Metagenomic insights into microorganisms and antibiotic resistance genes of waste antibiotic fermentation residues along production,storage and treatment processes

Antibiotic fermentation residue(AFR)is nutrient-rich solid waste generated from fermentative antibiotic production process.It is demonstrated that AFR contains high-concentration of remaining antibiotics,and thus may promote antibiotic resistance development in receiving environment or feeding farmed animals.However,the dominate microorganisms and antibiotic resistance genes(ARGs)in AFRs have not been adequately explored,hampering understanding on the potential antibiotic resistance risk development caused by AFRs.Herein,seven kinds of representative AFRs along their production,storage,and treatment processes were collected,and multiple methods including amplicon sequencing,metagenomic sequencing,and bioinformatic approaches were adopted to explore the biological characteristics of AFRs.As expected,antibiotic fermentation producer was found as the predominant species in raw AFRs,which were collected at the outlet of fermentation tanks.However,except for producer species,more environment-derived species persisted in stored AFRs,which were temporarily stored at a semi-open space.Lactobacillus genus,classified as Firmicutes phylum and Bacilli class,became predominant bacterial taxa in stored AFRs,which might attribute to its tolerance to high concentration of antibiotics.Results from metagenomic sequencing together with assembly and binning approaches showed that these newly-colonizing species(e.g.,Lactobacillus genus)tended to carry ARGs conferring resistance to the remaining antibiotic.However,after thermal treatment,remaining antibiotic could be efficiently removed from AFRs,and microorganisms together with DNA could be strongly destroyed.In sum,the main risk from the AFRs was the remaining antibiotic,while environment-derived bacteria which tolerate extreme environment,survived in ARFs with high content antibiotics,and may carry ARGs.Thus,hydrothermal or other harmless treatment technologies are recommended to remove antibiotic content and inactivate bacteria before recycling of AFRs in pharmaceutical industry.

Field-based evidence for the enrichment of intrinsic antibiotic resistome stimulated by plant-derived fertilizer in agricultural soil

Animal manures have been demonstrated to enhance antibiotic resistance in agricultural soils.However,little is known about the effects of plant-derived fertilizer on soil antibiotic resistome.Herein,metagenomic sequencing was used to investigate the effects of a plantderived fertilizer processed from sugarcane and beet on soil antibiotic resistance genes(ARGs)in a soybean field along crop growth stages.ARG profiles in the soils amended by plant-derived fertilizer were compared with those in the soils amended by chicken manure.The abundance and diversity of total ARGs in the soils amended by plant-derived fertilizer were significantly(P<0.05)elevated at the sprout stage,to a level comparable to that in the manured soils.Whereas,unlike chicken manure mainly introducing manure-borne ARGs to soil,the plant-derived fertilizer was indicated to mainly enrich multidrug resistance genes in soil by nourishing indigenous bacteria.ARGs with abundances in amended soils significantly(P<0.05)higher than in unamended soils at the sprout stage of soybean were considered as enriched ARGs.Decrease in the abundance of the enriched ARGs was observed in both the amended soils from the sprout to the harvest.Network analysis further identified Proteobacteria and Bacteroidetes as the primary bacterial taxa involved in the temporal variation of the enriched ARGs in the soils amended by plant-derived fertilizer,while in manured soils were Firmicutes and Actinobacteria.As revealed by multivariate statistical analyses,variation of the enriched ARGs in the soils amended by plant-derived fertilizer was majorly attributed to the response of co-occurred bacteria to depleting nutrients,which was different from the failed establishment of manure-borne bacteria in the manured soils.Our study provided field-based evidence that plant-derived fertilizer stimulated the intrinsic antibiotic resistome,and proposed attention to the un-perceived risk since some clinically relevant ARGs originate and evolve from natural resistome.

Detection Algorithm of Laboratory Personnel Irregularities Based on Improved YOLOv7

Due to the complex environment of the university laboratory,personnel flow intensive,personnel irregular behavior is easy to cause security risks.Monitoring using mainstream detection algorithms suffers from low detection accuracy and slow speed.Therefore,the current management of personnel behavior mainly relies on institutional constraints,education and training,on-site supervision,etc.,which is time-consuming and ineffective.Given the above situation,this paper proposes an improved You Only Look Once version 7(YOLOv7)to achieve the purpose of quickly detecting irregular behaviors of laboratory personnel while ensuring high detection accuracy.First,to better capture the shape features of the target,deformable convolutional networks(DCN)is used in the backbone part of the model to replace the traditional convolution to improve the detection accuracy and speed.Second,to enhance the extraction of important features and suppress useless features,this paper proposes a new convolutional block attention module_efficient channel attention(CBAM_E)for embedding the neck network to improve the model’s ability to extract features from complex scenes.Finally,to reduce the influence of angle factor and bounding box regression accuracy,this paper proposes a newα-SCYLLA intersection over union(α-SIoU)instead of the complete intersection over union(CIoU),which improves the regression accuracy while increasing the convergence speed.Comparison experiments on public and homemade datasets show that the improved algorithm outperforms the original algorithm in all evaluation indexes,with an increase of 2.92%in the precision rate,4.14%in the recall rate,0.0356 in the weighted harmonic mean,3.60%in the mAP@0.5 value,and a reduction in the number of parameters and complexity.Compared with the mainstream algorithm,the improved algorithm has higher detection accuracy,faster convergence speed,and better actual recognition effect,indicating the effectiveness of the improved algorithm in this paper and its potential for practical application

Based on the potential-energy-surface calculation,the impact of different deformation degrees of freedom on a single-particle structure and binding energies in nuclei around^(152)Nd,located on one of the hexadecapole-deformation islands,is analyzed in a multi-dimensional deformation space.Various energy maps,curves and tables are presented to indicate nuclear properties.The calculated equilibrium deformations and binding energies with different potential parameters are compared with experimental data and other theories.It is found that the inclusion of the hexadecapole deformations,especially the axial one,can improve the theoretical description of both nuclear shapes and masses.In addition,our calculated potential-energy curve shows that a critical deformation-point,β_(2)≈0.4,exists—the triaxial(hexadecapole)deformation effect can be neglectable but the hexadecapole(triaxial)one plays an important role before(after)such a critical point.

Construction of biomimetic proton transport channels in metal-organic framework

Construction of proton transport channels in metal-organic frameworks(MOFs)with simple synthesis processes,high proton conductivities and good performance stabilities has been of great interest for proton exchange membrane fuel cell(PEMFC).Herein,we mimic the proton transport behavior of amino acid residues in bacteriorhodopsin,select UiO-66-COOH as the host,glycine and aspartic acid as the functional guest molecules,and then functionalize the MOF framework with amino acids to obtain biomimetic proton transport channels.This strategy endows UiO-66-COOH-Asp a high proton conductivity of 1.19×10^(-2)S/cm at 70℃and 98%RH,excellent cycle stability of performances and performance durability,which can be comparable to the reported MOFs-based proton conductors.Moreover,the proton conduction mechanism in UiO-66-COOH-Asp is elaborated in detail due to its visual structure,which is also one of the advantages of adopting MOFs as research platform,making it possible to optimize the structure-activity relationship of advanced materials.Notably,this strategy has clear objectives and simple synthesis,which has made certain contributions to both theoretical research and future industrial production of proton conductors.

Construction of daily precipitation series and the observational characteristics of extreme precipitation in Tianjin,China during 1888-2022

Given the difficulties in rescuing and ensuring the quality of long-term climate data,current studies on century-scale climate change are usually limited to annual and monthly data,resulting in the poor detection of extreme climate events and their changes before 1950.In this study,we reconstructed a daily precipitation series for Tianjin from 15 September 1887 to 31 December 2022 on the basis of the most comprehensive daily precipitation records collected from the Tianjin Meteorological Archive,China,and in reference to the precipitation analysis results based on the datasets developed by the Climatic Research Unit Time-Series version 4.06,Global Precipitation Climatology Centre and University of Delaware along with the application of various homogenisation methods for climate series.Our approach provides a complete and reliable century-long daily precipitation series for the study of regional or local extreme weather and climate events.The reconstructed daily dataset reveals that the annual precipitation amount and R95 intensity in Tianjin during 1888-2022 lack significant trends and have values of 0.74±6.99 and-1.84±3.22 mm per decade,respectively.On the annual and seasonal scales,the precipitation amount and R95 intensity,particularly those in autumn,have increased since the latter half of the 20th century relative to those in 1888-1950.However,the increase in precipitation amount and R95 intensity is relatively limited compared with that in atmospheric water vapour content due to surface warming,indicating the highly sensitive response of extreme precipitation events to warming.In addition,the estimates for the return periods of 5,10,20,50 and 100 years covering 1888-2022,1888-1950 and 1951-2022 depict that the intensity of heavy rain and above magnitude was highest in 1888-1950 and decreased in 1951-2022.

Effect of Acupuncture on Endometrial Blood Flow in Women Undergoing in vitro Fertilization Embryo Transfer: A Single Blind, Randomized Controlled Trial

Objective: To evaluate the effect of manual acupuncture on endometrial blood flow parameters by three-dimensional(3D) power Doppler ultrasound in women undergoing in vitro fertilization embryo transfer(IVF-ET). Methods: Seventy patients undergoing IVF-ET were equally randomized into traditional or sham acupuncture treatment group for totally 4 days(from the day of oocyte aspiration to the day of embryo transfer) of treatment by random envelope method at the Reproductive Medicine Center and Outpatient Department of Integrated Traditional Chinese and Western Medicine, Tongji Hospital, Tongji Medicine College, Huazhong University of Science and Technology from January 2013 to December 2015. Patients in the traditional acupuncture group accepted traditional acupuncture methods with manual acupuncture, and Zhongji(CV3), Qihai(CV 6), Sanyinjiao(SP6), Taichong(LR 3), Tianshu(ST 25), Guilai(ST 29) and Zusanli(ST 36) were chosen. Patients at the sham acupuncture group accepted shallow acupuncture methods at 4 non-meridian points at each shoulder and upper arm. Outcome measures included endometrial ultrasonic indices such as vascularization index(VI), flow index(FI) and vascularization flow index(VFI), endometrial thickness and volume, subendometrial VI(sVI), subendometrial FI(sFI), subendometrial VFI(sVFI), implantation rate, clinical pregnancy rate, abortion rate, live birth rate and number of live births. Results: Finally, 34 patients in the traditional acupuncture group and 35 in the sham acupuncture group completed this trial. VI, FI and VFI of the traditional acupuncture group were significantly higher than those in the sham acupuncture group(P0.05). Global gene expression profiling of perirenal brown adipose tissue whitening in goat kids reveals novel genes linked to adipose remodeling

Background Brown adipose tissue(BAT)is known to be capable of non-shivering thermogenesis under cold stimulation,which is related to the mortality of animals.In the previous study,we observed that goat BAT is mainly located around the kidney at birth,and changes to white adipose tissue(WAT)in the perirenal adipose tissue of goats within one month after birth.However,the regulatory factors underlying this change is remain unclear.In this study,we systematically studied the perirenal adipose tissue of goat kids in histological,cytological,and accompanying molecular level changes from 0 to 28 d after birth.Results Our study found a higher mortality rate in winter-born goat kids,with goat birthing data statistics.Then we used thermal imaging revealing high temperature in goat hips at postnatal 0 d and gradually decrease during 28 d.This is consistent with the region of perirenal BAT deposition and highlights its critical role in energy expenditure and body temperature regulation in goat kids.Additionally,we found a series of changes of BAT during the first 28 d after birth,such as whitening,larger lipid droplets,decreased mitochondrial numbers,and down-regulation of key thermogenesis-related genes(UCP1,DIO2,UCP2,CIDEA,PPARGC1a,C/EBPb,and C/EBPa).Then,we used RNA-seq found specific marker genes for goat adipose tissue and identified 12 new marker genes for BAT and 10 new marker genes for WAT of goats.Furthermore,12 candidate genes were found to potentially regulate goat BAT thermogenesis.The mechanism of the change of this biological phenomenon does not involve a large-scale death of brown adipocytes and subsequent proliferation of white adipocytes.While apoptosis may play a limited role,it is largely not critical in this transition process.Conclusions We concluded that perirenal BAT plays a crucial role in thermoregulation in newborn goat kids,with notable species differences in the expression of adipose tissue marker genes,and we highlighted some potential marker genes for goat BAT and WAT.Additionally,the change fr

Dear Editor,I ridocorneal endothelial syndrome(ICE)is a rare ocular disease first proposed by Eagle et al[1]in 1979.It is mainly characterized by corneal endothelial abnormalities,accompanied by iris atrophy,iris pigmented nodules and peripheral anterior synechiae.ICE syndrome is typically unilateral,and classified into three clinical variants based on the changes of the cornea and iris,including progressive iris atrophy,Chandler’s syndrome,and Cogan-Reese syndrome.

A comprehensive review of recent advancements and developments in heat exchanger network synthesis techniques

The rapid development of computational technology and the increasing energy demand have improved heat exchanger network(HEN)synthesis.The HEN synthesis involves several optimizations of matches,distributions of heat loads,and stream splitting of heat units.Thus,obtaining good results at high efficiency has been the main standard for evaluating the techniques in the research area of HEN synthesis.This paper first summarizes and analyzes the main contributions of the existing HEN synthesis techniques.To compare related data quantitively,information on ten typical cases is presented in this paper.Furthermore,recently improved solutions for commonly encountered existing literature cases demonstrate the evolution and competition trends in the field of HEN synthesis.The comparison data presented in this paper not only provide a useful reference for future research but also present the optimization directions.Based on the findings of this study,it is noted that there is still a large room for improvement,and current approaches are incapable of dealing with all HEN cases.Moreover,it is still difficult to escape a local optimum and overcome structural constraints when seeking the global optimum.As a follow-up to the current work,the parallel computing mode and adaptively coordinating the ratio of global and local searching abilities are major development trends for future investigation.
