BACKGROUND Liuweiwuling Tablet(LWWL)is a Chinese patent medicine approved for the treatment of chronic inflammation caused by hepatitis B virus(HBV)*** studies have indicated an anti-HBV effect of LWWL,specifically in terms of antigen inhibition,but the underlying mechanism remains *** To investigate the potential mechanism of action of LWWL against *** In vitro experiments utilized three HBV-replicating and three non-HBV-replicating cell *** in vivo experiment involved a hydrodynamic injectionmediated mouse model with HBV *** and metabolomics were used to investigate the underlying mechanisms of action of *** In hepG2.1403F cells,LWWL(0.8 mg/mL)exhibited inhibitory effects on HBV DNA,hepatitis B surface antigen and pregenomic RNA(pgRNA)at rates of 51.36%,24.74%and 50.74%,*** inhibition rates of LWWL(0.8mg/mL)on pgRNA/covalently closed circular DNA in hepG2.1403F,hepG2.2.15 and hepG2.A64 cells were 47.78%,39.51%and 46.74%,*** of transcriptomics and metabolomics showed that the anti-HBV effect of LWWL was primarily linked to pathways related to apoptosis(PI3K-AKT,CASP8-CASP3 and P53 pathways).Apoptosis flow analysis revealed that the apoptosis rate in the LWWL-treated group was significantly higher than in the control group(CG)among HBV-replicating cell lines,including hepG2.2.15(2.92%±1.01%vs 6.68%±2.04%,PhepG2.A64(4.89%±1.28%vs 8.52%±0.50%,PhepG2.1403F(3.76%±1.40%vs 7.57%±1.35%,Phere were no significant differences in apoptosis rates between the non-HBV-replicating hepG2 cells(5.04%±0.74%vs 5.51%±1.57%,P>0.05),L02 cells(5.49%±0.80%vs 5.48%±1.01%,P>0.05)and LX2 cells(6.29%±1.54%vs 6.29%±0.88%,P>0.05).TUNEL staining revealed a significantly higher apoptosis rate in the LWWL-treated group than in the CG in the HBVreplicating mouse model,while no noticeable difference in apoptosis rates between the two groups

Improving flangeability of multiphase steel by increasing microstructural homogeneity

Multiphase microstructure significantly increases the strength,usually at the expense of flangeability because of lacking microstructure *** further improve the strength-flangeability of multiphase steel,the microstructural homogeneity was advanced by adjusting the hard martensite/austenite(M/A)*** strength-flangeability was measured via uniaxial tensile tests and hole expansion *** microstructures were characterized using a scanning electron microscope equipped with an electron backscatter diffraction detector and a transmission electron *** tests were supplementally used to quantitatively reveal the microstructural homogeneity of the *** show that the adjusted multiphase steel achieves an excellent ultimate tensile strength(~800 MPa)and flangeability(~135%hole expansion ratio).A promising homogeneous multiphase microstructure was obtained by controlling undercooled austenite transformed at about 600℃.This microstructure consists of soft polygonal ferrite,blocky bainitic ferrite,and hard M/A *** volume fraction of M/A islands is around 5%,and the average size is less than l *** nanoindentation analysis indicated that the participation of M/A islands impressively influenced the microstructural *** strain partition and better mechanical compatibility were present in the adjusted multiphase steel since the plasticity initiation started late,which resulted in a positive ***,avoiding M/A islands distributed in the chain along the rolling direction on the matrix hindered the possibility of voids coalescing into cracks and stabilized the flanging performance.

Blockchain Meets Generative Behavior Steganography:A Novel Covert Communication Framework for Secure IoT Edge Computing

The rapid development of Internet of things(Io T) and edge computing technologies has brought forth numerous possibilities for the intelligent and digital future. The frequent communication and interaction between devices inevitably generate a large amount of sensitive information. Deploying a blockchain network to store sensitive data is crucial for ensuring privacy and security. The openness and synchronicity of blockchain networks give rise to challenges such as transaction privacy and storage capacity issues, significantly impeding their development in the context of edge computing and Io T. This paper proposes a reliable fog computing service solution based on a blockchain fog architecture. This paper stores data files in the inter planetary file system(IPFS) and encrypts the file hash values used for retrieving data files with stream cipher encryption. It employs a steganographic transmission technique leveraging Alpha Zero's Gomoku algorithm to discretely transmit the stream cipher key across the blockchain network without a carrier, thus achieving dual encryption. This approach aims to mitigate the storage burden on the blockchain network while ensuring the security of transaction data. Experimental results demonstrate that the model enhances the transmission capacity of confidential information from kilobytes(KB) to megabytes(MB) and exhibits high levels of covert and security features.

A novel alternative for pyrogen detection based on a transgenic cell line

Pyrogen,often as a contaminant,is a key indicator affecting the safety of almost all parenteral drugs(including biologicals,chemicals,traditional Chinese medicines and medical devices).It has become a goal to completely replace the in vivo rabbit pyrogen test by using the in vitro pyrogen test based on the promoted‘reduction,replacement and refinement’principle,which has been highly considered by regulatory agencies from different *** used NF-κB,a central signalling molecule mediating inflammatory responses,as a pyrogenic marker and the monocyte line THP-1 transfected with a luciferase reporter gene regulated by NF-κB as an in vitro model to detect pyrogens by measuring the intensity of a fluorescence ***,we show that this test can quantitatively and sensitively detect endotoxin(lipopolysaccharide from different strains)and nonendotoxin(lipoteichoic acid,zymosan,peptidoglycan,lectin and glucan),has good stability in terms of NF-κB activity and cell phenotypes at 39 cell passages and can be applied to detect pyrogens in biologicals(group A&C meningococcal polysaccharide vaccine;basiliximab;rabies vaccine(Vero cells)for human use,freeze-dried;Japanese encephalitis vaccine(Vero cells),inactivated;insulin aspart injection;human albumin;recombinant human erythropoietin injection(CHO Cell)).The within-laboratory reproducibility of the test in three independent laboratories was 85%,80%and 80%and the interlaboratory reproducibility among laboratories was 83.3%,95.6%and 86.7%.The sensitivity(true positive rate)and specificity(true negative rate)of the test were 89.9%and 90.9%,*** summary,the test provides a novel alternative for pyrogen detection.

A correlation between the charge radii difference of mirror partner nucleiΔR_(ch) and the slope parameter L of symmetry energy has been built to ascertain the equation of state of isospin asymmetric nuclear *** this work,the influences of pairing correlations and isoscalar compression modulus on theΔRch are systematically investigated based on the Skyrme energy density functional *** calculated results suggest that the linear correlation betweenΔR_(ch) and L is decreased by the surface pairing *** slope parameter deduced from the difference of charge radii of mirror-pair nuclei ^(32)Ar-^(32)Si,^(36)Ca-^(36)S,^(38)Ca-^(38)Ar,and ^(54)Ni-^(54)Fe falls into the range of L=42.57−50.64 MeV,that is,the rather soft equation of state of asymmetric nuclear ***,the range of the slope parameter can also be influenced by the effective forces classified by various isoscalar incompressibility coefficients.

Previous carbene insertion to C–H bonds of 1,3-azoles relied on metal carbene species. herein, we report a metal-free C（sp2）–H bond functionalization of 1,3-azoles with trifluoroacetylsilanes. The reaction features mild conditions, broad substrate scope and wide functional group tolerance. The mechanistic study supports that the success of the reaction is probably attributed to the dual roles of trifluoroacetylsilanes under the photocatalyzed conditions: generating carbenes which undergo cyclopropanation and generating biradicals which promote ring-opening aromatization of the in situ generated fused cyclopropanes.

The integrated computational materials engineering(ICME)has achieved great success in accelerating the rational design and deployment of new *** is a new route of designing new materials and processes and highlighted by Materials Genome Initiative/Engineering that stresses the high-throughput computation in addition to high-throughput experimentation and materials *** article presents a brief review on the basic theories and multi-scale computational tools of ICME to design advanced steel grades,including the first-principles calculations,the CALPHAD method(i.e.,computational thermodynamics)fueled by dedicated databases,diffusion and phase-field simulations,as well as finite analysis methods and machine *** the ICME scheme to deal with steels,the CALPHAD method is considered as the core to readily consider multi-component systems and integrated to link the microscopic simulations(such as diffusion and phase field method to predict microstructure evolutions in response to external conditions)and macroscopic finite analysis method to deal with mechanical *** applications are also presented to address the new routes to carry out materials design,especially for advanced steels.
