MdbHLH162 connects the gibberellin and jasmonic acid signals to regulate anthocyanin biosynthesis in apple

Anthocyanins are secondary metabolites induced by environmental stimuli and developmental signals.The positive regulators of anthocyanin biosynthesis have been reported,whereas the anthocyanin repressors have been neglected.Although the signal transduction pathways of gibberellin(GA)and jasmonic acid(JA)and their regulation of anthocyanin biosynthesis have been investigated,the cross-talk between GA and JA and the antagonistic mechanism of regulating anthocyanin biosynthesis remain to be investigated.In this study,we identified the anthocyanin repressor MdbHLH162 in apple and revealed its molecular mechanism of regulating anthocyanin biosynthesis by integrating the GA and JA signals.MdbHLH162 exerted passive repression by interacting with MdbHLH3 and MdbHLH33,which are two recognized positive regulators of anthocyanin biosynthesis.MdbHLH162 negatively regulated anthocyanin biosynthesis by disrupting the formation of the anthocyanin-activated MdMYB1-MdbHLH3/33complexes and weakening transcriptional activation of the anthocyanin biosynthetic genes MdDFR and MdUF3GT by MdbHLH3 and MdbHLH33.The GA repressor MdRGL2a antagonized MdbHLH162-mediated inhibition of anthocyanins by sequestering MdbHLH162 from the MdbHLH162-MdbHLH3/33 complex.The JA repressors MdJAZ1 and MdJAZ2 interfered with the antagonistic regulation of MdbHLH162 by MdRGL2a by titrating the formation of the MdRGL2a-MdbHLH162 complex.Our findings reveal that MdbHLH162 integrates the GA and JA signals to negatively regulate anthocyanin biosynthesis.This study provides new information for discovering more anthocyanin biosynthesis repressors and explores the cross-talk between hormone signals.

Co/N co-doped flower-like carbon-based phase change materials toward solar energy harvesting

The photothermal conversion capacity of pristine organic phase change materials(PCMs)is inherently insufficient in solar energy utilization.To upgrade their photothermal conversion capacity,we developed bimetallic zeolitic imidazolate framework(ZIF)derived Co/N co-doped flower-like carbon(Co/N-FLC)-based composite PCMs toward solar energy harvesting.3D interconnected carbon framework with low interfacial thermal resistance,abundant carbon defects and high content of nitrogen doping,excellent localized surface plasmon resonance(LSPR)effect of Co nanoparticles,and light absorber Co_(3)ZnC in Co/N-FLC synergistically upgrade the photothermal capacity of(polyethylene glycol)PEG@Co/N-FLC composite PCMs with an ultrahigh photothermal conversion efficiency of 94.8%under 0.16 W/cm^(2).Uniformly anchored Co and Co_(3)ZnC nanoparticles in carbon framework guarantee excellent photon capture ability.Bridging carbon nanotubes(CNTs)in 2D carbon nanosheets further accelerate the rapid transport of phonons by constructing cross-connected heat transfer paths.Additionally,PEG@Co/N-FLC exhibits a thermal energy storage density of 100.69 J/g and excellent thermal stability and durable reliability.Therefore,PEG@Co/N-FLC composite PCMs are promising candidates to accelerate the efficient utilization of solar energy.

Hemolysis attributed to high dose vitamin C:Two case reports

BACKGROUND High-dose vitamin C treatment(HVCT)can reduce the adverse effect of chemotherapy and enhance the effect of antitumor therapy,which has been considered one of the safest alternative treatments.However,the severity of its adverse effects may have been underestimated.The most serious adverse effect is hemolysis,which may result in acute kidney injury or death.Although glucose-6-phosphate dehydrogenase(G6PD)deficiency is considered to be the main cause,the probability and pathological mechanism are not completely understood,leading to a lack of effective and standardized treatment methods.CASE SUMMARY Two patients with colorectal cancer developed hemolytic anemia after using 1 g/kg HVCT.In contrast to previous cases,the lowest hemoglobin level in the two cases was<50 g/L,which was lower than previously reported.This may be because Case 1 had chronic hepatitis B for many years,which caused abnormal liver reserve function,and Case 2 had grade II bone marrow suppression.Both patients improved and were discharged after blood replacement therapy.Our cases had the most severe degree of hemolysis but the best prognosis,suggesting that our treatment may be helpful for rescue of drug-induced hemolysis.This is the first review of the literature on hemolysis caused by HVCT,and we found that all patients with G6PD deficiency developed hemolysis after HVCT.CONCLUSION G6PD deficiency should be considered as a contraindication to HVCT,and it is not recommended for patients with bone marrow suppression,moderate-tosevere anemia,hematopoietic abnormalities,or abnormal liver and kidney function.Early blood purification and steroid therapy may avoid acute kidney injury or death caused by HVCT-related hemolytic anemia.

Analysis of China's Electricity Price and Electricity Burden of Basic Industries under the Carbon Peak Target before 2030

The Chinese government is deepening reformation of electricity prices during the 14th Five Year Plan period and has set a carbon emission reduction target of reaching carbon peak before 2030.In this context,will the carbon emission target influence electricity pricing and will electricity price influence competitiveness of Chinese main industries are two questions needing to be answered.This paper compares China's electricity price level with the selected major countries in the world,and four typical industries are selected to evaluate their electricity burden respectively.Then,the correlation between residential electricity price and industrial electricity price and the influencing factors is analyzed,from the perspectives of scale,structure and technology.According to the model obtained by regression analysis,the electricity price level and corresponding residential and industrial electricity burden in 2025 and 2030 are forecasted.Index Terms-Electricity burden,industrial electricity price,regression analysis,residential electricity price.

Hydroxysafflor Yellow A Promotes HaCaT Cell Proliferation and Migration by Regulating HBEGF/EGFR and PI3K/AKT Pathways and Circ_0084443

Objective:To investigate the effect and possible mechanism of hydroxysafflor yellow A(HSYA) on human immortalized keratinocyte cell proliferation and migration.Methods:HaCaT cells were treated with HSYA.Cell proliferation was detected by the cell counting kit-8 assay,and cell migration was measured using wound healing assay and Transwell migration assay.The mRNA and protein expression levels of heparin-binding epidermal growth factor(EGF)-like growth factor(HBEGF),EGF receptor(EGFR),phosphatidylinositol 3-kinase(PI3K),protein kinase B(AKT),mammalian target of rapamycin(mTOR),and hypoxia-inducible factor-1α(HIF-1α) were detected by quantitative real-time polymerase chain reaction(qRT-PCR) and Western blot,respectively.Circ_0084443-overexpressing HaCaT cells and empty plasmid HaCaT cells were constructed using the lentiviral stable transfection and treated with HSYA.The expression of circ_0084443 was detected by qRT-PCR.Results:HSYA(800 μmol/L) significantly promoted HaCaT cell proliferation and migration(P0.05).Conclusion:HSYA played an accelerative role in HaCaT cell proliferation and migration,which may be attributable to activating HBEGF/EGFR and PI3K/AKT signaling pathways,and had a particular inhibitory effect on the keratinocyte negative regulator circ_0084443.

Microneedle patch loaded with ferritin-nanocaged doxorubicin for locally targeted drug delivery and efficient skin cancer treatment

Ferritin has emerged as a promising nanocarrier for delivering therapeutic agents to tumours.However,the limited drug loading and the off-target impacts after systemic administration remain challenges for cancer treatment with ferritin-based agents.Herein,we develop a microneedle patch loaded with ferritin-nanocaged doxorubicin(DoxFe@Fn/MN)for skin cancer treatment.Briefly,doxorubicin(Dox)is encapsulated in ferritin(Fn)using an iron core-assisted strategy,which results in a 3.4-fold increase in Dox loading compared to the direct loading method.Then,a polyvinyl alcohol-based microneedle(MN)patch is used for the transdermal delivery of DoxFe@Fn,enabling targeted tumour accumulation of DoxFe@Fn and preventing off-target impacts.The released DoxFe@Fn can bind to CD71 highly expressed on skin cancer cells,facilitating its uptake.As a result,the DoxFe@Fn/MN therapy presents a robust antitumour effect in a melanoma tumour model,showing its potential as a promising therapeutic modality for skin cancer treatment.

Effect of oxide inclusions on MnS precipitates and tensile mechanical property of high-strength low-alloy steel

The key role of oxide inclusions on the microstructure and mechanical property of a high-strength low-alloy steel was investigated.The field emission scanning electron microscope equipped with energy-dispersive spectrometry was used to characterize MnS precipitates.Oxide inclusions play an important role in the shape control of MnS precipitates.More oxides fovored to decrease the size and the aspect ratio of MnS precipitates.With less oxide inclusions in the steel,approximately over 16.7%MnS precipitates were with aspect ratio a>5 and pure MnS precipitates accounted for 75.9%in number.However,with more oxide inclusions in the steel,only 7.4%MnS precipitates were with a>5 and pure MnS precipitates accounted for 60.1%in number.Refinement of MnS by oxide inclusions improved the strength and inhibited the anisotropy.More oxide inclusions in the steel increased the yield strength and tensile strength of the steel in both longitudinal and transverse directions,and lowered the anisotropy of the mechanical property.

Dual-channel mechano-phosphorescence:a combined locking effect with twisted molecular structures and robust interactions

Organic mechanoluminescence materials,featuring dual emission and ultralong phosphorescence characteristics,exhibit significant potential for applications in real-time stress sensing,pressure-sensitive lighting,advanced security marking techniques,and material breakage monitoring.However,due to immature molecular design strategies and unclear luminescence mechanisms,these materials remain rarely reported.In this study,we propose a valuable molecular design strategy to achieve dual-channel mechano-phosphorescence.By introducing the arylphosphine oxide group into a highly twisted molecular framework,enhanced intra-and intermolecular interactions could be achieved within rigid structures,leading to dual-channel mechanoluminescence with greatly promoted ultralong phosphorescence.Further investigations reveal the substantial boosting effect of intra-and intermolecular interactions on mechanoluminescence and ultralong phosphorescence properties by locking the highly twisted molecular skeleton.This work provides a concise and guiding route to develop novel smart responsive luminescence materials for widespread applications in material science.
