Induction of apoptosis of human gastric carcinoma SGC-7901 cell line by 5,7-dihydroxy-8-nitrochrysin in vitro

aiM： To investigate the effect of 5, 7-dihydroxy-8- nitrochrysin （NOChR） on apoptosis of human gastric carcinoma SGC-7901 cell line.METHODS： SGC-7901 cells were cultured in vitro and the inhibitory effect of NOChR on proliferation of SGC-7901 cells was measured by using an Ml-r assay. NOChR-induced apoptosis rate of SGC-7901 cells was detected using flow cytometry （FCM） with PI staining. DNA ladder bands were observed by DNA agarose gel electrophoresis. The influence of NOChR on the proxisome proliferator-activated receptor-γ （PPARγ）, Bcl-2 and Bax protein expression of SGC-7901 cells was analyzed by Western blot.RESULTS： MIF assay showed that NOChR markedly inhibited proliferation of SGC-7901 cells in a dose- dependent manner, and when ICso was 4.14 μmol/L, the potency of NOChR was 10 times than that of lead compound, chrysin （ChR, IC50 was 40.56 μmol/L）, and was similar to 5-fluorouracil （5-FU, IC50 was 4.51 μmol/L）. FCM with propidium iodide （PI） staining demonstrated that the apoptosis rates of SGC-7901 cells treated with 1.25, 5.00 and 20.00 μmol/L NOChR for 48 h were 9.8% 4- 0.2%, 36.8% 4- 1.9% and 45.5% 4- 3.5%, respectively, and were significantly higher when treated with 5.00 and 20.00 μmol/L NOChR than that with 20.00 μmol/L ChR （12.9% 4- 1.5%）. DNA agarose gel electrophoresis showed that treatment of SGC-7901 cells with 20.00 μmol/L NOChR for 48 h resulted in typical DNA ladder bands of DNA of SGC-7901 cells, which could be eliminated by treating with 10.00 μmol/L GW9662, a blocker of PPARy. Western blot analysis revealed that after 24 h of treatment with 20.00 μmol/L NOChR, PPARgamma and Bax protein expression of SGC-7901 cells increased but Bcl-2 expression decreased; however, pre-incubation with 10.00 μmol/L GW9662 could efficiently antagonize and weaken the regulatory effect of 20.00 μmol/L NOChR on Bax and Bcl-2 protein expression of SGC-7901 cells. CONCLUSION： NOChR induces apoptosis of SGO7901 cell lines by activating PPARy and decreasing rat

Attachment of tyrosinase on mixed self-assembled monolayers for the construction of electrochemical biosensor

A mixed self-assembled monolayers （SAMs） of thioctic acid （T-COOH） and thioctic acid amide （T-NH2） were used to immobilize tyrosinase for fabricating biosensor. The results showed that the mixed SAMs prepared from solution at the ratio of 1：4 provided an excellent microenvironment for enzymatic reaction between tyrosinase and substrate. The biosensor exhibited a fast response and high sensitivity for sensing substrate.

A Novel Divide and Conquer Solution for Long-term Video Salient Object Detection

Recently,a new research trend in our video salient object detection(VSOD)research community has focused on enhancing the detection results via model self-fine-tuning using sparsely mined high-quality keyframes from the given *** such a learning scheme is generally effective,it has a critical limitation,i.e.,the model learned on sparse frames only possesses weak generalization *** situation could become worse on“long”videos since they tend to have intensive scene ***,in such videos,the keyframe information from a longer time span is less relevant to the previous,which could also cause learning conflict and deteriorate the model ***,the learning scheme is usually incapable of handling complex pattern *** solve this problem,we propose a divide-and-conquer framework,which can convert a complex problem domain into multiple simple ***,we devise a novel background consistency analysis(BCA)which effectively divides the mined frames into disjoint *** for each group,we assign an individual deep model on it to capture its key attribute during the fine-tuning *** the testing phase,we design a model-matching strategy,which could dynamically select the best-matched model from those fine-tuned ones to handle the given testing *** experiments show that our method can adapt severe background appearance variation coupling with object movement and obtain robust saliency detection compared with the previous scheme and the state-of-the-art methods.

In this study,microstructure and mechanical behavior of two types of Mg-based bimetal plates with a high formability sleeve were systematically studied,with a great emphasis on the effect of the interface characteristic and the sleeve fraction on the plasticity of composite *** rule of mixtures(ROM)for elongation was also *** results show that when there is no or thin diffusion layer with thickness of about 3μm,Mg-based bimetal plates have a good plasticity with elongation of about 19-24%,and the ROM predicted elongations are very close to the experimental *** contrast,with a diffusion layer about 95-155μm thick,Mg-based bimetal plates exhibit a poor plasticity with elongation of about 11-17%,and the experimental elongations largely deviate from the ROM *** plasticity of Mg-based bimetal plates increases with increasing sleeve *** study provides new insights on the plastic deformation of Mg-based bimetal composites with a high formability sleeve.

In this paper,we discuss the related properties of some particular derivations in semihoops and give some characterizations of ***,we prove that every Heyting algebra is isomorphic to the algebra of all multiplicative derivations and show that every Boolean algebra is isomorphic to the algebra of all implicative ***,we show that the sets of multiplicative and implicative derivations on bounded regular idempotent semihoops are in oneto-one correspondence.

Hem-o-lok clip migration to duodenal bulb post-cholecystectomy:A case report

BACKGROUND Hem-o-lok clips are typically used to control the cystic duct and vessels during laparoscopic cholecystectomy(LC)and common bile duct exploration for stones in the bile duct and ***,we report a unique example of Hem-o-lok clip movement towards the duodenal bulb after LC,appearing as a submucosal tumor(SMT).Additionally,we provide initial evidence of gradual and evolving endoscopic manifestations of Hem-o-lok clip migration to the duodenal bulb wall and review the available *** SUMMARY A 72-year-old man underwent LC for gallstones,and Hem-o-lok clips were used to ligate both the cystic duct and cystic ***(EGD)2 years later revealed an SMT-like lesion in the duodenal *** to the symptomatology,the clinical examination did not reveal any major abnormalities,and the patient was followed up as an outpatient.A repeat EGD performed 5 months later revealed an SMT-like lesion in the duodenal bulb with raised edges and a central depression.A third EGD was conducted,during which a Hem-o-lok clip was discovered connected to the front side of the *** clip was extracted easily using biopsy forceps,and no complications *** months after the fourth EGD,the scar was surrounded by normal *** Clinicians should be aware of potential post-LC ***-o-lok clips should be removed if symptomatic.

Effect of oxide inclusions on MnS precipitates and tensile mechanical property of high-strength low-alloy steel

The key role of oxide inclusions on the microstructure and mechanical property of a high-strength low-alloy steel was *** field emission scanning electron microscope equipped with energy-dispersive spectrometry was used to characterize MnS *** inclusions play an important role in the shape control of MnS *** oxides fovored to decrease the size and the aspect ratio of MnS *** less oxide inclusions in the steel,approximately over 16.7%MnS precipitates were with aspect ratio a>5 and pure MnS precipitates accounted for 75.9%in ***,with more oxide inclusions in the steel,only 7.4%MnS precipitates were with a>5 and pure MnS precipitates accounted for 60.1%in *** of MnS by oxide inclusions improved the strength and inhibited the *** oxide inclusions in the steel increased the yield strength and tensile strength of the steel in both longitudinal and transverse directions,and lowered the anisotropy of the mechanical property.

Cardioprotective effects of diltiazem reevaluated by a novel myocardial ischemic model in Chinese miniature swine

aim:To develop a new model of myocardial ischemia in Chinese miniature swineand reevaluate the cardioprotective effects of ***:Myocardialischemia was induced by injecting self-embolus into the left anterior descending(LAD)coronary artery in qualified miniature ***(5 ***-1.d-1)wasorally administered to the swine by mixing into normal pig diet from 1 to 6 d afterself-embolus *** coronary angiography,30 point body surface electro-cardiogram(BS-ECG),hemodynamics,biochemistry,quantitative histology andpathohistology were determined 6 d after self-embolus ***:Embo-lization occurred in the LAD coronary artery of the Chinese miniature swine in-jected by *** were significant myocardial ischemia and largecardiac muscle infarction in the Chinese miniature swine,which were accompa-nied with increased BS-ECG,decreased hemodynamic indexes of the cardiac output,cardiac index,left cardiac work and left cardiac work index,and increased systemicvascular resistance *** analysis revealed myocardial degen-eration,necrosis,fibrosis,inflammatory cell infiltration and granulation tissuehyperblastosis(n=6).Diltiazem diminished the extent of the LAD embolism,ame-liorated myocardial ischemia,improved the hemodynamic indexes,increased theplasma superoxide dismutase activity,decreased the plasma malondialdehydecontent,narrowed the myocardial ischemic area and weakened the pathohistologicaldamage in the cardiac muscle(n=6).Conclusion:Myocardial ischemia induced byinjecting self-embolus into the LAD coronary artery in Chinese miniature swine isquite close to clinical pathophysiological *** is effective toinhibit the myocardial ischemia and restore the heart function in this novel model.
