This paper reviews the basic implications and policy orientation of the new development pattern and the impacts of constructing the new development pattern on the manufacturing transformation and upgrading,based on which the choice of manufacturing transformation and upgrading paths is further discussed under the new development pattern.This paper holds that the paths of manufacturing transformation and upgrading need to be adjusted and expanded under the conditions of constructing the new development pattern.The main ideas are as follows.Restructuring the manufacturing industry chain with the large domestic market as the main guide,creating the good environment conducive to independent innovation,coordinating the regional layout for the overall high-quality manufacturing development,optimizing the business environment and enhancing the vitality and driving force of the manufacturing’s development,relying on the Belt and Road Initiative(BRI)for a higher level of international circulation.The theoretical bases for adjusting and expanding the paths of manufacturing transformation and upgrading are:embracing the new requirements with domestic circulation as the mainstay,the characteristics with the mutual reinforcement of domestic and international circulations,and promoting the manufacturing’s high-quality development and the medium and high-speed growth.

Nanoparticles induced potent antitumor immunotherapy plays a significant role for enhancing conventional therapeutic effectiveness.However,revealing the pathway of how nanoagents themselves trigger the host immunity or how to maximize the immunotherapy efficacy still needs further exploration.Herein,rose-like MoS2 nanoflowers modified with 2-deoxy-D-glucose(2-DG)and glucose oxidase(GOx)(MPGGFs)have been successfully fabricated via a one-pot hydrothermal reaction and following one-by-one surface modification as a multifunctional nanocatalyst for photothermal therapy enhanced self-amplified chemodynamic immunotherapy(PTT-co-CDT).By introducing GOx,the obtained MPGGFs exhibited self-amplified chemodynamic therapeutic efficacy under hypoxia tumor microenvironment(TME)because of the raised intracellular H2O2 level via enzyme-catalysis of oxygen.Furthermore,combined with the intrinsic excellent photothermal conversion efficiency of MoS2 nanoflowers,PTT-co-CDT performances by MPGGFs could effectively induce the necroptosis of tumor cells both in vitro and in vivo.Then the induced necroptosis via PTT-co-CDT by MPGGFs could directly trigger host immunity by activating the antigen-specific T-cells(CD4^(+) and CD8^(+)).Finally,the excellent in vivo safety of MPGGFs makes us believe that the successful construction of rose-like multifunctional nanocatalyst not only has great potentials for self-amplified chemodynamic immunotherapy,but also provides a paradigm for exploring necroptosis triggered host immunity for cancer treatment.

Extraordinary thermal conductivity of polyvinyl alcohol composite by aligning densified carbon fiber via magnetic field

Thermal interface materials(TIMs)with high through-plane thermal conductivity are urgently desired to avoid overheating of high-power density electronics.Introducing and aligning fillers in polymer matrixes via magnetic field is a promising method to improve the thermal conductivity of the polymer.However,either the fillers need to be modified with magnetic particles or a strong magnetic field is needed for good alignment in high filler content.This prevents further improvement of the through-plane thermal conductivity.Herein,mesophase pitch-based carbon fibers(MPCFs)with a content as high as 76 wt.%are aligned vertically in water-soluble polyvinyl alcohol(PVA)under a low magnetic field(~0.4 T),forming a vertically aligned MPCF(VAMPCF)/PVA composite with an extraordinary through-plane thermal conductivity of 86 W/(m·K),which is higher than that of many alloys.In addition,both theoretical and experimental results demonstrate that the critical intensity of the magnetic field needed for good alignment of the fillers depends on their size and magnetic susceptibility.Furthermore,the water solubility of PVA makes it easy to recycle MPCFs.This study offers an inspired venue to develop excellent and eco-friendly TIMs to meet ever increasing demand in heat dissipation for electronics.

Staphylococcus aureus is a common marine foodborne pathogen.In this study,antibiotics ciprofloxacin and enrofloxacin were used to induce drug-resistance in S.aureus.The differentially expressed proteins(DEPs)were analyzed and compared with those in the bacteria cultured without antibiotics.The primary proteomic alterations were in the levels of cell membrane components and proteins related to lysine and folic acid biosynthesis,which were all significantly up-regulated.The minimal inhibitory concentrations(MIC)for both test drugs were elevated to 10μg m L^(-1)following serial passaging.These results indicated that,for both ciprofloxacin and enrofloxacin,drug-resistance were developed even in the subinhibitory levels and the primary response was a major alteration in the cell membrane proteome.These changes were similar to those observed in S.aureus cultured with super-MIC levels of these antibiotics.The current study provides a theoretical basis for in-depth study of the related changes of marine foodborne pathogens in subinhibitory concentrations that are commonly found in situ.

Hypoxia,as an important hallmark of the tumor microenvironment,is a major cause of oxidative stress and plays a central role in various malignant tumors,including glioblastoma.Elevated reactive oxygen species(ROS)in a hypoxic microenvironment promote glioblastoma progression;however,the underlying mechanism has not been clarified.Herein,we found that hypoxia promoted ROS production,and the proliferation,migration,and invasion of glioblastoma cells,while this promotion was restrained by ROS scavengers N-acetyl-L-cysteine(NAC)and diphenyleneiodonium chloride(DPI).Hypoxia-induced ROS activated hypoxia-inducible factor-1α(HIF-1α)signaling,which enhanced cell migration and invasion by epithelial-mesenchymal transition(EMT).Furthermore,the induction of serine protease inhibitor family E member 1(SERPINE1)was ROS-dependent under hypoxia,and HIF-1αmediated SERPINE1 increase induced by ROS via binding to the SERPINE1 promoter region,thereby facilitating glioblastoma migration and invasion.Taken together,our data revealed that hypoxia-induced ROS reinforce the hypoxic adaptation of glioblastoma by driving the HIF-1α-SERPINE1 signaling pathway,and that targeting ROS may be a promising therapeutic strategy for glioblastoma.

RNA modifications have been involved in numerous biological processes, and aberrations of these modifications are tightly associated with various diseases including cancer. Herein, we developed graphenebased solid-phase extraction and robust ultra performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS) combined with stable isotope-dilution for simultaneous enrichment and accurate determination of 17 modified nucleosides in human urine. We found graphene could effectively adsorb various modified nucleosides in human urine samples. With this method, we identified and quantified these modified nucleosides in urine samples collected from lung cancer patients and healthy controls.We revealed that the levels of 12 modified nucleosides were all diminished in urine from lung cancer patients, compared with healthy controls. It is worth noting that we demonstrated, for the first time, the presence of 5,2-O-dimethyluridine(m~5U_m) in human urine. Together, we established a robust analytical method for simultaneous determinations of 17 modified nucleosides in human urine, and our results revealed a close correlation between the concentrations of urinary modified nucleosides and the occurrence of lung cancer, implying the potential applications of these modified nucleosides as noninvasive biomarkers for the early detection of lung cancer. Moreover, this study will stimulate future investigations on the regulatory roles of RNA modifications in the initiation and progression of lung cancer.

A series of novel P-stereogenic monophosphorus ligands (Xie-phos) were synthesized via the hydrophosphinylation of alkynes with secondary phosphine oxides by palladium catalysis and the following basemediated cyclization. They are easily synthesized and modified. Their utility was demonstrated in catalyticasymmetric reactions, including α-arylation of amides and the domino Heck/Suzuki reaction with moderate enantioselectivity.

Antibody-mediated rejection(AMR)is one of the major causes of graft loss after transplantation.Recently,the regulation of B cell differentiation and the prevention of donor-specific antibody(DSA)production have gained increased attention in transplant research.Herein,we established a secondary allogeneic in vivo skin transplant model to study the effects of romidepsin(FK228)on DSA.The survival of grafted skins was monitored daily.The serum levels of DSA and the number of relevant immunocytes in the recipient spleens were evaluated by flow cytometry.Then,we isolated and purified B cells from B6mouse spleens in vitro by magnetic bead sorting.The B cells were cultured with interleukin-4(IL-4)and anti-clusters of differentiation 40(CD40)antibody with or without FK228 treatment.The immunoglobulin G1(IgG1)and IgM levels in the supernatant were evaluated by enzyme-linked immunosorbent assay(ELISA).Quantitative reverse transcription-polymerase chain reaction(RT-q PCR)and western blotting were conducted to determine the corresponding levels of messenger RNA(mRNA)and protein expression in cultured cells and the recipient spleens.The results showed that FK228 significantly improved the survival of allogeneic skin grafts.Moreover,FK228 inhibited DSA production in the serum along with the suppression of histone deacetylase 1(HADC1)and HDAC2 and the upregulation of the acetylation of histones H2A and H3.It also inhibited the differentiation of B cells to plasma cells,decreased the transcription of positive regulatory domain-containing 1(Prdm1)and X-box-binding protein 1(Xbp1),and decreased the expression of phosphorylated inositol-requiring enzyme 1α(p-IRE1α),XBP1,and B lymphocyte-induced maturation protein-1(Blimp-1).In conclusion,FK228 could decrease the production of antibodies by B cells via inhibition of the IRE1α-XBP1 signaling pathway.Thus,FK228 is considered as a promising therapeutic agent for the clinical treatment of AMR.

Research data infrastructures form the cornerstone in both cyber and physical spaces,driving the progression of the data-intensive scientific research paradigm.This opinion paper presents an overview of global research data infrastructure,drawing insights from national roadmaps and strategic documents related to research data infrastructure.It emphasizes the pivotal role of research data infrastructures by delineating four new missions aimed at positioning them at the core of the current scientific research and communication ecosystem.The four new missions of research data infrastructures are:(1)as a pioneer,to transcend the disciplinary border and address complex,cutting-edge scientific and social challenges with problem-and data-oriented insights;(2)as an architect,to establish a digital,intelligent,flexible research and knowledge services environment;(3)as a platform,to foster the high-end academic communication;(4)as a coordinator,to balance scientific openness with ethics needs.
