Regulation and function of endoplasmic reticulum autophagy in neurodegenerative diseases

The endoplasmic reticulum,a key cellular organelle,regulates a wide variety of cellular activities.Endoplasmic reticulum autophagy,one of the quality control systems of the endoplasmic reticulum,plays a pivotal role in maintaining endoplasmic reticulum homeostasis by controlling endoplasmic reticulum turnover,remodeling,and proteostasis.In this review,we briefly describe the endoplasmic reticulum quality control system,and subsequently focus on the role of endoplasmic reticulum autophagy,emphasizing the spatial and temporal mechanisms underlying the regulation of endoplasmic reticulum autophagy according to cellular requirements.We also summarize the evidence relating to how defective or abnormal endoplasmic reticulum autophagy contributes to the pathogenesis of neurodegenerative diseases.In summary,this review highlights the mechanisms associated with the regulation of endoplasmic reticulum autophagy and how they influence the pathophysiology of degenerative nerve disorders.This review would help researchers to understand the roles and regulatory mechanisms of endoplasmic reticulum-phagy in neurodegenerative disorders.

Scientific research is one of the primary ways to drive human civilization forward.Countless research institutions and researchers worldwide are dedicated to exploring the unknown,resulting in a flood of research findings[1].For instance,Web of Science alone collects over 10,000 academic papers on a daily basis.While each research field typically has around 10%of review articles,these mainly focus on a limited portion of the subject,failing to provide a comprehensive and real-time updated overview.As a result,researchers or students in a particular field may spend several months,or even longer,comprehending the state of research that has developed over just a few years.The lack of systematic background often leads to redundant or similar studies,which is highly detrimental to the progress of science and technology.

Analytical Solution and Simplified Formula for Added Mass of Horizontal and Vertical Motions of Truncated Cylinders Under Earthquake Action

This paper investigates the hydrodynamic characteristics of floating truncated cylinders undergoing horizontal and vertical motions due to earthquake excitations in the finite water depth.The governing equation of the hydrodynamic pressure acting on the cylinder is derived based on the radiation theory with the inviscid and incompressible assumptions.The governing equation is solved by using the method of separating variables and analytical solutions are obtained by assigning reasonable boundary conditions.The analytical result is validated by a numerical model using the exact artificial boundary simulation of the infinite water.The main variation and distribution characteristics of the hydrodynamic pressure acting on the side and bottom of the cylinder are analyzed for different combinations of wide-height and immersion ratios.The added mass coefficient of the cylinder is calculated by integrating the hydrodynamic pressure and simplified formulas are proposed for engineering applications.The calculation results show that the simplified formulas are in good agreement with the analytical solutions.

Numerical Study on the Aerodynamic and Fluid−Structure Interaction of An NREL-5MW Wind Turbine

A 5-MW wind turbine has been modeled and analyzed for fluid-structure interaction and aerodynamic performance.In this study,a full-scale model of a 5-MW wind turbine is first developed based on a computational fluid dynamics(CFD)approach,in which the unsteady,noncompressible Reynolds Averaged Navier‒Stokes(RANS)method is used.The main focus of the study is to analyze the tower shadow effect on the aerodynamic performance of the wind turbine under different inlet flow conditions.Subsequently,the finite element model is established by considering fluid/structure interactions to study the structural stress,displacement,strain distributions and flow field information of the structure under the uniform wind speed.Finally,the fluid-structure interaction model is established by considering turbulent wind and the tower shadow effect.The variation rules of the dynamic response of the one-way and twoway fluid−structure interaction(FSI)models under different wind speeds are analyzed,and the numerical calculation results are compared with those of the centralized mass model.The results show that the tower shadow effect and structural deformation are the main factors affecting the aerodynamic load fluctuation of the wind turbine,which in turn affects the aerodynamic performance and structural stability of the blades.The structural dynamic response of the coupled model shows significant similarity,while the structural displacement response of the former exhibits less fluctuation compared with the conventional centralized mass model.The one-way fluid-structure interaction(FSI)model shows a higher frequency of stress-strain and displacement oscillations on the blade compared with the twoway FSI model.

Probabilistic seismic inversion based on physics-guided deep mixture density network

Deterministic inversion based on deep learning has been widely utilized in model parameters estimation.Constrained by logging data, seismic data, wavelet and modeling operator, deterministic inversion based on deep learning can establish nonlinear relationships between seismic data and model parameters.However, seismic data lacks low-frequency and contains noise, which increases the non-uniqueness of the solutions. The conventional inversion method based on deep learning can only establish the deterministic relationship between seismic data and parameters, and cannot quantify the uncertainty of inversion. In order to quickly quantify the uncertainty, a physics-guided deep mixture density network(PG-DMDN) is established by combining the mixture density network(MDN) with the deep neural network(DNN). Compared with Bayesian neural network(BNN) and network dropout, PG-DMDN has lower computing cost and shorter training time. A low-frequency model is introduced in the training process of the network to help the network learn the nonlinear relationship between narrowband seismic data and low-frequency impedance. In addition, the block constraints are added to the PG-DMDN framework to improve the horizontal continuity of the inversion results. To illustrate the benefits of proposed method, the PG-DMDN is compared with existing semi-supervised inversion method. Four synthetic data examples of Marmousi II model are utilized to quantify the influence of forward modeling part, low-frequency model, noise and the pseudo-wells number on inversion results, and prove the feasibility and stability of the proposed method. In addition, the robustness and generality of the proposed method are verified by the field seismic data.

MRm-DLDet:a memory-resident malware detection framework based on memory forensics and deep neural network

Cyber attackers have constantly updated their attack techniques to evade antivirus software detection in recent years.One popular evasion method is to execute malicious code and perform malicious actions only in memory.Mali-cious programs that use this attack method are called memory-resident malware,with excellent evasion capability,and have posed huge threats to cyber security.Traditional static and dynamic methods are not effective in detect-ing memory-resident malware.In addition,existing memory forensics detection solutions perform unsatisfactorily in detection rate and depend on massive expert knowledge in memory analysis.This paper proposes MRm-DLDet,a state-of-the-art memory-resident malware detection framework,to overcome these drawbacks.MRm-DLDet first builds a virtual machine environment and captures memory dumps,then creatively processes the memory dumps into RGB images using a pre-processing technique that combines deduplication and ultra-high resolution image cropping,followed by our neural network MRmNet in MRm-DLDet to fully extract high-dimensional features from memory dump files and detect *** receives the labeled sub-images of the cropped high-resolution RGB images as input of ResNet-18,which extracts the features of the sub-images.Then trains a network of gated recurrent units with an attention mechanism.Finally,it determines whether a program is memory-resident malware based on the detection results of each sub-image through a specially designed voting layer.We created a high-quality dataset consisting of 2,060 benign and memory-resident programs.In other words,the dataset contains 1,287,500 labeled sub-images cut from the MRm-DLDet transformed ultra-high resolution RGB images.We implement MRm-DLDet for Windows 10,and it performs better than the latest methods,with a detection accuracy of up to 98.34%.Moreover,we measured the effects of mimicry and adversarial attacks on MRm-DLDet,and the experimental results demonstrated the robustness of MRm-DLDet.

The Chinese tree shrew(Tupaia belangeri chinensis),a member of the mammalian order Scandentia,exhibits considerable similarities with primates,including humans,in aspects of its nervous,immune,and metabolic systems.These similarities have established the tree shrew as a promising experimental model for biomedical research on cancer,infectious diseases,metabolic disorders,and mental health conditions.Herein,we used metatranscriptomic sequencing to analyze plasma,as well as oral and anal swab samples,from 105 healthy asymptomatic tree shrews to identify the presence of potential zoonotic viruses.In total,eight mammalian viruses with complete genomes were identified,belonging to six viral families,including Flaviviridae,Hepeviridae,Parvovirinae,Picornaviridae,Sedoreoviridae,and Spinareoviridae.Notably,the presence of rotavirus was recorded in tree shrews for the first time.Three viruses-hepacivirus 1,parvovirus,and picornavirus-exhibited low genetic similarity(94%)to known viral strains.Phylogenetic analyses also revealed that the rotavirus and mammalian orthoreovirus identified in this study may be novel reassortants.These findings provide insights into the diverse viral spectrum present in captive Chinese tree shrews,highlighting the necessity for further research into their potential for crossspecies transmission.

Co_(3)O_(4)as an efficient passive NO_(x) adsorber for emission control during cold-start of diesel engines

The Co_(3)O_(4)nanoparticles,dominated by a catalytically active(110)lattice plane,were synthesized as a low-temperature NO_(x) adsorbent to control the cold start emissions from vehicles.These nanoparticles boast a substantial quantity of active chemisorbed oxygen and lattice oxygen,which exhibited a NO_(x) uptake capacity commensurate with Pd/SSZ-13 at 100℃.The primary NO_(x) release temperature falls within a temperature range of 200-350℃,making it perfectly suitable for diesel engines.The characterization results demonstrate that chemisorbed oxygen facilitate nitro/nitrites intermediates formation,contributing to the NO_(x) storage at 100℃,while the nitrites begin to decompose within the 150-200℃range.Fortunately,lattice oxygen likely becomes involved in the activation of nitrites into more stable nitrate within this particular temperature range.The concurrent processes of nitrites decomposition and its conversion to nitrates results in a minimal NO_(x) release between the temperatures of 150-200℃.The nitrate formed via lattice oxygen mainly induces the NO_(x) to be released as NO_(2) within a temperature range of 200-350℃,which is advantageous in enhancing the NO_(x) activity of downstream NH_(3)-SCR catalysts,by boosting the fast SCR reaction pathway.Thanks to its low cost,considerable NO_(x) absorption capacity,and optimal release temperature,Co_(3)O_(4)demonstrates potential as an effective material for passive NO_(x) adsorber applications.

Liu-Jun-Zi decoction alleviates chemotherapy-induced anorexia by regulating gut microbiota and TLR4/MyD88/NF-κB p65 signaling pathway

Background:Liu-Jun-Zi decoction(LJZD),a classical nourishing formula in China,has been proven to be effective in treating chemotherapy-induced anorexia.In this study,the mechanism of LJZD in alleviating chemotherapy-induced anorexia was discussed from the aspects of regulating gut microbiota,repairing intestinal barrier injury and inhibiting inflammatory pathways.Methods:A rat model of chemotherapy-induced anorexia was established using cisplatin.The study evaluated the therapeutic effects of LJZD by observing the weight,food intake,and intestinal pathology of rats.The impact of LJZD on gut microbiota and metabolites,specifically short-chain fatty acids,was investigated through gut microbiota analysis and targeted metabolomics.The anti-inflammatory and intestinal protective effects of LJZD were assessed by examining the expression of intestinal tight junction proteins associated with the inflammatory pathway.Results:LJZD alleviated cisplatin-induced inflammation and intestinal barrier disruption,as evidenced by upregulated expression of tight junction protein 1(TJ-1)and occludin,along with reduced serum levels of interleukin 6(IL-6),interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α),and lipopolysaccharide.Additionally,LJZD alleviated microbiota imbalance and regulated the levels of short-chain fatty acids,especially increased the relative abundance of Coriobacteriales Incertae Sedis,Lactabacillus johnsonii F19785,Parasutterella,and reduced the Tyzzerella.In the hypothalamus,LJZD exerts suppressive effects on the toll-like receptor 4(TLR4)/myeloid differentiation factor 88(MyD88)/nuclear factor-κB(NF-κB)p65 signaling pathway,leading to a downregulation in the transcriptional activity of IL-6 and IL-1β,as well as Interleukin 6 receptors(IL-6R)and Interleukin-1βreceptors(IL-1R1)mRNA expression levels.Conclusion:In summary,LJZD alleviate chemotherapy-induced anorexia by modulating the gut microbiota,repairing the intestinal mechanical barriers,and suppressing the TLR4/MyD88/NF-κB p65 signaling pathway.
