Chronic pancreatitis(CP)is a progressive and irreversible fibroinflammatory disorder,accompanied by pancreatic exocrine insufficiency and dysregulated gut microbiota.Recently,accumulating evidence has supported a correlation between gut dysbiosis and CP development.However,whether gut microbiota dysbiosis contributes to CP pathogenesis remains unclear.Herein,an experimental CP was induced by repeated high-dose caerulein injections.The broad-spectrum antibiotics(ABX)and ABX targeting Gram-positive(G+)or Gram-negative bacteria(G-)were applied to explore the specific roles of these bacteria.Gut dysbiosis was observed in both mice and in CP patients,which was accompanied by a sharply reduced abundance for short-chain fatty acids(SCFAs)-producers,especially G+bacteria.Broad-spectrum ABX exacerbated the severity of CP,as evidenced by aggravated pancreatic fibrosis and gut dysbiosis,especially the depletion of SCFAs-producing G+bacteria.Additionally,depletion of SCFAs-producing G+bacteria rather than G-bacteria intensified CP progression independent of TLR4,which was attenuated by supplementation with exogenous SCFAs.Finally,SCFAs modulated pancreatic fibrosis through inhibition of macrophage infiltration and M2 phenotype switching.The study supports a critical role for SCFAs-producing G+bacteria in CP.Therefore,modulation of dietary-derived SCFAs or G+SCFAs-producing bacteria may be considered a novel interventive approach for the management of CP.

This paper presents a ranked differential evolution(RDE) algorithm for solving the identification problem of nonlinear discrete-time systems based on a Volterra filter model. In the improved method, a scale factor, generated by combining a sine function and randomness, effectively keeps a balance between the global search and the local search. Also, the mutation operation is modified after ranking all candidate solutions of the population to help avoid the occurrence of premature convergence. Finally, two examples including a highly nonlinear discrete-time rational system and a real heat exchanger are used to evaluate the performance of the RDE algorithm and five other approaches. Numerical experiments and comparisons demonstrate that the RDE algorithm performs better than the other approaches in most cases.

Objective: To investigate the anti-inflammatory activity of Radix Panacis quinquefolii extract(RPQE) and its therapeutic effects on inflammatory bowel disease(IBD). Methods: The 72-hour post-fertilization zebrafish was used to generate the local and systematic inflammation models through tail-amputation and lipopolysaccharide(LPS)-induction(100 μg/m L), respectively. The Tg(zlyz:EGFP) zebrafish was induced with 75 μg/m L 2,4,6-trinitrobenzene sulfonic acid(TNBS) for establishing the IBD model. The tail-amputated, LPS-, and TNBS-induced models were subjected to RPQE(ethanol fraction, 10–20 μg/m L) administration for 12 and 24 h, respectively. Anti-inflammatory activity of RPQE was evaluated by detecting migration and aggregation of leukocytes and expression of inflammation-related genes. Meanwhile, TNBS-induced fish were immersed in 0.2%(W/V) calcein for 1.5 h and RPQE for 12 h before photographing to analyze the intestinal efflux efficiency(IEE). Moreover, the expression of inflammation-related genes in these fish was detected by quantitative polymerase chain reaction. Results: Subject to RPQE administration, the migration and aggregation of leukocytes were significantly alleviated in 3 zebrafish models(P<0.01). Herein, RPQE ameliorated TNBS-induced IBD with respect to a significantly reduced number of leukocytes, improved IEE, and inhibited gene expression of pro-inflammatory factors(P<0.05 or P<0.01). Conclusion: RPQE exhibited therapeutic effects on IBD by inhibiting inflammation.

目的探讨miR-455-3p调控沉默信息调节因子2同源蛋白1(silent mating type information regulation 2 homolog 1,sirt1)表达对大鼠椎间盘髓核(nucleus pulposus,NP)细胞增殖及凋亡的影响。方法采用Ⅱ型胶原酶从大鼠腰椎椎间盘中分离NP细胞,并用甲苯胺蓝和Ⅱ型胶原(CollagenⅡ)免疫荧光染色对其鉴定。NP细胞分成Ctrl组、白细胞介素-1β(interleukin-1β,IL-1β)组、IL-1β+NC inhibitor组、IL-1β+miR-455-3p inhibitor组、IL-1β+miR-455-3p inhibitor+si-NC组、IL-1β+miR-455-3p inhibitor+si-sirt1组,除Ctrl组外的五组细胞均使用10 ng/mL的IL-1β诱导NP细胞退变模型。实时荧光定量聚合酶链式反应(real-time fluorescence quantitative polymerase chain reaction,qRT-PCR)检测miR-455-3p及sirt1 mRNA水平;细胞计数试剂盒8(cell counting kit 8,CCK-8)检测增殖;JC-1染色检测线粒体膜电位;Hoechst染色检测凋亡;蛋白免疫印迹检测sirt1、增殖(PCNA、Ki67、Cyclin D1)和凋亡(Bcl-2、Bax、Cleaved-caspase 3)相关蛋白及CollagenⅡ、Aggrecan表达;双荧光素酶报告分析实验验证miR-455-3p与sirt1的靶向关系。结果大鼠椎间盘NP细胞被成功分离。与IL-1β组、IL-1β+NC inhibitor组比较,IL-1β+miR-455-3p inhibitor组miR-455-3p表达、凋亡率、Bax、Cleaved-caspase 3表达及Bax/Bcl-2比值下降,细胞活力、PCNA、Ki67、Cyclin D1、Bcl-2、CollagenⅡ、Aggrecan表达升高(P<0.05)。miR-455-3p直接靶向负调控sirt1表达。干扰sirt1可逆转抑制miR-455-3p表达对NP细胞增殖、凋亡的影响。结论抑制miR-455-3p表达可通过靶向调控sirt1,抑制IL-1β诱导的NP细胞凋亡并促进NP细胞增殖。

Genotype analysis of rotaviruses isolated from children during a phase III clinical trial with the hexavalent rotavirus vaccine in China

The oral hexavalent live human-bovine reassortant rotavirus vaccine(RV6)developed by Wuhan Institute of Biological Products Co.,Ltd(WIBP)has finished a randomized,placebo-controlled phase III clinical trial in four provinces of China in 2021.The trail demonstrated that RV6 has a high vaccine efficacy against the prevalent strains and is safe for use in infants.During the phase III clinical trial(2019–2021),200 rotavirus-positive fecal samples from children with RV gastroenteritis(RVGE)were further studied.Using reverse transcription-polymerase chain reaction and high-throughput sequencing,VP7 and VP4 sequences were obtained and their genetic characteristics,as well as the differences in antigenic epitopes of VP7,were analyzed in detail.Seven rotavirus genotypes were identified.The predominant rotavirus genotype was G9P[8](77.0%),followed by prevalent strains G8P[8](8.0%),G3P[8](3.5%),G3P[9](1.5%),G1P[8](1.0%),G2P[4](1.0%),and G4P[6](1.0%).The amino acid sequence identities of G1,G2,G3,G4,G8,and G9 genotypes of isolates compared to the vaccine strains were 98.8%,98.2%–99.7%,88.4%–99.4%,98.2%,94.2%–100%,and 93.9%–100%,respectively.Notably,the vaccine strains exhibited high similarity in amino acid sequence,with only minor differences in antigenic epitopes compared to the Chinese endemic strains.This supports the potential application of the vaccine in preventing diseases caused by rotaviruses.

Outline-free floorplanning focuses on area and wirelength reductions, which are usually meaningless, since they can hardly satisfy modern design requirements. We concentrate on a more difficult and useful issue, fixed-outline floorplanning. This issue imposes fixed-outline constraints on the outline-free floorplanning, making the physical design more interesting and challenging. The contributions of this paper are primarily twofold. First, a modified simulated annealing(MSA) algorithm is proposed. In the beginning of the evolutionary process, a new attenuation formula is used to decrease the temperature slowly, to enhance MSA's global searching capacity. After a period of time, the traditional attenuation formula is employed to decrease the temperature rapidly, to maintain MSA's local searching capacity. Second, an excessive area model is designed to guide MSA to find feasible solutions readily. This can save much time for refining feasible solutions. Additionally, B*-tree representation is known as a very useful method for characterizing floorplanning. Therefore, it is employed to perform a perturbing operation for MSA. Finally, six groups of benchmark instances with different dead spaces and aspect ratios—circuits n10, n30, n50, n100, n200, and n300—are chosen to demonstrate the efficiency of our proposed method on fixed-outline floorplanning. Compared to several existing methods, the proposed method is more efficient in obtaining desirable objective function values associated with the chip area, wirelength, and fixed-outline constraints.

Bilateral central retinal artery occlusion （CRAO） has been rarely reported as the primary manifestation in patients with systemic lupus erythematosus （SLE）. The severe retinal vaso-occlusive diseases usually cause devastating and permanent damage to visual function in spite of vigorous treatment. A 42-year-old Chinese woman presented with abrupt bilateral vision loss. The diagnosis of bilateral CRAO was suggested by the ocular presentation and fluorescein angiography. Laboratory studies showed positive results of antinuclear antibody, anti-Ro/SSA anti-La/SSB; decreased levels of C3, C4 complement and normal levels of antiphospholipides antibodies （APAs）. Her visual acuity deteriorated despite systemic steroid and immunosuppressant treatment. Severe vaso-occlusive retinopathy may be an earlier manifestation of SLE without elevated level of APAs.

目的研究尼莫地平(Nimodipine)对脊髓损伤(SCI)大鼠及脊髓组织Ca^(2+)及细胞焦亡的影响。方法将36只SD雄性大鼠按照随机数表法分为假手术组(Sham组)、脊髓损伤组(SCI组)和SCI+Ca^(2+)抑制剂尼莫地平组(Nimodipine组)各12只。脊髓撞击法制备大鼠SCI模型,BBB评分评估大鼠运动功能,荧光探针检测脊髓组织Ca^(2+)含量,酶联免疫吸附法检测白介素1β(IL-1β)及白介素18(IL-18)含量,蛋白免疫印迹检测细胞焦亡相关蛋白NOD样受体蛋白3(NLRP3)及半胱氨酸/天冬氨酸蛋白酶1(Caspase-1)的表达水平。结果与Sham组比较,SCI组大鼠BBB评分降低,差异有统计学意义(P0.05),而14 d、21 d及28 d大鼠BBB评分明显增高,差异均有统计学意义(P<0.05);与SCI组比较,Nimodipine组大鼠Ca^(2+)(11.73±4.31)、IL-1β[(292.93±28.48)pg/mL]、IL-18[(279.81±22.52)pg/mL]含量和NLRP3(0.79±0.18)及Caspase-1(0.63±0.10)的表达水平明显减少,差异均有统计学意义(P<0.05)。结论尼莫地平可能通过抑制Ca^(2+)来改善SCI大鼠的细胞焦亡及运动功能。
