Paclitaxel-lipid prodrug liposomes for improved drug delivery and breast carcinoma therapy

Paclitaxel(PTX)is widely applied for the treatment of unresectable and metastasis breast carcinoma as well as other cancers,whereas its efficacy is always impeded by poor solubility.Liposomes are one kind of the most successful drug carriers which are capable of solubilizing PTX and improving patients’tolerance owing to excellent biocompatibility and biodegradability.However,poor compatibility between PTX and liposomes compromises the stability,drug loading and anti-tumor capacity of liposomal formulations.To address this issue,three lipids with various chain lengths,namely,myristic acid(MA,14C),palmitic acid(PA,16C)and stearic acid(SA,18C),were conjugated to PTX via ester bonds and the synthesized prodrugs with high lipophilicity were further formulated into liposomes,respectively.All liposomes show high stability and drug loadings,as well as sustained drug release.The chain lengths of lipids are negatively correlated with drug release and enzymatic conversion rates,which further impact the pharmacokinetics,tumor accumulation,and anti-tumor efficacy of liposomal PTX.Neither rapid nor slow drug release facilitates high tumor accumulation as well as anti-tumor efficacy of PTX.Among all liposomes,PTX-PA-loaded liposomes show the longest circulation and highest tumor accumulation of PTX and exert the most potent anti-tumor capacities in vivo,owing to its moderate drug release and enzymatic conversion rate.Witnessing its superior safety,PTX-PA liposomes hold potential for further clinical translation.

LSM14B coordinates protein component expression in the P-body and controls oocyte maturation

The generation of mature and healthy oocytes is the most critical event in the entire female reproductive process,and the mechanisms regulating this process remain to be studied.Here,we demonstrate that Smith-like(LSM)family member 14B(LSM14B)regulates oocyte maturation,and the loss of LSM14B in mouse ovaries leads to abnormal oocyte MII arrest and female infertility.Next,we find the aberrant transcriptional activation,indicated by abnormal non-surrounded nucleolus and surrounded nucleolus oocyte proportions,and abnormal chromosome assembly and segregation in Lsm14b-deficient mouse oocytes.The global transcriptome analysis suggests that many transcripts involved in cytoplasmic processing body(P-body)function are altered in Lsm14b-deficient mouse oocytes.Deletion of Lsm14b results in the expression and/or localization changes of P-body components(such as LSM14A,DCP1A,and 4E-T).Notably,DDX6,a key component of the P-body,is downregulated and accumulates in the nuclei in Lsm14b-deficient mouse oocytes.Taken together,our data suggest that LSM14B links mouse oocyte maturation to female fertility through the regulation of the P-body.

Yiwei Xiaoyu granules for treatment of chronic atrophic gastritis with deficiency syndrome of the spleen and stomach

BACKGROUND The Correa sequence,initiated by Helicobacter pylori(H.pylori),commonly progresses to gastric cancer through the stage of chronic atrophic gastritis(CAG).Although eradication of H.pylori only reduces the risk of gastric cancer,it does not eliminate the risk for neoplastic progression.Yiwei Xiaoyu granules(YWXY)are a commonly used composite preparation in Chinese clinics.However,the pursuit of excellence in clinical trials and the establishment of standardized animal experiments are still needed to contribute to full understanding and application of traditional Chinese medicine in the treatment of CAG.AIM To demonstrate the effectiveness of YWXY in patients with CAG and spleenstomach deficiency syndrome(DSSS),by alleviating histological scores,improving response rates for pathological lesions,and achieving clinical efficacy in relieving DSSS symptoms.METHODS We designed a double-blind,randomized,controlled trial.The study enrolled seventy-two H.pylori-negative patients(mean age,52.3 years;38 men)who were randomly allocated to either the treatment group or control group in a 1:1 ratio,and treated with 15 g YWXY or 0.36 g Weifuchun(WFC)tablet combined with the respective dummy for 24 wk.The pre-randomization phase resulted in the exclusion of 72 patients:50 participants did not meet the inclusion criteria,12 participants declined to participate,and 10 participants were excluded for various other reasons.Seven visits were conducted during the study,and histopathological examination with target endoscopic biopsy of narrow-band imaging was requested before the first and seventh visits.We also evaluated endoscopic performance scores,total symptom scores,serum pepsinogen and gastrin-17.RESULTS Six patients did not complete the trial procedures.Treatment with YWXY improved the Operative Link on Gastric Intestinal Metaplasia Assessment(OLGIM)stage,compared with WFC(P<0.05).YWXY provided better relief from symptoms of DSSS and better improvement in serum gastric function,compared with WFC(P<0.05).CONCLUSION YWXY

Mechanochemical route to fabricate an efficient nitrate reduction electrocatalyst

The electrochemical nitrate reduction reaction(NO_(3)RR)to ammonia under ambient conditions is a promising approach for addressing elevated nitrate levels in water bodies,but the progress of this reaction is impeded by the complex series of chemical reactions involving electron and proton transfer and competing hydrogen evolution reaction.Therefore,it becomes imperative to develop an electro-catalyst that exhibits exceptional efficiency and remarkable selectivity for ammonia synthesis while maintaining long-term stability.Herein the magnetic biochar(Fe-C)has been synthesized by a two-step mechanochemical route after a pyrolysis treatment(450,700,and 1000℃),which not only significantly decreases the particle size,but also exposes more oxygen-rich functional groups on the surface,promoting the adsorption of nitrate and water and accelerating electron transfer to convert it into ammonia.Results showed that the catalyst(Fe-C-700)has an impressive NH_(3)production rate of 3.5 mol·h^(−1)·gcat^(−1),high Faradaic efficiency of 88%,and current density of 0.37 A·cm^(−2)at 0.8 V vs.reversible hydrogen electrode(RHE).In-situ Fourier transform infrared spectroscopy(FTIR)is used to investigate the reaction intermediate and to monitor the reaction.The oxygen functionalities on the catalyst surface activate nitrate ions to form various intermediates(NO_(2),NO,NH_(2)OH,and NH_(2))and reduce the rate determining step energy barrier(*NO_(3)→*NO_(2)).This study presents a novel approach for the use of magnetic biochar as an electro-catalyst in NO_(3)RR and opens the road for solving environmental and energy challenges.

Principal component analysis(PCA)is employed to extract the principal components(PCs)present in nuclear mass models for the first time.The effects from different nuclear mass models are reintegrated and reorganized in the extracted PCs.These PCs are recombined to build new mass models,which achieve better accuracy than the original theoretical mass models.This comparison indicates that using the PCA approach,the effects contained in different mass models can be collaborated to improve nuclear mass predictions.

Stability of the Stratification of Water Bodies in the North Passage of the Yangtze River Estuary Based on the EFDC Model

To study the stratified stability of a water column in the North Passage of the Yangtze River Estuary,a numerical model of the hydrodynamics of this estuary is established using the EFDC model.On the basis of EFDC results,this paper derives and pro-vides the discriminative index of water body stability caused by salinity and analyzes the along-range variation in water body strati-fication stability in the North Passage of the Yangtze River Estuary and the periodic variation at a key location(bend area)based on the simulation results of the numerical model.This work shows that the water body in the bend area varies between mixed and strati-fied types,and the vertical average flow velocity has a good negative correlation with the differential velocity between the surface and bottom layers of the water body.The model simulation results validate the formulae for the stratified stability discriminant during spring tides.

Dissipation-Driven Superradiant Phase Transition of a Two-Dimensional Bose-Einstein Condensate in a Double Cavity

We study superradiant phase transitions in a hybrid system of a two-dimensional Bose–Einstein condensate of atoms and two cavities arranged with a tilt angle.By adjusting the loss rate of cavities,we map out the phase diagram of steady states within a mean field framework.It is found that when the loss rates of the two cavities are different,superradiant transitions may not occur at the same time in the two cavities.A first-order phase transition is observed between the states with only one cavity in superradiance and both in superradiance.In the case that both cavities are superradiant,a net photon current is observed flowing from the cavity with small decay rate to the one with large decay rate.The photon current shows a non-monotonic dependence on the loss rate difference,owing to the competition of photon number difference and cavity field phase difference.Our findings can be realized and detected in experiments.

Selection Method for New Energy Output Guaranteed Rates Considering Optimal Energy Storage Configuration

The output of new energy represented by wind power and photovoltaic power features volatility and randomness. It is a practical approach to use the guaranteed rate with statistical characteristics to analyze the output coefficient of new energy. However, there is a lack of analysis and demonstration on the value of the new energy output guaranteed rate. To solve this problem, a selection method for the new energy output guaranteed rate considering optimal energy storage configuration is proposed. First, the influence of the new energy output guaranteed rate on the new energy output coefficient is analyzed. Secondly, with the goal of minimizing the comprehensive costs, an optimal configuration model of energy storage based on the output guaranteed rate of new energy is constructed, and particle swarm optimization is used for the optimization solution. Then, according to the optimal energy storage configuration results under different guaranteed rates, a reasonable new energy output guaranteed rate is determined by maximizing the comprehensive benefits obtained from energy storage. Finally, an example applied to the power grid in an area of Guangdong in Southern China is provided to illustrate the effectiveness of the proposed method.

Organofluorine compounds are widely used in the realm of drug discovery and material science.Herein,we developed palladium catalyzed intermolecular aminofluorination and oxy-aminofluorination of gem‑difluoroalkenes with N-fluorobenzenesulfonimide(NFSI),in which NFSI was used as the nitrogen source and oxidant.The reaction provides an efficient and straightforward synthesis route of a series ofα-trifluoromethyl benzylic amines.Notably,three/four components oxy-aminofluorination processes were realized to giveα-trifluoromethyl benzylic ether with a terminal amino group,which proceed through C(sp^(3))-O bond cleavage of easily available ether and simultaneous introduced a fluorine,an amino and an oxy substituent in one pot with excellent regioselectivity.The divergent reactivity not only included the incorporation of one ether molecular,but also much more challenged two ether insertion with excellent selectivity through succession C(sp^(3))-O bonds cleavage.This protocol allows for concise synthesis of high value amines with fluoroalkyl-substituents and selectively transformation of easily available ethers by high-valent palladium catalysis.

Infrared and Visible Image Fusion Based on Res2Net-Transformer Automatic Encoding and Decoding

A novel image fusion network framework with an autonomous encoder and decoder is suggested to increase thevisual impression of fused images by improving the quality of infrared and visible light picture fusion. The networkcomprises an encoder module, fusion layer, decoder module, and edge improvementmodule. The encoder moduleutilizes an enhanced Inception module for shallow feature extraction, then combines Res2Net and Transformerto achieve deep-level co-extraction of local and global features from the original picture. An edge enhancementmodule (EEM) is created to extract significant edge features. A modal maximum difference fusion strategy isintroduced to enhance the adaptive representation of information in various regions of the source image, therebyenhancing the contrast of the fused image. The encoder and the EEM module extract features, which are thencombined in the fusion layer to create a fused picture using the decoder. Three datasets were chosen to test thealgorithmproposed in this paper. The results of the experiments demonstrate that the network effectively preservesbackground and detail information in both infrared and visible images, yielding superior outcomes in subjectiveand objective evaluations.
