Several studies have found that transplantation of neural progenitor cells(NPCs)promotes the survival of injured ***,a poor integration rate and high risk of tumorigenicity after cell transplantation limits their clinical *** extracellular vesicles(sEVs)contain bioactive molecules for neuronal protection and *** studies have shown that stem/progenitor cell-derived sEVs can promote neuronal survival and recovery of neurological function in neurodegenerative eye diseases and other eye *** this study,we intravitreally transplanted sEVs derived from human induced pluripotent stem cells(hiPSCs)and hiPSCs-differentiated NPCs(hiPSC-NPC)in a mouse model of optic nerve *** results show that these intravitreally injected sEVs were ingested by retinal cells,especially those localized in the ganglion cell *** with hiPSC-NPC-derived sEVs mitigated optic nerve crush-induced retinal ganglion cell degeneration,and regulated the retinal microenvironment by inhibiting excessive activation of *** analysis further revealed that hiPSC-NPC derived sEVs transported neuroprotective and anti-inflammatory miRNA cargos to target cells,which had protective effects on RGCs after optic nerve *** findings suggest that sEVs derived from hiPSC-NPC are a promising cell-free therapeutic strategy for optic neuropathy.

The extracellular vesicles show great potential as a noninvasive biomarker for the early detection of cancer. Hence, there is an urgent requirement to create biosensors that are time-saving, simple, and easily scalable in order to accomplish rapid, sensitive, and quantitative detection of extracellular vesicles. In this study, we present a self-propelled DNA walker powered by endonuclease *** CI, which enables the development of a “signal on” sensing platform for the rapid and highly sensitive detection of extracellular vesicles. The DNA motor employed tracks made of streptavidin magnetic beads, which consisted of substrate strands labeled with fluorescein and motor strands locked by aptamers. The aptamer recognition of the target protein on extracellular vesicles unlocked the motor strand, initiating the DNA motor process. After replacing the optimal buffer solution containing the endonuclease *** I, the motor strands autonomously moved along the streptavidin magnetic beads track, continuously releasing fluorescent molecules and producing detectable fluorescence signals. Under optimal conditions, the detection range was from 2×10～4particles/mL to 2×10～9particles/mL, with a detection limit of 2.9×10～3particles/mL,demonstrating excellent selectivity. This method has demonstrated good selectivity in different tumorderived extracellular vesicles and performs well in complex biological samples. The ability to effectively analyze surface proteins of extracellular vesicles in a short period of time gives our DNA walker a tremendous potential for developing simple and cost-effective clinical diagnostic devices.

Genome-Wide Identification and Expression Analysis of Calmodulin-Like Proteins in Tobacco

Calmodulin-like (CMLs) proteins are critical in calcium signaling and essential for plant growth, development, and stress responses. In many species, the CMLs families have been identified and described. However, the characterization and expression profiling of CMLs genes in tobacco is retrievable. In this study, a comprehensive whole-genome identification and analysis, and 75 NtCML genes were identified in tobacco, each containing two to four EF-hand domains. Most NtCML proteins exhibited conserved gene structures and motifs. Notably, most NtCML proteins were intron-less and distributed across 18 chromosomes. Two pairs of tandemly duplicated genes and seven pairs of segmentally duplicated genes were identified within the tobacco genome. Furthermore, 22 pairs of orthologous CMLs genes were discovered between Arabidopsis and tobacco. Cis-acting element analysis revealed that elements associated with hormones, stress responses, and plant growth and development were found in the promoter regions. Expression analysis indicated that some NtCML genes displayed tissue-specific expression patterns. Specifically, NtCML12, NtCML18, NtCML27, and NtCML28 showed significant upregulation during cold acclimation treatment. These results indicate that tobacco CMLs act as Ca2+ signal transducers, regulating plant growth and abiotic stress responses.

Benzotriazole(BTA)-based A_(2)-A_1-D-A_1-A_(2)type wide-bandgap(WBG)non-fullerene acceptors(NFAs)have shown promising potential in indoor photovoltaic,and in-depth investigation of their structure-property relationship is of great ***,we explored the chlorination effect of the side chain on the *** introduced Cl atoms into the benzyl side chains in parent BTA5 to synthesize two NFAs,BTA5-Cl with mono-chlorinated benzyl groups and BTA5-2Cl containing bi-chlorinated benzyl *** chose D18-Cl with deep-energy levels and strong crystallinity to pair with these three acceptors,affording high photovoltage and *** the stepwise chlorination,the open-circuit voltage(V_(OC))values decrease from 1.28,1.22,to 1.20 V,while the corresponding power conversion efficiencies(PCEs)improve from 5.07%,9.15%,to 10.96%.Compared with BTA5-based OSCs,introducing Cl atoms downshifts the energy levels and slightly increases the non-radiative energy loss(0.14<0.17<0.19 e V),resulting in a sequential decrease in VO ***,more chlorine atom replacements produce more effective exciton dissociation,higher charge transfer,and balanced carrier mobility in the blend films,ultimately achieving better *** work indicates that chlorination of the benzyl group on the terminals can improve the device's performance,implying good application potential in indoor photovoltaics.

Dual-Mode Colorimetric and Fluorescent Detection of Tumor Cells Based on Gold Nanoparticles-Loaded Phosphine Covalent Organic Frameworks

Circulating tumor cells(CTCs) are important markers for cancer. The part of tumor cells that are detached from the primary tumor or metastatic tumor and enter the blood circulation is called CTCs. It is crucial to develop a rapid, accurate, and easy-to-implement diagnostic system for CTCs for early tumor diagnosis and for monitoring progression of the disease. In this work, we reported a colorimetric and fluorescent dual-mode assay for the detection of CTCs. Our assay used magnetic nanoparticles and aptamer for CTCs capture and gold nanoparticles-loaded covalent organic frameworks(Au@COFs) for signal amplification, respectively. The magnetic nanoparticles were modified with folic acid to capture CTCs by interaction between folic acid and the folate receptor overexpressed on the surface of tumor cells. The covalent organic frameworks were engineered to have both nitro-reductase-like and glucose–oxidase-like activities. The nitro-reductase-like activity converted the substrate p-nitrophenol to p-nitroaniline for colorimetric detection, and the glucose-oxidase-like activity enabled fluorescence detection. Specifically, Au@COFs catalyzed glucose oxidation and generated hydrogen peroxide to oxidize Fe 2+to Fe 3+, which converted MIL(Al)-MOF to MIL(Fe)-MOF through ion exchange, resulting in the fluorescence quenching of MIL(Al)-MOF. Our assay showed high sensitivity with a detection limit of 17 cells/mL using MCF-7 cells as model cancer cells. This work provided an e fficient and ultrasensitive strategy for CTCs detection and has potential applications in cancer identification and diagnosis. Dual-mode detection system, combining colorimetric and fluorescent signals, integration of magnetic nanoparticles and aptamers for CTC capture, and utilization of covalent organic frameworks loaded with gold nanoparticles for signal amplification can improve sensitivity and accuracy.

To improve the wettability of hypereutectic Al−60Si alloy and enhance the mechanical properties of the joints,Al−60Si alloy was joined by ultrasonic soldering with Sn-9Zn solder,and a sound joint with in-situ Si particle reinforcement was *** oxide film of Al−60Si alloy at the interface was identified by transmission electron microscopy(TEM)analysis as amorphous Al_(2)O_(3).The oxide of Si particles in the base metal was also *** oxide film of Al−60Si alloy was observed to be removed by ultrasonic vibration instead of holding *** particle-reinforced joints(35.7 vol.%)were obtained by increasing the ultrasonication *** maximum shear strength peaked at 99.5 MPa for soldering at 330℃with an ultrasonic vibration time of 50 s.A model of forming of Si particles reinforced joint under the ultrasound was proposed,and ultrasonic vibration was considered to promote the dissolution of Al and migration of Si particles.

Recent developments in chirality chemistry have revealed remarkable instances of atropisomers featuring multiple chiral elements. Despite significant progress in the synthesis and application of atropisomers, developing a synthetic protocol for the simultaneous introduction of axial and point chirality is still a fascinating challenge. In this context, we present a convenient and versatile method for synthesizing novel atropisomers with axial and point chirality. This approach, based on a new reaction mechanism with nickel-controlled Si–C bond activation, facilitates the controllable migratory insertion of aldehydes into the Ni–C bond. We utilize multidimensional strategies of desymmetrization, kinetic resolution and dynamic kinetic resolution to complete enantioselective nickel-catalyzed ring expansion of benzosilacyclobutenes with aldehydes. The design of multidimensional kinetic resolution, desymmetrization, and dynamic resolution, features a straightforward catalytic system, mild reaction conditions, and broad functional group tolerances. Various axially chiral biaryl oxasilacycles bearing carbon-or siliconstereogenic centers are obtained with excellent diastereo-and enantio-selectivities(up to >20:1 dr and >99.9% ee) through the use of a chiral Ni complex with a large cavity-type 1,1,4,4-tetra-aryl-2,3-O-isopropylidene-L-threitol(TADDOL)-derived phosphoramidite ligand.
