The dearomative[5+1]annulation of 2-methylindoles with new five-membered synthons O-alkyl orthooxybenzaldehydes was developed unprecedentedly through cascade[1,5]-hydride transfer/dearomativecyclization in HFIP for the synthesis of spirochromanes bearing the 2-methylindolenine skeleton.Inaddition,the dual alkylation of the methyl group of 2-methylindolenines was achieved by sequential operation through the redox neutral[5+1]annulation with the second five-membered synthon N-alkylortho-aminobenzaldehyde,providing the chromane and tetrahydroquinoline fused spiroindolenines ingood yields.Furthermore,the auxiliary group that facilitates the hydride transfer process could be simplyremoved.

The dynamic impact force of debris flow on dams with a curved upstream face curved was investigated using laboratory experiments and a theoretical approach. Equations describing the impact force and maximum run-up height were derived. The experiments and theoretical considerations reveal that the impact force and maximum run-up height are mainly controlled by the Froude number(Fr) reduced by the cosine of the channel slope angle(cosa). Both the impact force and the maximum run-up height have a quadratic relation with the modified Froude number(Fr/(cosa)0.5).The experimental data and the results of the theoretical approach are in good agreement, indicating that the theoretical approach can be used in practical applications. It is concluded that the comparison between the curved-joint dam and the more conventional sharp-joint dam shows no differences in the maximum impact force and run-up height for the same modified Froude number.With the sharp-joint dam, the peak values of the impact force are reached more quickly than with the curved-joint dam.

Background:Time in range(TIR)refers to the time an individual spends within their target glucose range,which now has been popularized as an important metric to classify glycemic management and also recognized as an important outcome of current diabetes therapies.This study aimed to investigate the association between TIR and the severity of the urinary albumin excretion rate(UAER)in patients with type 2 diabetes mellitus(T2DM).Methods:We retrospectively analyzed the data of 1014 inpatients with T2DM at the Department of Endocrinology and Metabolism of Peking University International Hospital,China.TIR was defined as the percentage of blood glucose within the target range of 3.90-10.00 mmol/L.Urine samples for assessment of UAER were collected for 3 consecutive days from the start of hospitalization.Results:The TIR values for patients with normal urine levels of albumin,microalbuminuria,and macroalbuminuria were 70%±20%,50%±20%,and 30%±20%,respectively(allP83%.The incidences of microalbuminuria in Q1,Q2,Q3,and Q4 were 41.1%,21.6%,7.1%,and 5.5%(allP<0.001),respectively.The respective incidences of macroalbuminuria were 24.2%,1.1%,1.4%,and 0%(allP<0.001).In multinomial logistic regression analyses,TIR was significantly correlated with microalbuminuria(odds ratio[OR]0.58,95%confidence interval[CI]:0.52-0.65,P<0.001)and macroalbuminuria(OR 0.26,95%CI:0.18-0.38,P<0.001)after adjusting for age,sex,body mass index,diabetes duration,systolic blood pressure,and levels of triglycerides,glycosylated hemoglobin A1c,and creatinine.Conclusion:The proportion of blood glucose in TIR is closely related to the severity of UAER in patients with T2DM.

The intrinsic terminator in prokaryotic forms secondary RNA structure and terminates the transcription.However,leaking transcription is common due to varied terminator strength.Besides of the representative hairpin and U-tract structure,detailed sequence and thermodynamic features of terminators were not completely clear,and the effect of terminator on the upstream gene expression was unclearly.Thus,it is still challenging to use terminator to control expression with higher precision.Here,in E.Coli,we firstly determined the effect of the 3′-end sequences including spacer sequences and terminator sequences on the expression of upstream and downstream genes.Secondly,terminator mutation library was constructed,and the thermodynamic and sequence features differing in the termination efficiency were analyzed using the FlowSeq technique.The result showed that under the regulation of terminators,a negative correlation was presented between the expression of upstream and downstream genes(r=􀀀0.60),and the terminators with lower free energy corelated with higher upstream gene expression.Meanwhile,the terminator with longer stem length,more compact loop and perfect U-tract structure was benefit to the transcription termination.Finally,a terminator strength classification model was established,and the verification experiment based on 20 synthetic terminators indicated that the model can distinguish strong and weak terminators to certain extent.The results help to elucidate the role of terminators in gene expression,and the key factors identified are crucial for rational design of terminators,and the model provided a method for terminator strength prediction.

To assess the status and change trend of forest in China,an indicator framework was developed using SDG sub-indicators.In this paper,we propose an improved methodology and a set of workflows for calculating SDG indicators.The main modification include the use of moderate and high spatial resolution satellite data,as well as state-of-the-art machine learning techniques for forest cover classification and estimation of forest above-ground biomass(AGB).This research employs GF-1 and GF-2 data with enhanced texture information to map forest cover,while time series Landsat data is used to estimate forest AGB across the whole territory of China.The study calculate two SDG sub-indicators:SDG_(15.1.1) for forest area and SDG_(15.2.1) for sustainable forest management.The evaluation results showed that the total forest area in China was approximately 219 million hectares at the end of 2021,accounting for about 23.51%of the land area.The average annual forest AGB from 2015 to 2021 was estimated to be 105.01Mg/ha,and the overall trend of forest AGB change in China was positive,albeit with some spatial differences.

The ribosome-binding site(RBS)in the 5′untranslated region is recognized by 16S rRNA to start translation and is an essential element of the gene expression system.RBSs have been widely applied in regulating gene expression in various scenarios,including Gram-negative or Gram-positive bacteria.Here,we first rationally designed and constructed an RBS mutant library containing 66 RBSs.The strength of these RBSs in E.coli and C.glutamicum was characterized individually.The RBS strength spanned about 200 and 15 times in the two species,respectively.The strength of RBSs in C.glutamicum was generally lower than that of in E.coli.A total of 18 RBSs showed similar strength(within twofold differences)between the species in our study,and the correlation analysis of the strength of RBSs between E.coli and C.glutamicum(R^(2)=0.7483)revealed that these RBSs can be used across species.The sequence analysis revealed that the RBS region with two Ts stated was beneficial for RBS to function cross-species.The RBS characterized here can be used to precisely regulate gene expression in both hosts,and the characteristics of cross-species RBSs provide basic information for RBS rational design.

Introduction:Rabies is a viral zoonotic disease that causes progressive and fatal inflammation in the brain.Rabies has caused more than 5,000 human deaths in Shandong Province since 1955.This study aimed to analyze the epidemiological characteristics of human rabies in Shandong Province from 2010 to 2020 and to provide a scientific basis for policy changes.Methods:The data of reported human rabies cases from 2010 to 2020 were obtained from China’s National Notifiable Disease Reporting System,and data related to exposure and post-exposure prophylaxis(PEP)of the cases were acquired through case investigation.Results:A total of 414 human rabies cases were reported in Shandong Province from 2010 to 2020.Out of the 414 total cases,87.20%were primarily farmers;83.10%were over 40 years old.70.29%(265/377)belonged to categoryⅢexposure;and 96.67%(377/390)were exposed to the virus through infected dogs.The vaccine inoculation rate of these cases after exposure was only 8.85%;1.03%(4/390)had been vaccinated with rabies immunoglobulin,developing the disease 11 to 13 days after categoryⅢexposure.Conclusions:Dogs were still the primary animal hosts.Most rabies patients died due to no or inadequate post-exposure prophylaxis.Vaccine inoculation rates for dogs should reach the target of 70%as soon as possible.Health departments should improve the accessibility and quality of PEP;and improve the health literacy of the elderly in rural areas.

Background:Cardiovascular(CV)disease is the leading cause of morbidity and mortality in adults with type 2 diabetes(T2D).The aim of this study was to determine the CV risk in Chinese patients with T2D based on the 2019 European Society of Cardiology(ESC)and the European Association for the Study of Diabetes(EASD)guidelines on diabetes,pre-diabetes,and CV diseases.Methods:A total of 25,411 patients with T2D,who participated in the study of China Cardiometabolic Registries 3B study,were included in our analysis.We assessed the proportions of patients in each CV risk category according to 2019 ESC/EASD guidelines.Results:Based on the 2019 ESC/EASD guidelines,16,663(65.6%),1895(7.5%),and 152(0.6%)of patients were included in"very high risk,""high risk,"and"moderate risk"categories,respectively.The proportions of patients in each category varied based on age,sex,body mass index,and duration.While 58.7%(9786/16,663)of elderly patients were classified to"very high risk"group,89.6%(3732/4165)of patients with obesity were divided into"very high risk"group.Almost all patients with a duration of diabetes>10 years had"very high risk"or"high risk."However,6701(26.4%)of Chinese T2D patients,who had shorter duration,and one or two risk factors,could not be included in any category(the"unclear risk"category).Conclusions:In China,most patients with T2D have"very high"or"high"CV risk based on 2019 ESC/EASD guidelines.However,the risk of patients in"unclear risk"group needs to be further classified.

High harmonic generation(HHG)is an ideal probing source.In general,all harmonics are coupled with the corresponding input laser when generated,and for applications,they are separated using additional spectrometers.Herein,we report the angular isolation of relativistic harmonics at a predicted emission angle upon generation and,most importantly,a new phase-matching chain selection rule is derived to generate harmonics.Based on the laser plasma mechanism involving two non-collinear relativistic driving lasers,the nth harmonic carrying the information of both input lasers originates from its adjacent(n–1)th harmonic coupled with one of the input lasers.Meanwhile,the intensity and emission angle of the generated isolated harmonic are both greatly increased compared with those in the gas scheme.These results are satisfactorily verified by theoretical analysis and three-dimensional particle-in-cell simulations,which have physical significance and are essential for practical applications.
