Regulatory effect and mechanism of gastrin and its antagonists on colorectal carcinoma

AIM:To explore the effect and mechanism of gastrin and its antagonists prog lumide and somatostatin on colorectal carcinoma and their clinical ***:A model of transplanted human colonic carcinoma was established from SW480 cell line in gymnomouse *** volume and weight of transplanted carcinoma was observed under the effect of pentagatrin (PG), proglumide (PGL) and octapeptide somotostatin (SMS201-995, SMS). The cAMP content of carcinoma cell was determined by radioimmunoassay and the DNA, protein content and cell cycle were determined by flow-cytometry. The amount of viable cells was determined by MTT colorimetric analysis,IP(3) content was determined by radioimmunoassay, Ca(2+) concentration in cell by fluorometry and PKC activity by isotopic enzymolysis. The expression of gastrin, c-myc, c-fos and rasP21 in 48 cases of colorectal carcinoma tissue was detected by the immuno-cytochemistry SP method. Argyrophilia nucleolar organizer regions was determined with argyrophilia ***:The volume,weight, cAMP, DNA and protein content in carcinoma cell, cell amount and proliferation index of S and G(2)M phase in PG group were all significantly higher than those of control group. When PG was at the concentration of 25mg/L, the amount of viable cells, IP(3) content and Ca(2+) concentration in cell and membrane PKC activity in PG group were significantly higher than those in control group; when PGL was at a concentration of 32mg/L, they dropped to the lowest level in PG (25mg/L)+PGL group, but without significant difference from the control group. The positive expression rate of gastrin, c-myc, c-fos and rasP21 in carcinoma tissue was 39.6%, 54.2%, 47.9% and 54.2% respectively and significantly higher than that in mucosa 3cm and 6cm adjacent to carcinoma tissue and normal colorectal mucosa. Mechanistic investigation on Ce addition in tuning recrystallization behavior and mechanical property of Mg alloy

Constructing bimodal grain structure is a promising approach to achieve the high strength-ductility syn-ergy in Mg *** of bimodal grain is closely related to the dynamic and/or static recrystal-lization process,which has not been fully understood in the typical Mg-RE based *** this work,it is claimed for the first time that the minor Ce addition(∼0.3 wt%)into Mg matrix significantly pro-motes the pyramidaland non-basaldislocations at the early stage of extrusion,which con-sequently enhances the formation of sub-grain boundaries via the movement and recovery of pyramidal II-type*** this stage,fine sub-grain lamellae are widely observed predominantly due to the low migration rate of sub-grain boundary caused by the limited mobility of*** the later stage,the sub-grains continuously transform into dynamic recrystallized(DRXed)grains that have10¯10Taylor axis and also strong fiber texture,indicating substantial activation of pyramidal II-type*** low mobility ofdislocations,accompanied with the solute drag from grain boundary(Gb)segregation and pinning from nano-phases,cause a sluggish DRX process and thus a bimodal microstructure with ultra-fined DRXed grains,∼0.51μ*** resultant texture hardening and grain refinement hardening effects,originated from bimodal microstructure,result in a yield strength of∼352 MPa,which is exceptional in Mg-Ce dilute *** work clarifies the critical role of Ce addition in tuning recrystallization behavior and mechanical property of magnesium,and can also shed light on designing the other high-performance Mg alloys.

Electrochemical characteristics of ternary and quadruple lithium silicon nitrides as anode material for lithium ion batteries:the influence of precursors

Ternary and quadruple lithium silicon nitride anode materials for lithium ion batteries with different precursors were prepared by the simple process of high-energy ball milling. High capacity and excellent cyclability were obtained. The influence of precursor introduction on the electrochemical performance of products was investigated. This research reveals that the electrochemical performance of lithium silicon hitilde can be enhanced significantly by doping O. The cyclability of quadruple lithium silicon nitfide can be optimized remarkably by controlling the introduction quantity of the precursors. It is possible for the composite to be used as a capacity compensator within a wide voltage cut-off window.

Prismatic precipitate platelet is always purposefully designed in the microstructure of magnesium alloys due to its greater contribution to yield *** this study,with an introduction of In into Mg-Sm system,a category of novel{1010}_(α) prismatic platelets has replaced thoroughly the traditionalβ’precipitate formed in magnesium rare earth(Mg-RE)***,the microstructural characteristics of platelet are investigated particularly by atomic scale scanning transmission electron *** is confirmed that the platelet has a Mg_(2) InSm composition and can maintain a coherent relationship with α-Mg ***,on account of the similarities between In and Mg atoms,the Mg_(2) InSm prismatic platelet could be structurally categorized as a generalizedβ"precipitate with a(Mg_(2) In)Sm-type DO_(19) structure when both In and Mg are regarded as an equivalent ***,the addition of In into Mg-Sm alloy induces the formation ofβ"***,the formedβ"prismatic platelets generally have a large average aspect *** findings are of great significance to construct the effective precipitation strengthening phases and optimize the microstructure of Mg-based alloys.

Effects of Mulching Mode on Canopy Physiological, Ecological Characteristics and Yield of Upland Rice

The effects of mulching mode on population physiology and ecology of rice were studied using a combination P88S/1128 as the material under three mulching cultivation modes including plastic film mulching, straw mulching and liquid film mulching, as well as bare cultivation (control). The results indicated that mulching mode had significant effects on micro-meteorological factors and individual growth of rice, as shown by an increase of relative humidity, a better internal micro-meteorological environment of rice population, a significant reduction under the rice canopy temperature, especially during high-temperature periods. Rice plants under mulching cultivation conditions displayed a stronger transpiration and lower leaf temperature, thereby improving the ability of anti-high temperature stress and markedly increasing the photosynthetic rate. Furthermore, the yield components of rice were significantly optimized under mulching cultivation, of which with plastic film mulching displayed the highest grain number per panicle and seed-setting rate, and a yield increase of 16.81% compared with the control; and with straw mulching displayed an increase of effective panicle number and a 9.59% increase of total yield compared to the control.

Preparative isolation and purification of 12,13-dihydroxyeuparin from Radix Eupatorii Chinensis by high-speed counter-current chromatography

An efficient method for the isolation and purification of 12,13-dihydroxyeuparin from Radix Eupatorii Chinensis by high speed counter-current chromatography (HSCCC) was established in this paper. The ether extracts of Radix Eupatorii Chinensis were purified by HSCCC with a solvent system of hexyl hydride-ethyl acetate-methanol-water (1:2:1:2, v/v/v/v). The upper phase was used as the stationary phase and the lower phase as the mobile phase. About 8.4 mg of 12, 13-dihydroxyeuparin was obtained from 200 mg of ether extracts from Radix Eupatorii Chinensis in one-step HSCCC separation, with the purity of 96.71%, as determined by HPLC. After methanol- water recrystallization, the purity of 12,13-dihydroxyeuparin reached 99.83%. Such a simple and effective method was fairly useful to prepare pure compound as reference substances for related study on Radix Eupatorii Chinensis.

Cloning and characterization of 37 kDa laminin receptor precursor in pearl oyster,Pinctada fucata

A 1063 bp cDNA clone encoding a putative 37 kDa laminin receptor precursor (37 kDa LRP) is isolated from the mantle tissue of pearl oyster,Pinctada fucata. The amino acid sequence predicted from the cDNA sequence is 301 residues long,with a calculated molec-ular mass of 33.5 kDa. RT-PCR analysis shows that 37 kDa LRP mRNA is especially highly expressed in the mantle while widely expressed in several tissues. In situ hybridization analysis reveals that 37 kDa LRP is expressed in the outer epithelial cells of the mantle edge,suggesting its involvement in cell proliferation and secretion in P. fucata. The identification and characterization of 37 kDa LRP in the pearl oyster will help us to further understand the signal transduction in the processes of mantle epithelial cell proliferation and tissue formation.

Cytotoxicity and cellular imaging of quantum dots protected by polyelectrolyte

The nanocomposites of poly-diallyldimethylammonium chloride (PDADMAC) and CdTe quantum dots (QDs) (i.e. QDs-PDADMAC nanocomposites) have been prepared based on electrostatic interaction and their fluorescence stability in aqueous solution has been investigated. MTT method (3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide method) was used to study their cytotoxicity and A549 lung cancer cell as a model cell was also used to evaluate their cellular imaging. It was shown that the fluorescence stability of QDsPDADMAC nanocomposites was much better than that of bare QDs both in aqueous solution and cell. Meanwhile, QDs-PDADMAC nanocomposites display very low cytotoxicity in the low concentrations and better staining ability compared with QDs. QDs-PDADMAC nanocomposites will have great advantage on the cell analysis detection and imaging.

Covarying modes of the Pacific SST and northern hemispheric midlatitude atmospheric circulation anomalies during winter

The interannual-to-interdecadal relationship between the Pacific sea surface temperature (SST) and the northern hemispheric midlati- tude’s atmosphere represented by the circumpolar vortex was documented with the global oceanic and atmospheric reanalysis data of recent 50 *** covarying modes of the Pacific SST and northern circumpolar vortex anomalies during winter were examined using the sin- gular value decomposition and wavelet analysis *** is the interannual,ENSO-related mode and the other is the interdecadal, North Pacific SST-related mode with a period of around 20 *** two modes exhibit distinct spatial *** the interannual mode,the SST anomaly is characterized by a typical ENSO pattern with the principal signature in the tropical eastern Pacific and secondary one in the central North Pacific,while the atmospheric anomaly is regional,characterized by a Pacific-North American *** the inter- decadal mode,large SST anomaly is located in the central North Pacific,while the atmospheric anomaly is zonally global,associated with the midlatitute’s standing long-wave *** the central North Pacific is colder,the long-wave is stronger,and vice *** investigations suggest that the interdecadal mode could involve an interaction between "two oceans and an atmosphere".

Localization of calmodulin and calmodulin-like protein and their functions in biomineralization in P. fucata

Calmodulin (CaM) and calmodulin-like protein (CaLP) are two proteins involved in biomineralization. Their localizations in Pinct- ada fucata mantle epithelia were studied by Western blot (Wb) analysis of the nuclear/cytosol fraction of primary cultured P. fucata mantle cells and immunogold electron microscopy. The results showed a completely different distribution of these two proteins at the subcellular level. CaM was distributed throughout both the nucleus and cytoplasm of the mantle epithelium but CaLP was distributed only in the cytoplasm. The functions of these two proteins in biomineralization were investigated by shell regeneration. During this pro- cess, the expressions of CaM and CaLP were greatly enhanced in different organelles of the mantle epithelium. Overexpression of these two proteins and a mutant of calmodulin-like protein (M-CaLP) that lacks an extra C-terminal tail in MC3T3-E1 promoted the mRNA expression of osteopontin, a biomineralization marker for osteoblasts. All of the results indicated that CaM and CaLP have completely different distributions in the mantle epithelium and affect the biomineralization process at different levels. The extra C-terminal tail of CaLP is important for its functions in biomineralization in P. fucata.
