Bombyx mori cytoplasmic polyhedrosis virus （BmCPV） is one of the major viral pathogens for the silkworm. To date, the molecular mechanism of BmCPV invasion has been unclear. We cloned the full length complementary （c）DNA which encodes the ubiquitin-activating enzyme El-domain containing proteinl （UbE1DC1） ofBombyx mori by using suppression subtraefive hybridization （SSH） and rapid amplification of com- plementary （c）DNA ends （RACE）. The full-length eDNA of UbE1DClgene is 1 919 bp, consisting of a 100 bp 5＇ untranslated region, a 637 bp 3＇ untranslated region and an 1 182 bp open reading frame （ORF）, encoding a 393 amino acid protein. The protein contained the THiF_MoeB_hesA_family domain, an adenosine triphosphate binding site, which belongs to the family of ubiquitin-activating enzyme El. Reverse transcription - polymerase chain reaction analysis from the silkworm tissues, namely silk gland, hemo- cyte, fat body, gonad and midgut revealed that UbE1DC1 was expressed in all the five tissues. The real-time quantitative polymerase chain reaction analysis indicated that the relative expression of UbE1DC1 in the normal midgut was approximately 9.78-fold of that in the BmCPV-infected midgut. It is implicated that UbEIDCI may play an important role in the interaction between the host and BmCPV invasion.

Increased risk due to nonsteroidal anti-inflammatory drugs (NSAIDs) therapy has been observed in patients. Although diaphragm-like stricture in the small bowel and colon induced by NSAIDs therapy has been rarely reported, gastric body diaphragm-like stricture has not been reported. We describe the first case of gastric body diaphragm-like stricture due to NSAIDs in a 44-year-old male patient who was successfully treated by an endoscopic approach to avoid complicated surgery. This case highlights new insight into the disadvantages of NSAIDs and provides new data for future clinical studies.

The in vitro isolation and analysis of pancreatic stem/progenitor cells are necessary for understanding their properties and function; however, the preparation of high-quality single-cell suspensions from adult pancreas is pre- requisite. In this study, we applied a cold trypsin-ethylenediaminetetraacetic acid （EDTA） digestion method to disas- sociate adult mouse pancreata into single cells. The yield of single cells and the viability of the harvested cells were much higher than those obtained via the two commonly used warm digestion methods. Flow cytometric analysis showed that the ratio of ductal or BCRPl-positive cells in cell suspensions prepared through cold digestion was con- sistent with that found in vivo. Cell culture tests showed that pancreatic epithelial cells prepared by cold digestion maintained proliferative capacity comparable to those derived from warm collagenase digestion. These results indicate that cold trypsin-EDTA digestion can effectively disassociate an adult mouse pancreas into viable single cells with minimal cell loss, and can be used for the isolation and analysis of pancreatic stem/progenitor cells.

Background The mechanisms underlying postoperative pain remain unclear. Neurotransmitters of excitatory and inhibitory amino acids play an important role in the transmission and modulation of pain in the spinal dorsal horn. This study aimed to investigate the changes of release of excitatory and inhibitory amino acids in the spinal cord during postoperative pain and to provide a novel theoretical basis for postoperative pain management. Methods Loop microdialysis catheters were implanted subarachnoidally via the atlanto-occipital membrane in 16 healthy Sprague-Dawley rats. All rats without neural deficits were divided into two groups, Group A and Group B, following 5 days of recovery. The tubes for microdialysis were connected and 25 μl microdialysate sample for baseline value was collected after one-hour washout in each rat. A plantar incision in the right hind paws of rats in Group A were performed under 1.2% isoflurane. All rats in Group B were only anesthetized by 1.2% isoflurane for the same duration. The microdialysate samples were collected at 3 hours, 1 day, 2 days and 3 days after the incision （or isoflurane anesthesia in Group B） in both groups. The cumulative pain scores were also assessed at the above time-points. The amino acids in the microdialysate samples were tested using high performance liquid chromatography. Results Within Group A, the release of aspartate and glutamate at 3 hours after the incision was significantly higher than the baseline values and the release of glycine at 1 day after the incision significantly increased compared with the baseline values （P〈0.01）. Within Group B, the release of neurotransmitters at each time point had no significant difference compared with the baseline values （P〉0.05）. The release of aspartate and glutamate at 3 hours after the incision in Group A was significantly higher than that in Group B （P〈0.01）.
