Differentiation of human umbilical cord blood stem cells into hepatocytes in vivo and in vitro

AIM： To study the condition and potentiality of human umbilical cord blood stem cells （HUCBSC） to differentiate into hepatocytes in vivo or in vitro. METHODS： In a cell culture study of human umbilical cord blood stem cell （HUCBSC） differentiation, human umbilical cord blood mononuclear cells （HUCBMNC） were separated by density gradient centrifugation. Fibroblast growth factor （FGF） and hepatocyte growth factor （HGF） and the supernatant of fetal liver were added in the inducing groups. Only FGF was added in the control group. The expansion and differentiation of HUCBMNC in each group were observed. Human alpha fetoprotein （AFP） and albumin （ALB） were detected by immunohistochemistry. In the animal experiments, the survival SD rats with acute hepatic injury after carbon tetrachloride （CCL4） injection 48 h were randomly divided into three groups. The rats in group A were treated with human umbilical cord blood serum. The rats in group B were treated with HUCBMNC transplantation. The rats in group C were treated with HUCBMNC transplantation followed by intraperitoneal cyclophosphamide for 7 d. The rats were killed at different time points after the treatment and the liver tissue was histopathologically studied and human AFP and ALB detected by immunohistochemistry. The human X inactive-specific transcript gene fragment in the liver tissue was amplified by PCR to find human DNA. RESULTS： The results of cell culture showed that adherent cells were stained negative for AFP or ALB in control group. However, the adherent cells in the inducing groups stained positive for AFP or ALB. The result of animal experiment showed that no human AFP or ALB positive cells present in the liver tissue of group A （control group）. However, many human AFP or ALB positive cells were scattered around sinus hepaUcus and the central veins of hepatic Iobules and in the portal area in group B and group C after one month. The fragment of human X chromagene could be detected in the liver tissue

Despite the diverse roles of tripartite motif(Trim)-containing proteins in the regulation of autophagy,the innate immune response,and cell differentiation,their roles in skeletal diseases are largely *** recently demonstrated that Trim21 plays a crucial role in regulating osteoblast(OB)differentiation in ***,how Trim21 contributes to skeletal degenerative disorders,including osteoporosis,remains ***,human and mouse bone specimens were evaluated,and the results showed that Trim21 expression was significantly elevated in bone tissues obtained from osteoporosis ***,we found that global knockout of the Trim21 gene(KO,Trim2^(1-/-))resulted in higher bone mass compared to that of the control *** further demonstrated that loss of Trim21 promoted bone formation by enhancing the osteogenic differentiation of bone marrow mesenchymal stem cells(BMSCs)and elevating the activity of OBs;moreover,Trim21 depletion suppressed osteoclast(OC)formation of RAW264.7 *** addition,the differentiation of OCs from bone marrow-derived macrophages(BMMs)isolated from Trim21^(-/-)and Ctsk-cre;Trim21^(f/f)mice was largely compromised compared to that of the littermate control ***,YAP1/β-catenin signaling was identified and demonstrated to be required for the Trim21-mediated osteogenic differentiation of *** importantly,the loss of Trim21 prevented ovariectomy(OVX)-and lipopolysaccharide(LPS)-induced bone loss in vivo by orchestrating the coupling of OBs and OCs through YAP1 *** current study demonstrated that Trim21 is crucial for regulating OB-mediated bone formation and OC-mediated bone resorption,thereby providing a basis for exploring Trim21 as a novel dual-targeting approach for treating osteoporosis and pathological bone loss.

The neutron capture cross sections(n,γ)of bromine were obtained using the time-of-flight technique at the Back-n facility of the China Spallation Neutron ***γ-rays originating from neutron-induced capture events were detected using four C_(6)D_(6) *** pulse-height weighting technique and double-bunch unfolding method based on Bayesian theory were used in the data *** deductions,normalization,and corrections were carefully considered to obtain reliable measurement *** multilevel R-matrix Bayesian code SAMMY was used to extract the resonance parameters in the resolved resonance region(RRR).The average cross sections in the unresolved resonance region(URR)were obtained from 10 to 400 *** experimental results were compared with data from several evaluated libraries and previous experi-ments in the RRR and *** TALYS code was used to describe the average cross sections in the *** astrophysical Maxwell average cross sections(MACSs)of ^(79,81)Br from kT=5 to 100 keV were calculated over a sufficiently wide range of neutron *** a thermal energy of kT=30 keV,the MACS value for ^(79)Br 682±68 mb was in good agreement with the KADoNiS v1.0 recommended *** contrast,the value of 293±29 mb for ^(81)Br was substantially higher than that of the evaluated database and the KADoNiS v1.0 recommended value.

Understanding changes in pathogen behavior(*** virulence,a shift in transmission channel) is critical for the public health management of emerging infectious *** degradation via gene depletion or inactivation is recognized as a pathoadaptive feature of the pathogen evolving with the ***,little is known about the exact role of genome degradation in affecting pathogenic behavior,and the underlying molecular detail has yet to be *** large-scale global avian-restricted Salmonella genomes spanning more than a century,we projected the genetic diversity of Salmonella Pullorum(bvSP)by showing increasingly antimicrobial-resistant ST92 prevalent in Chinese *** phylogenomic analysis identified three lineages in bvSP,with an enhancement of virulence in the two recently emerged lineages(L2/L3),as evidenced in chicken and embryo infection ***,the ancestor L1 lineage resembles the Salmonella serovars with higher metabolic flexibilities and more robust environmental tolerance,indicating stepwise evolutionary trajectories towards avian-restricted ***-genome analysis pinpointed fimbrial degradation from a virulent *** later engineeredfim-deletion mutant,and all other five fimbrial systems,revealed behavior switching that restricted horizontal fecal-oral transmission but boosted virulence in *** depleting fimbrial appendages,bvSP established persistent replication with less proinflammation in chick macrophages and adopted vertical transovarial transmission,accompanied by ever-increasing intensification in the poultry ***,we uncovered a previously unseen paradigm for remodeling bacterial surface appendages that supplements virulence-enhanced evolution with increased vertical transmission.

Malnutrition is a common comorbidity among patients with ***,no nutrition-screening tool has been recognized in this population.A quick and easy screening tool for nutrition with high sensitivity and easy-to-use is *** on the previous 25 nutrition-screening tools,the Delphi method was made by the members of the Chinese Society of Nutritional Oncology to choose the most useful item from each *** to these results,we built a nutrition-screening tool named age,intake,weight,and walking(AIWW).Malnutrition was defined based on the scored patient-generated subjective global assessment(PG-SGA).Concurrent validity was evaluated using the Kendall tau coefficient and kappa consistency between the malnutrition risks of AIWW,nutritional risk screening 2002(NRS-2002),and malnutrition screening tool(MST).Clinical benefit was calculated by the decision curve analysis(DCA),integrated discrimination improvement(IDI),and continuous net reclassification improvement(c NRI).A total of 11,360 patients(male,n=6,024(53.0%)were included in the final study cohort,and 6,363 patients had malnutrition based on *** on AIWW,NRS-2002,and MST,7,545,3,469,and1,840 patients were at risk of malnutrition,*** sensitivities of AIWW,NRS-2002,and MST risks were 0.910,0.531,and 0.285,and the specificities were 0.768,0.946,and *** Kendall tau coefficients of AIWW,NRS-2002,and MST risks were 0.588,0.501,and 0.326,*** area under the curve of AIWW,NRS-2002,and MST risks were0.785,0.739,and 0.630,*** IDI,c NRI,and DCA showed that AIWW is non-inferior to NRS-2002(IDI:0.002(-0.009,0.013),c NRI:-0.015(-0.049,0.020)).AIWW scores can also predict the survival of patients with *** missed diagnosis rates of AIWW,NRS-2002,and MST were 0.09%,49.0%,and 73.2%,*** showed a better nutritionscreening effect than NRS-2002 and MST for patients with cancer and could be recommended as an alternative nutritionscreening tool

Realtime trigger and localization of bursts are the key functions of GECAM,an all-sky gamma-ray monitor launched on 2020 December *** developed a multifunctional trigger and localization software operating in the CPU of the GECAM Electronic *** onboard software has the following features:high trigger efficiency for real celestial bursts with a suppression of false triggers caused by charged particle bursts and background fluctuation,dedicated localization algorithm optimized for both short and long bursts,and low time latency of the trigger information which is downlinked through the Global Short Message Communication service of the global BeiDou navigation *** paper provides a detailed description of the design and development of the trigger and localization software system for *** covers the general design,workflow,the main functions,and the algorithms used in the *** paper also includes on-ground trigger tests using simulated gamma-ray bursts generated by a dedicated X-ray tube,as well as an overview of the performance for real celestial bursts during its in-orbit operation.
