The mechanisms of mushroom polysaccharides on immune functions and lipid metabolism of aged mammals have not been fully elucidated. In the present study， after assessing the impacts of one type of Lentinula edodes-derived polysaccharides， named L2， on immune functions and blood lipid profiles， isobaric tags for relative and absolute quantification (iTRAQ)-based proteomic profiling of the small intestinal tissues from aged mice treated with L2 was performed. L2 reversed immune function declines and modulated the lipid metabolism of aged mice evidenced by increased levels of serum TC， HDL-C， and LDL-C， and reduced levels of serum TG. Moreover， a total of 95 differentially regulated proteins (DRPs) were identified， of which 75 were up-regulated and 20 were down-regulated. Most of the DRPs were involved in intracellular and extracellular structure organization， and cellular and metabolic regulation. Particularly， approximately 16 and 9 DRPs participated in the regulation of immune functions and lipid metabolism， respectively. Furthermore， protein-protein interaction analysis highlighted that cadherin 1， plectin， cadherin-17， Ras GTPase-activating-like protein IQGAP2， and ezrin might be key proteins in response to L2 treatment. These findings provide new insights into the biological mechanisms of mushroom polysaccharides in anti-aging from a proteomic perspective.

Recently,immunotherapy has emerged as a promising and efective method for treating triple-negative breast cancer(TNBC).However,challenges still persist.Immunogenic cell death(ICD)is considered a prospective treatment and potential combinational treatment strategy as it induces an anti-tumor immune response by presenting the antigenic epitopes of dead cells.Nevertheless,the ICD process in TNBC and its impact on disease progression and the response to immunotherapy are not well understood.In this study,we observed dysregulation of the ICD process and verifed the altered expression of prognostic ICD genes in TNBC through quantitative real-time polymerase chain reaction(qRT-PCR)analysis.To investigate the potential role of the ICD process in TNBC progression,we determined the ICD-dependent subtypes,and two were identifed.Analysis of their distinct tumor immune microenvironment(TIME)and cancer hallmark features revealed that Cluster 1 and 2 corresponded to the immune“cold”and“hot”phenotypes,respectively.In addition,we constructed the prognostic signature ICD score of TNBC patients and demonstrated its clinical independence and generalizability.The ICD score could also serve as a potential biomarker for immune checkpoint blockade and may aid in the identifcation of targeted efective agents for individualized clinical strategies.

Due to the complex natures of dietary food components,it is difficult to elucidate how the compounds affect host health.Dietary food often selectively presents its mechanism of action on different cell types,and participates in the modulation of targeted cells and their microenvironments within organs.However,the limitations of traditional in vitro assays or in vivo animal experiments cannot comprehensively examine cellular heterogeneity and the tissue-biased influences.Single-cell RNA sequencing(sc RNA-seq)has emerged as an indispensable methodology to decompose tissues into different cell types for the demonstration of transcriptional profiles of individual cells.Sc RNA-seq applications has been summarized on three typical organs(brain,liver,kidney),and two representative immune-and tumor related health problems.The everincreasing role of sc RNA-seq in dietary food research with further improvement can provide sub-cellular information and the coupling between other cellular modalities.In this review,we propose utilizing sc RNAseq to more effectively capture the subtle and complex effects of food chemicals,and how they may lead to health problems at single-cell resolution.This novel technique will be valuable to elucidate the underlying mechanism of both the health benefits of food nutrients and the detrimental consequences food toxicants at the cellular level.

Creep damage evolution process of asphalt binder based on viscoelastic characteristics

The creep damage evolution of asphalt binder plays a significant role in investigating the formation mechanism of rutting. The rutting is the common distress at high temperatures for asphalt pavements. However, the reliability of existing creep damage parameters is under questioned, and these parameters cannot accurately illustrate the change of intrinsic microstructure for asphalt binder. In this paper, a new test protocol was given access to study the evolution of viscoelastic parameters during creep damage. It was completed by inserting the frequency sweep during creep test. The frequency sweep curve clusters were fitted by the generalized Kelvin-Voigt model for obtaining the change law of model parameters. Based on the change law and sensitivity analysis of model parameters,（E2+ E3）/2 was proposed as the creep damage variable. According to the curve of（E2+ E3）/2 versus loading time, two stages during the creep test could be identified: an approximate constant value in phase I and a linear decrease in phase II. Intrinsic differences about creep property of binders could be determined by this new proposed parameter. Above results not only ensure better understanding of the creep damage mechanism of binders, but also lay the theoretical foundation on predicting the anti-rutting performance of binders.

In this paper,we consider the-prize-collecting minimum vertex cover problem with submodular penalties,which generalizes the well-known minimum vertex cover problem,minimum partial vertex cover problem and minimum vertex cover problem with submodular penalties.We are given a cost graph and an integer.This problem determines a vertex set such that covers at least edges.The objective is to minimize the total cost of the vertices in plus the penalty of the uncovered edge set,where the penalty is determined by a submodular function.We design a two-phase combinatorial algorithm based on the guessing technique and the primal-dual framework to address the problem.When the submodular penalty cost function is normalized and nondecreasing,the proposed algorithm has an approximation factor of.When the submodular penalty cost function is linear,the approximation factor of the proposed algorithm is reduced to,which is the best factor if the unique game conjecture holds.

Background The sense of touch plays a crucial role in interactive behavior within virtual spaces,particularly when visual attention is absent.Although haptic feedback has been widely used to compensate for the lack of visual cues,the use of tactile information as a predictive feedforward cue to guide hand movements remains unexplored and lacks theoretical understanding.Methods This study introduces a fingertip aero-haptic rendering method to investigate its effectiveness in directing hand movements during eyes-free spatial interactions.The wearable device incorporates a multichannel micro-airflow chamber to deliver adjustable tactile effects on the fingertips.Results The first study verified that tactile directional feedforward cues significantly improve user capabilities in eyes-free target acquisition and that users rely heavily on haptic indications rather than spatial memory to control their hands.A subsequent study examined the impact of enriched tactile feedforward cues on assisting users in determining precise target positions during eyes-free interactions,and assessed the required learning efforts.Conclusions The haptic feedforward effect holds great practical promise in eyeless design for virtual reality.We aim to integrate cognitive models and tactile feedforward cues in the future,and apply richer tactile feedforward information to alleviate users'perceptual deficiencies.

Physics-informed neural network approach for heat generation rate estimation of lithium-ion battery under various driving conditions

Accurate insight into the heat generation rate(HGR) of lithium-ion batteries(LIBs) is one of key issues for battery management systems to formulate thermal safety warning strategies in advance.For this reason,this paper proposes a novel physics-informed neural network(PINN) approach for HGR estimation of LIBs under various driving conditions.Specifically,a single particle model with thermodynamics(SPMT) is first constructed for extracting the critical physical knowledge related with battery HGR.Subsequently,the surface concentrations of positive and negative electrodes in battery SPMT model are integrated into the bidirectional long short-term memory(BiLSTM) networks as physical information.And combined with other feature variables,a novel PINN approach to achieve HGR estimation of LIBs with higher accuracy is constituted.Additionally,some critical hyperparameters of BiLSTM used in PINN approach are determined through Bayesian optimization algorithm(BOA) and the results of BOA-based BiLSTM are compared with other traditional BiLSTM/LSTM networks.Eventually,combined with the HGR data generated from the validated virtual battery,it is proved that the proposed approach can well predict the battery HGR under the dynamic stress test(DST) and worldwide light vehicles test procedure(WLTP),the mean absolute error under DST is 0.542 kW/m^(3),and the root mean square error under WLTP is1.428 kW/m^(3)at 25℃.Lastly,the investigation results of this paper also show a new perspective in the application of the PINN approach in battery HGR estimation.

The development of receptive endometrium(RE) from pre-receptive endometrium(PE) for successful embryo implantation is a complex dynamic process in which the morphology and physiological states of the endometrial epithelium undergo a series of significant changes, including cell proliferation and apoptosis. However, the molecular mechanisms are not yet fully understood. In this study, a higher circRNA3669 level was observed in PE than in RE of goats. Functional assays revealed that this overexpression promoted the proliferation of goat endometrial epithelial cells(GEECs) by activating the expression of genes related to the PI3K/AKT-mTOR and MAPK pathways,thereby inhibiting apoptosis in vitro. Furthermore, circRNA3669 functioned as a competing endogenous RNA(ceRNA) to upregulate Reticulocalbin-2(RCN2) expression at the post-transcriptional level by interacting with and downregulating miR-26a in GEECs. In addition, RCN2, which is highly expressed in the PE of goats, was found to be regulated by β-estradiol(E2) and progesterone(P4). Our results demonstrated that RCN2 also affected the key proteins PI3K, AKT, mTOR, JNK, and P38 in the PI3K/AKT-mTOR and MAPK pathways, thereby facilitating GEECs proliferation and suppressing their apoptosis in vitro. Collectively, we constructed a new circRNA3669-miR-26aRCN2 regulatory network in GEECs, which further provides strong evidence that circRNA could potentially play a crucial regulatory role in the development of RE in goats.

The energy harvesting technology for the ubiquitous natural wind enables a desirable solution to the issue of distributed sensors in the bridge environmental sensing Internet of Things(Io T)system being restricted to conventional energy supply.In this work,a self-powered system based on a compact galloping piezoelectric-triboelectric energy harvester(GPTEH)is developed to achieve efficient wind energy harvesting.The GPTEH is constructed on the prototype of a cantilever structure with piezoelectric macro-fiber composite(MFC)sheets and a rectangular bluff body with triboelectric nanogenerators(TENGs).Through a special swing-type structural design with iron blocks inside the bluff body,the GPTEH exhibits preferable aerodynamic behavior and excellent energy conversion efficiency,compared to conventional cantilever kind of piezoelectric wind energy harvester(PWEH).The GPTEH also demonstrates the capability of high output power density(PEH of 23.65 W m^(-2)and TENG of 1.59 W m^(-2)),superior response wind speed(about 0.5 m s^(-1)),and excellent long-term stability(over 14000 cyclic tests).Furthermore,a power management system is developed to efficiently utilize the output energy from GPTEH to power the sensors and wirelessly transmit environmental data to the terminals.The proposed GPTEH-powered system exhibits a great potential for the bridge environmental monitoring and Io T technologies.
