AIM To investigate the role of apolipoprotein E (apoE) polymorphism in the lithogenesis of gallstone and the hereditary pathogenesis of the *** Polymerase chain reaction (PCR)was used to study apoE phenotypes and allelefrequencies in patients with gallstones and control, and the fasting serum lipids of subjectswere also measured by enzymatic *** The levels of triglyceride (TG) andvery low density lipoprotein cholesterol (VLDLC) were much higher in Ez/, patients than that inE,/, control. E,/, patients were accompanied withremarkably low levels of high density lipoproteincholesterol (HDLC) and its subforms. But in E,/#patients there were only slight changes in levelsof VLDLC and low density lipoprotein cholesterol (LDL--C).CONCluSION Different apoE phenotype patientswith gallstones have different cheracteristics ofdyslipidemia and the average level of serum lipids in patients with gallstones are higher thansubjects without gallstones in the same apoEgene phenotype. EZ allele is possibly one of thedangerous factors in the lithogenesis of cholecystolithiasis.

Diffuse-type tenosynovial giant cell tumor(dTGCT) is a destructive but rare benign proliferative synovial neoplasm. Although surgery is currently the main treatment modality for dTGCT, the recurrence risk is up to 50%. Therefore, there is a great need for effective drugs against dTGCT with minor side effects. The Janus kinase(JAK)/signal transducer and activator of transcription(STAT) signaling plays a central role in rheumatoid arthritis(RA), a disease with similar characteristics as dTGCT, but its function in dTGCT remains *** fibroblast-like synoviocytes(FLS) and macrophages were isolated from 10 synovial tissue samples from dTGCT patients for the screening and validation of the five clinically approved JAK inhibitors to treat RA against dTGCT. Cell viability, cell death, inflammation and the activity of the JAK family members of cultured dTGCT FLS(both 2-D and 3-D) and macrophages were investigated for the efficacy of the JAK inhibitors. Here, we found that similar to RA, JAK/STAT signaling was markedly activated in the dTGCT synovium. Of the 5 JAK inhibitors, peficitinib was shown to have the most potency in addressing some of the pathological responses of dTGCT FLS and *** potency of peficitinib was much higher than pexidartinib, which is the only FDA-approved drug for dTGCT. Mechanistically, peficitinib inhibited tyrosine kinase 2(TYK2), a JAK family member necessary for the pathological progression of dTGCT FLS and macrophages. In summary, we not only revealed JAK/STAT(especially TYK2) signaling as the major mechanism underlying dTGCT, but also identified peficitinib as a promising drug against dTGCT.

BACKGROUND： The elevation of neutrophil-lymphocyte ratio （NLR） has adverse effects on the prognosis of patients with hepatocellular carcinoma （HCC） who have received liver transplantation （LT）. The Hangzhou criteria are set for selecting HCC patients for LT. Pure silica zeolite has become an important porous material in the chemical industry due to its excellent stability and hydrophobicity. However, there are still some problems in the synthesis process of pure silica zeolite, such as environmental pollution, complex operation and high cost. How to effectively and environmentally synthesize pure silica zeolite still remains a significant challenge. This review summarizes the pure silica zeolitetype frameworks that have been discovered currently, introduces the progresses achieved in the synthesis of pure silica zeolite and prospects the areas for future exploration in the synthesis and development of pure silica zeolite.

AIM:To assess systematically the safety and efficacy of bile leakage test in liver ***:Randomized controlled trials and controlled clinical trials involving the bile leakage test were included in a systematic literature *** authors independently assessed the studies for inclusion and extracted the data.A meta-analysis was conducted to estimate postoperative bile leakage,intraoperative positive bile leakage,and *** used either the fixed-effects or random-effects ***:Eight studies involving a total of 1253 patients were included and they all involved the bile leakage test in liver *** bile leakage test group was associated with a significant reduction in bile leakage compared with the non-bile leakage test group(RR=0.39,95%CI:0.23-0.67;I2=3%).The white test had superiority for detection of intraoperative bile leakage compared with the saline solution test(RR=2.38,95%CI:1.24-4.56,P=0.009).No significant intergroup differences were observed in total number of complications,ileus,liver failure,intraperitoneal hemorrhage,pulmonary disorder,abdominal infection,and wound ***:The bile leakage test reduced postoperative bile leakage and did not increase incidence of *** emulsion is the best choice of solution for the test.

Dissecting the genetic basis of grain color and pre-harvest sprouting resistance in common wheat by association analysis

Pre-harvest sprouting(PHS) adversely affects wheat quality and yield, and grain color(GC) is associated with PHS resistance. However, the genetic relationship between GC and PHS resistance remains unclear. In this study, 168 wheat varieties(lines) with significant differences in GC and PHS resistance were genotyped using an Illumina 90K iSelect SNP array. Genome-wide association study(GWAS) based on a mixed linear model showed that 67 marker-trait associations(MTAs) assigned to 29 loci, including 17 potentially novel loci, were significantly associated with GC, which explained 1.1–17.0% of the phenotypic variation. In addition, 100 MTAs belonging to 54 loci, including 31 novel loci, were significantly associated with PHS resistance, which accounted for 1.1–14.7% of the phenotypic variation. Subsequently, two cleaved amplified polymorphic sequences(CAPS) markers, 2B-448 on chromosome 2B and 5B-301 on chromosome 5B,were developed from the representative SNPs of the major common loci ***-2B.3/***-2B.4controlling GC/PHS resistance and PHS resistance locus ***-5B.4, respectively. Further validation in 171 Chinese mini-core collections confirmed significant correlations of the two CAPS markers with GC and PHS resistance phenotypes under different environments(Plutaredoxin, as a potential candidate gene linked to ***-5B.4. The new CAPS marker CAPS-356 was then developed based on the SNP(T/C) in the coding sequences(CDS) region of TraesCS5B02G545100. The high-density linkage map of the Jing 411/Hongmangchun 21 recombinant inbred lines(RILs) constructed based on specific locus amplified fragment sequencing markers showed that CAPS-356 collocated with a novel QTL for PHS resistance, supporting the role of TraesCS5B02G545100 as the potential candidate gene linked to ***-5B.4. Development and validation of a predictive score for perioperative transfusion in patients with hepatocellular carcinoma undergoing liver resection

Liver resection is a major surgery requiring perioperative blood transfusion. Predicting the need for blood transfusion for patients undergoing liver resection is of great importance. The present study aimed to develop and validate a model for predicting transfusion requirement in HBV-related hepatocellular carcinoma patients undergoing liver resection. METHODS： A total of 1543 consecutive liver resections were included in the study. Randomly selected sample set of 1080 cases （70% of the study cohort） were used to develop a predic- tive score for transfusion requirement and the remaining 30% （n=463） was used to validate the score. Based on the preopera- tive and predictable intraoperative parameters, logistic regres- sion was used to identify risk factors and to create an integer score for the prediction of transfusion requirement. RESULTS： Extrahepatic procedure, major liver resection, hemoglobin level and platelets count were identified as inde- pendent predictors for transfusion requirement by logistic regression analysis. A score system integrating these 4 factors was stratified into three groups which could predict the risk of transfusion, with a rate of 11.4%, 24.7% and 57.4% for low, moderate and high risk, respectively. The prediction model appeared accurate with good discriminatory abilities, generat- ing an area under the receiver operating characteristic curve of 0.736 in the development set and 0.709 in the validation set. CONCluSIONS： We have developed and validated an integer- based risk score to predict perioperative transfusion for pa- tients undergoing liver resection in a high-volume surgicalcenter. This score allows identifying patients at a high risk and may alter transfusion practices.
