The plant hormone abscisic acid(ABA)is crucial for plant seed germination and abiotic stress ***,the association between ABA sensitivity and plant abiotic stress tolerance remains largely *** this study,436 rice accessions were assessed for their sensitivity to ABA during seed *** considerable diversity in ABA sensitivity among rice germplasm accessions was primarily reflected by the differentiation between the Xian(indica)and Geng(japonica)subspecies and between the upland-Geng and lowland-Geng *** upland-Geng accessions were most sensitive to ***-wide association analyses identified four major quantitative trait loci containing21 candidate genes associated with ABA sensitivity of which a basic helix-loop-helix transcription factor gene,OsbHLH38,was the most important for ABA *** functional analyses using knockout and overexpression transgenic lines revealed that OsbHLH38 expression was responsive to multiple abiotic *** of OsbHLH38 increased seedling salt tolerance,while knockout of OsbHLH38 increased sensitivity to salt stress.A salt-responsive transcription factor,OsDREB2A,interacted with OsbHLH38 and was directly regulated by ***,OsbHLH38 affected rice abiotic stress tolerance by mediating the expression of a large set of transporter genes of phytohormones,transcription factor genes,and many downstream genes with diverse functions,including photosynthesis,redox homeostasis,and abiotic stress *** results demonstrated that OsbHLH38 is a key regulator in plant abiotic stress tolerance.

Development of hybrid rice with high yield and grain quality is a goal of rice *** investigate the genetic mechanism of heterosis for rice milling and appearance quality in indica/xian rice,QTL mapping was conducted using 1061 recombinant inbred lines(RILs)derived from a cross of the xian rice cultivars Quan 9311B(Q9311B)and Wu-shan-si-miao(WSSM),and a backcross F_(1)(BC_(1)F_(1)) population developed by crossing the RILs with Quan 9311A(Q9311A),combined with phenotyping in two *** F_(1) hybrid(Q9311A×WSSM)showed various degrees of heterosis for milling and appearance quality.A total of 142 main-effect QTL(M-QTL)and 407 pairs of epistatic QTL(E-QTL)were identified for five milling and appearance quality traits and grain yield per plant(GYP)in the RIL,BC_(1)F_(1) and mid-parental heterosis(H_(MP)) *** detection of QTL in three populations revealed that most additive loci detected in the RILs did not show heterotic effects,but some of them did contribute to BC_(1)F_(1) trait *** heterosis of GYP,single-locus overdominance and epistasis were the main contributors to heterosis for milling and appearance *** contributed more to the heterosis for milling quality than to that for appearance ***(four)QTL regions harboring opposite(consistent)directions of favorable allele effects for GYP and grain quality were identified,indicating the presence of partial genetic overlaps between GYP and grain *** strategies are proposed to develop hybrid rice with high yield and good grain quality:1)pyramiding favorable alleles with consistent directions of gene effects for GYP and grain quality at the M-QTL on different chromosomes;2)introgressing favorable alleles for GYP and grain quality into the parents and then pyramiding and fixing these additive effects in hybrids;and 3)pyramiding overdominant and dominant loci and minimizing or eliminating underdominant loci from the parents.

Comparative analysis of metabolite changes in two contrasting rice genotypes in response to low-nitrogen stress

Identification of metabolites responsible for tolerance to low nitrogen availability(low-N)will aid in the genetic improvement of rice yield under nitrogen *** this study,a backcross introgression line(G9)and its recurrent parent Shuhui 527(SH527),which show differential responses to low-N stress,were used to identify metabolites associated with lowN tolerance in *** in metabolite contents in the leaves of G9 and SH527 at three growth stages under low-N stress were assessed by gas chromatography–mass *** metabolites,including amino acids and derivatives,were highly enriched in G9 compared with SH527 under the control condition,suggesting that the two genotypes had basal metabolite ***-N stress induced genotype-specific as well as growth stagedependent metabolite *** induced specifically in G9 that were involved in glycolysis and tricarboxylic acid metabolism were enriched at the tillering and grain filling stages,and metabolites involved in nitrogen and proline metabolism were enriched at the booting *** of pyroglutamate,glutamate,2-oxoglutarate,sorbose,glycerate-2-P,and phosphoenolpyruvic acid in G9 suggests that these metabolites could be involved in low-N stress *** results presented here provide valuable information for further elucidation of the molecular mechanisms of low-N tolerance in crops.

Arbuscular mycorrhizal fungi promote functional gene regulation of phosphorus cycling in rhizosphere microorganisms of Iris tectorum under Cr stress

The mutualistic symbiotic system formed by clumping arbuscular mycorrhizal fungi(AMF)and plants can remediate heavy metal-contaminated ***,the specific mechanisms underlying the interaction between AMF and inter-root microbial communities,particularly their impact on organic phosphorus(P)cycling,remain *** study investigated the gene regulation processes involved in inter-root soil phosphorus cycling in wetland plants,specifically Iris tectorum,following inoculation with AMF under varying concentrations of chromium(Cr)*** macro-genome sequencing,we analyzed the composition and structure of the inter-root soil microbial community associated with Iris tectorum under greenhouse pot *** results demonstrated significant changes in the diversity and composition of the inter-root soil microbial community following AMF inoculation,with Proteobacteria,Actinobacteria,Chloroflexi,Acidobacteria,and Bacteroidetes being the dominant *** Cr stress,species and gene co-occurrence network analysis revealed that AMF promoted the transformation process of organic phosphorus mineralization and facilitated inorganic phosphorus ***,network analysis of functional genes indicated strong aggregation of(pstS,pstA,pstC,***,phoR,pp-gppA)genes,which collectively enhanced phosphorus uptake by *** findings shed light on the inter-root soil phosphorus cycling process during the co-remediation of Cr-contaminated soil by AMF-Iris tectorum symbiosis,providing valuable theoretical support for the application of AMF-wetland plant symbiosis systems to remediate heavy metal-contaminated soil.

Differential microRNA expression between shoots and rhizomes in Oryza longistaminata using high-throughput RNA sequencing

Plant microRNAs(miRNAs)play important roles in biological processes such as development and stress *** the diverse functions of miRNAs in model organisms have been well studied,their function in wild rice is poorly *** this study,high-throughput small RNA sequencing was performed to characterize tissue-specific transcriptomes in Oryza longistaminata.A total of 603 miRNAs,380 known rice miRNAs,72 conserved plant miRNAs,and151 predicted novel miRNAs were identified as being expressed in aerial shoots and ***,99 and 79 miRNAs were expressed exclusively or differentially,respectively,in the two tissues,and 144 potential targets were predicted for the differentially expressed miRNAs in the *** annotation of these targets suggested that transcription factors,including squamosa promoter binding proteins and auxin response factors,function in rhizome growth and *** expression levels of several miRNAs and target genes in the rhizomes were quantified by RT-PCR,and the results indicated the existence of complex regulatory mechanisms between the miRNAs and their *** target cleavage sites were verified by RNA ligase-mediated rapid 5′end *** results provide valuable information on the composition,expression and function of miRNAs in ***,and will aid in understanding the molecular mechanisms of rhizome development.

DNA methylation changes detected by methylation-sensitive amplified polymorphism in two contrasting rice genotypes under salt stress

DNA methylation,one of the most important epigenetic phenomena,plays a vital role in tuning gene expression during plant development as well as in response to environmental *** the present study,a methylation-sensitive amplified polymorphism（MSAP） analysis was performed to profile DNA methylation changes in two contrasting rice genotypes under salt *** with visibly different phenotypes in response to salt stress,epigenetic markers classified as stable inter-cultivar DNA methylation differences were determined between salt-tolerant FL478 and salt-sensitive *** addition,most tissue-specific DNA methylation loci were conserved,while many of the growth stage-dependent DNA methylation loci were dynamic between the two ***,salt stress induced a decrease in DNA methylation specifically in roots at the seedling stage that was more profound in IR29 than in the *** result may indicate that demethylation of genes is an active epigenetic response to salt stress in roots at the seedling stage,and helps to further elucidate the implications of DNA methylation in crop growth and development.

Associations between Mycobacterium tuberculosis Beijing geno- type and drug resistance to four first-line drugs： a survey in China

Investigations on the genetic diversity of Mycobacterium tuberculosis in China have shown that Beijing genotype strains play a dominant role. To study the assoeiation between the M. tuberculosis Beijing genotype and the drug-resistance phenotype, 1286 M. tuberculosis clinical isolates together with epidemiologieal and clinical information of patients were collected from the center for tuberculosis （TB） prevention and control or TB hospitals in Beijing municipality and nine provinces or autonomous regions in China. Drug resistance testing was conducted on all the isolates to the four first-line anti-TB drugs （isoniazid, rifampicin, streptomycin, and ethambutol）. A total of 585 strains were found to be resistant to at least one of the four anti-TB drugs. The Beijing family strains consisted of 499 （53.20%） drug-sensitive strains and 439 （46.80%） drug-resistant strains, whereas the non-Beijing family strains comprised 202 （58.05%） drug-sensitive strains and 146 （41.95%） drug-resistant strains. No significant difference was observed in prevalence （X2=2.41, P 〉 0.05） between the drug-resistant and drug- sensitive strains among the Beijing family strains. Analysis of monoresistance, multidrug-resistant TB, and geographic distribution of drug resistance did not find any relationships between the M. tuberculosis Beijing genotype and drug-resistance phenotype in China. Results confirmed that the Beijing genotype, the predominant M. tuberculosis genotype in China, was not associated with drug resistance.

目的　通过探讨广东部分登革Ⅰ型病毒的变异性 ,初步了解我国毒株的可能来源。方法　对来自广东省不同年代的登革Ⅰ型毒株LD 2 5 (1985年 )、LD 36 (1991年 )和登革Ⅰ型血清 95 142(1995年 ) ,采用RT PCR方法扩增其E/NS1部分基因片段 ,然后克隆测序。将所测序列中的 2 40bp的片段与国际上已知的多个登革Ⅰ型毒株的相应序列进行比较 ,并以 6 %的核苷酸差异 (divergence)作为分亚型的标准。结果　LD 2 5与LD 36、95 142分别属于两个不同的亚型 ,其中LD 2 5与泰国 1980年毒株及中国台湾 1987年、1988年毒株的同源性高达 (97 1～ 98 8) % ,属于一个亚型。LD 36和 95 142则与菲律宾 (1988年 )、澳大利亚 (1981年 )、印度尼西亚 (1978年 )及瑙鲁 (1974年 )等地的毒株同源性高达(97 5～ 10 0 ) % ,同属于另一个亚型。结论　广东登革Ⅰ型病毒可能不仅是从一个地方传入 ,来源比较复杂。
