Heart failure(HF),which is a common and serious complication of diabetes,has received more attention in recent years.One reason for this focus on HF is that,with a large reduction in atherothrombotic events after acute myocardial infarction,HF is a relatively more frequently encountered cardiac outcome in patients with type 2 diabetes(T2DM).Another reason is the unexpected reduction in hospitalization for HF observed in cardiovascular outcome trials with sodium-glucose co-transporter-2 inhibitors(SGLT2is).[1-3]These findings led to speculation by clinicians and researchers on whether more effort should be made to differentiate patients with diabetes at a high risk of developing HF.

Rhodioloside has been shown to protect cells from hypoxia injury,and bone marrow mesenchymal stem cells have a good effect on tissue repair.To study the effects of rhodioloside and bone marrow mesenchymal stem cells on spinal cord injury,a rat model of spinal cord injury was established using the Infinite Horizons method.After establishing the model,the rats were randomly divided into five groups.Rats in the control group were intragastrically injected with phosphate buffered saline(PBS)(5μL).PBS was injected at 6 equidistant points around 5 mm from the injury site and at a depth of 5 mm.Rats in the rhodioloside group were intragastrically injected with rhodioloside(5 g/kg)and intramuscularly injected with PBS.Rats in the mesenchymal stem cell(MSC)group were intramuscularly injected with PBS and intramuscularly with MSCs(8×10^6/mL in a 50-μL cell suspension).Rats in the Ad-HIF-MSC group were intragastrically injected with PBS and intramuscularly injected with HIF-1 adenovirus-infected MSCs.Rats in the rhodioloside+Ad-HIF-MSC group were intramuscularly injected with MSCs infected with the HIF-1 adenovirus and intragastrically injected with rhodioloside.One week after treatment,exercise recovery was evaluated with a modified combined behavioral score scale.Hematoxylin-eosin staining and Pischingert’s methylene blue staining were used to detect any histological or pathological changes in spinal cord tissue.Levels of adenovirus IX and Sry mRNA were detected by real-time quantitative polymerase chain reaction and used to determine the number of adenovirus and mesenchymal stem cells that were transfected into the spinal cord.Immunohistochemical staining was applied to detect HIF-1 protein levels in the spinal cord.The results showed that:(1)compared with the other groups,the rhodioloside+Ad-HIF-MSC group exhibited the highest combined behavioral score(P<0.05),the most recovered tissue,and the greatest number of neurons,as indicated by Pischingert’s methylene blue staining.(2)Compared with the PBS group,HIF-1 protein ex

With the increasing use of immune checkpoint inhibitors(ICI)including anti-cytotoxic T lymphocyte associated antigen-4(CTLA-4)and anti-programmed cell death-1(PD-1)in cancers,ICI-induced type 1 diabetes has been reported throughout the world.In this review,we aim to summarize the characteristics of this disease and discuss the mechanism of it.As an immune-related adverse event,type 1 diabetes developed after the administration of anti-PD-1 or anti-PD-ligand 1(PD-L1)in the combination with or without anti-CTLA-4.It usually presented with acute onset,and 62.1%of the reported cases had diabetic ketoacidosis.Only a third of them had positive autoantibodies associated with type 1 diabetes.Susceptible HLA genotypes might be associated.T-cell-stimulation by blocking of the interaction of PD-1 and PD-L1 in pancreatic β cells was the main mechanism involved in the pathology.Insulin was the only effective treatment of ICI-induced type 1 diabetes.In conclusions,ICI-induced type 1 diabetes is a potentially life-threating adverse event after the immunotherapy of cancers.Screening and early recognition is important.Further investigation of the mechanism may help to better understand the pathology of type 1 diabetes.

Background:The incidence of liver injury caused by anti-tuberculous(TB)drugs is very high.However,owing to a lack of sufficient evidence,preventive use of hepatoprotective drugs is not yet recommended.Therefore,we aimed to assess the protective effect of hepatoprotective drugs for anti-TB drug-induced liver injury.Methods:We conducted a literature search in China Biology Medicine disc,China National Knowledge Infras-tructure,WanFang,Chinese Scientific and Technological Journal,PubMed,Cochrane Library,Web of Science,and Embase.We performed meta-analysis using R 4.0 and Review Manager 5.3 software.Results:A total of 18 studies involving 3589 patients from 2 groups were included.Use of hepatoprotective drugs contributed to a lower incidence of liver injury as compared with conventional anti-TB treatment alone(relative risk[RR]=0.39,95%confidence interval[CI]:0.28-0.53,p<0.001).In subgroup analysis,significant protective effects were noted for mild liver injury(RR=0.30,95%CI 0.15-0.58),moderate(or severe)liver injury(RR=0.35,95%CI 0.19-0.65),and liver injury within 2-4 weeks(RR=0.37,95%CI 0.19-0.71).We also found a statistically significant difference in the incidence of drug withdrawal(RR=0.58,95%CI 0.34-0.97,p=0.040).Conclusions:Our results demonstrate that hepatoprotective drugs are effective in preventing liver injury in pa-tients receiving anti-TB treatment,to some extent.

As an active metabolite of venlafaxine and emerging antidepressant,Odesmethylvenlafaxine(ODVEN) was widely detected in different water bodies,which caused potential harm to human health and environmental safety.In this study,the comparative work on the ODVEN degradation by UV(254 nm) and UV-LED(275 nm) activated sodium percarbonate(SPC) systems was systematically performed.The higher removal rate of ODVEN can be achieved under UV-LED direct photolysis(14.99%) than UV direct photolysis(4.57%) due to the higher values of photolysis coefficient at the wavelength 275nm.Significant synergistic effects were observed in the UV/SPC(80.38%) and UV-LED/SPC(53.57%) systems and the former exhibited better performance for the elimination of ODVEN.The degradation of ODVEN all followed the pseudo-first-order kinetics well in these processes,and the pseudo-first-order rate constant(kobs) increased with increasing SPC concentration.Radicals quenching experiments demonstrated that both ·OH and CO_(3)·-were involved in the degradation of ODVEN and the second-order rate constant of ODVEN with CO_(3)·-(1.58 × 10^(8)(mol/L)-1sec-1) was reported for the first time based on competitive kinetic method.The introduction of HA,Cl-,NO_(3)-and HCO_(3)-inhibited the ODVEN degradation to varying degrees in the both processes.According to quantum chemical calculation,radical addition at the ortho-position of the phenolic hydroxyl group was confirmed to be the main reaction pathways for the oxidation of ODVEN by·OH.In addition,the oxidation of ODVEN may involve the demethylation,H-abstraction,OH^(-)addition and C-N bond cleavage.Eventually,the UV-LED/SPC process was considered to be more cost-effective compared to the UV/SPC process,although the UV/SPC process possessed a higher removal rate of ODVEN.

To the Editor:The prevalence of type 2 diabetes mellitus(T2DM)in older Chinese adults(≥60 years)is over 20%.[1]In comparison to younger patients,older diabetes patients tend to have lower rate of adequate glycemic control(<7.0%hemoglobin A1c[HbA1c])and a higher risk of hypoglycemia.[2]Currently,there are limited data on the clinical characteristics and treatment patterns of older T2DM patients in China.We conducted a nation-wide cross-sectional study to examine the clinical characteristics,anti-diabetic treatment patterns,and glycemic control in older(≥60 years)Chinese patients.Factors associated with poor glycemic control were identified with multivariate regression analysis.

Background Despite extensive research evaluating the association between prenatal exposure to secondhand smoke(SHS)and the development of autism spectrum disorders(ASD),no study has investigated the association by considering the pre-conceptional period.This study aimed to investigate the associations of pre-conceptional and prenatal SHS exposure and the development of ASD among toddlers.Methods In this cross-sectional study,parents of 6049 toddlers aged 16–30 months were recruited from 7 tertiary hospitals,21 communities,and 7 kindergartens located in seven cities in six provinces from five geographical regions of China.We analyzed the associations of SHS exposure and the odds of ASD among toddlers in different exposure windows(pre-conceptional and/or prenatal periods).Data were analyzed from November 2021 to January 2022.Results Among the 6049 toddlers included in the analysis[22.7(4.1)months;44.8%girls],71 were identified and diagnosed with ASD.Compared with the unexposed toddlers,toddlers with pre-conceptional SHS exposure had higher odds of ASD(OR 2.30,95%CI 1.36–3.84),while we observed a non-significantly positive association regarding prenatal SHS exposure.When considering both pre-conceptional and prenatal periods,toddlers who were continuously exposed to SHS during these two periods had higher odds than those without SHS exposure(OR 2.32,95%CI 1.24–4.14).Conclusion We reported positive SHS–ASD associations when exposed during the pre-conceptional period and continuously exposed during pre-conceptional and prenatal periods,emphasizing the critical window of pre-conception for targeted intervention on smoking.

Dysregulated RUNX1 Predicts Poor Prognosis by Mediating Epithelialmesenchymal Transition in Cervical Cancer

Objective Runt-related transcription factor 1(RUNX1)has been proven to be over-expressed and vital in many malignancies.However,its role in cervical cancer is still unclear.Methods Some online databases(Oncomine,GEPIA,UALCAN,LinkedOmics,and others)were used to explore the expression level,prognostic significance,and gene mutation characteristics of RUNX1 in cervical cancer.The protein levels of RUNX1 in cervical cancer were measured by immunohistochemistry(IHC).The functional changes of cervical cancer cells were measured in vitro after decreasing RUNX1.Results Bioinformatic results revealed that RUNX1 was upregulated in cervical cancer compared to normal tissues.Moreover,over-expression of RUNX1 was significantly correlated with cervical cancer patients’clinical parameters(e.g.,individual cancer stages,patients’age,nodal metastasis status,and others).Meanwhile,functional enrichment analysis of RUNX1-related genes indicated that RUNX1 was mainly involved in the epithelial-mesenchymal transition(EMT)process in cervical cancer.Furthermore,RUNX1 may be upregulated by hsamiR-616-5p and hsa-miR-766 identified by miRDB,TargetScan,and miRWalk.Finally,RUNX1 was upregulated in cervical cancer compared to normal tissues by IHC in collected cervical cancer samples.The invasion and migration abilities of cervical cancer cells were significantly reduced by repressing EMT after knocking down RUNX1 in vitro.Conclusion RUNX1 was highly expressed in cervical cancer,and upregulated RUNX1 could significantly promote the invasive abilities of cervical cancer cells by inducing EMT.Therefore,RUNX1 may be a potential biomarker for early diagnosis and targeted therapy of cervical cancer.

Primary pigmented nodular adrenocortical disease （PPNAD） causes adrenocorticotropic hormone （ACTH）-independent Cushing＇s syndrome （CS）,which is the most frequent endocrine manifestation of Carney complex （CNC）.[1] In the disease process of PPNAD,both presadrenal glands are involved and feature small brown-black nodules separated by the atrophic adrenal cortex.
