miR-18a has been identified as a significantly expressed microRNA(miRNA)in non-small cell lung cancer(NSCLC)and plays a vital role in cancer cell transformation,metastasis,and carcinogenesis.Herein,a pair of binary probes from numerous probe pairs based on single nucleotide polymorphism analyses of miR-18a and miR-18b was first designed and screened to develop a Y-shaped ratio biosensor for accurate detection of serum miR-18a in NSCLC.The special structure of the binary probes combined with the hairpin showed strong specificity for miR-18a,which was confirmed by polyacrylamide gel electrophoresis assay and square wave voltammetry assay.Furthermore,it is beneficial to immobilize single-stranded DNA(ssDNA)probes due to the large specific surface area of nanoporous gold,thereby improving the sensitivity of the biosensor.The Y-shaped ratio biosensor exhibited a wide detection range and can quantify the concentration of miR-18a in the range of 10 fmol/L–100 pmol/L,with a limit of detection of 0.211 fmol/L(S/N=3).Moreover,it exhibits excellent detection capabilities in serum samples since the biosensor showed a high selectivity toward the coexistence of miR-18a and miR-18b.Therefore,the prepared Y-shaped ratio biosensor is a highly sensitive and specific miR-18a detection tool,capable of identifying microscopic amounts of miR-18a in serum samples,providing great potential for early screening of NSCLC.

Free-moving-state microscopic imaging of cerebral oxygenation and hemodynamics with a photoacoustic fiberscope

We report the development of a head-mounted photoacoustic fiberscope for cerebral imaging in a freely behaving mouse.The 4.5-gram imaging probe has a 9-µm lateral resolution and 0.2-Hz frame rate over a 1.2-mm wide area.The probe can continuously monitor cerebral oxygenation and hemodynamic responses at single-vessel resolution,showing significantly different cerebrovascular responses to external stimuli under anesthesia and in the freely moving state.For example,when subjected to high-concentration CO_(2) respiration,enhanced oxygenation to compensate for hypercapnia can be visualized due to cerebral regulation in the freely moving state.Comparative studies exhibit significantly weakened compensation capabilities in obese rodents.This new imaging modality can be used for investigating both normal and pathological cerebrovascular functions and shows great promise for studying cerebral activity,disorders and their treatments.

Submerged entry nozzle(SEN)clogging during continuous casting of Ti-stabilized ultra-pure ferritic stainless(Ti-UPFS)steels was systematically investigated via cross-sectional analysis and acid dissolution treatment.The SEN deposit profile was characterized as occurring in three major layers:(1)an eroded refractory layer;(2)an initial adhesive layer comprised an Al_(2)O_(3)-ZrO_(2) composite sub-layer and a dense Al_(2)O_(3)-based deposit sub-layer;and(3)a porous multiphase deposit layer mainly consisting of MgO·Al_(2)O_(3),CaO-Al_(2)O_(3),and CaO-TiOx.The MgO·Al_(2)O_(3)-rich inclusions did not adhere directly to the eroded refractory but were entrapped during the deposit growth.Results of inclusion characterization in the tundish revealed that the MgO·Al2O3-rich particles present in the tundish served as the primary source of clogging deposits.Furthermore,a novel cavity-induced adhesion model by circular approximation was established to explain the effects of complex inclusion characteristics and refractory material type on adhesion force.A high number of small MgO·Al_(2)O_(3) inclusions were expected to accelerate the buildup of clogging deposits.Improving the modification of MgO·Al_(2)O_(3)-rich inclusions in the size range of 2-4μm by Ca treatment was crucial to minimizing the risk of SEN clogging during the continuous casting of Ti-UPFS steels.

Improving the catalytic activity of non-noble metal single atom catalysts(SACs)has attracted considerable attention in materials science.Although optimizing the local electronic structure of single atom can greatly improve their catalytic activity,it often involves in-plane modulation and requires high temperatures.Herein,we report a novel strategy to manipulate the local electronic structure of SACs via the modulation of axial Co-S bond anchored onto graphitic carbon nitride(C_(3)N_(4))at room temperature(RT).Each Co atom is bonded to four N atoms and one S atom(Co-(N,S)/C_(3)N_(4)).Owing to the greater electronegativity of S in the Co-S bond,the local electronic structure of the Co atoms is available to be controlled at a relatively moderate level.Consequently,when employed for the photocatalytic hydrogen evolution reaction,the adsorption energy of intermediate hydrogen(H*)on the Co atoms is remarkably low.In the presence of the Co-(N,S)/C_(3)N_(4)SACs,the hydrogen evolution rates reach up to 10 mmol/(g·h),which is nearly 10 and 2.5 times greater than the rates in the presence of previously reported transition metal/C_(3)N_(4)and noble platinum nanoparticles(PtNPs)/C_(3)N_(4)catalysts,respectively.Attributed to the tailorable axial Co-S bond in the SAC,the local electronic structure of the Co atoms can be further optimized for other photocatalytic reactions.This axial coordination engineering strategy is universal in catalyst designing and can be used for a variety of photocatalytic applications.

Since electromagnetic pollution is detrimental to human health and the environment, numerous efforts have been successively made to achieve excellent electromagnetic interference shielding effectiveness(EMI SE) via designing the hierarchical structures for electromagnetic interference(EMI) shielding polymer composites. Among the plentiful structures, the asymmetric structures are currently a hot spot, principally categorizing into multi-layered, porous, fibrous, and segregated asymmetric structures, which endows the high EMI shielding performance for polymer composites incorporated with magnetic, conductive, and/or dielectric micro/nano-fillers, due to the “absorption-reflection-reabsorption” shielding mechanism. Therefore, this review provides the retrospection and summary of the efforts with respect to abundant asymmetric structures and multifunctional micro/nano-fillers for enhancing EMI shielding properties, which is conducive to the booming development of polymeric EMI shielding materials for the promising prospect in modern electronics and 5-generation(5G) technology.

In this paper, the problem of time-varying aerodynamic parameters identification under measurement noises is studied. By analyzing the key aerodynamic parameters that affect the aircraft control system, a system model with extended states for identifying equivalent aerodynamic parameters is established, and error parameters are extended to the system state, avoiding the difficulty caused by the unknown dynamic in the system. Furthermore, an identification algorithm based on extended state Kalman filter is designed, and it is proved that the algorithm has quasi-consistency, thus, the estimation error can be evaluated in real time. Finally, the simulation results under typical flight scenarios show that the designed algorithm can accurately identify aerodynamic parameters, and has desired convergence speed and convergence precision.

In-situ upgrading by heating is feasible for low-maturity shale oil, where the pore space dynamically evolves. We characterize this response for a heated substrate concurrently imaged by SEM. We systematically follow the evolution of pore quantity, size(length, width and cross-sectional area), orientation, shape(aspect ratio, roundness and solidity) and their anisotropy—interpreted by machine learning. Results indicate that heating generates new pores in both organic matter and inorganic minerals. However, the newly formed pores are smaller than the original pores and thus reduce average lengths and widths of the bedding-parallel pore system. Conversely, the average pore lengths and widths are increased in the bedding-perpendicular direction. Besides, heating increases the cross-sectional area of pores in low-maturity oil shales, where this growth tendency fluctuates at 300°C. In addition, the orientation and shape of the newly-formed heating-induced pores follow the habit of the original pores and follow the initial probability distributions of pore orientation and shape. Herein, limited anisotropy is detected in pore direction and shape, indicating similar modes of evolution both bedding-parallel and bedding-normal. We propose a straightforward but robust model to describe evolution of pore system in low-maturity oil shales during heating.

The combination of donor-acceptor(D-A)structures presents a viable strategy for fabricating covalent organic frameworks(COFs)with exceptional photocatalytic performances.Nevertheless,the selection of functional groups on donor or acceptor building blocks and their effect on the macroscopic properties of COFs are ambiguous.In this study,we tactfully synthesized a pair of Py-DBT-COFs from the same pyrene(Py)donor and 4,7-diphenylbenzo[c][1,2,5]thiadiazole(DBT)acceptor cores with distinct primitive functional groups.The primitive functional groups of building units determine the photocatalytic properties of corresponding Py-DBT-COFs.Specifically,Py-C-DBT-COF synthesized from Py-4CHO and DBT-2NH_(2)showcases a splendid H_(2)evolution rate as high as 21,377.7μmol/(g·h)(with 5 wt.%Pt)originating from better charge transfer capacity,which is significantly superior to that of Py-N-DBT-COF constructed from Py-4NH_(2)and DBT-2CHO.The distinct photocatalytic performances of the two COFs are demonstrated to originate from the different charge separation and transfer capabilities.This work supplies a new avenue for optimizing the photocatalytic performance of D-A COFs from the perspective of primitive functional group selections.

BACKGROUND MicroRNAs(miRNAs)regulate gene expression and play a critical role in cancer physiology.However,there is still a limited understanding of the function and regulatory mechanism of miRNAs in gastric cancer(GC).AIM To investigate the role and molecular mechanism of miRNA-145-5p(miR145-5p)in the progression of GC.METHODS Real-time polymerase chain reaction(RT-PCR)was used to detect miRNA expression in human GC tissues and cells.The ability of cancer cells to migrate and invade was assessed using wound-healing and transwell assays,respectively.Cell proliferation was measured using cell counting kit-8 and colony formation assays,and apoptosis was evaluated using flow cytometry.Expression of the epithelial-mesenchymal transition(EMT)-associated protein was determined by Western blot.Targets of miR-145-5p were predicated using bioinformatics analysis and verified using a dual-luciferase reporter system.Serpin family E member 1(SERPINE1)expression in GC tissues and cells was evaluated using RT-PCR and immunohistochemical staining.The correlation between SERPINE1 expression and overall patient survival was determined using Kaplan-Meier plot analysis.The association between SERPINE1 and GC progression was also tested.A rescue experiment of SERPINE1 overexpression was conducted to verify the relationship between this protein and miR-145-5p.The mechanism by which miR-145-5p influences GC progression was further explored by assessing tumor formation in nude mice.RESULTS GC tissues and cells had reduced miR-145-5p expression and SERPINE1 was identified as a direct target of this miRNA.Overexpression of miR-145-5p was associated with decreased GC cell proliferation,invasion,migration,and EMT,and these effects were reversed by forcing SERPINE1 expression.Kaplan-Meier plot analysis revealed that patients with higher SERPINE1 expression had a shorter survival rate than those with lower SERPINE1 expression.Nude mouse tumorigenesis experiments confirmed that miR-145-5p targets SERPINE1 to regulate extracellular signal-regu

Heterogeneous body compositions and all-cause mortality in acute coronary syndrome patients:a ten-year retrospective cohort study

BACKGROUND The association of different body components,including lean mass and body fat,with the risk of death in acute coronary syndrome(ACS)patients are unclear.METHODS We enrolled adults diagnosed with ACS at our center between January 2011 and December 2012 and obtained fol-low-up outcomes via telephone questionnaires.We used restricted cubic splines(RCS)with the Cox proportional hazards model to analyze the associations between body mass index(BMI),predicted lean mass index(LMI),predicted body fat percentage(BF),and the value of LMI/BF with 10-year mortality.We also examined the secondary outcome of death during hospitalization.RESULTS During the maximum 10-year follow-up of 1398 patients,331 deaths(23.6%)occurred,and a U-shaped relationship was found between BMI and death risk(P_(nonlinearity)=0.03).After adjusting for age and history of diabetes,the overweight group(24≤BMI<28 kg/m^(2))had the lowest mortality(HR=0.53,95%CI:0.29-0.99).Predicted LMI and LMI/BF had an inverse linear relationship with a 10-year death risk(P_(nonlinearity)=0.24 and P_(nonlinearity)=0.38,respectively),while an increase in BF was associ-ated with increased mortality(P_(nonlinearity)=0.64).During hospitalization,31 deaths(2.2%)were recorded,and the associations of the indicators with in-hospital mortality were consistent with the long-term outcome analyses.CONCLUSION Our study provides new insight into the“obesity paradox”in ACS patients,highlighting the importance of considering body composition heterogeneity.Predicted LMI and BF may serve as useful tools for assessing nutritional status and predicting the prognosis of ACS,based on their linear associations with all-cause mortality.
