Expression of oestrogen receptor-α and oestrogen receptor-β in prostate cancer

Background Recent studies have suggested that estrogens are involved in normal and abnormal prostate growth, though their exact role is still controversial. Oestrogens exert inhibitory and stimulatory effects on prostate gland, but the expression of oestrogen receptor-α （ERα） and oestrogen receptor-β （ERβ） in malignant prostate tissue remains unresolved. We determined ERa and ERβ in prostate cancer and investigated the relationship between expression of ER and pathological features of prostate carcinoma. Methods Thirty-two cases of prostate cancer, 12 cases of normal prostate tissue and 32 cases of benign prostate hyperplasia were analyzed for the expression of ERa and ERβ using semiquantitative, reverse transcription polymerase chain reaction （RT-PCR） and the products sequenced. Results Comparisons of the normal, hyperplastic and tumour prostate tissues indicated an overexpression of ERa in tumour specimens （P〈0.01）. However, the expression of ERβ significantly reduced in tumour tissues compared with normal and hyperplastic specimens （P〈0.01）, suggesting that severe pathological features of prostate cancer were associated with lower ERβ expression. Spearman analysis showed negative correlation between ERβ expression and tumour stage, grade （-0.67, -0.43, respectively, both P〈0.05）, and a positive correlation between ERα expression and tumour stage, grade （0.51, 0.57, respectively, both P〈0.01）. Our analysis also showed that hormone refractory, prostate cancer, compared with hormone dependent, prostate cancer, displayed a decreased expression of ERβ （P〈0.01） and an increased expression of ERa. Conclusions ERα and ERβ may play important roles in the development of prostate cancer. The decrease in ERβ expression is associated with higher Gleason grade tumours and prostate cancer with higher metastatic potential. The loss of ERβ could be one of the key processes leading to uncontrolled growth of prostate epithelial cells.

BACKGROUND:This study aimed to establish an eff ective nomogram to predict the survival of heat stroke(HS)based on risk ***:This was a retrospective,observational multicenter cohort *** analyzed patients diagnosed with HS,who were treated between May 1 and September 30,2018 at 15 tertiary hospitals from 11 cities in Northern ***:Among the 175 patients,32 patients(18.29%)died before hospital *** the univariate analysis,mechanical ventilation,initial mean arterial pressureping validation(B=1,000).Furthermore,the nomogram performed better when predicting 14-d survival,compared to 7-d *** prognostic index cut-off value was set at 2.085,according to the operating characteristic curve for overall survival *** model showed good calibration ability in the internal and external validation ***:A novel nomogram,integrated with prognostic factors,was proposed;it was highly predictive of the survival in HS patients.

We investigate high time resolution data obtained by the Gravitational wave high-energy Electromagnetic Counterpart All-sky Monitor(GECAM)during the flare event on 2022 April 21 at 01:52 *** subpeaks with durations of 4-6 s have been detected in the hard X-ray precursor phase,and the key feature is that they appear in pairs and seem like double-peak *** subpeaks are rarely observed in hard X-ray band and confirmed by the microwave obtained by Nobeyama Radio Polarimeters(NoRP)and Radio Solar Telescope Network(RSTN).While an exponential function can describe the continuum component of the time profile from the precursor to part of the impulsive *** periods of quasi-periodic pulsations(QPPs)are detected to be about 7.3 and 12.8 s for the precursor and impulsive phase,respectively,with at least 95%confidence *** paired QPPs are assumed to be double-peak QPPs and then the scenario of current loop coalescence model is found to be in good agreement with our *** precursor phase can be interpreted as the oscillating coalescence of two islands,while the impulsive phase can be interpreted as more islands to coalesce one by one to form larger islands.

The Moon provides a unique environment for investigating nearby astrophysical events such as *** samples retain valuable information from these events,via detectable long-lived“fingerprint”radionuclides such as^(60)*** this work,we stepped up the development of an accelerator mass spectrometry(AMS)method for detecting^(60)Fe using the HI-13tandem accelerator at the China Institute of Atomic Energy(CIAE).Since interferences could not be sufficiently removed solely with the existing magnetic systems of the tandem accelerator and the following Q3D magnetic spectrograph,a Wien filter with a maximum voltage of±60 kV and a maximum magnetic field of 0.3 T was installed after the accelerator magnetic systems to lower the detection background for the low abundance nuclide^(60)Fe.A 1μm thick Si_(3)N_(4) foil was installed in front of the Q3D as an energy *** particle detection,a multi-anode gas ionization chamber was mounted at the center of the focal plane of the ***,an^(60)Fe sample with an abundance of 1.125×10^(-10)was used to test the new AMS *** results indicate that^(60)Fe can be clearly distinguished from the isobar^(60)*** sensitivity was assessed to be better than 4.3×10^(-14)based on blank sample measurements lasting 5.8 h,and the sensitivity could,in principle,be expected to be approximately 2.5×10^(-15)when the data were accumulated for 100 h,which is feasible for future lunar sample measurements because the main contaminants were sufficiently separated.

The Bayan Obo supergiant carbonatite-related rare-earth-element-niobium-iron(REE-Nb-Fe) endogenetic deposit(thereafter as the Bayan Obo deposit), located at 150 km north of Baotou City in the Inner Mongolia Autonomous Region, is the largest rare-earth element(REE) resource in the world. Tectonically,this deposit is situated on the northern margin of the North China Craton and adjacent to the Xing’anMongolian orogenic belt to the south. The main strata within the mining area include the Neoarchean Se’ertengshan Group and the Mesoproterozoic Bayan Obo Group. Generally, the rare earth, niobium, and iron mineralization within the deposit are intrinsically related to the dolomite carbonatites and the extensive alteration of the country rocks caused by the carbonatite magma intrusion. The alteration of country rocks can be categorized into three types: contact metasomatism(anti-skarn and skarn alteration), fenitization,and hornfelsic alternation. As indicated by previous studies and summarized in this review, the multielement mineralization at Bayan Obo is closely associated with the metasomatic replacement of siliceous country rocks by carbonatite magmatic-hydrothermal fluids. The metasomatic process is comparable to the conventional skarnification that formed due to the intrusion of intermediate-acid magmatic rocks into limestone strata. However, the migration pattern of Si O2, Ca O, and Mg O in this novel metasomatic process is opposite to the skarn alteration. Accordingly, this review delineates, for the first time, an antiskarn metallogenic model for the Bayan Obo deposit, revealing the enigmatic relationship between the carbonatite magmatic-hydrothermal processes and the related iron and rare earth ***, this study also contributes to a better understanding of the REE-Nd-Fe metallogenetic processes and the related fluorite mineralization at the Bayan Obo deposit.

Methods: Thirty-two cases of PCa, 12 cases of normal prostate tissue (NOR) and 32 cases of benign prostate hyperplasia (BPH) were analyzed for the expression of ERα and ERβ by using semi-quantitative reverse transcription polymerase chain reaction (RT-PCR), and the products were sequenced. Results: The comparison of the normal, hyperplastic and tumor prostate tissues indicated an overexpression of ERa in tumor specimens (P<0.01) . On the contrary, the expression of ERβ was significantly reduced in tumor tissues compared with that in normal and hyperplastic specimens (P<0.01), suggesting that severe clinico-pathologic features of PCa were associated with lower ERβ expression. Spearmen analysis showed a significant negative correlation between ERβ expression and tumor stage and grade (P<0.05 ), and a significant positive correlation between ERα expression and tumor stage, grade, respectively (P<0.01) . Our analysis also showed that hormone-refractory PCa (HRPC), compare to hormone-dependent PCa (HDPC), displayed a decreased expression of ERβ (P<0.01) and an increased expression of ERα. Conclusions: The results suggest that ERα and ERβ may play an important role in the development of PCa. The loss or decrease of ERβ expression is associated with higher Gleason grade tumors and PCa with higher metastatic potential. Our data show that the loss of ERβ could be one of the key elements leading to uncontrolled growth of prostate epithelial cells.

Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains *** A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of *** Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two *** Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. Women with comorbidities were more likely to have severe disease, while men with comorbidities were more likely to have unfavorable treatment outcomes.

Hard X-ray Imager(HXI)is one of the three scientific instruments onboard the Advanced Spacebased Solar Observatory(ASO-S)mission,which is proposed for the 25th solar maximum by the Chinese solar *** is designed to investigate the non-thermal high-energy electrons accelerated in solar flares by providing images of solar flaring regions in the energy range from 30 keV to 200 *** imaging principle of HXI is based on spatially modulated Fourier synthesis and utilizes about 91 sets of bi-grid sub-collimators and corresponding LaBr3 detectors to obtain Fourier components with a spatial resolution of about 3 arcsec and a time resolution better than 0.5 *** engineering prototype has been developed and tested to verify the feasibility of *** this paper,we present background,instrument design and the development and test status of the prototype.

Recent evidence suggests that CD147 serves as a novel receptor for severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)*** CD147 via anti-CD147 antibody could suppress the in vitro SARS-CoV-2 *** is a humanized anti-CD147 IgG_(2) monoclonal antibody,which may effectively prevent SARS-CoV-2 infection in coronavirus disease 2019(COVID-19)***,we conducted a randomized,double-blinded,placebo-controlled phase 1 trial to evaluate the safety,tolerability,and pharmacokinetics of meplazumab in healthy subjects,and an open-labeled,concurrent controlled add-on exploratory phase 2 study to determine the efficacy in COVID-19 *** phase 1 study,59 subjects were enrolled and assigned to eight cohorts,and no serious treatment-emergent adverse event(TEAE)or TEAE grade≥3 was *** serum and peripheral blood Cmax and area under the curve showed non-linear pharmacokinetic *** obvious relation between the incidence or titer of positive anti-drug antibody and dosage was observed in each *** biodistribution study indicated that meplazumab reached lung tissue and maintained>14 days stable with the lung tissue/cardiac blood-pool ratio ranging from 0.41 to *** the exploratory phase 2 study,17 COVID-19 patients were enrolled,and 11 hospitalized patients were involved as concurrent *** meplazumab treatment significantly improved the discharged(P=0.005)and case severity(P=0.021),and reduced the time to virus negative(P=0.045)in comparison to the control *** results show a sound safety and tolerance of meplazumab in healthy volunteers and suggest that meplazumab could accelerate the recovery of patients from COVID-19 pneumonia with a favorable safety profile.

There is considerable potential for integrating transarterial chemoembolization(TACE),programmed death-(ligand)1(PD-[L]1)inhibitors,and molecular targeted treatments(MTT)in hepatocellular carcinoma(HCC).It is necessary to investigate the therapeutic efficacy and safety of TACE combined with PD-(L)1 inhibitors and MTT in real-world *** this nationwide,retrospective,cohort study,826 HCC patients receiving either TACE plus PD-(L)1 blockades and MTT(combination group,n=376)or TACE monotherapy(monotherapy group,n=450)were included from January 2018 to May *** primary endpoint was progression-free survival(PFS)according to modified *** secondary outcomes included overall survival(OS),objective response rate(ORR),and *** performed propensity score matching approaches to reduce bias between two *** matching,228 pairs were included with a predominantly advanced disease *** PFS in combination group was 9.5 months(95%confidence interval[CI],8.4-11.0)versus 8.0 months(95%CI,6.6-9.5)(adjusted hazard ratio[HR],0.70,P=0.002).OS and ORR were also significantly higher in combination group(median OS,19.2[16.1-27.3]vs.15.7 months[13.0-20.2];adjusted HR,0.63,P=0.001;ORR,60.1%vs.32.0%;P<0.001).Grade 3/4 adverse events were observed at a rate of 15.8%and 7.5%in combination and monotherapy groups,*** results suggest that TACE plus PD-(L)1 blockades and MTT could significantly improve PFS,OS,and ORR versus TACE monotherapy for Chinese patients with predominantly advanced HCC in real-world practice,with an acceptable safety profile.
