Psoriasis is a chronic,recurrent,inflammatory systemic disease that is not only characterized by skin manifestations but may also be accompanied by various comorbidities,imposing a heavy burden on patients’physical and mental health and affecting their quality of *** the continuous approval of various biological agents for clinical use has provided more effective treatment options to patients with psoriasis,no consensus on the evaluation of comprehensive treatment goals has yet been *** treat-to-target(T2T)strategy requires consideration of multiple dimensions of treatment outcomes,the development of long-term management goals,and regular assessments of treatment conditions,which are often used in the management of chronic ***,based on the latest consensuses and guidelines,research data,and clinical experience as well as the combination of survey results and expert group discussions,the present consensus focuses on 4 dimensions of short-and long-term integrated management goals for biological agents:alleviating skin lesions,improving quality of life,screening and managing psoriasis comorbidities,and ensuring drug *** implementation methods,evaluation time,treatment monitoring,and program adjustments are also herein described to achieve comprehensive management of psoriasis to the maximum *** consensus provides a reference for clinical practice.

Vaccination against Schistosoma japonicum Infection by DNA Vaccine Encoding Sj22.7 Antigen

To observe the in vitro expression of DNA vaccine pcDNA3-Sj22.7 and its immunologicaleffect in mice,the recombinant plasmid pcDNA3-Sj22.7 was used to transfect HeLa cells with liposome-mediated method and the expression of Sj22.7 mRNA and protein was examined using reversetranscription-polymerase chain reaction, sodium dodecylsulfate-polyacrylamide gel electrophoresis andWestern ***,the ability of pcDNA3-Sj22.7 to protect against Schistosoma japonicum challengeinfections was analyzed according to worm reduction rate and egg reduction rate after vaccination of *** serum levels of specific IgG antibody and T lymphocyte proliferation response were also *** the challenge infection,Sj22.7-driven interferon (IFN)-γand interleukin (IL)-4 was also *** showed that pcDNA3-Sj22.7 could express Sj22.7 mRNA and protein in *** in a worm reduction rate of 29.70%,egg reduction rate of 47.25% (liver) and 51.73% (intestine),and egg reduction rate of 25.90% (eggs per female),suggesting induction of significant anti-fecundityin the pcDNA3-Sj22.7 group. Enzyme-linked immunosorbent assay and Western blot analysis indicatedthat immunized mice generated specific IgG against Sj22.7.T lymphocytes from mice immunized withpcDNA3-Sj22.7 showed a significant proliferation response to *** culture of spleen cells showedthat secretion of IFN-γ increased but IL-4 decreased. The results indicate that DNA vaccination bypcDNA3-Sj22.7 is sufficient to elicit significant levels of protective immunity against *** *** DNA vaccine could induce significant cellular and humoral immune response,and display predominantT helper cell type 1 type immune responses,which contribute to the protective immunity against challengeinfection in mice.

Genetic and Epigenetic Alterations of DLC-1,a Candidate Tumor Suppressor Gene,in Nasopharyngeal Carcinoma

The DLC-1 gene,located at the human chromosome region 8p22,behaves like a tumorsuppressor gene and is frequently deleted in diverse *** deletion of 8p22 is not an uncommon eventin nasopharyngeal carcinoma(NPC),therefore we explored the expression levels of the DLC-1 gene in NPCsand NPC cell lines by reverse transcription-polymerase chain *** results showed the mRNA levelof DLC-1 was *** identify the mechanism of DLC-1 downregulation in NPC,we investigatedthe methylation status of the DLC-1 gene using methylation-specific PCR,and found that 79%(31 of 39)ofthe NPC tissues and two DLC-1 nonexpressing NPC cell lines,6-10B and 5-8F,were methylated in theDLC-1 CpG *** PCR was also carried out,and loss of heterozygosity was found at fourmicrosatellite sites(D8S552,D8S 1754,D8S 1790 and D8S549)covering the whole DLC-1 gene with ratiosof 33%(4 of 12 informative cases),18%(2 of 11),5%(1 of 18),and 25%(3 of 12),***,our results suggest that DLC-1 might be an NPC-related tumor suppressor gene affected by aberrantpromoter methylation and gene deletion.

Effect of DPC4 Gene on Invasion and Metastasis of Colorectal Carcinoma Cells

To investigate the effect of DPC4 gene on invasion and metastasis of colorectal carcinomacells,the expression of DPC4 was detected in sixty-three samples of colorectal tumors and seven cases ofcolorectal *** biological behavior of tumors expressing DPC4 was evaluated (including tumorstaging,differentiation degree and metastasis).pcDNA3.1-DPC4 plasmid was constructed and transferredinto HCT116 cells not expressing *** cell models (DPC4~+-HCT116) steadily expressing DPC4 *** with HCT116 and pcDNA3.1-HCT116 cells,the doubling time of DPC4~+-HCT116 cellswas lengthened obviously (P<0.01),the apoptosis rate of DPC4~+-HCT 116 cells was significantly increased(P<0.01),the cloning efficiency,cell adherency,migration and invasion ability of DPC4~+-HCT116 cells weredropped obviously (P<0.01).The number of cancer nodules was decreased significantly in abdominal cavityand liver of the nude mice inoculated with DPC4~+-HCT116 *** activity of MMP-9 and MMP-2 wasdetected by gelatin *** comparison with HCT116 and pcDNA3.1-HCT116 cells,the activity ofMMP-9 was decreased in DPC4~+-HCT116 ***,the down-regulation of DPC4 expression may beassociated with the carcinogenesis of colorectal ***4 may inhibit the proliferation of coloncancer cell by restraining growth and inducing apoptosis,and the invasion and metastasis of colorectalcarcinoma ***-9 may be one of the downstream target genes regulated by DPC4.

＜正＞Objective To investigate the expression of synaptophysin（SYN） in rat retina following acute high intraocular pressure（HIOP）.Methods The intraocular pressure of all left eyes was increased until b wave of flash electroretinogragh （fERG） disappeared and such pressure maintained for 60 *** right eyes served as normal *** were sacrificed at 1,3,7 or 14 d following operation *** and in situ hybridization were used to detecting the expression of *** In normal condition,SYN was mainly located in the outer plexiform layer（OPL） and the inner plexiform layer（IPL）.SYN expression was extended to the inner part of the outer nuclear layer（ONL） after *** was the widest distribution of SYN in the OPL and ONL on the 7th after *** the 14th day,the distribution of SYN was similar to that of the normal *** in situ *** positive neurons were located in the inner nuclear layer（INL） and the ganglion cell layer（GCL）.Expression of SYN mRNA in the INL was increased following HIOP and reached its peak on the 7th day,then decreased to a level lower than normal at the 14th *** SYN expression was firstly up-regulated then down-regulated following *** was indicated that there may exist a phenomenon of synaptogenesis in the early stage while synapse degeneration in the later stage in rat retina following HIOP.
