Bone implantation surgery is often accompanied by bacterial infection,resulting in infectious bone non-union,pathological fracture and other serious consequences,which will aggravate the pain of patients.A non-antibiotic coating consisting of sodium dodecyl sulphate(sDs)and levulinic acid(LA)with different concentrations was prepared by the authors on the zinc–aluminium alloy(ZA6-1)using a wet chemistry treatment for orthopaedic *** influence of sDs/LA concentrations on the surface morphology,composition and performance of the developed coating was *** results showed that as-prepared coating on a zinc alloy surface could improve the substrate's corrosion resistance and increase the degradation rate from 0.82 to 19.70μm/year upon raising the sDs/LA ***,higher hydrophilicity(100%)and morphology,as well as good cell adhesion and differentiation(ALP>95%for 7 days)were observed on coated zinc *** increased sDs/LA concentration slightly weakens the biocompatibility and enhances the antibacterial performance of coated zinc alloys due to the synergistic effect of sDs/***,the coating comprising 6 wt.%sDs and 9 wt.%LA showed excellent antibacterial action with a high level of biocompatibility,confirming its potential application for orthopaedic implants.

Based on recent molecular data, it has been suggested that sporothrix globosa is the main causal agent of sporotrichosis in China. The objective of this study was to compare the morphology, growth characteristics, patterns of carbon source usage, and susceptibility to antifungal agents among sporothrix strains. A total of 15 clinical strains confirmed to be s. globosa, from three different regions of China, and 11 ex-type strains from the CBs-KNAW biodiversity center were obtained. The elongated conidia of s. pal/ida, s. variecibatus, s. schenckii, and s. schenckii luriei were clearly different from the subglobose and globose conidia of s. globosa strains. s. schenckii is able to assimilate sucrose, raffinose, and ribitol. susceptibility profiles of these sporothrix species were evaluated by measuring minimum inhibitory concentrations （MICs）. Fluconazole, itraconazole, terbinafine, and amphotericin B showed good activity against most s. globosa clinical isolates from China. Potassium iodide also showed a low MIC against s. pal/ida, while fluconazole showed a high MIC for s. mexicana, s. humicola, s. g/obosa, s. schenckii, and s. inflata; these strains might be considered tolerant. The species showed differences in susceptibility to antifungal drugs and should therefore be properly identified during diagnosis prior to designing therapeutic strategies.

Objectives: To quantitatively study the adhesive pro- perties of hepatoma cells to collagen Ⅳ coated artifi- cial basement membrane and to investigate the rele- vance of cell adhesive forces to the concentration of collagen Ⅳ. Methods: synchronous G1 and s phase cells were a- chieved using thymine-2-desoxyriboside and cochicine sequential blockage method and double thymine-2- desoxyriboside blockage method respectively. The adhesive forces of hepatoma cells were investigated by micropipette aspiration technique. Results: The adhesive forces of hepatoma cells to ar- tificial basement membrane were (107.78±65.44) ×10^(-10)N, (182.60±107.88)×10^(-10)N, (298.91± 144.13)×10^(-10)N when the concentration of the membrane coated by 1, 2, 5μg/ml collagen Ⅳ re- spectively (Psive forces of G1 and s phases hepatoma cells to artificial basement membrane were (275.86±232.80)×10^(-10)N and (161.16±120.40)×10^(-10)N respectively when the concentration of the membrane coated by 5μg/ml collagen Ⅳ (Psions: The adhesive forces of hepatoma cells to artifical basement membrane in direct proportion to the concentration of collagen Ⅳ suggests that the in- crease of basement membrane might be conducive to the chemotactic motion and adhesiveness of tumor cells. G1 phase cells are more capable of adhering to basement membrane than s phase cells. Hepatoma cells, especially G1 phase cells, may survive in blood circulation, and sequest and adhere in microcircula- tion, and get through basement membrane for re- mote metastasis.

Lactate is present at a high level in the microenvironment of mammalian preimplantation embryos in vivo and in vitro. However, its role in preimplantation development is unclear. Here, we report that lactate is highly enriched in the nuclei of early embryos when major zygotic genome activation(ZGA) occurs in humans and mice. The inhibition of its production and uptake results in developmental arrest at the 2-cell stage, major ZGA failure, and loss of lactate-derived H3K18lac, which could be rescued by the addition of Lac-Co A and recapitulated by overexpression of H3K18R mutation. By profiling the landscape of H3K18lac during mouse preimplantation development, we show that H3K18lac is enriched on the promoter regions of most major ZGA genes and correlates with their expressions. In humans, H3K18lac is also enriched in ZGA markers and temporally concomitant with their expressions. Taken together, we profile the landscapes of H3K18lac in mouse and human preimplantation embryos, and demonstrate the important role for H3K18lac in major ZGA, showing that a conserved metabolic mechanism underlies preimplantation development of mammalian embryos.
