Xuefu Zhuyu decoction improves neurological dysfunction by increasing synapsin expression after traumatic brain injury

Xuefu Zhuyu decoction has been used for treating traumatic brain injury and improving post-traumatic dysfunction, but its mechanism of action needs further investigation. This study established rat models of traumatic brain injury by controlled cortical impact. Rat models were intragastrically administered 9 and 18 g/kg Xuefu Zhuyu decoction once a day for 14 or 21 days. Changes in neurological function were assessed by modified neurological severity scores and the Morris water maze. Immunohistochemistry, western blot assay, and re- verse-transcription polymerase chain reaction were used to analyze synapsin protein and mRNA expression at the injury site of rats. Our results showed that Xuefu Zhuyu decoction visibly improved neurological function of rats with traumatic brain injury. These changes were accompanied by increased expression of synaptophysin, synapsin I, and postsynaptic density protein-95 protein and mRNA in a dose-de- pendent manner. These findings indicate that Xuefu Zhuyu decoction increases synapsin expression and improves neurological deficits alder traumatic brain injury.

Boosting thermal stability and crystallization of closed-loop-recyclable biodegradable poly(p-dioxanone) by end-group regulation

A chemically closed-loop-recyclable biodegradable polymer,poly(p-dioxanone)(PPDO),is one of the ideal candidates for single-use plastic products due to its suitability for different application scenarios.Fascinatingly,when PPDO wastes can be collected,its monomer p-dioxanone(PDO) will be obtained through chemical recycling of these wastes;when cannot be collected,the wastes are able to be biodegraded into harmless substances.However,unsatisfied thermal stability and low crystallization rate of PPDO restrict its wider applications.Herein,based on end-group regulation,we simultaneously realized the significant enhancement of thermal stability and crystallization of PPDO through the simple melt processing with tributyl phosphite(TBP) or triphenyl phosphite(TPP).The model reactions were conducted to investigate the reaction mechanism and theoretical products during the preparation of PPDO/phosphite compounds.Two kinds of phosphites were proved to act as the end-capped reagent and chain extender in the melt processing,while TBP presented better reactivity.As a result,the activation energy of thermal decomposition was largely elevated,and the unprecedented T_(5%)(the temperature at a weight loss of 5%) and Tmax(the temperature at a maximum rate of weight loss) of PPDO were obtained,i.e.,T_(5%)of ~330 ℃ and Tmaxof ~385 ℃ in N_(2) atmosphere,T_(5%)of ~240 ℃ and Tmaxof ~317 ℃ in air atmosphere,respectively.Furthermore,the increased crystallization rate,crystallinity,crystalline orderliness,and realizable monomer recovery(yield >90%,purity >99.9%) of PPDO/phosphite compounds were confirmed.

Invasive fungal infections are life-threatening,and neutrophils are vital cells of the innate immune system that defend against them.The role of LTA4H-LTB_(4)-BLT1 axis in regulation of neutrophil responses to fungal infection remains poorly understood.Here,we demonstrated that the LTA4H-LTB_(4)-BLT1 axis protects the host against Candida albicans and Aspergillus fumigatus,but not Cryptococcus neoformans infection,by regulating the antifungal activity of neutrophils.Our results show that deleting Lta4h or Blt1 substantially impairs the fungal-specific phagocytic capacity of neutrophils.Moreover,defective activation of the spleen tyrosine kinase(Syk)and extracellular signal-related kinase(ERK1/2)pathways in neutrophils accompanies this impairment.Mechanistically,BLT1 regulates CR3-mediated,β-1,3-glucan-induced neutrophil phagocytosis,while a physical interaction with CR3 with slight influence on its dynamics is observed.Our findings thus demonstrate that the LTA4H-LTB_(4)-BLT1 axis is essential for the phagocytic function of neutrophils in host antifungal immune response against Candida albicans and Aspergillus fumigatus.

Hole mobility enhancement in monolayer WSe2 p-type transistors through molecular doping

Two-dimensional （2D） transition metal dichalcogenide （TMDC） semiconductor materials exhibit extraordinary electrical properties,holding promise for the realization of next-generation complementary metal-oxide-semiconductor （CMOS） devices at ultimate scaling.However,constrained by effective device doping strategies,the hole mobility and device performance of tungsten diselenide （WSe2） p-type transistors,especially monolayer chemical vapor deposition （CVD）-grown WSe2,have not met expectations.In this paper,an effective performance enhancement of monolayer WSe2p-type transistor was achieved through a molecular doping strategy.Synthesizing monolayer WSe2directly on SiO2back-gated substrates and leveraging energy band alignment design,4-nitrobenzenediazonium tetrafluoroborate （4-NBD） molecular dopant with a concentration of 10 m M was utilized to modulate the Fermi level position of monolayer WSe2for hole doping.The devices demonstrated a more than 98%increase in hole mobility,reaching up to 97 cm2·V-1·s-1while maintaining the current on/off ratio of 108.Monolayer p-type WSe2transistors with 1μm channel length exhibit a high drive current surpassing 176μA·μm-1,exceeding previous CVD-WSe2devices with similar channel length.This straightforward and effective approach to improving the electrical performance of WSe2transistors paves the way for advanced logic technologies based on transition metal dichalcogenide semiconductors.

Mutual antagonism of mouse-adaptation mutations in HA and PA proteins on H9N2 virus replication

Avian H9N2 viruses have wide host range among the influenza A viruses.However,knowledge of H9N2 mammalian adaptation is limited.To explore the molecular basis of the adaptation to mammals,we performed serial lung passaging of the H9N2 strain A/chicken/Hunan/8.27 YYGK3W3-OC/2018(3W3)in mice and identified six mutations in the hemagglutinin(HA)and polymerase acidic(PA)proteins.Mutations L226Q,T511I,and A528V of HA were responsible for enhanced pathogenicity and viral replication in mice;notably,HA-L226Q was the key determinant.Mutations T97I,I545V,and S594G of PA contributed to enhanced polymerase activity in mammalian cells and increased viral replication levels in vitro and in vivo.PA-T97I increased viral polymerase activity by accelerating the viral polymerase complex assembly.Our findings revealed that the viral replication was affected by the presence of PA-97I and/or PA-545V in combination with a triple-point HA mutation.Furthermore,the double-and triple-point PA mutations demonstrated antagonistic effect on viral replication when combined with HA-226Q.Notably,any combination of PA mutations,along with double-point HA mutations,resulted in antagonistic effect on viral replication.We also observed antagonism in viral replication between PA-545V and PA-97I,as well as between HA-528V and PA-545V.Our findings demonstrated that several antagonistic mutations in HA and PA proteins affect viral replication,which may contribute to the H9N2 virus adaptation to mice and mammalian cells.These findings can potentially contribute to the monitoring of H9N2 field strains for assessing their potential risk in mammals.

In this editorial,we offer our perspective on the groundbreaking study entitled“Hypoxia and inflammatory factor preconditioning enhances the immunosup-pressive properties of human umbilical cord mesenchymal stem cells”,recently published in World Journal of Stem Cells.Despite over three decades of research on the clinical application of mesenchymal stem cells(MSCs),only a few therapeutic products have made it to clinical use,due to multiple preclinical and clinical challenges yet to be addressed.The study proved the hypoxia and inflammatory factor preconditioning led to higher immunosuppressive effects of MSCs without damaging their biological characteristics,which revealed the combination of inflammatory factors and hypoxic preconditioning offers a promising approach to enhance the function of MSCs.As we delve deeper into the intricacies of pretreat-ment methodologies,we anticipate a transformative shift in the landscape of MSC-based therapies,ultimately contributing to improved patient outcomes and advancing the field as a whole.

Regulation of AMPK activation by extracellular matrix stiffness in pancreatic cancer

The adenosine monophosphate(AMP)-activated protein kinase(AMPK)sits at a cen-tral node in the regulation of energy metabolism and tumor progression.AMPK is best known to sense high cellular ADP or AMP levels,which indicate the depletion of energy stores.Previous studies have shown that the low expression of phosphorylated AMPK is associated with a poor prognosis of pancreatic cancer.In this study,we report that AMPK is also highly sensitive to extracellular matrix(ECM)stiffness.We found that AMPK is activated in cells when cultured under low ECM stiffness conditions and is functionally required for the metabolic switch induced by ECM stiffness.This regulation of AMPK requires the Hippo kinases but not LKB1/CaMKKp.Hippo kinases directly phosphorylate AMPKa at Thr172 to activate AMPK at low ECM stiffness.Furthermore,we found AMPK activity is inhibited in patients with pancreatic ductal adenocarcinoma(PDAC)with high ECM stiffness and is associated with a poor survival outcome.The activation of Hippo kinases by ROCK inhibitor Y-27632 in combination with the mitochondrial inhibitor metformin synergistically activates AMPK and dramatically inhibits PDAC growth.Together,these findings establish a novel model for AMPK regulation by the mechanical properties of ECMs and provide a rationale for simultaneously targeting the ECM stiffness-Hippo kinases-AMPK signaling and low glucose-LKB1-AMPK signaling pathways as an effective therapeutic strategy against PDAC.

Polyoxometalates-derived ternary metal oxides electrocatalyst for N_(2) reduction under ambient conditions

Accelerating the breaking of the nitrogen nonpolar bond(N≡N)is an important factor to improve the effi ciency of the electro-catalytic nitrogen reduction reaction(e-NRR).In this work,polyoxometalates-derived FeMo-based ternary oxide materials MoO_(2)-Mo_(4)O_(11)-FeMoO_(4)@X(abbreviated as MoFeO@X,X represents the synthesis temperature of 650,750 and 850℃)were designed and synthesized for e-NRR under ambient conditions.The scanning electron microscopy images of MoFeO@750 show an ellipsoidal-like structure(0.86×0.6μm).The relatively large specifi c surface area,formation of multiple interfaces,together with the synergistic eff ect of iron and molybdenum bimetals,would make MoFeO@X catalyst more easily absorb and activate N_(2)in the e-NRR.Expectedly,the synthesized MoFeO@750 shows an optimal NH_(3)production rate of 16.57μg·h^(−1)·mg cat.^(−1)and Faradaic effi ciency of 12.33%at−0.3 V versus reversible hydrogen electrode(νs.RHE),with outstanding electrochemical and structural stability.

Light-controlled soft bio-microrobot

Micro/nanorobots hold exciting prospects for biomedical and even clinical applications due to their small size and high controllability.However,it is still a big challenge to maneuver micro/nanorobots into narrow spaces with high deformability and adaptability to perform complicated biomedical tasks.Here,we report a light-controlled soft bio-microrobots(called“Ebot”)based on Euglena gracilis that are capable of performing multiple tasks in narrow microenvironments including intestinal mucosa with high controllability,deformability and adaptability.The motion of the Ebot can be precisely navigated via light-controlled polygonal flagellum beating.Moreover,the Ebot shows highly controlled deformability with different light illumination duration,which allows it to pass through narrow and curved microchannels with high adaptability.With these features,Ebots are able to execute multiple tasks,such as targeted drug delivery,selective removal of diseased cells in intestinal mucosa,as well as photodynamic therapy.This light-controlled Ebot provides a new bio-microrobotic tool,with many new possibilities for biomedical task execution in narrow and complicated spaces where conventional tools are difficult to access due to the lack of deformability and bio-adaptability.

Geomorphology-oriented theoretical framework and construction method for value-added DEM

Digital elevation model(DEM)plays a fundamental role in the study of the earth system by expressing surface configuration,understanding surface process,and revealing surface mechanism.DEM is widely used in analysis and modeling in the field of geoscience.However,traditional DEM has the defect of single attribute,which is difficult to support the research in earth system science oriented to geoscience process and mechanism mining.Hence,realizing the value-added data model on the basis of traditional DEM is necessary to serve digital elevation modeling and terrain analysis under the background of a new geomorphology research paradigm and earth observation technology.A theoretical framework for value-added DEM that mainly includes concept,connotation,content,and categories,is constructed in this study.The relationship between different types of value-added DEMs as well as the research significance and application category of this theoretical framework are also proposed.The following are different methods of value-added DEMs:(1)value-added methods of DEM space and time dimensions that emphasize the integration of the ground and underground as well as coupling of time and space,(2)attribute-based value-added methods composed of material(including underground,surface,and ground)and morphological properties,and(3)value-added methods of features and physical elements that consider geographical objects and landform features formed by natural processes and artificial effects.The digital terrace,slope,and watershed models are used as examples to illustrate application scenarios of the three kinds of value-added methods.This study aims to improve expression methods of DEMs under the background of new surveying and mapping technologies by adding value to the DEM at three levels of dimensions,attributes,and elements as well as support knowledge-driven digital geomorphological analysis in the era of big data.
