Neuroprotective effect of the ethanol extract of Artemisia capillaris on transient forebrain ischemia in mice via nicotinic cholinergic receptor

Artemisia capillaris Thunberg is a medicinal plant used as a traditional medicine in many cultures. It is an effective remedy for liver problems including hepatitis. Recent pharmacological reports have indicated that Artemisia species can exert various neurological effects. Previously, we reported a memory-enhancing effect of Artemisia species. However, the mechanisms underlying the neuroprotective effect of A. capillaris(AC) are still unknown. In the present study, we investigated the effect of an ethanol extract of AC on ischemic brain injury in a mouse model of transient forebrain ischemia. The mice were treated with AC for seven days, beginning one day before induction of transient forebrain ischemia. Behavioral deficits were investigated using the y-maze. Nissl and Fluoro-jade B staining were used to indicate the site of injury. To determine the underlying mechanisms for the drug, we measured acetylcholinesterase activity. AC(200 mg·kg-1) treatment reduced transient forebrain ischemia-induced neuronal cell death in the hippocampal CA1 region. The AC-treated group also showed significant amelioration in the spontaneous alternation of the y-maze test performance, compared to that in the untreated transient forebrain ischemia group. Moreover, AC treatment showed a concentration-dependent inhibitory effect on acetylcholinesterase activity in vitro. Finally, the effect of AC on forebrain ischemia was blocked by mecamylamine, a nonselective nicotinic acetylcholine receptor antagonist. Our results suggested that in a model of forebrain ischemia, AC protected against neuronal death through the activation of nicotinic acetylcholine receptors.

Ecological evaluation of marine macroalgal communities on five islands of Korea in the yellow Sea

Macroalgae have long been used as biological indicators of marine ecosystem health worldwide due to their ecological importance and sensitivity to environmental stress.A number of previous studies have utilized macroalgal communities in monitoring surveys of environmental *** study examined the characteristics and patterns of marine macroalgal communities in the yellow Sea off the western coast of *** were analyzed for the number of species,biomass,and coverage ratio by macroalgal *** the study period,82 macroalgal species(10 green algae,17 brown algae,and 55 red algae)were identified at the five study sites,with the highest number of species found at Gwanrido and Uido(both containing 41 species)and the lowest at Daeijakdo(27 species).The average biomass(via dry weight)was 98.63 g/m^(2),consisting of green algae(8.39 g/m^(2)),brown algae(35.08 g/m^(2)),and red algae(55.16 g/m^(2)).The dominant macroalgae species in terms of biomass were Corallina pilulifera,Sargassum thunbergii,and Ulva australis in the intertidal zones,and Botryocladia wrightii and Gelidium elegans in the subtidal ***,evenness,and diversity indices based on the biomass of abundant species were 5.08,0.65,and 2.30,respectively,over the entire study *** on the evaluation of the environmental states by the community indices,overall,the Ecological Evaluation Index of macroalgae communities in the study area was marked as“Good-Moderate”,but was determined as“ModerateLow”at several sites during *** results can be a direct approach in the assessment of coastal habitats in which anthropogenic as well as climate change influences persist.

Chikungunya fever is a vector-borne viral disease transmitted to humans by chikungunya virus(CHIKV)-infected *** have been many outbreaks of CHIKV infection worldwide,and the virus poses ongoing risks to global *** prevent and control CHIKV infection,it is important to improve the current CHIKV diagnostic approaches to allow for the detection of low CHIKV concentrations and to correctly distinguish CHIKV infections from those due to other mosquito-transmitted viruses,including dengue virus(DENV),Japanese encephalitis virus(JEV),and Zika virus(ZIKV).Here,we produced monoclonal antibodies(mAbs)against the CHIKV envelope 2 protein(CHIKV-E2)and compared their sensitivity and specificity with commercially available mAbs using enzyme-linked immunosorbent assays(ELISA).Two anti-CHIKV-E2 mAbs,19-1 and 21-1,showed higher binding affinities to CHIKV-E2 protein than the commercial mAbs *** particular,the 19-1 mAb had the strongest binding affinity to inactivated ***,the 19-1 mAb had very little cross-reactivity with other mosquito-borne viruses,such as ZIKV,JEV,and *** results suggest that the newly produced anti-CHIKV-E2 mAb,19-1,could he used for CHIKV diagnostic approaches.

Pollen tube growth is essential for successful double fertilization, which is critical for grain yield in crop plants. Rapid alkalinization factors(RALFs) function as ligands for signal transduction during fertilization. However, functional studies on RALF in monocot plants are ***, we functionally characterized two pollen-specific RALFs in rice(Oryza sativa) using multiple clustered regularly interspaced palindromic repeats(CRISPR)/CRISPR-associated protein 9-induced loss-of-function mutants,peptide treatment, expression analyses, and tag reporter lines. Among the 41 RALF members in rice, OsRALF17 was specifically expressed at the highest level in pollen and pollen *** applied OsRALF17 or OsRALF19peptide inhibited pollen tube germination and elongation at high concentrations but enhanced tube elongation at low concentrations, indicating growth regulation. Double mutants of OsRALF17 and OsRALF19(ralf17/19) exhibited almost full male sterility with defects in pollen hydration, germination, and tube elongation,which was partially recovered by exogenous treatment with OsRALF17 peptide. This study revealed that two partially functionally redundant OsRALF17 and OsRALF19 bind to Oryza sativa male-gene transfer defective 2(OsMTD2) and transmit reactive oxygen species signals for pollen tube germination and integrity maintenance in rice. Transcriptomic analysis confirmed their common downstream genes, in osmtd2 and ralf17/19. This study provides new insights into the role of RALF, expanding our knowledge of the biological role of RALF in regulating rice fertilization.

Evaluation of Estrogenic Activity and Measurement of EDCs in Wastewater Treatment Plants

Correlations between estrogenic activity and DOC/UV260 ratio in wastewater treatment processes were investigated to propose a simple, reliable and comprehensive indicator for the presence of estrogenic substances. Contrary to this, when short-term bioassays such as the E-SCREEN, receptor binding and reporter gene expression assays are used for detecting estrogenic activity in the wastewater sample, they require a long time, at least a few days. The major factors contributing to the estrogenic activity were found to be 17β-estradiol （E2） and estrone （El）. A good relationship between the DOC/ UV260 ratio and the concentration of estrogens （El and E2） in the effluent of the activated sludge process was found： the E2 concentration increased as the DOC/UV260 ratio increased while the E1 concentration decreased. The relative estrogenic activity and DOC/UV260 ratio showed a good correlation （R^2 = 0.84） for all sewage samples except the ozonized samples in the sewage treatment plants. This study shows that the estrogenic compounds are hard to be mineralized by the conventional biological processes. Advanced oxidation processes are required to further remove estrogenic substances in the secondary effluent. By analysis of DOC and UV260, the estrogenic activity in the wastewater can be rapidly estimated.

Lithospheric thinning and ignition of a Cordilleran magmatic flare-up:Geochemical and O-Hf isotopic constraints from Cretaceous plutons in southern Korea

Northeast Asian continental margins contain the products of magma emplacement driven by prolonged subduction of the(paleo-)Pacific *** observed in many Cordilleran arcs,magmatic evolution in this area was punctuated by high-volume pulses amid background *** present study investigates the early evolution of the Cretaceous magmatic flare-up using new and published geochronological,geochemical,and O-Hf isotope data from plutonic rocks in the southern Korean *** a long(~50 m.y.)magmatic hiatus and the development of the Honam Shear Zone through flat-slab subduction,the Cretaceous flare-up began with the intrusion of monzonites,granodiorites,and granites in the inboard Gyeonggi Massif and the intervening Okcheon *** to Jurassic granitoids formed during the former flare-up,Albian(~111 Ma)monzonites found in the Eopyeong area of the Okcheon Belt have distinctly higher zirconε_(Hf)(t)(-7.5±1.3)andδ^(18)O(7.78‰±0.25‰)values and lower wholerock La/yb and Sr/y *** voluminous coeval granodiorite and granite plutons in the Gyeonggi Massif are further reduced in Sr/y and to a lesser extent,in La/yb,and have higher zirconε_(Hf)(t)values(-13 to-19)than the Precambrian basement(ca.-30).These chemical and isotopic features indicate that Early Cretaceous lithospheric thinning,most likely resulting from delamination of tectonically and magmatically overthickened lithospheric keel that was metasomatized during prior subduction episodes,and consequent asthenospheric upwelling played vital roles in igniting the magmatic *** O-Hf isotopic ranges of synmagmatic zircons from the Albian plutons and their Paleoproterozoic and Jurassic inheritance attest to the involvement of lithospheric mantle and crustal basement in magma generation during this decratonization *** magmatism then migrated trenchward and culminated in the Late Cretaceous,yielding widespread granitoid rocks emplaced at shallow crustal *** early Late Cretaceous(94-8

Thyroid peroxidase antibody positivity and triiodothyronine levels are associated with pediatric Graves'ophthalmopathy
