Overview of epigenetic degraders based on PROTAC, molecular glue, and hydrophobic tagging technologies

Epigenetic pathways play a critical role in the initiation, progression, and metastasis of cancer. Over the past few decades, significant progress has been made in the development of targeted epigenetic modulators(e.g., inhibitors). However, epigenetic inhibitors have faced multiple challenges,including limited clinical efficacy, toxicities, lack of subtype selectivity, and drug resistance. As a result,the design of new epigenetic modulators(e.g., degraders) such as PROTACs, molecular glue, and hydrophobic tagging(Hy T) degraders has garnered significant attention from both academia and pharmaceutical industry, and numerous epigenetic degraders have been discovered in the past decade. In this review,we aim to provide an in-depth illustration of new degrading strategies(2017-2023) targeting epigenetic proteins for cancer therapy, focusing on the rational design, pharmacodynamics, pharmacokinetics, clinical status, and crystal structure information of these degraders. Importantly, we also provide deep insights into the potential challenges and corresponding remedies of this approach to drug design and development. Overall, we hope this review will offer a better mechanistic understanding and serve as a useful guide for the development of emerging epigenetic-targeting degraders.

地下矿山采用充填法进行开采需要在采空区充填之前构筑封闭挡墙,高质量安全封闭技术是充填采空区的重要环节。研制了装配式柔性脱水充填挡墙,墙体骨架呈弧形网格状曲面结构,采用装配式建构方式,能够适应巷道规格变化,密封层使用透水织物,具有全断面脱水特征。设计了3.5 m×4.0 m规格试验挡墙,并在阶段空场充填采矿法和进路式充填采矿法充填采场开展了工业试验。结果表明:试验挡墙在过墙期间具有较好的脱水效果,高浓度料浆多次充填试验测得的墙体变形量未超过10 mm,膏体料浆一次充全高,最大变形量为34 mm,均在允许变形范围内。扩大试验安装成本分析表明,挡墙的综合换算成本约为435元/m^(3),验证了装配式柔性脱水充填挡墙的技术经济综合优势。

为在实验室模拟矿山原位充填环境的压力、温度等因素对胶结充填体的固结影响,本文设计了一种考虑压力与温度协同控制的胶结充填体养护方法及试验装置。采用该试验装置开展了养护压力与养护温度对胶结充填体早期固结强度的影响研究。结果表明:养护压力、养护温度对胶结充填体早期强度有明显影响;养护压力对充填体早期强度发展有利;随着养护温度升高,胶结充填体强度呈先升高后降低的趋势,养护温度为55℃时,胶结充填体强度呈下降趋势。在养护压力和养护温度的耦合影响下,高温有利于胶结充填体的早期强度发展。

目的:随着开采年限增加,安庆铜矿已逐步面临尾矿库库容不足难题。为此开展了安庆铜矿尾矿综合利用工艺研究,以期实现尾砂资源化利用,降低排尾量,延长尾矿库使用年限。方法:通过安庆铜矿尾砂产出工艺调研,结合尾砂物化特性分析,研究形成了基于旋流分级联合震动脱水的尾砂综合利用工艺技术。并通过工业试验研究,分析了分级粗尾砂产品质量指标与利用效率。结果:试验结果表明,粗尾砂产品-74μm颗粒平均含量低于15%,含水率约18%,硫元素含量低于0.5%,均满足建筑用砂标准。工业试验的尾砂综合利用能力为30 t/h,利用效率为48%,每年可减少约20万t尾矿库排尾量,能有效缓解安庆铜矿尾矿库库容不足难题。意义:相关成果对于面临类似难题的矿山具有较好的指导意义和实用价值,为我国金属矿尾矿固废综合利用技术的发展起到促进作用。

大直径深孔采矿是基于凿岩设备、爆破技术、机械化铲装等多方面技术发展而形成的高效采矿方法。在分析安庆铜矿开采技术现状基础上,阐述了矿山采用高阶段大直径深孔采矿技术的工艺布置。在原浅部采场VCR采矿法的基础上,按照大直径深孔采矿工艺要求对孔网布置、爆破方式、采场边界控制爆破等进行优化,提出了VCR法拉槽侧向分段崩落的回采方式。通过在矿山深部采场应用,取得了采场单日最大出矿量超过900 t、大块率控制在5%以内、充填体混入率小于8%的效益。安庆铜矿的大直径深孔采矿实践证明,该项技术是增加采矿量,扩大回采规模,提高劳动生产率和经济效益的有效途径,为国内外类似矿山应用该方法提供了借鉴。

Eucalyptus grandis *** ex Maiden bark was liquefied in glycerol with two types of *** chemical components of the residues with respect to temperature were examined to investigate the liquefaction behavior of *** results reveal that sulfuric acid was more efficient in converting bark into fragments in glycerol at low temperatures B 433.15 K,equivalent to 160C than phosphoric *** liquefaction order of chemical components was lignin,hemicelluloses,and *** decrease of liquefaction yields at high temperatures(≤453.15 K)catalyzed by sulfuric acid was possibly a result of the recondensation of lignin and/or hemicelluloses.
