Soil acidifications become one of the main causes restricting the sustainable development of agriculture and causing issues of agricultural product safety.In order to explore the effect of different acidification on soil cadmium(Cd)availability,soil pot culture and hydroponic(soil potting solution extraction)were applied,and non-invasive micro-test technique(NMT)was combined.Here three different soil acidification processes were simulated,including direct acidification by adding sulfuric acid(AP1),acid rain acidification(AP2)by adding artificial simulated acid rain and excessive fertilization acidification by adding(NH_(4))_(2)SO_(4)(AP3).The results showed that for direct acidification(AP1),DTPA-Cd concentration in field soils in Liaoning(S1)and Zhejiang(S2)increased by 0.167-0.217 mg/kg and 0.181-0.346 mg/kg,respectively,compared with control group.When soil pH decreased by 0.45 units in S1,the Cd content of rice stems,leaves and roots increased by 0.48 to 6.04 mg/kg and 2.58 to 12.84mg/kg,respectively,When the pH value of soil S1 and S2 decreased by 0.20 units,the average velocity of Cd^(2+)at 200μm increased by 10.03-33.11 pmol/cm~2/sec and 21.33-52.86pmol/cm^(2)/sec,respectively,and followed the order of AP3>AP2>AP1.In summary,different acidification measures would improve the effectiveness of Cd,under the same pH reduction condition,fertilization acidification increased Cd availability most significantly.

Antihypertensive effects of whey protein hydrolysate involve reshaping the gut microbiome in spontaneously hypertension rats

Novel angiotensin-converting enzyme(ACE)inhibitory peptides were identified from whey protein hydrolysates(WPH)in vitro in our previous study and the antihypertensive abilities of WPH in vivo were further investigated in the current study.Results indicated that WPH significantly inhibited the development of high blood pressure and tissue injuries caused by hypertension.WPH inhibited ACE activity(20.81%,P<0.01),and reduced renin concentration(P<0.05),thereby reducing systolic blood pressure(SBP)(12.63%,P<0.05)in spontaneously hypertensive rats.The increased Akkermansia,Bacteroides,and Lactobacillus abundance promoted high short chain fatty acid content in feces after WPH intervention.These changes jointly contributed to low blood pressure.The heart weight and cardiomyocyte injuries(hypertrophy and degeneration)were alleviated by WPH.The proteomic results revealed that 19 protein expressions in the heart mainly associated with the wingless/integrated(Wnt)signaling pathway and Apelin signaling pathway were altered after WPH supplementation.Notably,WPH alleviated serum oxidative stress,indicated by the decreased malondialdehyde content(P<0.01),enhanced total antioxidant capacity(P<0.01)and superoxide dismutase activity(P<0.01).The current study suggests that WPH exhibit promising antihypertensive abilities in vivo and could be a potential alternative for antihypertensive dietary supplements.

Resurvey After 20 Years:Updating the Distribution,Population,and Potential Threats of the Chinting Alpine Toad

Considering the increased anthropogenic impacts,species with a limited range and low detectability often lack fundamental information and conservation actions,placing them at a high risk of endangerment.The Chinting alpine toad Scutiger chintingensis is a rare mountain amphibian endemic to the eastern margin of the Qinghai-Xizang Plateau in China.Within its whole distribution range,only three known populations(Wolong,Emei,and Wawu)exist and no recent population status report has been documented for this species over the past two decades.From 2020 to 2023,we investigated the species distribution,and assessed the risk factors for the main populations.We recorded this species in all distribution areas,and updated a new distribution site with a lower elevation limit.The relative population density was 0.024±0.012 ind./m^(2)on Mount Emei,whereas only 0.008±0.017 ind./m^(2)on Mount Wawu.No significant difference was observed in the number of individuals between the two populations;however,the relative population density was significantly different.Sewage and waste discharge resulting from the construction of scenic areas,as well as disturbances from tourism,were the primary anthropogenic factors that influenced the survival of this species.Our results provide the updated information on the distribution and population status of the Chinting alpine toad,and suggest that unrecorded populations,as well as a wider elevation range,may exist for this species.Our findings emphasise the importance of timely updates of species distribution and population information and offer a basis for the future conservation of endangered amphibians.

Salt stress-induced changes in soil metabolites promote cadmium transport into wheat tissues

Soil salinity is known to improve cadmium(Cd)mobility,especially in arid soils.However,the mechanisms involved in how salt stress-associated metabolic profiles participate in mediating Cd transport in the soil-plant system remain poorly understood.This study was designed to investigate the effects of salinity-induced changes in soil metabolites on Cd bioavailability.Sodium salts in different combinations according to molar ratio(NaCl:Na_(2)SO_(4)=1:1;NaCl:Na_(2)SO_(4):NaHCO_(3)=1:2:1;NaCl:Na_(2)SO_(4):NaHCO_(3):Na_(2)CO_(3)=1:9:9:1;NaCl:Na_(2)SO_(4):NaHCO_(3):Na_(2)CO_(3)=1:1:1:1)were applied to the Cd-contaminated soils,which increased soil Cd availability by 22.36%and the Cd content in wheat grains by 36.61%,compared to the control.Salt stress resulted in soil metabolic reprogramming,which might explain the decreased growth of wheat plants and increased Cd transport from the soil into wheat tissues.For example,down-regulation of starch and sucrose metabolism reduced the production of sugars,which adversely affected growth;up-regulation of fatty acid metabolism allowed wheat plants to maintain a normal intracellular environment under saline conditions;up-regulation of the tricarboxylic acid(TCA)cycle was triggered,causing an increase in organic acid synthesis and the accumulation of organic acids,which facilitated the migration of soil Cd into wheat tissues.In summary,salt stress can facilitate Cd transport into wheat tissues by the direct effect of salt-based ions and the combined effect of altered soil physicochemical properties and soil metabolic profiles in Cd-contaminated soils.

Research on the influence of flexible wheelset rotation effect on wheel rail contact force

Purpose-Under the high-speed operating conditions,the effects of wheelset elastic deformation on the wheel rail dynamic forces will become more notable compared to the low-speed condition.In order to meet different analysis requirements and selecting appropriate models to analyzing the wheel rail interaction,it is crucial to understand the influence of wheelset flexibility on the wheel-rail dynamics under different speeds and track excitations condition.Design/methodology/approach-The wheel rail contact points solving method and vehicle dynamics equations considering wheelset flexibility in the trajectory body coordinate system were investigated in this paper.As for the wheel-rail contact forces,which is a particular force element in vehicle multibody system,a method for calculating the Jacobian matrix of the wheel-rail contact force is proposed to better couple the wheel-rail contact force calculation with the vehicle dynamics response calculation.Based on the flexible wheelset modeling approach in this paper,two vehicle dynamic models considering the wheelset as both elastic and rigid bodies are established,two kinds of track excitations,namely normal measured track irregularities and short-wave irregularities are used,wheel-rail geometric contact characteristic and wheel-rail contact forces in both time and frequency domains are compared with the two models in order to study the influence of flexible wheelset rotation effect on wheel rail contact force.Findings-Under normal track irregularity excitations,the amplitudes of vertical,longitudinal and lateral forces computed by the flexible wheelset model are smaller than those of the rigid wheelset model,and the virtual penetration and equivalent contact patch are also slightly smaller.For the flexible wheelset model,the wheel rail longitudinal and lateral creepages will also decrease.The higher the vehicle speed,the larger the differences in wheel-rail forces computed by the flexible and rigid wheelset model.Under track short-wave irregularity excitations,the ve

目前政府回应公众诉求具有明显的单一结果导向特征,具体表现为将平息突发事件及其舆情作为回应的首要目标,且倾向于采取权宜式的回应手段。然而,这种政府回应方式容易带来回应目标的错位,消解地方政府的公信力,增加社会稳定的长期风险。因此,政府回应公众诉求亟需实现从单一结果导向向“过程—结果”导向的转型。“过程—结果”导向式政府回应并不以快速平息特定突发事件为目标,而是致力于归纳与提炼突发事件背后深层次的社会问题及其根源,并根据问题的合理性与优先度对其进行符合程序正义的回应与解决。公众参与和民主协商是“过程—结果”导向式政府回应的核心,这不仅能提升公众对决策过程的认同度和对决策结果的接受度,还有助于推动多元主体的参与与合作,实现长效的社会治理。

Betaine-homocysteine methyltransferase（BHMT） regulates protein methylation and is correlated with tumorigenesis;however,the effects and regulation of BHMT in hepatocarcinogenesis remain largely unexplored.Here,we determined the clinical significance of BHMT in the occurrence and progression of hepatocellular carcinoma（HCC） using tissue samples from 198 patients.BHMT was to be frequently found（86.6%） expressed at relatively low levels in HCC tissues and was positively correlated with the overall survival of patients with HCC.Bhmt overexpression effectively suppressed several malignant phenotypes in hepatoma cells in vitro and in vivo,whereas complete knockout of Bhmt(Bhmt-/-) produced the opposite effect.We combined proteomics,metabolomics,and molecular biological strategies and detected that Bhmt-/-promoted hepatocarcinogenesis and tumor progression by enhancing the activity of glucose-6-phosphate dehydrogenase（G6PD）and PPP metabolism in DEN-induced HCC mouse and subcutaneous tumor-bearing models.In contrast,restoration of Bhmt with an AAV8-Bhmt injection or pharmacological inhibition of G6PD attenuated hepatocarcinogenesis.Additionally,coimmunoprecipitation identified monomethylated modifications of the G6PD,and BHMT regulated the methylation of G6PD.Protein sequence analysis,generation and application of specific antibodies,and site-directed mutagenesis indicated G6PD methylation at the arginine residue 246.Furthermore,we established bidirectionally regulated BHMT cellular models combined with methylation-deficient G6PD mutants to demonstrate that BHMT potentiated arginine methylation of G6PD,thereby inhibiting G6PD activity,which in turn suppressed hepatocarcinogenesis.Taken together,this study reveals a new methylation-regulatory mechanism in hepatocarcinogenesis owing to BHMT deficiency,suggesting a potential therapeutic strategy for HCC treatment.

Relapse is the most important cause of treatment failure in acute leukemia(AL).Thus,how to predict relapse is critical for improving the survival of patients with AL.Measurable residual diseases(MRDs;previously termed minimal resid-ual diseases),refering to the presence of remaining leukemia cells after the declaration of complete remission(CF)detected by morphological analysis,is the most important biomarker forrelapseprediction.'Several methods,including multiparameter flow cytometry(MFC),real-time quantitative polymerase dhain reaction(qPCR),digital PCR(dPCR),and next-generation sequencing(NGS),are used to monitor MRD after treatment,reaching a sensitvityof 10^(-4)to 10^(-6).

外阴硬化性苔藓345例临床病理分析及维持治疗频率的初步探讨

目的:探讨外阴硬化性苔藓(VLS)的临床病理特征及治疗,并初步探讨其维持治疗频率。方法:收集2018年至2021年于复旦大学附属妇产科医院诊断为VLS的患者共345例,回顾性分析其临床病理特征、治疗方法及效果,对病情稳定的患者进行维持治疗并定期随访,探究维持治疗的最低用药频率。结果:(1)一般情况:345例VLS患者的确诊年龄为(50.4±14.7)岁(范围:8~84岁),其中50~59岁年龄段的占比最高(30.1%,104/345)。177例有合并疾病和相关家族史记录的患者中,18.6%(33/177)合并自身免疫性疾病,24.3%(43/177)合并过敏性皮肤疾病,5.6%(10/177)患者的女性直系亲属有长期外阴瘙痒或皮肤色素减退病史。(2)临床特征:204例有症状记录的患者中,最常见的症状为外阴瘙痒(96.1%,196/204);214例有体征记录的患者中,最常见体征为外阴色素减退(96.3%,206/214);202例有病变累及部位记录的患者中,常见累及部位依次为小阴唇(70.3%,142/202)、大阴唇(67.8%,137/202)、阴唇沟(59.4%,120/202)。62例外阴萎缩患者的病变累及部位数[(2.7±1.1)个]显著多于152例非萎缩患者[(2.2±1.0)个;t=3.48, P=0.001];外阴萎缩患者的病程为(9.3±8.5)年,显著长于非萎缩患者[(6.6±5.6)年;t=2.04, P=0.046]。(3)病理特征:286例有电子病理图片的患者中,表皮层中最常见的病理特征为上皮钉突钝化(71.3%,204/286),真皮层中常见的病理特征依次为间质胶原化(84.6%,242/286)、炎细胞浸润(73.8%,211/286)。(4)治疗:177例患者在确诊后进行了规范治疗并在本院定期随访。初始治疗阶段,26.0%(46/177)患者使用0.05%丙酸氯倍他索乳膏,74.0%(131/177)患者使用0.1%糠酸莫米松软膏治疗;两种药物的完全缓解率分别为80.4%(37/46)、74.0%(97/131),两者比较,差异无统计学意义( χ2=0.76, P=0.385)。维持治疗期间,27.1%(48/177)的患者每周2次用药,35.0%(62/177)每周1次用药,37.9%(67/177)每10天1次用药;维持治疗6个月后随访,无瘙痒复发或外阴体征进展患者。结论:绝大多数VLS患者有外阴瘙痒、色素减退、大小阴唇受累、进行性外阴萎缩、表皮层和真皮层炎症浸润后改变,初始治疗局部使用糠酸莫米松和丙酸氯倍他索治疗均具有良好的疗效,个体化的维持治疗频率在保证患者病情稳定的情况下可最大程度减少副反应的发生。
