uptake and extracellular adsorption experiments using ICP-MS indicated that the amounts of Cd accumulated on the cell surface of P. tricornutum and inside the cell decreased along with the increase of EDTA concentration,which conformed to the prediction of the Free Ion Activity Model (FIAM). However,extracellular adsorption of Cd increased at first and then decreased along with the increase in the concentration of Cys,while intracellular uptake increased under Cys concentrations from the blank value to 4.45 μmol/L,and then tended to remain at the same level when the Cys concentration was greater than 4.45 μmol/L,and this deviated remarkably from the FIAM. The interactions of Cd with _Si_OH,_C_OH and NH2(CO) _OH on the cell wall were confirmed using FT-IR and XPS studies. The results obtained using HPLC of the phytochelatins (PCs) produced by P. tricornutum under CdCl2,Cd_EDTA and Cd_Cys stress suggested that the main reason for the different effects of EDTA and Cys on the bioaccumulation and transformation of Cd by P. tricornutum was that Cys is not only a complexing ligand to Cd,as is EDTA,but also it is a precursor of the intracellular synthesizing PCs participating in the cellular defense mechanism against Cd. Furthermore,the discovery of in vivo PCs and oxidized_PCs as well as Cd-PC2 in P. tricornutum using ESI-IT-MS provided the evidence for deactivation of Cd by the PCs,reducing Cd-toxicity to P. tricornutum.

Portable multi-amplified temperature sensing for tumor exosomes based on MnO_(2)/IR780 nanozyme with high photothermal effect and oxidase-like activity

Efficient determination of tumor exosomes using portable devices is crucial for the establishment of facile and convenient early cancer diagnostic methods. However, it is still challenging to effectively amplify the detection signal to achieve tumor exosomes detection with high sensitivity by portable devices. To address this issue, we developed a portable multi-amplified temperature sensing strategy for highly sensitive detecting tumor exosomes based on multifunctional manganese dioxide/IR780 nanosheets(MnO_(2)/IR780 NSs) nanozyme with high oxidase-like activity and enhanced photothermal ***, MnO_(2)/IR780 NSs were synthesized via a facile one-step method with mild experimental conditions, which not only exhibited a stronger photothermal effect than that of MnO_(2) but also showed excellent oxidase-like activity that can catalyze the oxidation of 3',3',5',5'-tetramethylbenzidine(TMB) to generate TMB oxide(oxTMB) with a robust photothermal property, thus conjoining with MnO_(2)/IR780 NSs to further enhance the temperature signal. The present assay enables highly sensitive determination of tumor exosomes with the detection limit down to 5.1×10~3 particles/mL, which was comparable or superior to those of the most previously reported sensors. Furthermore, detection of tumor exosomes spiked in biological samples was successfully realized. More importantly, our method showed the recommendable portability, robust applicability, and easy manipulation. By taking advantages of these features,this high-performance photothermal sensor offered a promising alternative means for nondestructive early cancer diagnosis and treatment efficacy evaluation.

Dengue virus(DENV)is a mosquito-borne virus with a rapid spread to humans,causing mild to potentially fatal illness in hundreds of millions of people each *** to the large number of serotypes of the virus,there remains an unmet need to develop protective vaccines for a broad spectrum of the ***,we constructed a modified mRNA vaccine containing envelope domain III(E-DIII)and non-structural protein 1(NS1)coated with lipid *** multi-target vaccine induced a robust antiviral immune response and increased neutralizing antibody titers that blocked all four types of DENV infection in vitro without significant antibodydependent enhancement(ADE).In addition,there was more bias for Th1 than Th2 in the exact E-DIII and NS1-specific T cell responses after a single ***,intramuscular immunization limited DENV transmission in vivo and eliminated vascular *** findings highlight that chimeric allogeneic structural and non-structural proteins can be effective targets for DENV vaccine and that they can prevent the further development of congenital DENV syndrome.

Selection and Verification of Reference Genes for qRT-PCR Analysis in Iris domestica under Drought

Studies of biouptake and transformation of mercury by a typical unicellular diatom Phaeodactylum tricornutum

Mercury(Hg) is a toxic heavy metal with its biogeochemical cycling in the ocean depending on the type and behavior of the oceanic *** present work aimed to evaluate bioaccumulation and transformation of Hg by Phaeodactylum tricornutum,a typical unicellular diatom,when exposed to the extremely high level of Hg in order to understand the possible mechanisms of acute stress *** can accumulate Hg(its bioaccumulation factor is at 104 level),and the 96 h EC 50 was estimated to be 145μg *** amounts of surface-bound Hg being about 1.2 to 4.8 times higher than those of intracellular Hg under exposure to HgCl 2(from 20 to 120μg L-1 concentrations) suggested that the cell wall of *** is an important "fence" towards *** entering the *** cell,Hg underwent transformation in its chemical form via interactions with high molecular weight sulfur-containing proteins(accounting for 68% of the intracellular Hg),and glutathione as well as the induced phytochelatins(PCs)(24% Hg) which alleviated the toxicity of *** addition,the existence of organic ligands greatly influenced the uptake and transformation behavior of *** towards HgCl2,especially in the case of cysteine(Cys),which increased the uptake of Hg,but alleviated the toxicity of Hg towards *** due to the fact that Cys is an important precursor for the synthesis of PCs inside the *** uptake process of Hg by *** was in agreement with the Freundlich isotherm,suggesting a typical heterogeneous sorption *** importantly,we observed the conversion of HgCl2 into methylmercury inside the *** cells and its release into the culture solution using HPLC/CVG-AFS and GC-MS,although the mechanism needs to be further investigated.
