Asymptotic false discovery control of the Benjamini-Hochberg procedure for pairwise comparisons

In a one-way analysis-of-variance(ANOVA) model,the number of pairwise comparisons can become large even with a moderate number of *** by this,we consider a regime with a growing number of groups and prove that,when testing pairwise comparisons,the Benjamini-Hochberg(BH) procedure can asymptotically control false discoveries,despite the fact that the involved t-statistics do not exhibit the wellknown positive dependence structure required for exact false discovery rate(FDR) *** Tukey's perspective that the difference between the means of any two groups cannot be exactly zero,our main result provides control over the directional false discovery rate and directional false discovery proportion.A key technical contribution of our work is demonstrating that the dependence among the t-statistics is sufficiently weak to establish the convergence result typically required for asymptotic FDR *** analysis does not rely on conventional assumptions such as normality,variance homogeneity,or a balanced design,thereby offering a theoretical foundation for applications in more general settings.

This study evaluated toxic efficacy of Eupatorium adenophorum extracts,against the Kunming *** acute study,we firstly tested median lethal dose(LD50)in mice of three cadinene sesquiterpenes 2-deoxo-2-(acetyloxy)-9-oxoageraphorone(DAOA),9-oxo-agerophorone(OA)and 9-oxo-10,11-dehydro-agerophorone(ODA)from Eupatorium adenophorum(Ea).DAOA(215–4640 mg/kg BW,given orally)showed lowest LD50 at 926 mg/kg BW for male mice in contrast with OA(1470 mg/kg BW)and ODA(1470 mg/kg BW).In sub-acute study,repeated doses(75–300 mg/kg BW,for 7 days)of DAOA/OA increased blood parameters,liver and spleen index in dose dependent relationship,along with decrease in thymus *** blood biochemical and histopathological examination showed that DAOA/OA dose 300 mg/kg BW significantly causes pathological changes of hepatic lobules and hepatocytes,which are consistent with cholestasis and hepatic injury.75 mg/kg dose of DAOA/OA was found to be approximately/totally safe over the span of 7 days treatment showing no change in all above described *** sesquiterpenes guarantee low risk to environment as a type of low toxic botanical components,which may find potential application in biopesticides development field.

ERK1/2 Activities Are Dispensable for Oocyte Growth but Are Required for Meiotic Maturation and Pronuclear Formation in Mouse

Previous studies revealed that extracellular regulated kinase-1 and -2 （ERK1/2） cascade plays pivotal roles in regulating oocyte meiotic cell cycle progression. However, most knowledge about the in vivo function of ERK1/2 in mammalian oocytes was indirectly obtained from analyzing the phenotypes of Mos knockout mice. In this study, we knocked out Erkl and Erk2 in mouse oocytes as early as the primordial follicle stage using the well-characterized Gdp-Cre mouse model, and for the first time directly investigated the in vivo function of ERK1/2 in mouse oocytes. In this novel mouse model, we observed that ERK1/2 activities in oocyte are dispensable for primordial follicle maintenance, activation and follicle growth. Different from the Mos null oocytes, the ERK1/2-deleted oocytes had well-assembled spindles at metaphase I （MI）, extruded polar body-I （PB1） with normal sizes, and did not undergo a full parthenogenetic activation characterized for pronuclear formation. However, the ovulated ERK1/2-deleted oocytes had poorly-assembled metaphase II （MII） spindles, spontaneously released polar body-2 （PB2）, and were arrested at another metaphase called metaphase III （MIII）. In addition, ERK1/2 deletion prevented male pronuclear formation after fertilization, and caused female infertility. In conclusion, these results indicate that ERK1/2 activities are required for not only MII-arrest maintenance, but also efficient pronuclear formation in mouse oocytes.

Objective:To investigate the effects of Alpiniae oxyphyllae Fructus(AOF)on renal lipid deposition in diabetic kidney disease(DKD)and elucidate its molecular ***:The mechanism of AOF in treating DKD was explored by network pharmacological enrichment analysis,molecular docking,and molecular dynamics *** effects of AOF on renal function and lipid deposition were assessed in a mouse model of DKD and high glucose-stressed HK-2 *** viability and lipid accumulation were detected by CCK8 and oil red O *** expressions of PPARαand fatty acid oxidation-related genes(ACOX1 and CPT1A)were detected by quantitative RT-PCR,Western blot,and ***,PPARαknockdown was performed to examine the molecular mechanism of AOF in treating ***:Network pharmacological enrichment analysis,molecular docking,and molecular dynamics simulation showed that the active compounds in AOF targeted PPARαand thus transcriptionally regulated ACOX1 and *** lowered blood glucose,improved dyslipidemia,and attenuated renal injury in DKD ***-containing serum accentuated high glucose-induced decrease in cell viability and ameliorated lipid ***,it significantly upregulated the expression of PPARα,ACOX1,and CPT1A in both in vivo and in vitro experiments,which was reversed by PPARα***:AOF may promote fatty acid oxidation via PPARαto ameliorate renal lipid deposition in DKD.

Predicting the efficiency of arsenic immobilization in soils by biochar using machine learning

Vitamin C(L-ascorbic acid;VC) plays an important role in many biological processes,through donating electrons to act as a potent water-soluble anti-oxidant and acting as a cofactor for the family of Fe(ll) 2-oxoglutarate dependent *** catalyze the demethylation of5-methylcytosine(5 mC) in DNA,N-methyladenosine(m A) in RNA and lysine residues in histone proteins,and thereby dynamically regulate epigenetic modification of local chromatin and RNA structures,including ten-eleven-translocation 1-3(Tet1-3) enzymes,fat mass and obesity-associated(Fto),AlkB homologue 5(Alkbh5) AlkB enzymes,and Jumonji C-domain-containing *** studies demonstrated that VC supplement during in vitro maturation enhances cytoplasmic maturation and developmental competence of porcine oocytes derived from cumulus-oocyte complexes(COCs),however,the underlying molecular mechanism remains *** the present study,we reported that addition of one derivative of VC,250μM magnesium ascorbyl phosphate(AA2 P),greatly enhanced the nuclear maturation(as indicated by higher rate of first polar body extrusion and increased mRNA level of Bmpl5),cytoplasmic maturation(reduced reactive oxygen species level) and developmental potency(higher cleavage and blastotocyst rates of parthenotes and decreased mRNA levels of Bax and Caspase3) of pig oocytes derived from COCs after 44 h in vitro *** found that 250 u.M AA2 P supplementation in maturation culture of porcine COCs induced dynamic erasure of 5 mC,m A and H3 K27 me3,but establishment of H3 K4 me3 and K3 K36 me3 in mature porcine *** then found that 250 u.M AA2 P could decrease mRNA and protein abundances of DNMT3 a,but increase levels of ***,we showed that mRNA abundance of HIF-1 alpha,HIF-2 alpha,TET3,METTL14,KDM5 b and EED were suppressed by 250μM AA2 P treatment,while elevating DNMT3 b to a higher level in mature *** findings show VC can regulate epigenetic maturation of oocytes,and thus
